Developing A Multifunctional Thermo-Responsive Viral Like Particle (Vlp) Platform With New Physicochemical Characteristics, Sepehr Dejdar

ETD Archive

Smart materials are a class of material whose physical characteristics change significantly in a controllable fashion once exposed to an external stimulus such as stress, pH, light, temperature, etc. If the temperature is the external stimuli (control variable) the material is termed thermo-responsive. Out of numerous thermo-responsive materials, elastin-like polypeptide (ELP) is a well-known protein-based biopolymer. Protein cages are well-defined biological nanostructures which are highly symmetrical and monodisperse. This class of proteins are both chemically and genetically tunable, making them a suitable recipient of new properties. Protein re-engineering is the basis of developing a hybrid platform (e.g. polymer-protein) to deliver …

Development Of Photoacoustic Tomographic Systems For Brain Hemorrhage Detection, Karl Kratkiewicz Jan 2020

Development Of Photoacoustic Tomographic Systems For Brain Hemorrhage Detection, Karl Kratkiewicz

Wayne State University Dissertations

Preterm neonates (age) and/or with low birth weight (

Photoacoustic imaging (PAI) is an imaging modality which derives its contrast from differences in optical absorption coefficients of tissue through laser irradiation and acoustic detection. Therefore, PAI derives its signal directly from optical absorbers such as blood; the critical component in a hemorrhage. We discuss operation and processing methods of our Verasonics Vantage system. We then investigate thermal safety of PAI in mouse model for various laser pulse repetitions rates and illumination duration. We then develop two photoacoustic tomographic systems for eventual translation to clinic for hemorrhage detection. One system being …

Spatiotemporal Release Of Growth Factors In A Biomaterial-Focused Organotypic Spinal Cord Injury Model, Elizabeth Mays Jan 2020

Spatiotemporal Release Of Growth Factors In A Biomaterial-Focused Organotypic Spinal Cord Injury Model, Elizabeth Mays

Wayne State University Dissertations

Over 17,000 people per year in the United States sustain a spinal cord injury (SCI) for which there is no gold standard of care and life-long complications. SCI is a complex wound environment with various growth factors (GFs), cellular activity, and scar formation at various timepoints. For example, basic fibroblast growth factor (bFGF) is released immediately to protect local support cells, namely oligodendrocytes (OLs). Brain derived neurotrophic factor (BDNF) has peak expression at 2-3 days in mice and 5-7 days in humans, and aids axonal growth across the injury site. Incidentally this is around the same time as M1 macrophage …

In Vitro Atherosclerosis Disease Model Via The Ring Stacking Method, Cameron Brandon Pinnock Jan 2020

In Vitro Atherosclerosis Disease Model Via The Ring Stacking Method, Cameron Brandon Pinnock

Wayne State University Dissertations

Creation of an in vitro atherosclerotic disease model using the novel Ring Stacking Method. Singular self-assembling tissue rings made up smooth muscle cells and fibrin hydrogel are stacked on one another to create a tissue engineered vessel. These biologically engineered blood vessels are then seeded with endothelial cells via combined static rotational and dynamic bioreactor in order to create a functional intima layer. Early stage atherosclerosis was induced via the addition of oxidized low-density lipoproteins (ox-LDL) to the fibrin hydrogel that creates the media layer of the engineered vessel. After the creation of the intima layer the engineered vessel was …

Development Of A Novel Cardiac Ischemia-Reperfusion Model In The Axolotl, Jeremy Tolentino Llaniguez Jan 2020

Development Of A Novel Cardiac Ischemia-Reperfusion Model In The Axolotl, Jeremy Tolentino Llaniguez

Wayne State University Dissertations

The Center for Disease Control’s National Center for Health Statistics data for mortality from diseases of the heart show the age-adjusted death rate has fallen from almost 600 deaths in the 1950s to just over 190 deaths per 100,000 U.S. residents today. With the recognized limitations of pharmacotherapy of myocardial infarction (MI), cell-based therapies have been undergoing rapid development and clinical testing. However, there is still no consensus about cell types, delivery routes, dosing and treatment schedules and pretreatment conditioning of cells prior to administration. Furthermore, a fundamental question remains unanswered about the reasons for the poor capacity for myocardial …

Electrochemical Detection Of Reactive Oxygen Species Via A Platinum Microelectrode Array, Victor M. Carriere Jr. Jan 2020

Electrochemical Detection Of Reactive Oxygen Species Via A Platinum Microelectrode Array, Victor M. Carriere Jr.

