Multiple Myeloma (MM) is a treatable, yet incurable, malignancy of bone marrowplasma cells. This cancer affects many patients and many succumb to relapse of tumor burden despite a large number of available chemotherapeutic agents developed for therapy. This is because MM tumors are heterogeneous and receive protection from therapeutic agents by the microenvironment and other mechanisms including homologous MM-MM aggregation. Therefore, therapy failure and frequent patient relapse is due to the evolution of drug resistance, not a lack of available drugs. Traditional bacterial identification methods take one to two days to complete, relying on large bacteria colonies for visual identification. In order to decrease this analysis time in a cost-effective manner, a method to sort and concentrate bacteria based on the bacteria's characteristics itself is needed. One example of such a method is dielectrophoresis, which has been used by researchers to separate bacteria from sample debris and sort bacteria according to species. This work presents variations in which dielectrophoresis can be performed and their associated drawbacks and benefits specifically to bacterial identification. In addition, a potential microfluidic design will be discussed.

Elastin-like peptides (ELPs) are stimulus-responsive protein-based polymers which are attractive material for biomedical research due to their biocompatibility and unique properties. The physical properties of ELPs are dependent on the chain length and the chosen amino acid at the guest residue position. This imparts unlimited flexibility in designing ELP based biomaterials with the desired physical properties.
We have shown that in addition to their physical properties, ELPs have biological activities that are conducive to tissue regeneration. Specifically, we found that ELPs induce fibroblast proliferation via cell surface heparan sulfate proteoglycans (HSPG). Furthermore, our data suggests that ELP based materials with …

Application of tissue-engineered vascular grafts (TEVGs) for the replacement of small-diameter arteries is limited due to thrombosis and intimal hyperplasia. Previous studies have attempted to address the limitations of TEVGs by developing scaffolds that mimic the composition (collagen and elastin) of native arteries to better match the mechanical properties of the graft with the native tissue. However, most existing scaffolds do not recapitulate the aligned topography of the collagen fibers found in native vessels. Electroretinagram (ERG) is the recording of electrical activity of retinal cells elicited by light stimulation, which has been widely used to help diagnose different types of retinal dysfunctions. The ERG response signal is a short non-stationary signal that contains overlapping components. Different Digital Signal Processing (DSP) techniques are investigated using MATLAB to study the time-frequency responses of the ERG signal such as Short Time Fourier Transform (STFT), Continuous Wavelet Transform (CWT) and Discrete Wavelet Transform (DWT). Over the past few years, researchers have focused their attention on the development of targeted cancer therapies to minimize the side effects associated with non-targeted treatments such as chemotherapy. Specifically, these approaches have focused on blocking growth factor receptors (GFR) that are overexpressed in cancer cells. In this thesis, we also focus on targeting overexpressed GFR; however, instead of blocking the GFR, our novel approach aims at using them to selectively enhance the endocytotic process of macropinocytosis to deliver peptides that either disrupts the mitochondria or inhibits glycolysis.

BACKGROUND:
The developing visual system in Drosophila melanogaster provides an excellent model with which to examine the effects of changing microenvironments on neural cell migration via microfluidics, because the combined experimental system enables direct genetic manipulation, in vivo observation, and in vitro imaging of cells, post-embryo. Exogenous signaling from ligands such as fibroblast growth factor (FGF) is well-known to control glia differentiation, cell migration, and axonal wrapping central to vision.

