The objective of the project is to use aptamers, oligonucleotides designed to selectively bind target molecules, to develop a methodology for building an enzyme-linked aptasorbent assay (ELASA). This assay is designed to detect the presence of a target protein nucleolin that is overexpressed on the surface of cancer cells, and it would act as a novel diagnostic method for the disease. In both developed and developing countries, there is a need for a fast diagnostic system to detect pathogens within a fluid sample. In developing a microfluidic platform, which utilizes a microfluidic chip and an optical detection method, doors may be opened for new methods of determining pathogen concentration in fluid. Most biological reactions are not instantaneous. A flow-controlling mechanism with no power requirement may be implemented in the microfluidic platform. Pregnant women in low-income communities often lack access to the necessary healthcare for successful births. This is frequently due to the high costs of medical care, the remote location of patients, and the infrequency of primary care medical visits. To address this inequity, we have created a mobile application and imaging unit that allows for the low-cost implementation of urinalysis testing, which will aid in the detection of warning signs for prenatal health risks. More than 300,000 total hip replacement surgeries are performed in the US each year. Minimally invasive total hip replacement surgery requires modified tools to access the smaller incision. An offset hip impactor was redesigned to improve tool lifetime, ease of use, and manufacturing cost. The redesigned tool had an increased lifetime and a significant reduction in the number of parts. The report contains detail drawings, a manufacturing plan, and physical testing results.

Background

Improved strategies for stent‐based treatment of coronary artery disease at bifurcations require a greater understanding of artery morphology.

Objective

We developed a workflow to quantify morphology in the left main coronary (LMCA), left anterior descending (LAD), and left circumflex (LCX) artery bifurcations.

Methods

Computational models of each bifurcation were created for 55 patients using computed tomography images in 3D segmentation software. Metrics including cross‐sectional area, length, eccentricity, taper, curvature, planarity, branching law parameters, and bifurcation angles were assessed using open‐sources software and custom applications. Geometric characterization was performed by comparison of means, correlation, and linear discriminant analysis (LDA).

Iliac artery aneurysms are considered rare and difficult to detect and treat. Prompt diagnosis and timely intervention are essential, because the incidence of rupture is as high as 50%. The reported mortality rate for patients who undergo surgery for ruptured iliac artery aneurysm ranges from 50% to 70%. This study developed an in-vitro mechanical model of an iliac artery aneurysm to verify the accuracy of computer simulation software. The characterization of postsynaptic potentials, in terms of neurotransmitter receptor activation, is of clinical significance because information associated with receptor activation can be used in the diagnosis and study of neurological disorders. Single-unit recordings provide a method of measuring postsynaptic potentials in neurons using a microelectrode system, but yield no detailed information regarding the neurotransmitter receptors that contribute to the potential. Purpose: To evaluate how metrics used to describe the cone mosaic change in response to simulated photoreceptor undersampling (i.e., cell loss or misidentification).

Methods: Using an adaptive optics ophthalmoscope, we acquired images of the cone mosaic from the center of fixation to 10° along the temporal, superior, inferior, and nasal meridians in 20 healthy subjects. The World Health Organization estimates that over 30 million people require some sort of prosthetic technology. However, traditional prosthetic fitting practices take a lot of time and cost a lot of money, making them inaccessible to millions of people around the world. A portable near-infrared spectral tomography (NIRST) system was developed with simultaneous frequency domain (FD) and continuous-wave (CW) optical measurements for efficient characterization of breast cancer in a clinical oncology setting. Simultaneous FD and CW recordings were implemented to speed up acquisition to 3 minutes for all 9 wavelengths, spanning a range from 661nm to 1064nm. An adjustable interface was designed to fit various breast sizes and shapes. The robotic lower extremity exoskeletons address the ambulatory problems confronting individuals with paraplegia. Paraplegia due to spinal cord injury (SCI) can cause motor deficit to the lower extremities leading to inability to walk. Raman spectroscopy was applied to nail clippings from 633 postmenopausal British and Irish women, from six clinical sites, of whom 42% had experienced a fragility fracture. The objective was to build a prediction algorithm for fracture using data from four sites (known as the calibration set) and test its performance using data from the other two sites (known as the validation set). Results from the validation set showed that a novel algorithm, combining spectroscopy data with clinical data, provided area under the curve (AUC) of 74% compared to an AUC of 60% from a reduced QFracture score (a clinically accepted …

The first part of the dissertation is on Finite Difference Time Domain (FDTD) optimization and experimental demonstration of a nano-scale instrument that allows sensing at the cellular and subcellular levels. Basal release of nitric oxide from endothelial cells modulates contractile activity in the corpus cavernosum via inhibition of the RhoA/Rho-kinase signaling pathway. We aimed to investigate nitric oxide bioavailability, oxidative stress and the Rho-kinase pathway in the relaxation of the corpus cavernosum of an obese and diabetic model of mice (db/db mice). We hypothesized that in db/db mice impaired relaxation induced by Rho-kinase inhibitor is accompanied by diminished NO bioavailability, increased oxidative stress and upregulation of the RhoA/Rho-kinase signalling pathway. Less than half of patients suffering from major depressive disorder, a leading cause of disability worldwide, achieve remission with current antidepressants, making it imperative to develop more effective treatment. A new therapeutic direction is emerging from the increased understanding of natural resilience as an active stress-coping process. It is known that potassium (Kþ) channels in the ventral tegmental area (VTA) are an active mediator of resilience. However, no druggable targets have been identified to potentiate active resilience mechanisms.
