One of the hallmarks of cancer is increased vessel growth to feed the tumor’s rapid expansion. The newly formed blood vessels are immature and often leaky due to structural and functional abnormalities. This leads to ineffective anti-cancer drug delivery and provides tumor cells with additional opportunities to metastasize. We propose to develop an in vitro vascular endothelium model and introduce aggressive triple-negative breast cancer (TNBC) cells to mimic the tumor environment, utilizing a Transwell migration assay. Many hydrogel systems designed to mimic the extracellular matrix (ECM) employ thiol-based chemistries with undesirable functional groups, including vinyl sulfones and methacrylates. In downstream applications, any unreacted functional groups are problematic and render hydrogels non-viable for human use. Here, we introduce modifications to the Native Chemical Ligation (NCL) mechanism, exploring the elimination of cytotoxic byproducts and increased reaction rate by using a β-thiolactone. The inclusion of the β-thiolactone results in retention of the thiol within the gel, as well as enhanced kinetics due to the thermodynamic favorability of the ring opening and amide formation. This study investigated chitin extraction from the stalks and atypical mushrooms, the often-produced waste in the mushroom production process, using a simultaneous application of protease and glucanase with or without an alkaline pretreatment stage. A commercial chitin, chemically extracted from A. bisporus was used as the control. The purity of extracted chitin (98.92–99.36 %) was slightly lower than commercial chitin (99.80 %), but with comparable physicochemical properties (p < 0.05). All enzymatically extracted chitin in this study exhibited an agglomerated, microfibrillar and irregular structure, while the control showed standalone granules with weathering and cracks on the surfaces. The XRD and FTIR spectra indicated that the extracted chitin and the control belong to the α-allotrope group. The degree of acetylation was higher in the extracted chitin (76.97–78.22 %) than in the control (75.05 %). The highest molecular weight obtained for the extracted chitin was 4.53 × 102 KDa (W0548). The study demonstrated the possibility of producing pure, high molecular weight chitin from the waste produced during the commercial production of button mushrooms, using a potentially more sustainable approach than the traditional one. This enzymatic approach reduces chemical waste, as shown by the excellent results obtained for samples without the pre-treatment stage, and improves environmental safety compared to traditional methods.

This work reports the development and evaluation of dendrimer-based nanogels based on Poly amidoamine (PAMAM) dendrimer generation 5, engineered to act as a carrier with reactive oxygen species (ROS)-scavenging capabilities. We developed a cross-linking reaction-enabled flash nanoprecipitation method in which the cross-linking reaction occurs during the flash nanoprecipitation process to form a cross-linked nanostructure. Using this approach, an N-hydroxy succinimide (NHS)-functionalized ROS-responsive thioketal cross-linker (TK-NHS) was synthesized and utilized to cross-link DAB-core PAMAM dendrimer G5, resulting in the formation of G5-TK nanogels. Nanoparticle-based therapeutics hold promise for the treatment of atherosclerosis, but challenges such as low drug-loading capacity and a lack of scalable, controllable production hinder their clinical translation. Flash nanoprecipitation, a continuous synthesis method, offers a potential solution for scalable and reproducible nanoparticle production. In this study, we employed a custom-designed multi-inlet vortex mixer to perform cross-linking reaction-enabled flash nanoprecipitation, facilitating controlled and scalable synthesis of cross-linked Poly amidoamine (PAMAM) dendrimer nanoparticles. Here we present a simple and efficient method for fabricating laser-induced graphene (LIG) electrodes for the electrochemical detection of acetaminophen (AAP), a crucial analgesic and antipyretic that can become toxic at elevated concentrations. Unlike conventional sensors that require intricate chemical modifications, the fabricated LIG-based sensor eliminates the need for functionalization, offering a streamlined sensing solution. The LIG electrodes were characterized using scanning electron microscopy, energy-dispersive spectroscopy, and Raman spectroscopy, which confirmed the formation of a highly porous graphene network with excellent purity and conductivity. In stressful environments, optimizing team performance requires not only assessing communication but also monitoring physiological indicators of stress. Electrodermal activity (EDA) and heart rate variability (HRV) are commonly used to capture autonomic responses, offering insight into emotional arousal and cardiovascular regulation.

