Satellite telemetry tags are used to track the migration patterns of large cetaceans. These tags penetrate the dermis and remain embedded in the underlying blubber tissue. As the dermis of cetaceans is host to a diverse microbiome, and it is impossible to clean the skin before implanting the devices, the potential for infection is increased when the tags penetrate through the skin. H2O2 is a potential antimicrobial agent that, in addition to showing broad-spectrum efficacy against gram-negative and gram-positive bacteria, can promote wound healing outcomes by promoting proliferative factors and peptides that protect against oxidative stress. Cancer is a cluster of diseases, and 1.8 million Americans are newly diagnosed each year. Treatment issues such as drug instability, the occurrence of severe side effects, as well as resistance make the need for solutions to improve conventional methods, like chemotherapy, apparent. Breast cancer is one of the most common types of cancer worldwide, and standard-of-care for early-stage disease typically involves a lumpectomy or breast-conserving surgery (BCS). BCS involves the local resection of cancerous tissue, while sparring as much healthy tissue as possible. State-of-the-art methods for intraoperatively evaluating BCS margins are limited. Approximately 20% of BCS cases result in a tissue resection with cancer at or near the resection surface (i.e., a positive margin). A two-fold increase in ipsilateral breast cancer recurrence is associated with the presence of one or more positive margins. Consequently, positive margins often necessitate costly re-excision procedures to …

One approach to interrogating the complexities of human systems in their well-regulated and dysregulated states is through the use of digital twins. Digital twins are virtual representations of physical systems that are descriptive of an individual's state of health, an object fundamentally related to precision medicine. A key element for building a functional digital twin type for a disease or predicting the therapeutic efficacy of a potential treatment is harmonized, machine-parsable domain knowledge. Hypothesis-driven investigations are the gold standard for representing subsystems, but their results encompass a limited knowledge of the full biosystem. Multi-omics data is one rich source of …

จุดประสงค์ของงานวิจัยนี้ เพื่อสร้างแม่แบบภาพเพทมาตรฐานบริเวณสมองขึ้นมาใหม่ และเพื่อเปรียบเทียบประสิทธิภาพในการกระบวนการซ้อนทับภาพเพทระหว่างแม่แบบภาพเพทบริเวณสมองที่สร้างขึ้นกับแม่ภาพเพทบริเวณสมองแบบเดิมที่มีการใช้งานอยู่ในปัจจุบัน แม่แบบภาพเพทมาตรฐานบริเวณสมองสร้างขึ้นมาจากภาพเพท กับภาพเอ็มอาร์ไอแบบ DTI จากข้อมูลภาพถ่ายทางรังสีจากกลุ่มอาสาสมัครที่มีภาวะความจำปกติ จำนวน 24 คน อายุระหว่าง 42-79 ปี ซึ่งกลุ่มอาสาสมัครดังกล่าวจะต้องทำการถ่ายเพทบริเวณสมองที่มีการฉีดสารเภสัชรังสี 11C-PIB เข้าไปในร่างกาย และทำการถ่ายภาพเอ็มอาร์ไอบริเวณสมองด้วยเทคนิค T1 และเทคนิค DTI จากนั้นทำการแก้ไขค่ากระแสไฟฟ้ารบกวน การกำจัดคลื่นสัญญาณรบกวน และการสร้างแผนที่โดยวิธี Whole-brain probabilistic tractography จากภาพถ่ายเอ็มอาร์ไอเทคนิค DTI โดยแผนที่ดังกล่าวที่ถูกสร้างขึ้นมาจะถูกนำไปใช้ในขั้นตอนการซ้อนทับ และขั้นตอนการ Normalize กับภาพเพทเพื่อช่วยในการกำหนดขอบเขตภายในบริเวณเนื้อสมอง หลังจากนั้นทำการประเมินความถูกต้องของการซ้อนทับของภาพแม่แบบภาพเพทมาตรฐาน โดยการประเมินการซ้อนทับกับภาพเพทในกลุ่มผู้ป่วยที่มีภาวะสมองเสื่อมในระยะเริ่มต้นที่เข้ารับการถ่ายภาพเพทบริเวณสมองด้วยการตรวจแบบเดียวกัน จากนั้นทำการประเมินและให้คะแนนความถูกต้องในการซ้อนทับของภาพเพทกับแม่แบบภาพเพทมาตรฐานบริเวณสมองโดยรังสีแพทย์ผู้เชี่ยวชาญทางด้านเวชศาสตร์นิวเคลียร์จำนวน 3 คน ด้วยวิธีการแบบปกปิดข้อมูล แล้วทำการเปรียบเทียบค่าคะแนนดังกล่าวโดยใช้การทดสอบทางสถิติแบบ Wilcoxon