Semi-Interpenetrating Network Gelatin Fiber Sca Old For Oral Mucosal Delivery Of Insulin, Leyuan Xu Jul 2013

Semi-Interpenetrating Network Gelatin Fiber Sca Old For Oral Mucosal Delivery Of Insulin, Leyuan Xu

Theses and Dissertations

Common therapy for diabetes mellitus is subcutaneous administration of insulin that is subject to serious disadvantages, such as patient noncompliance and occasional hypoglycemia. Hence, oral administration of insulin could be more convenient and serve as a desired route. However, oral administration of insulin is severely limited by the low bioavailability of insulin through the gastrointestinal tract. In this study, a semi-interpenetrating network gelatin fiber scaffold (sIPN GF) was fabricated for oral mucosal delivery of insulin as an alternative route. This sIPN GF was engineered from an electrospun gelatin fiber scaffold (GF), which was further crosslinked with polyethylene glycol diacrylate (PEG-DA) …

Efferent Copy And Corollary Discharge Motor Control Behavior Associated With A Hopping Activity, Wangdo Kim, António P. Veloso, Filipa João, Sean S. Kohles Jul 2013

Efferent Copy And Corollary Discharge Motor Control Behavior Associated With A Hopping Activity, Wangdo Kim, António P. Veloso, Filipa João, Sean S. Kohles

Mechanical and Materials Engineering Faculty Publications and Presentations

Hoppers respond not only to stimuli from the ground surfaces but also to cues generated by their own behaviors. This leads to desensitization because although the afferent and reafferent signals have distinct causes, they are carried by the same sensory channels. From a behavioral viewpoint, it may be necessary to distinguish between signals from the two causes especially when monitoring changes in the external environment separate from those due to self-movement. We were able to separate afferent sensory stimuli from self-generated, reafferent signals using an action oriented perception system and dynamic programming approach. This effort addressed the question of how …

Footwear-Centric Body Area Network With Directional Uwb Antenna, Domenico Gaetano, Vit Sipal, Patrick Mcevoy, Max Ammann, C Brannigan, Louise Keating, Frances Horgan

Articles

A footwear-centric body area network employing a directional antenna is compared with waist-centric systems using omnidirectional and directional antennas. The impact of body movements on path gain is analysed for two bands at 3.99 GHz and 7.99 GHz. The path gain and data rate results demonstrate that footwear-centric configurations are equivalent or better than waist-centric body area networks.

A Computational Model Of Nanoparticle Transport And Delivery In Tumor Tissue, Vishwa Priya Podduturi Jul 2013

A Computational Model Of Nanoparticle Transport And Delivery In Tumor Tissue, Vishwa Priya Podduturi

Doctoral Dissertations

Determining the factors that influence the delivery of nanoparticles to tumors and understanding the relative importance of each of these factors is fundamental to optimize the drug delivery process. In this research, a model that combines random walk with the pressure driven flow of nanoparticles in a tumor vasculature is modeled. Nanoparticle movement in a cylindrical tube with dimensions similar to the tumor's blood capillary with a single pore is simulated. Nanoparticle velocities are calculated as a pressure driven flow over imposed to Brownian motion. During the study, the effect of red blood cells (RBC) is also studied by comparing …

Hip Proprioceptors Preferentially Modulate Reflexes Of The Leg In Human Spinal Cord Injury, Tanya Onushko, Allison S. Hyngstrom, Brian D. Schmit Jul 2013

Hip Proprioceptors Preferentially Modulate Reflexes Of The Leg In Human Spinal Cord Injury, Tanya Onushko, Allison S. Hyngstrom, Brian D. Schmit

Biomedical Engineering Faculty Research and Publications

Stretch-sensitive afferent feedback from hip muscles has been shown to trigger long-lasting, multijoint reflex responses in people with chronic spinal cord injury (SCI). These reflexes could have important implications for control of leg movements during functional activities, such as walking. Because the control of leg movement relies on reflex regulation at all joints of the limb, we sought to determine whether stretch of hip muscles modulates reflex activity at the knee and ankle and, conversely, whether knee and ankle stretch afferents affect hip-triggered reflexes. A custom-built servomotor apparatus was used to stretch the hip muscles in nine chronic SCI subjects …

Zein: Novel Natural Polymer For Nanoparticle- And Film-Mediated Gene Delivery, Jessica D. Taylor Jul 2013

