Analysis Of Arterial Compliance Using A Surrogate Arm Bench Top Model For The Validation Of Oscillometric Blood Pressure Methods, Christopher J. Cunningham
Analysis Of Arterial Compliance Using A Surrogate Arm Bench Top Model For The Validation Of Oscillometric Blood Pressure Methods, Christopher J. Cunningham
Master's Theses
A study was performed on a recently developed prototype of the Yong-Geddes surrogate arm design to collect compliance data of the various system components and determine the accuracy of measurements made through the bench top model. The study was performed to perceive the effectiveness of the model as a tool for validating non-invasive blood pressure detection monitors. Three stages of testing were performed to gather pressure and volume data from an artificial artery component, a sphygmomanometer, and the surrogate arm system to produce compliance estimations. Mathematical equations from supported arterial hemodynamics studies and clinical trials were applied to the pressure …

Microneedles For Wound Healing, Maya Matheny, Maggie Dubus, Lily Eribes
Bioengineering Senior Theses
Microneedles are a virtually painless and minimally invasive transdermal delivery system comprised of an array of micron-sized needles that can be used to deliver cells, drugs, proteins, and vaccines. However, a major challenge to this delivery method is controlling puncture depth. To advance the microneedle landscape, our project aims to identify and characterize the biomechanical cues between a microneedle puncture and a hydrogel skin phantom for pre-clinical testing A stopper microneedle design that controlled puncture depth into the phantom skin model patch to 150μm was tested on millimeter-thick mold-casted alginate slabs. The hysteresis loop differs in the presence and absence …

Reversing the pulse polarity, i.e., changing the electric field direction by 180°, inhibits electroporation and electrostimulation by nanosecond electric pulses (nsEPs). This feature, known as “bipolar cancellation,” enables selective remote targeting with nsEPs and reduces the neuromuscular side effects of ablation therapies. We analyzed the biophysical mechanisms and measured how cancellation weakens and is replaced by facilitation when nsEPs are applied from different directions at angles from 0 to 180°. Monolayers of endothelial cells were electroporated by a train of five pulses (600 ns) or five paired pulses (600 + 600 ns) applied at 1 Hz or 833 kHz. Reversing …

Biomechanical Knee Joint For Exoskeleton, Gabriel Ireton Ahern, Calloway Miller, Christianna Altamura, Eric Mailes
Biomechanical Knee Joint For Exoskeleton, Gabriel Ireton Ahern, Calloway Miller, Christianna Altamura, Eric Mailes
Mechanical Engineering
Our senior design project consisted of designing and manufacturing a biomechanically accurate, actuated knee joint to be integrated into an exoskeleton being developed by the Lower Limb Exoskeleton Assist Project (LLEAP), a part of the EMPOWER student association at Cal Poly, San Luis Obispo. As the human knee flexes and extends throughout gait motion, the center of rotation changes. Currently marketed exoskeletons have one point of rotation, which over constrains the knee and causes misalignment between the user and the suit [1]. Our goal was to mimic natural knee joint motion by changing the center or rotation, thus reducing misalignment …

Produced water (PW) generated from hydraulic fracturing operations was treated using an integrated electrocoagulation, ultrafiltration, membrane distillation, and crystallization process (EC UF MDC). The aim was to determine the viability of this integrated process for maximizing water recovery. The results obtained here indicate that optimizing the various unit operations could lead to increased recovery of PW. Membrane fouling limits all membrane separation processes. A pretreatment step to suppress fouling is essential. Here, removal of total suspended solids (TSS) and total organic carbon (TOC) was achieved by electrocoagulation (EC) followed by ultrafiltration (UF). The hydrophobic membrane used in membrane distillation may …

Dissertations
The measurement of vital signs ? such as peripheral capillary oxygen saturation (SpO2) and heart rate (HR) levels ? by a pulse oximeter is studied. The pulse oximeter is a non-invasive device that measures photoplethysmography (PPG) signals and extracts vital signs from them. However, the quality of the PPG signal measured by oximetry sensors is known to deteriorate in the presence of substantial human and sensor movements contributing to the measurement noise. Methods to suppress such noise from PPG signals measured by an oximeter and to calculate the associated vital signs with high accuracy even when the wearer …

Development Of Non-Viral Nano-Carriers For Gene Delivery And Covid-19 Vaccines, Zhongyu Li
Dissertations
Gene therapy is one of the most promising medical fields with the potential to effectively advance the treatment of difficult diseases by producing therapeutic proteins or repairing defective gene sequences. However, gene therapy presents severe challenges in delivery including renal clearance, phagocytosis, enzymatic degradation, protein absorption, and cellular internalization barriers, which have been barriers for translation into the clinic. Naked nucleic acids, with their intrinsic negative charge, electrostatically repulse the anionic cell membrane, preventing cellular uptake. Also, they are considered foreign genetic material when introduced into the body and are rapidly cleared by the reticuloendothelial system or degraded by nucleases, …

