We report a miniature, visible to near infrared G-Fresnel spectrometer that contains a complete spectrograph system, including the detection hardware and connects with a smartphone through a microUSB port for operational control. The smartphone spectrometer is able to achieve a resolution of ~5 nm in a wavelength range from 400 nm to 1000 nm. We further developed a diffuse reflectance spectroscopy system using the smartphone spectrometer and demonstrated the capability of hemoglobin measurement. Proof of concept studies of tissue phantoms yielded a mean error of 9.2% on hemoglobin concentration measurement, comparable to that obtained with a commercial benchtop spectrometer. The …
Far-Field Optical Microscopy Based On Stimulated Emission Depletion, Yunxia Wang
Far-Field Optical Microscopy Based On Stimulated Emission Depletion, Yunxia Wang
Theses and Dissertations
Conventional lens-based (far-field) fluorescence microscopy is a widely used imaging technique with spatial resolution up to 150–350 nm. However, this technology cannot discern very small structural features, because the spatial resolution is limited by diffraction to about half of the wavelength of light (λ/2,λ is the wavelength of light). Hence, most of the developments in microscopy aim at improving resolution. In the past decades, stimulated emission depletion (STED) microscopy has been developed to bypass the diffraction limit for the application in biological imaging with resolution approaching the nanoscale. The basic principle of STED microscopy is to employ a doughnut-shape laser …

Background—Prior studies have reported differences in lumbo-pelvic kinematics during a trunk forward bending and backward return task between individuals with and without chronic low back pain; yet, the literature on lumbo-pelvic kinematics of patients with acute low back pain is scant. Therefore, the purpose of this study was set to investigate lumbo-pelvic kinematics in this cohort.
Methods—A case-control study was conducted to investigate the differences in pelvic and thoracic rotation along with lumbar flexion as well as their first and second time derivatives between females with and without acute low back pain. Participants in each group completed one …
Modified Pamam Dendrimers In Tunable Drug-Delivery Systems: A Sustained-Release Dendrimer Hydrogel For Anti-Glaucoma Drugs And Surface-Engineered Macrophages As Nanoparticle Carriers For Targeted Anti-Cancer Therapy, Christopher A. Holden
Theses and Dissertations
Two specific drug-delivery applications were sought in this work using polyamidoamine (PAMAM) dendrimers. One drug-delivery system used a novel dendrimer hydrogel (DH) for sustained delivery of anti-glaucoma drugs. In this work, PAMAM G3.0 dendrimers were covalently bonded with poly(ethylene glycol) (PEG12000) molecules which were subsequently acrylated, resulting in photocurable DH conjugates. For pharmacological studies, DH were loaded with a solution of intraocular pressure lowering drugs, brimonidine and timolol maleate, and were characterized for in vitro release and ex vivo transport and uptake. DH formulations were shown to increase the loading of drug molecules, increase transcorneal drug delivery, and …

Development And Validation Of A Novel Resonant Energy Transfer (Fret) Biosensor To Measure Tensile Forces At The Linc Complex In Live Cells, Paul Arsenovic
Theses and Dissertations
There is a large body of evidence supporting the theory that cell physiology largely depends on the mechanical properties of its surroundings or micro-environment. More recently studies have shown that changes to intra-cellular mechanical properties can also have a meaningful impact on cell function and in some cases lead to the progression of ailments or disease. For example, small changes to the protein sequence of a structural nuclear envelope protein called lamin-A is known to cause a variety of neurological and musculoskeletal diseases referred to as laminopathies. Currently, there is little incite into how these mutations lead to disease progression …
Electrospraying Extracellular Matrix To Form Nanoparticles, Patrick Link
Electrospraying Extracellular Matrix To Form Nanoparticles, Patrick Link
Theses and Dissertations
Chronic Obstructive Pulmonary Disease (COPD) is a leading cause of death worldwide. Alveolar wall destruction is a significant contributor to COPD. Inflammatory macrophages are a major source of the Extracellular Matrix (ECM) proteolysis. ECM breakdown causes air to get trapped in the alveoli, obstructing airflow. One step in curing COPD may be to convert inflammatory to pro-regenerative macrophages. Recently, decellularized ECM scaffolds have shown the ability to induce a pro-regenerative phenotype.
Yet these scaffolds are incapable for reaching the alveolar region of the lungs. To reach the alveolar region particles need a diameter of 1-5 μm or smaller than 300 …
The Effect Of Vibrotactile Feedback On Remote Manual Task Performance, Matthew S. Standard
The Effect Of Vibrotactile Feedback On Remote Manual Task Performance, Matthew S. Standard
Theses and Dissertations
Vibrotactile feedback offers a unique opportunity to augment or reconstruct impaired tactile sensations, whether that be in the form of enhancing prosthetics or specialized protective clothing. Important information about temperature and object slippage serve to endanger the human operator or equipment. This thesis presents three experiments which investigate amplitude modulated vibrotactile signals as a scalar dimension of roughness, the effect those signals and their locations (finger pad, forearm, bicep) have on the performance of two tasks: the sensing of temperatures simulated by vibrotactile signals and gripping an object of simulated surface texture. The results show task performance increase when the …
Altering A Runner’S Foot Strike Using A Modified Elliptical Trainer, Daniel Shull
Altering A Runner’S Foot Strike Using A Modified Elliptical Trainer, Daniel Shull
Theses and Dissertations
One possible solution to common running related injuries is to transition runners from a rearfoot strike during initial contact to a midfoot strike. Natural rearfoot strike runners were studied to see if a modified elliptical trainer could be used to alter their running pattern to that of a midfoot strike runner. Their results were compared to subjects who ran on a non-modified elliptical trainer. After training on the modified elliptical trainer, subjects demonstrated a decrease in foot angle at initial contact when attempting to run with a midfoot strike. Training did not affect all kinetic metrics or stride frequency. However, …