Functional magnetic resonance imaging (fMRI) studies have demonstrated alterations during task-induced brain activation in spinal cord injury (SCI) patients. The interruption to structural integrity of the spinal cord and the resultant disrupted flow of bidirectional communication between the brain and the spinal cord might contribute to the observed dynamic reorganization (neural plasticity). However, the effect of SCI on brain resting-state connectivity patterns remains unclear. We undertook a prospective resting-state fMRI (rs-fMRI) study to explore changes to cortical activation patterns following SCI. To replace photoreceptors lost to disease or trauma and restore vision, laboratories around the world are investigating photoreceptor replacement strategies using subretinal transplantation of photoreceptor precursor cells (PPCs) and retinal progenitor cells (RPCs). Significant obstacles to advancement of photoreceptor cell-replacement include low migration rates of transplanted cells into host retina and an absence of data describing chemotactic signaling guiding migration of transplanted cells in the damaged retinal microenvironment. Clinical evidence links arterial calcification and cardiovascular risk. Finite-element modelling of the stress distribution within atherosclerotic plaques has suggested that subcellular microcalcifications in the fibrous cap may promote material failure of the plaque, but that large calcifications can stabilize it. Yet the physicochemical mechanisms underlying such mineral formation and growth in atheromata remain unknown. Here, by using three-dimensional collagen hydrogels that mimic structural features of the atherosclerotic fibrous cap, and high-resolution microscopic and spectroscopic analyses of both the hydrogels and of calcified human plaques, we demonstrate that calcific mineral formation and maturation results from a series of events involving the …

Study Design. Fusion success with rhBMP-2 and autograft in titanium or PEEK corpectomy devices was evaluated in a sheep lumbar corpectomy model. The 6 treatment groups included titanium mesh or PEEK corpectomy devices filled with rhBMP-2 on a compression-resistant matrix (CRM) carrier; rhBMP-2 in a morselized absorbable collagen sponge (ACS) carrier combined with resorbable ceramic granules; and autograft.
Objective. The aim of this study was to determine fusion rates associated with 2 different preparations of rhBMP-2 as well as autograft in an instrumented ovine lumbar corpectomy model 6 months postoperatively.

Additionally, the fear of falling can be a major obstacle to independent living for otherwise self-sufficient individuals. This fear is significantly heightened in individuals who have sustained a fall and often results in self-imposed restrictions on mobility and exercise, causing weakening in these individuals and further exacerbating the danger. The mechanics of the running stride was altered around the 1970s when shoes were released with an elevated heel (McDougall 2009). At this point in time a heel strike running style became increasingly common and has remained the popular pattern when running in traditional shoes. In fact, the exact same person has been seen to alter their stride when running in a traditional shoe where they heel strike verses barefoot where they take on a forefoot strike (Lieberman 2010).

Walkers provided stability to the elderly but cannot assist a person from sitting to standing. The objective of this project is to present the design and analysis of a lift assist walker. This report discusses the design and analysis of a collapsible lift assist walker capable of lifting a patient up to 250 lbs. from seated to standing in under 10 seconds. The designed walker utilized a two stage scissor mechanism with a gas spring assisted embedded linear actuator.

The goal of this project was to determine whether i) fused deposition modeling could be employed to manufacture molds for vascular constructs, ii) whether vascular constructs could be created from these molds, and iii) to verify practical equivalence between observed fluid velocities. Dye tracking was to be employed to characterize fluid velocity profiles through the in vitro vascular constructs, including a half-vessel model and a full vessel model of an iliac artery aneurysm. Elastin like polypeptides (ELPs) are a class of naturally derived biomaterials that are non-immunogenic, genetically encodable, and biocompatible making them ideal for a variety of biomedical applications, ranging from drug delivery to tissue engineering. Also, ELPs undergo temperature-mediated inverse phase transitioning, which allows them to be purified in a relatively simple manner from bacterial expression hosts. Being able to genetically encode ELPs allows for the incorporation of bioactive peptides and functionalization of ELPs. This work utilizes ELPs for regenerative medicine and drug delivery.

Background: The objective of this study is to assess standardized histograms of signal intensities of T1 signal and T2 signal on sagittal view without enhancement during (1) acute stage, and (2) convalescence stage of pediatric patients with Enterovirus 71 related brainstem encephalitis (BE), and with respect to (3) healthy normal.
Methods: Our subjects were hospitalized between March 2010 and October 2012, and underwent pre- and post-contrast MRI studies. Feb. 22, 2016 – A Utah State University researcher has taken a big step toward making a safer, more natural dye that can be used in the food, textile, cosmetic and other industries.
Dr. Jixun Zhan, an associate professor of biological engineering at USU, has secured a patent for an innovative method to produce the deep blue dye known as indigoidine. The tint was originally synthesized from a bacterial strain found inRhode Island and offered a promising alternative to the synthetic dyes used to color jeans, leather, food and paper.