Signed Ranks test การประเมินความสอดคล้องภายในโดยใช้สถิติ Fleiss Kappa test และสถิติ Cohen’s weighted Kappa test ผ่านโปรแกรม SPSS ที่ระดับความเชื่อมั่น ร้อยละ 95 นอกจากนี้ในงานวิจัยได้ทำการหาค่า Dice similarly coefficient เพิ่มเติม เพื่อเปรียบเทียบความถูกต้องของการซ้อนทับดังกล่าว ผลการศึกษาวิจัยพบว่าคะแนนประเมินความถูกต้องในการซ้อนทับของภาพเพทกับแม่แบบมาตรฐานภาพเพทบริเวณสมองที่สร้างขึ้นด้วยวิธีการใหม่มีค่าสูงกว่าแม่แบบมาตรฐานภาพเพทบริเวณสมองที่สร้างขึ้นด้วยวิธีการเดิมอย่างมีนัยสำคัญทางสถิติ ซึ่งสอดคล้องกับค่า Dice similarly coefficient ที่พบว่ามีค่าสูงสุดในแม่แบบมาตรฐานภาพเพทบริเวณสมองที่สร้างขึ้นด้วยวิธีการใหม่ อย่างไรก็ตามพบว่าผลการประเมินค่าความสอดคล้องภายในอยู่ในระดับสอดคล้องกันแบบเล็กน้อย ซึ่งจากผลการศึกษาวิจัยดังกล่าวสรุปได้ว่า แม่แบบมาตรฐานภาพเพทบริเวณสมองที่สร้างขึ้นมาใหม่โดยใช้ภาพเอ็มอาร์ไอเทคนิค DTI มาร่วมด้วย มีการซ้อนทับของภาพที่ดีกว่าเดิมและเหมาะสมในการที่จะนำไปใช้ในการช่วยวินิจฉัยผู้ป่วยที่มีสภาวะโรคสมองเสื่อม

การศึกษานี้มีวัตถุประสงค์เพื่อพัฒนาอุปกรณ์ฝึกเดินที่มีการนำอุปกรณ์วัดระยะทางด้วยแสง หรือ อุปกรณ์ไลดาร์ มาประยุกต์ใช้ในการเก็บข้อมูลการเดิน และนำข้อมูลการเดินที่อุปกรณ์ไลดาร์เก็บได้ มาทำการประมวลผลเป็นตัวแปรการเดินต่าง ๆ ได้อย่างแม่นยำโดยไม่จำเป็นต้องทำการปรับเทียบทุกครั้งก่อนใช้งาน และ ไม่จำเป็นต้องติดวัตถุลงบนร่างกายของผู้ใช้งานก็สามารถเก็บข้อมูลได้อย่างแม่นยำ โดยในการศึกษาครั้งนี้ ได้ทำการทดสอบความถูกต้องและความแม่นยำในการเก็บข้อมูลโดยเปรียบเทียบกับอุปกรณ์ถ่ายภาพความเคลื่อนไหวทางคลินิก โดยจากการทดสอบพบว่า จากความคลาดเคลื่อนในการวิเคราะห์ตัวแปรเชิงระยะทาง และ ความคลาดเคลื่อนในการวิเคราะห์ตัวแปรเชิงเวลา ที่ต่ำกว่า 10% โดยสรุป จากการทดสอบกับระบบเก็บข้อมูลการเดินมาตรฐานทางการแพทย์ มีความเป็นไปได้ที่จะนำอุปกรณ์ฝึกเดินพร้อมระบบวัดระยะทางด้วยแสงที่พัฒนาขึ้นนี้ มาใช้งานในการเก็บข้อมูลการเดินจริงได้ ซึ่งระบบฝึกเดินที่พัฒนาขึ้นนี้ จะมีประโยชน์ในการกายภาพบำบัดและการฟื้นฟูร่างกายแบบทางไกล

งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาการประยุกต์ใช้ไบโอเซนเซอร์ชนิดควอตซ์คริสตัลไมโครบาลานซ์ (quartz crystal microbalance, QCM) ในการวิเคราะห์อันตรกิริยาระหว่างแบคทีเรียก่อโรคปริทันต์ชนิด Aggregatibacter actinomycetemcomitans กับโมโนโคลนอลแอนติบอดีที่จำเพาะเพื่อการตรวจวัดแบคทีเรียก่อโรคปริทันต์ ไบโอเซนเซอร์สำหรับการตรวจวัดถูกสร้างขึ้นโดยใช้คิวซีเอ็มเซนเซอร์ ชนิด 30 เมกะเฮิรตซ์ ที่ผ่านการปรับสภาพพื้นผิวอิเล็กโทรดทองด้วย 11-mercaptoundecanoic acid (11-MUA) และได้ประเมินความหนาแน่นการเรียงตัวของชั้น 11-MUA ด้วยเทคนิคไซคลิกโวลแทมเมตรี จากนั้นทำการตรึงโมโนโคลนอลแอนติบอดีชนิด Anti-A. actinomycetemcomitans กับ 11-MUA โดยใช้ 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide/N-hydroxysuccinimide (EDC/NHS) เพื่อใช้แอนติบอดีทำหน้าที่เป็นสารรู้จำทางชีวภาพ (biorecognition element) สำหรับการตรวจวัด A. actinomycetemcomitans การวิเคราะห์อันตรกิริยาระหว่างแบคทีเรียกับแอนติบอดีชนิด anti-A. actinomycetemcomitans ทำด้วยกัน 4 รูปแบบ ได้แก่ รูปแบบที่ 1 ใช้ข้อมูลการเปลี่ยนแปลงความถี่ (∆F) ซึ่งผลการศึกษาบ่งชี้ว่าสามารถใช้ข้อมูล ∆F ในการจำแนกความสามารถในการจับกันระหว่างแอนติบอดีและเซลล์แบคทีเรียชนิดต่าง ๆ ได้ โดยคู่แอนติบอดีและเซลล์แบคทีเรียที่มีความจำเพาะต่อกันมีรูปแบบ ∆F ที่แสดงให้เห็นแนวโน้มการเปลี่ยนแปลงของ ∆F ที่ขึ้นกับความเข้มข้น รูปแบบที่ 2 ใช้ค่าอนุพันธ์อันดับหนึ่งของการเปลี่ยนแปลงความถี่ (dF/dT) ซึ่งผลการศึกษาบ่งชี้ว่าสามารถใช้วิธีนี้จำแนกความจำเพาะของแอนติบอดีชนิด Anti-A. actinomycetemcomitans กับแบคทีเรียต่างชนิดได้ โดยเมื่อเกิดการจับกันระหว่างแอนติบอดีและเซลล์ที่มีความจำเพาะกัน ค่า dF/dT จะเปลี่ยนแปลงเป็นลบ และค่า dF/dT แสดงให้เห็นแนวโน้มการเปลี่ยนแปลงเป็นรูปแบบที่ขึ้นกับความเข้มข้นเช่นเดียวกับข้อมูล ∆F รูปแบบที่ 3 ใช้ข้อมูลเวลาการตอบสนอง (τ) ซึ่งผลการศึกษาบ่งชี้ว่าการจับกันระหว่างแอนติบอดีกับเซลล์ A. actinomycetemcomitans ที่ความเข้มข้นสูงสุด (1.16 × 108 เซลล์/มิลลิลิตร) ให้ค่า τ เฉลี่ยเพียง 143 วินาที ทำให้ทราบผลลัพธ์ของการจับกันระหว่างแอนติบอดีกับเซลล์ A. actinomycetemcomitans รวดเร็วกว่าการติดตามจากข้อมูล ∆F ถึง 3 เท่า รูปแบบที่ 4 ใช้ข้อมูลความชันของการตอบสนอง …