The Effect Of Spinal Cord Stimulation And Video Games Training On Body-Machine Interface Control, Jie Fei
McKelvey School of Engineering Graduate Student Theses & Dissertations
Damage to the spinal cord causes long-lasting loss of motor and sensory function, and currently, there is no ‘cure’ for paralysis. However, even people with severe spinal cord injuries (SCI) have some residual mobility. Studies have shown that transcutaneous electrical spinal cord stimulation (tSCS) combined with functional training targeting residual mobility can further improve the motor function of individuals with SCI. In this study, we present a technical framework that aims to enhance rehabilitation outcomes by targeting residual mobility through a motor training-based approach. Our technical framework centers around a non-invasive body-machine interface (BoMI) that relies on the use of …

Novel Microfluidic Devices To Model The Interactions Between Lymphatics And Breast Cancer, Jade Weber
McKelvey School of Engineering Graduate Student Theses & Dissertations
The lymphatic system is responsible for immune circulation and fluid balance in the body. It accomplishes this by draining interstitial fluid from local tissue and transferring it to lymph nodes and back into blood circulation. However, this process is implicated in many pathologies, one of the most dangerous being breast cancer metastasis to the lymph nodes. The largest factor in breast cancer patient mortality is metastasis. Lymphangiogenesis, the growth of new lymphatic vessels, has been thought to play a dynamic role in aiding breast cancer metastasis. Breast cancer tumor cells have been shown to remodel the functionality of local lymph …

Reflectance Quantitative Polarized Light Imaging For Microstructural Evaluation Of Musculoskeletal Soft Tissues, Leanne Elizabeth Iannucci
Reflectance Quantitative Polarized Light Imaging For Microstructural Evaluation Of Musculoskeletal Soft Tissues, Leanne Elizabeth Iannucci
McKelvey School of Engineering Graduate Student Theses & Dissertations
Musculoskeletal soft tissue injury is common and can lead to debilitating pain, loss of tissue function, and overall decrease in quality of life. Proper biomechanical function of musculoskeletal soft tissues is dictated by their hierarchically organized collagenous extracellular matrices, which are disrupted in periods of disease or injury. To best understand dynamic structure-function relationships of these tissues in normal and pathological conditions, it is imperative to be able to visualize collagen microstructure using a quantitative, high spatiotemporal resolution imaging modality. The innate birefringence associated with collagen fibers permits the use of biomedical polarimetry techniques to probe collagen in a specific …

Single-Molecule Super-Resolution Imaging Of Geobacter Sulfurreducens Under Anaerobic Conditions, Ziyi Hu
McKelvey School of Engineering Graduate Student Theses & Dissertations
Geobacter sulfurreducens are anaerobic bacteria capable of making electrical contacts with other organisms and extracellular electron acceptors. The challenge of imaging live Geobacter bacteria is maintaining anaerobic conditions during the imaging process. In this thesis, we augment a single-molecule localization microscope (SMLM) with a home-built anaerobic imaging chamber and use constant argon bubbling to maintain oxygen-free imaging conditions. To validate the imaging protocol, we use the transient binding of Nile red to resolve the spherical morphology of lipid-coated glass spheres with nanoscale resolution. However, when imaging Geobacter, the distribution of Nile red localizations is non-uniform, both between different cells …

Piezoelectric And Conductive Polymer Based Flexible Devices Enabling Cardiovascular Health Sensing And Energy Harvesting, Andrew Closson
Dartmouth College Ph.D Dissertations
Piezoelectric materials show great promise for low-power wearable and implantable sensing, but their rigidity makes it challenging to integrate them with biological tissue. To address this, researchers have started exploring polymer-based functional materials that offer flexibility and are suitable for interfacing with the human body. However, these materials are still in their early stages, and a framework is necessary to illustrate how these materials, in conjunction with novel fabrication techniques and device designs, can enable the development of multi-functional sensing and energy harvesting devices.
This thesis utilizes highly scalable fabrication methods for functional polymers to build and test a flexible …

Inflammatory skin diseases are estimated to impact 20% of the global population and are the fourth leading cause of nonfatal disability worldwide. Diagnosis and management are predominantly based on clinician visual assessment of disease related changes in skin morphology. The qualitative nature of this method can result in misdiagnosis and underdiagnoses of treatable diseases. There persists systematic undertreatment of skin of color patients given the more subtle presentation of erythema against pigmented skin. There exists a clinical need for a quantitative and objective inflammation assessment tool that meets the needs of a diverse patient population. The accumulation of interstitial fluid …
A General-Purpose Software Platform For Closed-Loop Neuromodulation, William Engelhardt
A General-Purpose Software Platform For Closed-Loop Neuromodulation, William Engelhardt
McKelvey School of Engineering Graduate Student Theses & Dissertations
Implementing closed-loop neuromodulation therapies is a challenging and expensive endeavor. It requires developing software capable of acquiring signals from a bio-signal amplifier, analysis of these signals, and initiation of precisely timed stimulation, all of which need to be accomplished in real-time with very low latency. Developing this software is difficult, as it requires a wide range of expertise ranging from interfacing with hardware to real-time signal processing. Even when successfully implementing such a system for one set of hardware, it often then only works within the laboratory that conceived it. This is because of the inherent heterogeneity in the devices …
