Effects Of Hard-To-Soft Segment Ratios On The Synthesis And Physico-Mechanical Properties Of Polyurethane Films, Aaron C. Wilson May 2020

Effects Of Hard-To-Soft Segment Ratios On The Synthesis And Physico-Mechanical Properties Of Polyurethane Films, Aaron C. Wilson

Mechanical Engineering Theses

Blood-contacting cardiovascular stents often induce a secondary clotting event due to unrestricted enzymatic activities. The use of hemocompatible polyurethane coatings on these implantable devices is one of the most promising methods to reduce device rejection. In this study, four commercial polyurethane films of various thicknesses and compositions were evaluated for their anticoagulation properties. Results suggested that these films exhibited excellent thermal and physico-mechanical properties while capable of increasing contact time with blood plasma by over a thousand-fold as compared to a control surface. Due to the unknown structure and composition of these commercial films, polyurethane samples were synthesized from toluene …

Optimized Xanthene-Based Probes For Pancreatic Cancer Imaging, Ian Ruramai Munhenzva May 2020

Optimized Xanthene-Based Probes For Pancreatic Cancer Imaging, Ian Ruramai Munhenzva

Dissertations and Theses

Pancreatic Ductal Adenocarcinoma (PDAC) is one of the deadliest human malignancies with an extremely poor 5-year survival rate of below 5%. Surgical resection is the most effective treatment of choice because chemotherapy and radiation therapy do not improve life expectancy. Residual tumor after PDAC surgery is common due to a lack of PDAC targeted intraoperative contrast agents to confirm clear margins.

Fluorescence imaging has the potential to improve surgery outcome and PDAC patients' survival rate via the use of highly PDAC-specific molecular probes to facilitate tumor identification. This thesis describes the application of a focused library of benzoxanthene fluorophores for …

A Microfluidic Platform For High-Throughput Screening Of Aquaporin Performance, Adriana Del Pino Herrera, Jordan Hoydick, Rachel Rauh, Elyssa El-Hajj, Madison Burchfield

Undergraduate Research and Scholarship Symposium

Aquaporins are a family of small integral membrane proteins that transport water across cell membranes in response to osmotic gradients. They facilitate fluid secretion and absorption across epithelial surfaces in kidney tubules, exocrine glands, and gastrointestinal tract. Here, we describe a novel microfluidic method to evaluate and screen for aquaporin-based transmembrane permeability in mammalian cells. A microfluidic device was designed and fabricated for the encapsulation of single mammalian and yeast cells in micron-sized droplets. For this purpose, Chinese Hamster Ovarian (CHO) cells were used. CHO cells express AQP1 (aquaporin-1) homologous to human kidney aquaporins. The cells were cultivated and exposed …

Development Of Fluorescent Hyaluronic Acid Nanoparticles For Intraoperative Tumor Detection, Nicholas E. Wojtynek May 2020

Development Of Fluorescent Hyaluronic Acid Nanoparticles For Intraoperative Tumor Detection, Nicholas E. Wojtynek

Theses & Dissertations

Surgical resection remains to be the primary treatment for the majority of solid tumors, including breast cancer. The complete removal of the primary tumor, local metastases, and metastatic lymph nodes dramatically improve a patient’s treatment outcome and prognosis. Nevertheless, surgeons are limited to tactile and visual cues in distinguishing malignant and healthy tissue. This can result in a positive surgical margin (PSM), which occurs when tumor goes undetected and is left behind in the surgical cavity. PSMs decreases a patient’s prognosis and necessitate additional treatment in the form of surgery, radiation, and chemotherapy. An emerging imaging modality, known as fluorescence-guided …

Heel Down And Toe-Off Time Measured With Ultrasonic Doppler System And Force Plate Sensor, Sabin Timsina May 2020

Heel Down And Toe-Off Time Measured With Ultrasonic Doppler System And Force Plate Sensor, Sabin Timsina

Honors Theses

Collie Box is a medical device that measures the gait parameters of the person walk- ing in front of it. This device uses the Ultrasonic Doppler system to extract the heel-contact and toe-off times of a person walking within the range of 2-10 meters. These times are used to determine the leg’s swing phase and double stance times. The ultrasonic transducer of 10mm diameter is driven at 40kHz. At the time of the heel-contact and toe-off, foot velocity is zero while the torso part of the human body is still in motion. The wide directivity of 10mm diameter ultrasonic transducer …