Master's Theses

Oxidative stress, an excess of endogenous or exogenous reactive oxygen species (ROS) in the body, is closely aligned with inflammatory responses. ROS such as hydrogen peroxide, superoxide, and radical hydroxyl ion serve essential functions in fighting infection, but chronic elevation of these species irreversibly damages cellular components. Given the central role of inflammation in a variety of diseases, including Alzheimer’s Disease, atherosclerosis, and rheumatoid arthritis, a low-cost, extracellular, non-invasive assay of ROS is needed.

This work reports the use of a platinum microelectrode array (Pt MEA)-based ceramic probe to detect time- and concentration-dependent variations in hydrogen peroxide (H2O2) production by …

Incorporating Cardiac Substructures Into Radiation Therapy For Improved Cardiac Sparing, Eric Daniel Morris Jan 2020

Incorporating Cardiac Substructures Into Radiation Therapy For Improved Cardiac Sparing, Eric Daniel Morris

Wayne State University Dissertations

Growing evidence suggests that radiation therapy (RT) doses to the heart and cardiac substructures (CS) are strongly linked to cardiac toxicities, though only the heart is considered clinically. This work aimed to utilize the superior soft-tissue contrast of magnetic resonance (MR) to segment CS, quantify uncertainties in their position, assess their effect on treatment planning and an MR-guided environment.

Automatic substructure segmentation of 12 CS was completed using a novel hybrid MR/computed tomography (CT) atlas method and was improved upon using a 3-dimensional neural network (U-Net) from deep learning. Intra-fraction motion due to respiration was then quantified. The inter-fraction setup …

Computational Analysis Of Oxidative Stress In Endothelial Dysfunction: Insights On The Role Of Tetrahydrobiopterin, Ascorbate And Glutathione, Sheetal Kedar Panday Jan 2020

Computational Analysis Of Oxidative Stress In Endothelial Dysfunction: Insights On The Role Of Tetrahydrobiopterin, Ascorbate And Glutathione, Sheetal Kedar Panday

Wayne State University Dissertations

Oxidative stress and endothelial dysfunction are reported in the cardiovascular and neurovascular diseases. Oxidative stress is caused due to an increase in the generation of reactive oxygen (ROS) and nitrogen species (RNS) and incapacity of antioxidant systems to eliminate ROS and RNS. Endothelial dysfunction is characterized by a reduction in nitric oxide (NO) bioavailability. NO is constitutively produced by enzyme endothelial nitric oxide synthase (eNOS). A reduction in tetrahydrobiopterin (BH4), which is an essential cofactor of eNOS, can lead to eNOS uncoupling. There is complex interplay between the ROS/RNS and antioxidant system underlying pathophysiologies of vascular diseases, however our quantitative …

Nanoparticle-Mediated Controlled Myocardial Drug Delivery: A New Treatment For Hypertrophic Cardiomyopathy, Michelle Katherine Mcgrath Copeland Jan 2020

Nanoparticle-Mediated Controlled Myocardial Drug Delivery: A New Treatment For Hypertrophic Cardiomyopathy, Michelle Katherine Mcgrath Copeland

Bioengineering Dissertations - Archive

Hypertrophic cardiomyopathy (HCM) is a genetic disease of the sarcomere, resulting in overgrowth of the septum that separates the left and right ventricles. HCM can affect as many as 1/200 people, making it the most common genetic cardiomyopathy in the general population, as well as the most common cause of sudden cardiac death (SCD). In severe HCM cases, cardiothoracic surgeons perform septal myectomy to remove the excess tissue. However, many patients are poor surgical candidates and require another, less invasive treatment option. In these cases, alcohol septal ablation (ASA) is performed. With ASA, cardiologists deliver pure alcohol through a catheter …

C-Molecules: Molecular-Guided Surgery With Fluorescence Imaging, Riley Karp, Joshua Gutow, William Wolfe-Mcguire Jan 2020

C-Molecules: Molecular-Guided Surgery With Fluorescence Imaging, Riley Karp, Joshua Gutow, William Wolfe-Mcguire

ENGS 89/90 Reports

Surgeries may cost lives if performed inadequately, which is why tools such as fluorescence guided surgery exist to improve surgical outcomes. Fluorescence guided surgery (FGS) is the practice of injecting patients with a fluorescent molecule that, in combination with highly sensitive cameras, allows surgeons to clearly identify complex biological structures. While beneficial in procedures such as endoscopy and gallbladder surgery, this technique is most notable for its role in complex cancer surgeries where misidentifying the boundaries of a tumor can have deadly consequences. Although FGS has the potential to revolutionize cancer treatment, highly sensitive imaging systems are necessary to identify …