This pilot study compares a commercial wearable device, the EmbracePlus, with a research-grade device, the Shimmer3 GSR+, to evaluate differences in physiological signal acquisition. Magnetic heating mediated by magnetic iron oxide nanoparticles (MION) has several clinical applications including cancer thermal therapy, tissue thawing for organ preservation, and magnetogenetics. However, current models of magnetic heating, including the widely used linear response theory, inadequately predict MION heating efficiency. An experimental study covering alternating magnetic field (AMF) strengths and frequencies ranging from 3.98 – 27.85 kA/m and 103.6 – 984.1 kHz, respectively, with a range of MION core diameters 9.5- 30.3 nm was conducted to elucidate the relationships between these AMF parameters and MION sizes and the resulting heating efficiencies. This dissertation investigates the hemodynamics of pulsating blood flow in curved vessels through direct numerical simulations (DNS), with the overarching aim of uncovering fluid dynamic mechanisms linked to the onset and progression of aortic dissection in humans. The work is structured in two parts: (i) fundamental studies in idealized geometries and (ii) preliminary investigations in patient-specific anatomies. Long-term effects of this include joint instability, limited movement, and arthritis. Bone infections (osteomyelitis), reactive oxygen species (ROS) generation via excessive inflammation, and underlying conditions such as osteoporosis can contribute to suboptimal bone tissue regeneration. Ultra-high molecular weight polyethylene (UHMWPE) is a linear long chain homopolymer valued for its toughness, wear resistance, and chemical inertness. In the medical field, these properties make it the material of choice for bearing surfaces despite recognized tradeoffs between wear resistance, toughness, and oxidation resistance. Electrically conductive composites of UHMWPE show promise to enable new types of smart and active load-bearing implants in a future of personalized medicine, if beneficial properties can be maintained with the addition of solid-state additives.

Evidence in existing literature suggests that peripheral mutant Huntingtin protein may be involved and deteriorate HD. Red blood cells (RBCs), being the most abundant type of cells in the peripheral blood, can show detectable changes in their biophysical properties due to subtle changes in cell membranes. Epilepsy is increasingly recognized as a disorder not only of seizures but also of widespread network dysfunction, impacting cognitive and emotional processing. One critical cognitive process affected in epilepsy is emotional conflict processing, which relies on the interplay of multiple brain networks. However, the spatiotemporal dynamics of emotional conflict processing in epilepsy, particularly in adolescents, have remained poorly understood. Traumatic brain injury (TBI) is a major cause of neurological impairment, often leading to variable recovery and uncertain prognosis in the neurocritical care setting. There is a pressing clinical need for robust, physiologically grounded biomarkers to inform prognosis and therapeutic decision-making in acute TBI. This thesis investigates neurovascular coupling (NVC), the physiological coordination between neuronal activity and cerebral blood flow, as a candidate biomarker for brain function and recovery after injury.

Mobulas (manta and devil rays) are large-scale ram filter feeders that separate planktonic food particles from large volumes of water with minimal clogging. This contrasts with most human-made filters that can suffer from problematic clogging requiring additional mechanisms for clearing blocked surfaces and maintaining performance. Prior studies have shown that mobulas employ a unique mechanism referred to as ricochet separation to filter feed, whereby captive vortices in filter pores cause particles to bounce off the filter surfaces and away from the filter pores. Such mechanobiological relationships are especially critical in the cardiovascular system, where endothelial cells lining the vasculature align in response to fluid shear stresses from blood flow (Sinha, 2016). Diabetes mellitus patients must regularly monitor their blood glucose levels to manage glycaemia, typically requiring capillary tests at least three times daily and laboratory tests one to two times per month. These conventional methods involve finger pricking, causing significant discomfort and stress. This study introduces an innovative non-invasive glucose monitoring approach by integrating photoplethysmography (PPG) technology with an artificial intelligence (AI) algorithm, complemented by a mobile application using Flutter, IoT systems, and Firebase cloud for real-time data access. Congenital heart disease (CHD) stands as the leading congenital anomaly which affects pediatric populations throughout the world. The effectiveness of treatment depends on both early diagnosis and accurate identification but echocardiography requires manual interpretation which proves time-consuming and inconsistent especially when examining pediatric patients with their distinct cardiac systems. The research aims to create a deep learning-based diagnostic framework which uses ECG data to identify coronary artery disease subtypes in pediatric patients. Despite the outstanding electric properties and cost-effectiveness of poly- (3,4-ethylenedioxythiophene) (PEDOT) and its derivatives, their performance as hole transport layer (HTL) materials in conventional perovskite solar cells (PSCs) has lagged behind that of widely used spirobifluorene-based molecules or poly(triaryl amine). This gap is mainly from their poor solubility and energy alignment mismatch. In this work, the design and synthesis of a pyrrole-modified HTL (PPr) based on 3,4-propylenedioxythiophene (ProDOT) are presented for efficient and stable PSCs. Magnetic nanoparticles (MNPs) are highly versatile nanomaterials in nanomedicine, owing to their diverse magnetic properties, which can be tailored through variations in size, shape, composition, and exposure to inductive magnetic fields. Over four decades of research have led to the clinical approval or ongoing trials of several MNP formulations, fueling continued innovation. Beyond traditional applications in drug delivery, imaging, and cancer hyperthermia, MNPs have increasingly advanced into molecular medicine. Wastewater-based epidemiology (WBE) is a promising tool for improving health outcomes through early detection and cost-effective pathogen surveillance. Long-term care facilities (LTCFs) serve and employ vulnerable populations that may particularly benefit from the use of WBE, but financial and technical costs associated with standard sampling methods limit the feasibility of WBE in the LTCF setting. In this work, we used passive sampling to simplify the wastewater analysis process and compared its performance to the standard composite sampling method.