A Comparison Of Neural Activity For Peripersonal And Extrapersonal Viewing In Real And Virtual Environments, Olivia Eustice May 2020

A Comparison Of Neural Activity For Peripersonal And Extrapersonal Viewing In Real And Virtual Environments, Olivia Eustice

Honors Theses

This study involved a series of tests comparing the similarities and differences in neural activity at a subject’s peripheral and extrapersonal space in the real environment and virtual reality. We hypothesized that there would be similar brain activity at each of these environments depending on the focal distance of an object from the participant. Peripheral space is the visual space that is a reachable distance from a person. It has an overall neural pattern in the dorsal stream of the brain. Extrapersonal space is the visual space that does not directly surround a person and cannot be directly acted on. …

Investigatin Actin-Myosin Mechanics To Model Heart Disease Using Fluorescence Microscopy And Optical Trapping, Justin Edward Reynolds May 2020

Investigatin Actin-Myosin Mechanics To Model Heart Disease Using Fluorescence Microscopy And Optical Trapping, Justin Edward Reynolds

Honors Theses

Hypertrophic cardiomyopathy (HCM) is a hereditary disease in which the myocardium becomes hypertrophied, making it more difficult for the heart to pump blood. HCM is commonly caused by a mutation in the β-cardiac myosin II heavy chain. Myosin is a motor protein that facilitates muscle contraction by converting chemical energy from ATP hydrolysis into mechanical work and concomitantly moving along actin filaments. Optical tweezers have been used previously to analyze single myosin biophysical properties; however, myosin does not work as a single unit within the heart. Multiple myosin interacts to displace actin filaments and do not have the same properties …

From Branches To Fibers - Investigating F-Actin Networks With Biochemistry And Mathematical Modeling, Melissa A. Riddle May 2020

From Branches To Fibers - Investigating F-Actin Networks With Biochemistry And Mathematical Modeling, Melissa A. Riddle

Senior Honors Projects, 2020-current

F-actin networks have different structures throughout the cell depending on their location or mechanical role. For example, at the leading edge of a migrating cell, F-actin is organized in a region called the lamellipodia as a branched network responsible for pushing the membrane outwards. Behind the lamellipodia is a lamellar actin network where focal adhesions and stress fibers originate, and then within the cell cortex, actin is arranged in a gel-like network. Stress fibers are an important organization of F-actin and how they arise from either the branched lamellipodia network or the gel-like cortex network is poorly understood. Our approach …

Implementation Of A Computer-Vision System As A Supportive Diagnostic Tool For Parkinson’S Disease, Diego Machado Reyes

Honors Theses

Parkinson’s disease is the second most common neurodegenerative disorder, affecting nearly 1 million people in the US and it is predicted that the number will keep increasing. Parkinson’s disease is difficult to diagnose due to its similarity with other diseases that share the parkinsonian symptoms and the subjectivity of its assessment, thus increasing the probabilities of misdiagnosis. Therefore, it is relevant to develop diagnostic tools that are quantitatively based and monitoring tools to improve the patient’s quality of life. Computer-based assessment systems have shown to be successful in this field through diverse approaches that can be classified into two main …

Characterization Of Photoacoustic Flow Cytometry Signals, Craig Benzinger May 2020

Characterization Of Photoacoustic Flow Cytometry Signals, Craig Benzinger

Electronic Theses and Dissertations

Photoacoustic flow cytometry has been utilized to clinically determine the presence of melanoma circulating tumor cells (CTCs). Further investigation was conducted into the morphology of detection signals and how they could be manipulated to allow for further classification. Novel features were extracted from waveforms that appear to have strong classification ability. Neural networks were also used to determine classification potential and the creation of feature mapping for future unsupervised classification. Detections were expanded from single waves to a time dependent multiwave event. Waveforms were also determined to be of non-parametric distribution, allowing for classification by neural network but not allowing …

Pilot Study Exploring The Effect Of Targeted Cox-2 Inhibition In Macrophages Responding To Neuronal Injury; Promoting Enhanced Axonal Regeneration, Alyssa Brauckmann