Assessing 3d Printability Of Bioinks, Nicole Ramirez Jan 2020

Assessing 3d Printability Of Bioinks, Nicole Ramirez

Honors Undergraduate Theses

The field of tissue engineering (TE) is continuously improving through the use of additive manufacturing techniques (AM) such as three-dimensional (3D) bioprinting. The 3D bioprinter has significantly gained attention in the TE field because it is more efficient than regenerative medicine and is readily available as opposed to organ transplants. Working like a conventional 3D printer, the 3D bioprinter is able to dispense material layer by layer from the bottom up with the printing head able to move in the X, Y, and Z direction. This movement allows for the fabrication of structures with complex geometries. In this study, the …

Driver drowsiness has been a significant hazard resulting in various traffic accidents. Therefore, monitoring this condition is crucial not only in alerting drivers, but also in avoiding fatal accidents. Many research studies propose new systems to reduce the number of drowsiness-related injuries and fatalities. The ultimate goal for a drowsiness detection system is to detect the drowsiness on time and minimize the system or environment errors to avoid false readings, such as studying physiological signal processing patterns. These potentially life-saving systems must operate in a timely manner with the highest precision. Researchers proposed various methods based on driving pattern changes, …

Evaluation Of Far-Side Occupant Safety Based On Numerical Modeling, Syed Akhtar Imam Jan 2020

Evaluation Of Far-Side Occupant Safety Based On Numerical Modeling, Syed Akhtar Imam

Wayne State University Dissertations

The objective of this study was to augment the knowledge of the far-side occupant injury biomechanics in side impact vehicle crashes. Most research studies conducted to investigate the far-side occupant injuries are through the field crash data for a better understanding of human impact responses, injury mechanisms, and injury tolerance levels. The data obtained from field data is also used in the development of injury mitigation technologies, such as safety belts, airbags, etc. A field data represents the injury outcome of an automotive crash, but it doesn’t leave behind enough information for in-depth knowledge. The use of cadaver is the …

Development Of Rat Head Finite Element Model And Tissue Level Biomechanical Threshold For Traumatic Axonal Injury, Runzhou Zhou Jan 2020

Development Of Rat Head Finite Element Model And Tissue Level Biomechanical Threshold For Traumatic Axonal Injury, Runzhou Zhou

Wayne State University Dissertations

Traumatic brain injury (TBI) is caused by local tissue deformation at the time of trauma, leading to neurological dysfunction. In the United States alone, 2.87 million people sustain a TBI each year, of which one-fifth results in death. Traumatic axonal injury (TAI) is a well-recognized consequence of every fatal head injury and more than 85% of vehicular crash-related blunt head injuries. The most common and important pathologic feature of TBIs are multifocal changes to axons in the white matter produced by rapid head acceleration/deceleration during a traumatic event with consequent local shear/tension on neural tissue and axons contributing to secondary …

Risks And Benefits Of Using A Commercially Available Ventricular Assist Device For Failing Fontan Cavopulmonary Support: A Modeling Investigation, Masoud Farahmand, Minoo N. Kavarana, Ethan Kung

Publications

Fontan patients often develop circulatory failure and are in desperate need of a therapeutic solution. A blood pump surgically placed in the cavopulmonary pathway can substitute the function of the absent sub-pulmonary ventricle by generating a mild pressure boost. However, there is currently no commercially available device designed for the cavopulmonary application; and the risks and benefits of implanting a ventricular assist device (VAD), originally designed for the left ventricular application, on the right circulation of failing Fontan patients is not yet clear. Moreover, further research is needed to compare the hemodynamics between the two clinically-considered surgical configurations for cavopulmonary …

Bioelectrics Publications

Increased levels of heat shock protein 90 (HSP90) have been recently implicated in the pathogenesis of pulmonary fibrosis and the use of HSP90 inhibitors constitutes a potential therapeutic approach. Similarly, acute exposure to nitrogen mustard (NM) is related to the development of chronic lung injury driven by TNF-α, TGF-β, ERK and HSP90. Thus, we developed a murine model of NM-induced pulmonary fibrosis by instilling C57BI/6J mice with 0.625 mg/kg mechlorethamine hydrochloride. After 24 h, mice began receiving AUY-922, a second generation HSP90 inhibitor, at 1 mg/kg 2 times per week or 2 mg/kg 3 times per week, for either 10 …

Extracellular Matrix Nanoparticles Effects On The Lung In Vivo, Brittaney E. Ritchie Jan 2020

Extracellular Matrix Nanoparticles Effects On The Lung In Vivo, Brittaney E. Ritchie

Theses and Dissertations

Acute respiratory distress syndrome (ARDS) is a life-threatening condition that causes diffuse alveolar damage and a loss of the extracellular matrix (ECM). This leads to pulmonary edema and lung function deterioration. Our lab has created decellularized porcine lung, electrosprayed ECM nanoparticles that have been previously shown to have pro-regenerative capabilities in vitro.