Zein: Novel Natural Polymer For Nanoparticle- And Film-Mediated Gene Delivery, Jessica D. Taylor

Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research

Gene delivery, the introduction of DNA into cells, is applicable to gene therapy, DNA vaccination, functional genomics and diagnostics, tissue engineering, and drug-eluting medical devices. Particulates incorporating DNA are promising vehicles for gene delivery and overcome some of the barriers that hinder successful gene transfer, with the ability to protect DNA and provide for controlled, localized, and sustained release and transfection. Furthermore, innovative new gene delivery strategies that incorporate DNA particulates or complexes within films or coatings for devices and scaffolds could further provide for controlled and sustained transfection at the site of implant. Zein, a hydrophobic protein from corn, …

The Effects Of Separating Visual And Motor Workspaces On The Generalization Of Visuomotor Adaptation Across Movement Conditions, Yuming Lei Jul 2013

The Effects Of Separating Visual And Motor Workspaces On The Generalization Of Visuomotor Adaptation Across Movement Conditions, Yuming Lei

Master's Theses (2009 -)

Separating visual and proprioceptive information in terms of workspace locations during reaching movement has been shown to disturb transfer of visuomotor adaptation across the arms. Here, we investigated whether separating visual and motor workspaces would also disturb generalization of visuomotor adaptation across movement conditions within the same arm. In our behavioral study, subjects were divided into four experimental groups (plus three control groups). The first two groups adapted to a visual rotation under a "dissociation" condition in which the targets for reaching movement were presented in midline while their arm performed reaching movement laterally. Following that, they were tested in …

An Fmri Study On Supra-Spinal Contributions To Upper And Lower Limb Motor Control, Shancheng Bao Jul 2013

An Fmri Study On Supra-Spinal Contributions To Upper And Lower Limb Motor Control, Shancheng Bao

Master's Theses (2009 -)

The differences in the neural mechanisms contributing to upper and lower extremity movement have not been fully elucidated, and this might be a factor that leads to the ineffectiveness of rehabilitation techniques for most stroke survivors. It is unclear whether therapies designed for upper extremities should also be used for the lower extremities, and vice versa. In this study, fMRI was used to examine the supraspinal control of UE and LE movement in both neurologically intact individuals and people with post-stroke hemiparesis. We compared the location, volume, and intensity of brain activity associated with upper and lower extremity pedaling and …

Surgical Tooling Designed For The Direct Anterior Approach To Total Hip Arthroplasty, Jon-Peter Meckel Jul 2013

Surgical Tooling Designed For The Direct Anterior Approach To Total Hip Arthroplasty, Jon-Peter Meckel

Master's Theses

Surgical Tooling Designed for the Direct Anterior Approach to Total Hip Arthroplasty Jon-Peter Meckel

Total hip arthroplasty (THA) is becoming more and more common in the US as people continue to live longer and more active lives. The main reason that a THA is required is due to the “wear and tear” affliction of osteoarthritis, which in the year 2000 had at least 3% of the population over 30 showing symptoms. A revitalized approach to THA is the direct anterior approach, or Smith-Petersen approach, which limits the amount of musculature affected by the surgery and creates a very stable joint …

Optical Imaging Of Lipopolysaccharide-Induced Oxidative Stress In Acute Lung Injury From Hyperoxia And Sepsis, Reyhaneh Sepehr, Said H. Audi, Sepideh Maleki, Kevin Staniszewski, Annie L. Eis, Girija G. Konduri, Mahsa Ranji Jul 2013

Optical Imaging Of Lipopolysaccharide-Induced Oxidative Stress In Acute Lung Injury From Hyperoxia And Sepsis, Reyhaneh Sepehr, Said H. Audi, Sepideh Maleki, Kevin Staniszewski, Annie L. Eis, Girija G. Konduri, Mahsa Ranji

Biomedical Engineering Faculty Research and Publications

Reactive oxygen species (ROS) have been implicated in the pathogenesis of many acute and chronic pulmonary disorders such as acute lung injury (ALI) in adults and bronchopulmonary dysplasia (BPD) in premature infants. Bacterial infection and oxygen toxicity, which result in pulmonary vascular endothelial injury, contribute to impaired vascular growth and alveolar simplification seen in the lungs of premature infants with BPD. Hyperoxia induces ALI, reduces cell proliferation, causes DNA damage and promotes cell death by causing mitochondrial dysfunction. The objective of this study was to use an optical imaging technique to evaluate the variations in fluorescence intensities of the auto-fluorescent …