Electronic Theses and Dissertations

Celecoxib nanoemulsion (CXB-NE) has been developed as a macrophage targeted analgesics by Dr. Janjic and her team at Duquesne University, (Janjic et al, 2018; Liu et al, 2020; Saleem et al, 2019b; Vasudeva et al, 2014). The CXB-NE nanoemulsion carrying a Nonsteroidal Anti-inflammatory (NSAID) inhibitor of COX-2 activity result in a reduction in PGE2 expression in macrophages. Using CXB-NE in rats that have peripheral nerve injury constricting the sciatic nerve relieves hypersensitivity, a pain-like behavior. The treatment also decreases inflammation associated with this chronic constriction injury (Janjic et al, 2018; Saleem et al, 2019b; Stevens et al, 2019). In this …

Identification Of Human Adrenal Carcinoma Cells Using Photoacoustic Flow Cytometry, Alexis Stahl May 2020

Identification Of Human Adrenal Carcinoma Cells Using Photoacoustic Flow Cytometry, Alexis Stahl

Electronic Theses and Dissertations

With consistently high mortality rates in patients with adrenal cancer, lack of early detection and diagnosis are an ever increasing problem in fighting the deadly disease. Currently, very few patients are diagnosed with adrenal cancer within the early stages due to the absence of symptoms until large tumors are formed, which is in most cases is fatal. Because of this, early detection equipment is crucial to optimizing the survival rate in patients. We studied the absorbance wavelength of human adrenal carcinoma cells, as well as the minimum cell concentration needed in samples using photoacoustic flow cytometry (PAFC). PAFC generates ultrasonic …

Design Of A 3d Printed Bioreactor For Bone Cancer Research, Brooklyn K. Vanderwolde, Katelyn Hillson May 2020

Design Of A 3d Printed Bioreactor For Bone Cancer Research, Brooklyn K. Vanderwolde, Katelyn Hillson

The Journal of Undergraduate Research

Bone cancer is an aggressive disease and has peak occurrence during physiological stimulation of growth and aging. Astronauts who undergo long-term space missions also acquire an increased risk of bone tissue degeneration and cancer. Few in-vitro models currently exist capable of reproducing the complex microenvironment of bone tissue to support multicellular activity in a three-dimensional structure. This limits the ability to understand disease progression and develop suitable treatment strategies. Therefore, developing a bone tissue model and an associated bioreactor is critical to understand the risks associated with cancer progression and improve treatment and preventions related to those risks. The goal …

Rapid Antibiotic Susceptibility Testing Platform For Direct Clinical Samples, Terrance Zhang May 2020

Rapid Antibiotic Susceptibility Testing Platform For Direct Clinical Samples, Terrance Zhang

Honors Scholar Theses

Infectious diseases and septicemia are two of the major causes of death in the U.S., necessitating rapid treatment of septic patients with proper, efficacious antibiotics. Unfortunately, the emergence and spread of multidrug-resistant bacteria are continuously being aggravated by an abuse in antibiotic prescription at a clinical and agricultural level. It is known that antibiotic resistance evolves through the sequential accumulation of multiple mutations in bacteria, which is accentuated by prolonged exposure of bacteria to ineffective antibiotics when implementing traditional septicemia treatment. The goal of this project is to develop a novel, easy-to-use AST platform for rapid antimicrobial susceptibility profiling to …

Concentrating Photovoltaic Retrofit For Existing Parabolic Trough Solar Collectors: Design, Experiments, And Levelized Cost Of Electricity, Todd P. Otanicar, Rhetta Wingert, Matthew Orosz, Clay Mcpheeters

Mechanical and Biomedical Engineering Faculty Publications and Presentations

Photovoltaics and concentrating solar thermal power are two ways for generating electricity from sunlight, albeit through different methods. Parabolic trough style powerplants represent 3.6 GW of electricity production, but many of these plants are aging and being replaced with photovoltaics. An alternative option that could be employed to leverage the sunk capital cost associated with the primary optics would be the design of a pure photovoltaic retrofit working within the existing plant architecture. Here, a secondary optical concentrator is designed to use the existing primary optics of a parabolic trough type solar thermal powerplant. The design is a v-shaped secondary …

Stirred Suspension Bioreactor Differentiation Of Human Mesenchymal Stem Cells Into Smooth Muscle Cells, Chris Slavin May 2020

Stirred Suspension Bioreactor Differentiation Of Human Mesenchymal Stem Cells Into Smooth Muscle Cells, Chris Slavin