Spinal surgical procedures often require release of intervertebral discs and ligaments to optimally achieve postural correction on a patient-specific basis. In this paper, a T7-T8 Finite Element (FE) model is utilized to examine internal load sharing during resection steps performed in a Ponte osteotomy. The FE model was rotated bidirectionally along three anatomical planes using an externally applied moment. In each step, the Ranges of Motion (RoM), Instantaneous Centers of Rotation (ICR), and forces from ligaments, discs, facet, and costovertebral joints were calculated. The first purpose of this thesis was to compare the amount of micromotion seen in the femoral stem in a revision total hip arthroplasty between simple loading conditions and loading conditions derived from activities of daily living, through the use of experimental and computational methods. The amount of micromotion at the bone-implant interface was larger for activities of daily living, with ranges of 200μm more than the largest simple loading conditions. Motor skills are fundamental for the development of children. Neurodevelopmental tests currently used by professionals for measuring motor control maturity exhibit several limitations. To address some of these, we have designed the Lognometer, a tablet-based device that can run computerized neuromotor tests. To normalize this tool against a representative population, we collected handwritten triangles from 780 children. We used the Sigma-Lognormal model and a prototype-based parameter estimation algorithm to analyze these movements. To ensure clinical acceptance, we developed an explainable solution relying on statistical regression. It is important to accurately evaluate the motor control maturity to help physicians diagnose delayed or abnormal motor development in children. Traditionally, it has been challenging to design assessment methods that are practical and accurate at the same time. This study aims to develop an effective algorithm to predict motor control maturity based on the Kinematic Theory of rapid human movements. The study is used to understand the working procedure of the Olympus PSD-30 Electrosurgical Unit which a high-frequency alternating current is to measure the voltage and power output from the unit when used for a surgical operation to determine the extent of tissue damage. The output power and voltage were as examined with the stopwatch, then with an Arduino time based for 1, 2, and 3 seconds to understand the different modes of the cut and coagulation feedback with an RCC circuit used to mimic the human body. However, playgrounds are reported to be the leading location of unintentional recreation injuries to children aged 1-10. Over 185,000 children annually are being treated in emergency departments for playground injuries. These play-related injuries have remained stagnant according to epidemiology studies of the past three decades. Neuromotor dysfunction after a concussion is common, but balance tests used to assess neuromotor dysfunction are typically subjective. Current objective balance tests are either cost- or space-prohibitive, or utilize a static balance protocol, which may mask neuromotor dysfunction due to the simplicity of the task. Predicting functional outcomes from spinal cord injury (SCI) at the acute setting is important for patient management. This work investigated the relationship of early magnetic resonance imaging (MRI) biomarkers in a rat model of cervical contusion SCI with long-term