Protein Loop Length Estimation From Medium Resolution Cryoem Images, Andrew R. Mcknight Jul 2013

Protein Loop Length Estimation From Medium Resolution Cryoem Images, Andrew R. Mcknight

Computer Science Theses & Dissertations

In the post-genomic era, proteomics research presents a new frontier in life science. Proteins play roles in virtually every biological process, and understanding their atomic structures is the key to unraveling how they carry out their work. Compared to the over half million protein sequences in UniProt, only around 25,000 unique sequences have been atomically modeled and deposited to PDB (Protein Databank). Cryoelectron Microscopy (cryoEM) is an important biophysical technique that produces 3D subnanometer resolution images of molecules not amenable to past approaches like x-ray crystallography or nuclear magnetic resonance. De novo modeling is becoming a promising approach to derive …

Subtalar Joint Instability: Diagnosis And Conservative Treatment, Julie Choisne Jul 2013

Subtalar Joint Instability: Diagnosis And Conservative Treatment, Julie Choisne

Mechanical & Aerospace Engineering Theses & Dissertations

Subtalar instability may be caused by various ligamentous injuries. Combined instability at the ankle and subtalar joint is not adequately diagnosed. Further, isolated subtalar instability is usually misdiagnosed which may lead to long term damage to the joint. Developing a non-invasive and clinically practical tool to diagnose subtalar joint instability would be an important asset. The ability of an ankle brace, a common treatment for hindfoot instability, to promote stability for the subtalar joint was not well established. The purposes of this study were to 1) assess the kinematics of the subtalar, ankle, and hindfoot in the presence of isolated …

Quantitative Assessment Of Program Outcomes Using Longitudinal Data From The Fe Exam, Joe C. Guarino, James R. Ferguson, V. Krishna C. Pakala

Mechanical and Biomedical Engineering Faculty Publications and Presentations

There have been many studies providing details on using results from the Fundamentals of Engineering (FE) exam as metrics for meeting ABET program outcomes1. However, implementing an FE-based set of metrics poses challenges not limited to assessing validity of results. Programs using FE-based metrics must also determine the position of the metrics in the overall assessment process. We present a method for using FE-based metrics as an integral part of the ABET program assessment process. The principle issues we address are: (1) The validity of using FE metrics for a group of graduating students when not all of …

Spatiotemporal Fluorescent Detection Measurements Using Embedded Waveguide Sensors, Mark C. Harrison, Andrea M. Armani Jun 2013

Spatiotemporal Fluorescent Detection Measurements Using Embedded Waveguide Sensors, Mark C. Harrison, Andrea M. Armani

Engineering Faculty Articles and Research

Integrated waveguide biosensors, when combined with fluorescent labeling, have significantly impacted the field of biodetection. While there are numerous types of waveguide sensors, the fundamental excitation method is fairly consistent: the evanescent field of the waveguide excites a fluorophore whose emission is detected, either directly via imaging or indirectly via a decrease in power transfer. Recently, a sensor device was demonstrated which is able to back-couple the emitted light into the waveguide, allowing the signal to be detected directly. However, this previous work focused on the development of an empirical model, leaving many theoretical questions unanswered. Additionally, the results from …

Amperometric Detection Of Bioamines In Cancer Borealis Using Microchip Capillary Electrophoresis Integrated With Micellar Chromatography, Ajay Fernandez, Jason Howard, Christina Shuh Jun 2013

Amperometric Detection Of Bioamines In Cancer Borealis Using Microchip Capillary Electrophoresis Integrated With Micellar Chromatography, Ajay Fernandez, Jason Howard, Christina Shuh

Bioengineering Senior Theses

In order to study neuromodulation in Cancer borealis, commonly known as the Jonah crab, we designed a microchip capillary electrophoretic (CE) device with micellar electrokinetic chromatography (MEKC) to separate bioamines and detect their concentrations using amperometric detection (AD). The analysis will be used to characterize the bioamines: dopamine, octopamine, tyramine, serotonin, norepinephrine, and gammaaminobutyric acid (GABA). The device measures the concentration of bioamines in microdialysate samples taken directly from the pericardial cavity of the crab. The research on Jonah crabs will be applied to enhance our understanding of the crab neurology. Since the structure and in some instances, the function …

Microfluidic Detection Of Arsenic Contamination In Groundwater, Mary Reynolds, Kyle Perricone Jun 2013