Biomedical Engineering Undergraduate Honors Theses

Human mesenchymal stem cells (hMSCs) are a promising candidate for cellular therapies due to their multipotency, self-renewal capacity, and immunomodulatory properties. However, their isolation is a difficult and potentially painful process with very low yield, and traditional static mammalian cell culture techniques are too slow and expensive for large scale growth and differentiation of stem cells to be practical. Current research is focused on improving methods for cultivating hMSCs in stirred suspension bioreactors (SSBs), but little work has been done with regard to their differentiation in dynamic conditions such as those in SSBs. Differentiation at a large scale would increase …

Photobiomodulation And Stem Cell Differentiation: Application To Modulate Adipogenesis Through Cellular And Nuclear Morphology, Andrew Redmond Mccolloch

Bioengineering Dissertations - Archive

Mesenchymal stem cells (MSCs) are of great interest in regenerative medicine due to their capability to self-renew and differentiate to various lineages. Typically, differentiation to specific cell types has relied on the use of growth factors and other reagents to provide an environment conducive to the desired cell type. More recently, orthogonal cues other than chemical factors have been elucidated to regulate the lineage commitment. For example, the cell shape, cellular mechanics and substrate stiffness have been found to play a role in determining the fate of MSCs. However, it remains still elusive how the morphological and mechanical changes in …

Study Of Upregulation Of Drug Resistance In H460 Lung Cancer Cells Via Microchannel Device, Nowmi Haider May 2020

Study Of Upregulation Of Drug Resistance In H460 Lung Cancer Cells Via Microchannel Device, Nowmi Haider

Bioengineering Theses - Archive

Metastasis is the main reason of high mortality rates in cancer patients. Treating these patients becomes difficult once the cancer has metastasized, especially due to their drug resistance. Drug resistance is a common problem that occurs when treating cancer patients, as in most cancer types, the cells are capable of drug efflux. This makes treatments like chemotherapy ineffective. Therefore, it is important to find alternative methods of treatment that can effectively kill drug resistant cancer cells. Lung cancer is one type of cancer that is the second most diagnosed cancer in the U.S. In this study, microchannels were used to …

Fabrication And Examination Of Magnetic Bacterial Cellulose As A Material For Biomedical Engineering, Xi Chen May 2020

Fabrication And Examination Of Magnetic Bacterial Cellulose As A Material For Biomedical Engineering, Xi Chen

Bioengineering Theses - Archive

Bacterial cellulose (BC) is one of the most abundant biopolymers in nature. Several attributes such as facilitative modification, high pore size/porosity, able to mass produce as well as biocompatibility renders BC a promising candidate for many fronts of biomedical engineering. One consideration for BC as a biomedical material over conventional plant cellulose is the ease in which BC can be obtained to provide high purity cellulose. In comparison to collagen, another biopolymer with considerable prospects for biomedical engineering, BC possesses a higher degree of elasticity, water retention, and lower degradability. The high affinity towards water and high porosity provide BC …

Examination Of The Effects Of Confined Migration On Androgen Resistant Prostate Cancer And Potential Treatment Option For Drug Resistant Pc3 Cells With Calcitriol, Emily Torrey Ann Hills May 2020

Examination Of The Effects Of Confined Migration On Androgen Resistant Prostate Cancer And Potential Treatment Option For Drug Resistant Pc3 Cells With Calcitriol, Emily Torrey Ann Hills

Bioengineering Theses - Archive

When localized to the prostate, prostate cancer (PCa) has a near 100% 5-year survival rate, however, when prostate cancer metastasizes the survival rate drastically drops to around 30%. The low death rate of primary PCa is due to the wealth of knowledge we have of non-metastatic cancer and the sufficient treatment regimens. However, this understanding does not apply to metastatic cancer, so therapies are greatly lacking. To gain a better understanding of the invasion process for PCa we utilized microchannel technology to uncover the differences between migratory and non-migratory PCa. Specifically, the following study examines the effects of confined migration …

Optical Metabolic Imaging As A Predictor Of Patient Tumor Response To Therapy And Apoptosis, Andrew Larey May 2020

Optical Metabolic Imaging As A Predictor Of Patient Tumor Response To Therapy And Apoptosis, Andrew Larey