Sleep analysis in animal models typically involves recording an electroencephalogram (EEG) and electromyogram (EMG) and scoring vigilance state in brief epochs of data as Wake, REM (rapid eye movement sleep) or NREM (non-REM) either manually or using a computer algorithm. Computerized methods usually estimate features from each epoch like the spectral power associated with distinctive cortical rhythms and dissect the feature space into regions associated with different states by applying thresholds, or by using supervised/unsupervised statistical classifiers; but there are some factors to consider when using them:

Revision of orthopedic surgeries is often expensive and involves higher risk from complications. Since most total joint replacement devices use a polyethylene bearing, which serves as a weak link, the assessment of damage to the liner due to in vivo exposure is very important. The failures often are due to excessive polyethylene wear. The glenoid liners are complex and hemispherical in shape and present challenges while assessing the damage. Instantaneous tensile testing and stress-relaxation testing are forms of mechanical testing used to determine the elastic and viscoelastic properties of biological tissue. Equilibrium testing is a form of testing that combines both of these testing approaches at different strain points to determine the elastic properties of a material and also assess their viscoelastic properties in the same test. This testing method is commonly used on highly viscoelastic materials such as cartilage but has never been fully described in dense collagenous materials such as tendon or ligament. Background
Gaucher Disease type 1 (GD1) often manifests in childhood. Early treatment with enzyme replacement therapy (ERT) may prevent disease complications. We report the assessment of velaglucerase alfa ERT in pediatric GD1 patients who participated in a long-term extension study (HGT-GCB-044, ClinicalTrials.gov Identifier NCT00635427).
Methods
Safety and efficacy were evaluated in pediatric patients receiving velaglucerase alfa 30–60 U/kg by intravenous infusion every other week. In addition to key hematological and visceral efficacy assessments, exploratory assessments conducted specifically in pediatric patients included evaluation of height, bone age, bone marrow burden, and Tanner stage of puberty.

Application of nonlinear control theory to biomedical systems involves tackling some unique and challenging problems. The mathematical models that describe biomedical systems are typically large and nonlinear. In addition, biological systems exhibit dynamics which are not reflected in the model (so-called 'un-modeled dynamics') and hard constraints on the states and control actions, which exacerbate the difficulties in designing model-based controllers or observers.

Background: Exposure to tensile loading has been shown to maintain tendon/ligament homeostasis, with the tensile modulus of the tissue increasing in response to greater loading frequency, and even decreasing in the absence of load. However after injury or surgical repair the effect of tensile load is controversial, with many studies implicating mechanical loading with impaired healing. Purpose: To characterize the effects of intraframe distortion due to involuntary eye motion on measures of cone mosaic geometry derived from adaptive optics scanning light ophthalmoscope (AOSLO) images.
Methods: We acquired AOSLO image sequences from 20 subjects at 1.0, 2.0, and 5.08 temporal from fixation. An expert grader manually selected 10 minimally distorted reference frames from each 150-frame sequence for subsequent registration. Cone mosaic geometry was measured in all registered images (n ¼ 600) using multiple metrics, and the repeatability of these metrics was used to assess the impact of the distortions from each reference frame. Bioactive glasses have found applications in diverse fields, including orthopedics and dentistry, where they have been utilized for the fixation of bone and teeth and as scaffolds for drug delivery. The present work outlines the characterization of two novel titanium-containing glass series, one silica-based and one borate-based. For the silica-based series, titanium is added at the expense of silicon dioxide whereas for the borate-based series, it is added at the expense of boron oxide as confirmed by Energy Dispersive Spectroscopy.