Cardiovascular diseases are disorders affecting the blood vessels and the heart. According to the World Health Organization, cardiovascular diseases are one of the leading causes of death worldwide. They are responsible for over 17.1 million deaths per year worldwide, representing 31.5% of deaths 1, 2. Atherosclerosis, a chronic inflammatory disorder affecting large arteries, is the underlying cause of many cardiovascular diseases. Plaque rupture is a serious complication of advanced atherosclerosis, often leading to life-threatening clinical consequences such as myocardial infarction (heart attack) or stroke. 75% of newly developed myocardial infarction cases are caused by atherosclerotic plaque rupture. It affects approximately …
Dynamic Model And Estimator Development For A Smart Refractory Brick With Embedded Wireless Sensors For Gasifier Applications, Qiao Huang
Graduate Theses, Dissertations, and Problem Reports (ETD)
The operating temperature is one of the most important variables for gasifier operation. A higher temperature can shorten the service life of the refractory lining. A lower temperature can reduce the extent of carbon conversion and disrupt the flow of molten slag in a slagging gasifier. Therefore temperature is one of the most important variable in a gasifier. In slagging gasifiers, molten slag can penetrate into the refractory lining leading to refractory spallation, undesired downtime, and costly replacement. Even though the temperature and extent of slag penetration are extremely important for the gasifier operation, they cannot be measured reliably by …
A Comprehensive Analysis On Eeg Signal Classification Using Advanced Computational Analysis, Kaushik Bhimraj
A Comprehensive Analysis On Eeg Signal Classification Using Advanced Computational Analysis, Kaushik Bhimraj
College of Graduate Studies: Theses & Dissertations
Electroencephalogram (EEG) has been used in a wide array of applications to study mental disorders. Due to its non-invasive and low-cost features, EEG has become a viable instrument in Brain-Computer Interfaces (BCI). These BCI systems integrate user's neural features with robotic machines to perform tasks. However, due to EEG signals being highly dynamic in nature, BCI systems are still unstable and prone to unanticipated noise interference. An important application of this technology is to help facilitate the lives of the tetraplegic through assimilating human brain impulses and converting them into mechanical motion. However, BCI systems are remarkably challenging to implement …
3d Bioprinting Hydrogel For Tissue Engineering An Ascending Aortic Scaffold, Benjamin Stewart
3d Bioprinting Hydrogel For Tissue Engineering An Ascending Aortic Scaffold, Benjamin Stewart
Electronic Theses and Dissertations
The gold standard in 2016 for thoracic aortic grafts is Dacron®, polyethylene terephthalate, due to the durability over time, the low immune response elicited and the propensity for endothelialization of the graft lumen over time. These synthetic grafts provide reliable materials that show remarkable long term patency. Despite the acceptable performance of Dacron® grafts, it is noted that autographs still outperform other types of vascular grafts when available due to recognition of the host's cells and adaptive mechanical properties of a living graft. 3-D bioprinting patient-specific scaffolds for tissue engineering (TE) brings the benefits of non-degrading synthetic …
The Effect Of Twist On The Mechanical Properties Of The Intervertebral Disc, Maxine Harvey-Burgess
The Effect Of Twist On The Mechanical Properties Of The Intervertebral Disc, Maxine Harvey-Burgess
Theses and Dissertations (Comprehensive)
Introduction: Intervertebral disc (IVD) herniation is a common injury to the IVD and a frequent source of low back pain. IVD herniation occurs when the nucleus pulposus migrates through both the inter- and intralamellar matrices of the annulus fibrosus (AF). There are a number of mechanical risk factors associated with herniation, including repetitive flexion and twist. In vivo, twist combined with repetitive flexion has been associated with increased experience of low back pain and herniation. Additionally, in vitro, when repetitive flexion is combined with twist, IVD herniation occurs more easily than repetitive flexion alone. However, the mechanisms behind …
An Enhanced Spring-Particle Model For Red Blood Cell Structural Mechanics: Application To The Stomatocyte–Discocyte– Echinocyte Transformation, Mingzhu Chen, Fergal Boyle
An Enhanced Spring-Particle Model For Red Blood Cell Structural Mechanics: Application To The Stomatocyte–Discocyte– Echinocyte Transformation, Mingzhu Chen, Fergal Boyle
Articles
Red blood cells (RBCs) are the most abundant cellular element suspended in blood. Together with the usual biconcave-shaped RBCs, i.e., discocytes, unusual-shaped RBCs are also observed under physiological and experimental conditions, e.g., stomatocytes and echinocytes. Stomatocytes and echinocytes are formed from discocytes and in addition can revert back to being discocytes; this shape change is known as the stomatocyte–discocyte–echinocyte (SDE) transformation. To-date, limited research has been conducted on the numerical prediction of the full SDE transformation. Spring- particle RBC (SP-RBC) models are commonly used to numerically predict RBC mechanics and rheology. However, these models are incapable of predicting the full …