Biomedical Engineering Undergraduate Honors Theses

Despite a plethora of medical advancements, malignant tumors remain difficult to treat due to complex cancer biology and interpatient heterogeneity. Phenotypically similar tumors may respond differently to a given treatment. In clinical oncology, it is paramount to quantify tumor response to therapy for predicting treatment outcome, monitoring tumor changes, identifying effective therapies, and avoiding unnecessary cost and toxicity to the patient. Current standards of monitoring tumor response to therapy include CT, MRI, PET, and histopathology; these methods either take a merely anatomical approach or are invasive and time consuming, necessitating a functional biomarker approach. MPM represents a metabolic-level method of …

Redesign And Quantitative Assessment Of An Accelerated Venous Valve Fatigue Apparatus, Megan Kueh May 2020

Redesign And Quantitative Assessment Of An Accelerated Venous Valve Fatigue Apparatus, Megan Kueh

Biomedical Engineering Undergraduate Honors Theses

Chronic venous insufficiency (CVI) is a form of cardiovascular disease that is caused when valves in the leg become incompetent. Current treatment methods serve to manage symptoms, but there are currently no ways to treat the underlying cause of CVI. A venous valve prosthetic made from a xenograft of a bovine jugular vein is one possible treatment method currently in the research phase. Prosthetic valves must be tested with an accelerated wear tester prior to approval for clinical testing. Although such testers exist for heart valves, physiological differences between heart and venous valves restrict the use those testers on venous …

The Utilization Of Autofluorescence To Study The Effects Of L-Buthionine-Sulfoximine On Cellular Metabolism In Vitro, Madison Belew

Biomedical Engineering Undergraduate Honors Theses

Oxidative damage and stress impacts metabolic activity and has been implicated in a number of diseases . These medical conditions have been found to be associated with a lack of glutathione (GSH). There is a need to study the effects that the absence of glutathione has in order to expand knowledge of indicators that cause conditions like cataracts, cancer, or impaired wound healing. L-buthionine-sulfoximine (BSO) has been used in past studies to hinder GSH production and induce oxidative stress within a cell. This study explores the effects of BSO and induced oxidative stress on the metabolic activity and processes within …

Development Of An Inertial Measurement-Based Assessment Of Disease Severity In Chronic Fatigue Syndrome, Turner Palombo, Suzanne Vernon, Shad Roundy May 2020

Development Of An Inertial Measurement-Based Assessment Of Disease Severity In Chronic Fatigue Syndrome, Turner Palombo, Suzanne Vernon, Shad Roundy

Utah Space Grant Consortium

While myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is relatively new and poorly understood, a recent upsurge in research has identified the disease’s core symptoms, including post-exertional malaise and orthostatic intolerance. The FDA has yet to approve any treatments for ME/CFS, partially due to a lack of validated efficacy endpoints.

The central focus of this research is to develop ME/CFS efficacy endpoints using a non-invasive, inertial measurement-based approach. Accessible endpoints will provide a way to properly evaluate potential treatments for ME/CFS. Using a Kalman filter, inertial measurement unit (IMU) data can be converted to optimized leg angle estimates. These angle estimates can …

Improving Technologies In Anesthesia, Patrick Kolbay, Joseph Orr May 2020

Improving Technologies In Anesthesia, Patrick Kolbay, Joseph Orr

Utah Space Grant Consortium

General anesthesia is well known to offer physicians access to a broad variety of invasive procedures otherwise deemed too risky. Anesthesia machines provides the means for anesthetizing patients safely in the hospital operating room. However, these devices are increasingly unable to meet the demands and needs outside of the hospital. Developing countries struggle to purchase and maintain these costly devices, leading to a 40-fold increase in anesthesia-related deaths compared to developed countries. Small-office practices in the United States experience significantly poorer anesthesia outcomes and increased legal claims versus their larger hospital counterparts, resulting in 60% more anesthesia-related deaths. Environmental impacts …

Mechanical Analysis Of Double Network Hydrogels To Determine Suitability As An Articular Cartilage Substitute, Casey Virginia Young

All Theses

The Center for Disease Control has found that between 2013-2015, 26% of women and 19% of men reported doctor-diagnosed cartilage disease or damage. Currently, there exists a substantial population with cartilage damage or disease needing a solution to increasing pain and decreasing function and mobility. Many of these individuals are young and thus looking for a treatment to delay a total knee replacement. Many others are elderly and thus looking to avoid major surgeries, as the riskiness of these procedures increases with age. As researchers look towards a method of cartilage replacement, hydrogels show measurable potential as a synthetic substitute …