functional outcome and tissue sparing. Forty rats with contusion injury at C5 at either the spinal cord midline (bilateral) or over the lateral cord (unilateral) were examined using in vivo multi-modal quantitative MRI at 1 day post-injury. Plasma-activated water (PAW) is considered to be an effective anticancer agent due to the diverse aqueous reactive oxygen and nitrogen species (RONS: ROS and RNS), but the drawback of low dose and short duration of RONS in acidified PAW limits their clinical application. Herein, this Letter presents an innovative therapeutic avenue for cancer treatment with highly-effective alkaline PAW prepared by air surface plasma. Extracellular-regulated kinase 3 (ERK3) is overexpressed in a variety of cancers, including lung cancer. This atypical mitogen-activated protein kinase (MAPK) has a unique structure which includes a C34 domain and C-terminus and about which relatively little is known. ERK3 has been demonstrated to promote cell migration and metastasis in multiple cancer types. A yeast two-hybrid assay using ERK3 as bait indicated Diacylglycerol kinase ζ (DGKζ) is a binding partner. Among these, diacetylene-containing gelators are especially important because they can form photopolymerizable gels, which are useful stimuli-responsive materials. Cancer is the second leading cause of death in women and late stage (metastatic) cancers have abysmal survival rates compared to early stage regional cases (27% vs 86%). As a tumor grows, the surrounding extracellular matrix (ECM) is reorganized into a dense, collagen rich matrix. The new matrix of aligned collagen fibers provides unique mechanical cues such as anisotropic stiffness and contact guidance. In 2019, approximately 50.2 million adults (20.4% of the US population) experienced chronic pain, contributing to $560-635 billion in direct medical costs. In addition, the worldwide prevalence of diabetes mellitus has reached epidemic proportions and is set to increase to 629 million by 2045. Gene expression in mammalian cells requires complex nuclear choreography, and there is increasing evidence that spatiotemporal organization of chromatin and nuclear compartments plays an important role in gene expression. In this dissertation, I examined the function of SON, a splicing factor with a known role in nuclear organization, in chromatin-mediated gene expression. SON association with a transcriptionally inactive U2OS 2-6-3 reporter gene array provided a useful model to study SON’s chromatin dynamics. In October 2021, 59 scientists from 14 countries and 13 U.S. states collaborated virtually in the Third Annual Baylor College of Medicine & DNANexus Structural Variation hackathon. The goal of the hackathon was to advance research on structural variants (SVs) by prototyping and iterating on open-source software. This led to nine hackathon projects focused on diverse genomics research interests, including various SV discovery and genotyping methods, SV sequence reconstruction, and clinically relevant structural variation, including SARS-CoV-2 variants. Naturalistic stimuli, such as movies, contain interacting, multimodal and semantic features and allow for free exploration through eye movements. The full extent of neural responses to features such as motion, film cuts and eye movement behavior has not been established. The main hypothesis of this thesis is that complex multimodal and semantic stimuli in naturalistic movies engage a widespread ensemble of locations across the entire brain. To address this question I analyzed simultaneous intracranial and eyetracking data from over 6,000 electrodes across 23 patients with intractable epilepsy.