3d Bioprinting A Pcl/13-93b3 Glass Composite And Its Potential Use As A Bio-Ink, Caroline Blair Murphy
Masters Theses
"A major limitation of using synthetic scaffolds in tissue engineering is little growth of incorporated cells in the interior of the scaffold, resulting in insufficient angiogenesis in the scaffold interior. Recently, cells have been 3D bioprinted concurrently with biomaterials to produce a cellularized, bioactive, angiogenic 3D environment. This thesis describes a novel solvent-extrusion method for printing polycaprolactone (PCL)/bioactive borate glass composite as a biomaterial for a cell-laden scaffold.
Bioactive borate glass was added to a mixture of PCL and organic solvent to make an extrudable paste, creating scaffolds measuring 10×10×1 mm3 in overall dimensions with pore sizes ranging from …
Combination Of Bmp-2 And Relaxin To Induce Osteogenesis In Vivo, Sahitya Injamuri
Combination Of Bmp-2 And Relaxin To Induce Osteogenesis In Vivo, Sahitya Injamuri
Masters Theses
"Bone grafts are a common procedure for treating craniofacial injuries such as trauma, surgery, and reconstruction. Autografts and allografts have limitations which encourages the use of synthetic bone grafts. While synthetic bone grafts, such as hydroxyapatite (HA) microspheres, are advantageous for their cost and reproducibility, they lack the cells and growth factors needed for proper osteoinduction. Bone morphogenetic protein-2 (BMP-2) has been shown to increase the osteoinduction of grafts but the effective dose is above physiological levels and causes undesired effects on the body. Relaxin has been shown to enhance BMP-2's ability to induce bone formation in vitro and thus …
Segmental Movement Compensations In Patients With Transtibial Amputation Identified Using Angular Momentum Separation, Brecca M. M. Gaffney
Segmental Movement Compensations In Patients With Transtibial Amputation Identified Using Angular Momentum Separation, Brecca M. M. Gaffney
Electronic Theses and Dissertations
Patients with unilateral dysvascular transtibial amputation (TTA) adopt movement compensations that are required to maintain balance and achieve ambulation in the absence of ankle plantar flexion, and result in increased and asymmetric joint loading patterns. As a result, this population is at an increased risk of overuse injuries, such as low back pain (LBP). Clinical gait analysis is used to guide diagnostics in movement retraining following amputation, and is performed using instrumented (research based) or observational analyses (clinically based). However, instrumented analyses are currently impractical in most clinical settings due to expense and computational limitations. This dissertation presents the use …
Mechanical Interventions In Soft Tissue Repair, Elizabeth Marie Meier
Mechanical Interventions In Soft Tissue Repair, Elizabeth Marie Meier
Wayne State University Dissertations
This body of work sets to investigate some of these mechanical interventions that are designed to promote wound healing, repair, or even replace an injured tissue. By investigating three separate tissues and three separate mechanical interventions, we can draw conclusions about the implications of including mechanical interventions in biomedical research and clinical treatments. The use of sutures to close wounds is highly common, however the effects of sutures on the tensile mechanics of human skin are largely unknown. To evaluate how sutures may affect uniaxial tensile mechanics, human skin samples were sutured and loaded in tension in multiple orientations. The …
Development Of A Programmed Electrospun Three Dimensional (3d) Nanofiber Collecter And It’S Application To Orthopedic Implant Coatings, Liang Chen
Wayne State University Dissertations
Orthopedic implants might not directly unite with bones especially in compromised patients even if they have been appropriately fixed. The lack of early osseointegration would lead to the failure of the orthopedic implant. A “bone-like” implant surface is urgently needed to accelerate osseointegration. Electrospun nanofiber (NF) is a promising implant coating due to its highly porous nanoscale structure. It mimics the collagen I nanofibrous network of bone tissue; meanwhile it has been widely used as a drug delivery device. However, its compact and dense structure is not ideal for cell growth. Our strategy was to develop a functional three-dimensional (3D) …
Six Degrees Of Freedom: Kinematics Of The Healthy Ankle Syndesmosis Joint, Veronica A. Hogg-Cornejo
Six Degrees Of Freedom: Kinematics Of The Healthy Ankle Syndesmosis Joint, Veronica A. Hogg-Cornejo
Electronic Theses and Dissertations
Syndesmotic injury, more commonly known as a "high ankle sprain", accounts for over 12% of all ankle sprain incidents in the US; of which, over 25% occur during a sporting activity. Typically, harm to the syndesmosis occurs in sports such as football, soccer, lacrosse, and hockey where it is common for an athlete to experience rapid and extreme dorsiflexion-external rotations of the foot. Severe syndesmotic sprains have been noted by clinicians as the most difficult ankle injury to accurately diagnose and treat, require the most recuperation time, and often results in life-long dysfunction. Even more problematic, 40% of patients suffering …

