Advancing Mitochondrial Dna Visualization And Dynamics In Plants: Application Of The Mt-Hi-Ness Fluorescent Marker, Madeleine H. Faber

Mitochondrial DNA (mtDNA) plays a crucial role in cellular energy production and inheritance, yet its dynamics and exchange remain poorly understood in plant systems due to limitations in visualization techniques. Traditional visualization methods such as exogenous DNA stains and fluorescent protein markers present challenges in specificity and live-cell imaging. To address this challenge, we adapted the MT-HI-NESS fluorescent marker, originally developed for mammalian cells, for use in plant mitochondrial studies. Using Arabidopsis thaliana as a model system, we employed mitochondrial targeting sequences (IVD and AOX) to ensure accurate localization and to confirm mtDNA binding specificity. The production of RNA is vital for many commercial and research purposes, with one of the more notable reasons being vaccine production. The RNA polymerase, derived from T7 bacteriophage, is commonly used for RNA production due to its ability to produce full-length RNA transcripts. However, researchers have faced limitations in optimizing this polymerase. For example, the polymerase generates heterogeneous RNA products that require additional purification steps during RNA production. Here, we selected three amino acid point substitutions based on a recent publication from Moderna that showed increased RNA purity compared to the wild type T7 RNA polymerase sequence. We created …

Insects are often overlooked in science curricula when talking about conservation due to the surrounding fear and disgust regarding insects. This disgust not only permeates the mindsets of students, but also teachers, which leads to a hesitance to incorporate an insect-based curriculum or club into a lesson. In response to this, I have created a four-session club curriculum that can be used across four two hour long sessions spread out across four weeks. Aristotle’S Poetics: A Biologist’S Approach To Tragedy, James J. Clauss

The paper describes a class that introduces Aristotle’s Poetics from the perspective of his scientific, especially biological, treatises: a systematic analysis of what emotions a tragedy is supposed to elicit and how they are resolved by the audience (homoeopathy). Students read 11 ancient and modern plays and view one film, observing whether his theory of the Tragic explains our responses. The final project includes the writing of a tragic story (not a drama) evincing the students’ own understanding of the Tragic based on the readings.

Nebraska is mostly an agricultural state with a non-Hodgkin lymphoma (NHL) incidence rate of 20.1 per 100,000 individuals, which is greater than the US incidence rate of 19 per 100,000 individuals. It is possible that the higher incidence rate in Nebraska is due to an increase in pesticide usage. Pesticide exposure is linked to t(14;18), a genetic translocation involving the BCL2 gene that regulates apoptosis. The dysregulation of apoptosis could contribute to the proliferation of NHL. This exploratory study utilized the Nebraska Lymphoma Study Group, which consists of patients with non-Hodgkin lymphoma, Hodgkin disease, or some other neoplastic hematologic disorder …

Heavy metals are commonplace in nature, and are typically found in scarce quantities in all drinking sources. However, in high enough concentrations, they can have a myriad of negative health effects. This study seeks to find the concentrations of such metals in the Lincoln water supply utilizing the resources of the Water Center at the University of Nebraska at Lincoln. To test for discrete concentrations of trace elements, a method of using Inductively Coupled Plasma Mass Spectrometry (ICP-MS) was implemented. All populations display patterns in allele frequencies over time. Some alleles cease to exist, while some grow to become the norm. These frequencies can shift or stay constant based on the conditions the population lives in. If in Hardy-Weinberg equilibrium, the allele frequencies stay constant. Most populations, however, have bias from environmental factors, sexual preferences, other organisms, etc. We propose a stochastic Markov chain model to study allele progression across generations. In such a model, the allele frequencies in the next generation depend only on the frequencies in the current one.

Fertility rates in a country vary and can be influenced by numerous different aspects of an individual’s life and the society in which they live. One aspect that may influence fertility rates at the national level is the gross domestic product (GDP). GDP is an indicator of a country’s economic productivity. Because fertility is linked to both economic development and individual resources, GDP may be a useful indicator of fertility rates. The purpose of this study was to determine how well students are being prepared in high school for introductory college biology courses.