Microfluidic Detection Of Arsenic Contamination In Groundwater, Mary Reynolds, Kyle Perricone

Bioengineering Senior Theses

The difficulty of detecting small quantities of arsenic in water currently threatens the health of millions of people worldwide, as long-term exposure to arsenic has been associated with both cancerous and noncancerous health risks. Existing technologies make it possible to very accurately quantify arsenic levels in water; however the expense, extensive training, and off-site analysis required by these methods impede wide scale use. Here, we report on research to develop an affordable and point-of-use microfluidic platform capable of detecting trace amounts of arsenic in groundwater samples. We intend this device to meet the World Health Organization's (WHO's) ASSURED criteria for …

The Fabrication & Characterization Of An Electrokinetic Microfluidic Pump From Su-8, A Negative Epoxy-Based Photoresist, Nash Anderson Jun 2013

The Fabrication & Characterization Of An Electrokinetic Microfluidic Pump From Su-8, A Negative Epoxy-Based Photoresist, Nash Anderson

Master's Theses

Microfluidics refers to manipulation, precise control, and behavior of fluids at the micro and nanoliter scales. It has entered the realm of science as a way to precisely measure or mix small amounts of fluid to perform highly controlled reactions. Glass and polydimethylsiloxane (PDMS) are common materials used to create microfluidic devices; however, glass is difficult to process and PDMS is relatively hydrophobic. In this study, SU-8, an epoxy based (negative) photoresist was used to create various electrokinetic microfluidic chips. SU-8 is commonly used in microelectromechanical design. Spin coating of various SU-8 formulations allows for 1 μm to 100 μm …

Evaluation Of Corrosion Properties Of Proprietary Metal Alloy Stents For In-Vivo Use, Michael David Bremner Jun 2013

Evaluation Of Corrosion Properties Of Proprietary Metal Alloy Stents For In-Vivo Use, Michael David Bremner

Materials Engineering

To assess corrosion rates of metal alloy bio-absorbable stents an experimental set-up was designed to mimic the coronary artery environment. The artery was modeled using 4mm diameter Tecoflex tubing and the metal alloy stents were inserted into the tubing using a catheter. As is the case in cardiac surgery, the catheter with the stent and a microballoon were maneuvered to the desired position. The microballoon was then slowly inflated to expand the stent and compress it against the tubing walls. The catheter and microballoon were then withdrawn. A circulating pump system was set up to cycle fetal bovine serum (FBS) …

Fluorescence Characterization Of Quantum Dots For Use As Biomarkers, Logan M. Grimes

Materials Engineering

Fluorescence profiles of quantum dots (QDs) were characterized to select the ideal QDs for encapsulation in phospholipids for use as biomarkers to selectively adhere to cancer cells. QDs were synthesized and extracted 0, 30, 60, and 90 seconds after precursor compounds were mixed. These extractions were isolated by extraction time. Portions from each vial were coated in a zinc sulfide shelling procedure, leaving at least half of the QD solution unshelled. These samples were characterized over four days to monitor fluctuations in fluorescence. This was done utilizing an Ocean Optics spectrometer in conjunction with Spectra Suite software. The central wavelength, …

Nitric oxide (NO) is an endogenous vasodilator as well as natural inhibitor of platelet adhesion and activation that can be released from a NO donor species, such as diazeniumdiolated dibutylhexanediamine (DBHD/N2O2) within a polymer coating. In this study, various Food and Drug Administration approved poly(lactic-co-glycolic acid) (PLGA) species were evaluated as additives to promote a prolonged NO release from DBHD/N2O2 within a plasticized poly(vinyl chloride) (PVC) matrix. When using an ester-capped PLGA additive with a slow hydrolysis time, the resulting coatings continuously release between 7 and 18 × 10−10 mol cm−2 min−1 NO for 14 days at 37 °C in …

Millions of cases of bone injury or loss due to trauma, osteoporosis, and cancer occur in the United States each year. Because bone is limited in its ability to regenerate, alternative therapy approaches are needed. Bone tissue engineering has the potential to correct musculoskeletal disorders through the development of cell-based substitutes for osteogenic tissue replacement. Multiple medical imaging techniques such as magnetic resonance microscopy (MRM) were investigated recently; these techniques are able to provide useful information on the anatomical and structural changes of developing bone. However, there is a need for noninvasive approaches to evaluate biochemical constituents and consequent compositional …
