Histology performed by Cal Poly students for research projects and labs are currently performed by hand using a dipping technique. This technique is time consuming, expensive, imprecise, and often unrepeatable. Central Coast Pathology donated a Dako Autostainer to the Biomedical Engineering Department that can perform immunohistological stains of up to 48 samples simultaneously using tightly controlled reagent volumes and incubation times. Protocols are programmed into the machine and can be repeated under the same conditions every time with minimal preparation and exposure to potentially dangerous reagents. This project’s goal was to analyze the properties of the cortical femur on 100 mice bones from Lawrence Livermore National Laboratory. Analysis was limited to imaging which determined the ratio of bone volume to total volume (BV/TV) and osteocyte lacunae density. Mice were altered to knock out their SOST gene: a negative regulator of bone formation. Twelve groups were created to differentiate their treatment, duration, and phenotype. Transgenic (TG) mice had an overexpression of the SOST gene: they carried a bacterial artificial chromosome. This study evaluates the potential effects of silver salts on biocompatible metals used for prostheses during the chemical reduction process to produce a silver antimicrobial layer on the metal’s surface. Samples of two biocompatible metals were obtained: Stainless Steel 316L and ASTM F75 Cobalt Chromium Alloy. Three different silver salts were also acquired: silver nitrate, silver sulfadiazine, and silver chloride. Specimens of each metal were cut to size using a 4-1/2 inch aluminum oxide, 40 grit, cut off wheel for metal, attached to a Dewalt Angle Grinder. This report is documentation of a Senior Project completed by Dylan Reinsdorf and Kendall Smith, in partial fulfillment of a Bachelor of Science in Mechanical Engineering at Cal Poly, San Luis Obispo. Medtronic Inc. approached the team with a need to quantify proximal conformity of stent grafts in the thoracic aorta. A solution was designed that included an aorta model that simulates clinical conditions, and a measurement method for stent graft conformability. This report documents all phases of the project, from sponsor requirements to final design to product realization and testing.

The purpose of this thesis project was to demonstrate the ability to utilize the capabilities of aptamers so that they may act as capture reagents for paper microfluidic devices. Several characterization experiments were conducted as a precursor before the final experimentation was performed. Paper characterization, manufacturing protocols for printing and heating, as well as 3D chip fabrication were all performed and analyzed. The results confirmed that the control of fluid through a 3D microfluidic device based in nitrocellulose is possible.

This thesis utilizes previously described manufacturing and design techniques for the creation of a PDMS-glass bonded microfluidic device, capable of electrochemical impedance spectroscopy (EIS). EIS has been used across various fields of research for different diagnostic needs. This project encompasses the implementation of a computational model to simulate the microfluidic separation of like-charged particles in a continuous flow environment. By accomplishing this task the model can be used to optimize future fractionations by tailoring the process parameters to the properties of the target particles. The purpose of this study is to characterize the compact bone remodeling of the ovariectomized ewe and its effectiveness as an animal model for studies of postmenopausal osteoporosis. The use of animal models is a beneficial way to evaluate the effectiveness of medical devices and therapeutic agents for treating diseases. The ovine model is an attractive option because of its large size and similar bone remodeling to humans. Peripheral Arterial Occlusive Disease (PAOD) is an acquired inflammatory disease where a peripheral artery becomes occluded due to the buildup of atherosclerotic plaques. In patients that possess collateral arteries, an occlusion can lead to shear induced outward remodeling, arteriogenesis, of these collaterals, partially restoring blood flow. However, newly remodeled collaterals exhibit reduced functional vasodilation, which may impair normal activity, such as ambulation. To model chronic ischemia and arteriogenesis in collaterals, a femoral artery ligation in a murine hindlimb is commonly performed. This dissertation examines manipulation of visual feedback in virtual reality (VR) to increase excitability of distinct neural networks in the sensorimotor cortex. The objective is to explore neural responses to visual feedback of motor activities performed in complex virtual environments during functional magnetic resonance imaging (fMRI), and to identify sensory manipulations that could further optimize VR rehabilitation of persons with hemiparesis. Recovery of upper extremity function remains particularly resistant to intervention, with 80% to 95% of persons demonstrating residual upper extremity impairments lasting beyond six months after the stroke. Over the past few decades, tissue engineering approaches have pushed their way to the forefront of biotechnological research and development, serving a wide variety of applications. This research utilizes quantitative methods to analyze the outcome of two bone tissue engineering studies. In an in vivo study, the use of Negative Pressure Wound Therapy (NPWT) in combination with stem cell-loaded scaffolds was investigated for the repair of large bone defects. This study describes the analysis of reaching and grasping abilities of the hemiparetic arm and hand of patients post stroke after a series of interactive virtual reality (VR) simulated training sessions and conventional physical therapy of similar intensity. Six subjects participated in VR training and five subjects in clinical rehabilitation for two weeks. Subjects’ finger joint angles were measured during a kinematic reach to grasp test using CyberGlove® and arm joint angles were measured using the trackSTARTM system prior to training and after training. The long-term impairment of the SCI patients has led researchers to investigate different rehabilitation and treatment efforts that require careful observations to be made on reliable markers.