In this study, the ECM nanoparticle effects on young murine lungs were tested in vivo. An ECM nanoparticle suspension, previously used for the in vitro studies, was aerosolized intratracheally into the lungs using a microsprayer. 24 hours later, the lung mechanics, bronchoalveolar lavage fluid, and histology …

Development Of A Single Use Device Intended To Wash Blood Clot Debris From A Stent Retriever In The Operating Room, Shane Diller Jan 2020

Development Of A Single Use Device Intended To Wash Blood Clot Debris From A Stent Retriever In The Operating Room, Shane Diller

Theses and Dissertations

Ischemic stroke is a life-threatening condition that requires immediate surgical intervention to remove the blood clot that is lodged in the blood vessels of the brain. Mechanical thrombectomy, performed by an endovascular surgeon, involves inserting a stent retriever, which deploys distally and attempts to capture the clot when dragged backwards. A complication prevalent in 50% - 67% of cases is that the blood clot is only partially removed and clings to the mesh framework of the stent. The operating surgeon must either clean the debris from the stent retriever, to be reinserted or throw the stent retriever away and open …

Automatic Hip Analysis Suite, Nathan J. Veilleux Jan 2020

Automatic Hip Analysis Suite, Nathan J. Veilleux

Theses and Dissertations

Image recognition and computer vision are becoming increasingly useful for diagnostic and surgical planning purposes. Analyses of patient morphology can be performed much more quickly and reliably with algorithmic assistance. Traditional measurements of the hip involve visual inspection of patient radiographs or hand calculation with software assistance. In the most extreme cases, estimates of bone orientation are made via palpation of a patient’s hip. There is the possibility for significant improvements in reliability if measurements can be made in the absence of human interpretation. As many of these techniques require CT scans as a prerequisite, all of the necessary information …

Development Of Cellulose Acetate-Based Scaffold For Bone Tissue Engineering Applications, Ruhit Sinha Jan 2020

Development Of Cellulose Acetate-Based Scaffold For Bone Tissue Engineering Applications, Ruhit Sinha

Electronic Theses and Dissertations

Cellulose Acetate (CA) based biomaterials are being used as substrates for bone ingrowth applications due to their nontoxic and nonirritant nature coupled with optimum morphology and stiffness. Electrospinning with additives and/or post-treatment has emerged as a viable protocol to further improve mechanical properties of CA and expand its utility. Herein, we highlight the role of potassium chloride (KCl) in association with airdrying to enhance the elastic modulus and tensile strengths of CA fibers. Salt aggregation in between fibers is observed through Scanning Electron Microscopy (SEM), however, Fourier-transform infrared (FTIR) analysis signifies the interactions between K+ ions and acetyl groups. The …

Comparison Of Hybridizing Options For Solar Heat, Biomass And Heat Storage For Electricity Generation In Spain, Cm Iftekhar Hussain, Brian Norton, Aidan Duffy Jan 2020

Comparison Of Hybridizing Options For Solar Heat, Biomass And Heat Storage For Electricity Generation In Spain, Cm Iftekhar Hussain, Brian Norton, Aidan Duffy

Articles

Diurnal and seasonal variations of direct solar energy circumscribe locations suitable for sole use of concentrated solar power for electricity generation, whereas to generate electricity at night and during winter months, biomass systems can be used that incur fuel expenditures and may encounter fuel supply discontinuities. Combining solar heat, biomass conversion and heat storage in a hybrid electricity generation plant may overcome the limitations of sole use of any one of these energy sources in specific geographic and/or economic conditions. In this work standalone biomass, hybrid concentrated solar power-biomass and hybrid concentrated solar power-biomass-thermal energy storage power generation systems are …

Increasing Performance Of Classifiers For Ssvep-Based Brain-Computer Interfaces Using Extension Methods, Ethan Douglas Webster Jan 2020

Increasing Performance Of Classifiers For Ssvep-Based Brain-Computer Interfaces Using Extension Methods, Ethan Douglas Webster

Legacy Theses & Dissertations (2009 - 2024)

Brain-computer interfaces (BCI) provide an alternative communication method that does not require standard physical mediums (speech, typing, etc.). These systems have been implemented to provide additional communication and control options for people with certain motor disabilities. Classification is an important part of BCI systems and consists of inferring user commands from brain activity. Supervised classification methods often achieve higher accuracy, but unsupervised classification methods are useful when training is not practical for the user. This thesis focuses on unsupervised classification algorithms used for a BCI speller application and presents extensions for two existing classifiers that improve classification accuracy and thus …