Who says that learning about the various properties of Biomaterials is boring? Clearly, they haven't taken Dr. Yu Huang's BENG 2330 Properties of Biomaterials course. Not only do these students know how to distinguish the differences between natural and synthetic biomaterials. They know how to have a good time bringing the mannequin challenge into their lab clean up.

Bone development and length relies on the growth plate formation, which is dependent on degradative enzymes such as MMPs. Indeed, deletion of specific members of this enzyme family in mice results in important joint and bone abnormalities, suggesting a role in skeletal development. As such, the control of MMP activity is vital in the complex process of bone formation and growth. We generated a transgenic mouse line to overexpress TIMP3 in mouse chondrocytes using the Col2a1-chondrocyte promoter. This overexpression in cartilage resulted in a transient shortening of growth plate in homozygote mice but bone length was restored at eight weeks …

Activity Preservation Of Plasmonic Biosensors With A Metal-Organic Framework, Lu Wang
McKelvey School of Engineering Graduate Student Theses & Dissertations
Antibody-antigen recognition enables antibody-conjugated nanostructures to serve as plasmonic biosensors with tunable specificity. However due to the instability of antibodies, these biosensors are susceptible to changes in the environment such as heat and aridity, leading to constraints on the transportation and handling of these sensors. Here we establish a method using a metal-organic framework crystal to preserve biosensor activity under severe environmental conditions, including exposure to high temperatures, an organic solvent and a proteolytic agent. After zeolitic imidazolate framework-8 (ZIF-8) crystals formed for 12 hours on a biosensor of gold nanorods conjugated with a model antibody, rabbit IgG, 80% of …

Slight alterations in nanoparticles' surface properties can significantly influence the corona composition which may alter their interaction with the biological milieu. Size and porosity of silica nanoparticles (SNPs) are known to be predominant factors influencing their dose-dependent toxicity. Little is known however about the extent and type of protein adsorption on SNPs as a function of physicochemical properties and the role this might play on mechanisms of cellular uptake and toxicity. In this work we investigated the influence of size and porosity of SNPs on protein adsorption, cellular uptake, and toxicity in RAW 264.7 macrophages. Toxicity of the SNPs was …
