Background/aim: Brain microvascular endothelial cells (BMECs), which constitute the blood–brain barrier (BBB), are essential for maintaining central nervous system homeostasis. Like BMECs, multipotent mesenchymal stem cells (MSCs) originate from the mesodermal lineage. Thus, MSCs may serve as a direct and efficient cellular source for BMEC-like differentiation. Notably, differentiation of human induced pluripotent stem cells (hiPSCs) into BMECs typically involves a 2-step protocol: inducing mesodermal commitment followed by endothelial specification. In contrast, direct differentiation from MSCs could bypass the initial mesodermal induction step, offering a streamlined alternative. Artificial Intelligence (AI) is transforming Supply Chain Management (SCM), yet many organizations struggle to assess their readiness for AI adoption and to understand how AI capabilities develop across maturity stages. This dissertation addresses this gap by developing a Capability Maturity Model (CMM) for AI integration in SCM, grounded in Organizational Information Processing Theory (OIPT), the Resource-Based View, and related capability frameworks. The model provides a structured approach for evaluating an organization's information-processing requirements, resource configurations, and alignment needed for effective AI-enabled supply chain operations.

Pancreatic ductal adenocarcinoma (PDAC) is the most lethal form of pancreatic cancer, with poor prognosis driven by late diagnosis, therapeutic resistance, and an immunosuppressive tumor microenvironment. Interactions between tumor cells and immune cells promote immune evasion and tumor progression, limiting the efficacy of immune checkpoint blockade and other immunotherapies. Given the high expression of trophoblast cell surface antigen 2 (TROP2) in PDAC, we developed a TROP2-targeted Toll-like receptor 7 (TLR7) agonist (E104) designed for selective accumulation within the tumor microenvironment to activate antitumor immunity. Background/aim: Pyrazole derivatives are of growing interest due to their diverse pharmacological activities. However, their biological activity is often highly sensitive to subtle structural modifications. Existing quantitative structure–activity relationships (QSAR) approaches frequently fail to capture the conformational flexibility and nonlinear structure–activity relationships (SAR) of such heterocyclic scaffolds, creating a gap in the accurate prediction of their biological profiles. Role Of Piezo1 Channels Expressed In Pvn In Regulating Sympathetic Nerve Activity And Arterial Blood Pressure In Rats, Yue Chen, Wei Guo, Jichun Wang, Min Wang, Yuying Yang, Gregory Miodonski, Enshe Jiang, Qing Hui Chen, Yi Yang, Renjun Wang

Mechanosensitive Piezo1 channels participate in regulating pain sensitivity, insulin secretion, and vascular tension; however, their expression in the autonomic paraventricular nucleus (PVN) and role in modulating sympathetic outflow and cardiovascular function remain unstudied. In this study, unilateral PVN microinjection of the Piezo1 channel blocker Dooku1 (0.1, 1, 10, 100, and 200 pmol) administered to anesthetized male rats increased renal sympathetic nerve activity (RSNA) and mean artery pressure (MAP) in a dose-dependent manner, with maximum increases of 93 ± 30% (p < 0.0001) and 21 ± 5 mmHg (p < 0.0001), respectively, elicited by Dooku1 at 100 pmol. Similarly, PVN microinjection of the peptide Piezo1 channel blocker GsMTx4 (1 nmol) significantly increased RSNA (p < 0.001) and MAP (p < 0.0001). Conversely, PVN-microinjected Piezo1 channel activators Yoda1 (5 nmol) and Jedi2 (5 nmol) did not significantly alter RSNA or MAP. Western blot and qRT-PCR analyses of the hypothalamic PVN showed abundant Piezo1 mRNA and protein expression. Immunofluorescence detection showed that Piezo1 was expressed in pre-sympathetic PVN neurons with axons projecting to the rostral ventrolateral medulla. We conclude that Piezo1 channels expressed in the autonomic PVN neurons play an important role in regulating sympathetic outflow and cardiovascular function.

Background/aim: High-grade serous ovarian cancer (HGSOC) remains one of the most aggressive forms of ovarian malignancy and frequently shows resistance to conventional therapies. This study aimed to synthesize a novel series of purine analogues, 6-[(4-substituted benzyl amine)/(4-substituted aniline)]-9-cyclopentyl purines, and evaluate their anticancer efficacy against HGSOC cell lines.

Materials and methods: We assessed the biological effects of the synthesized purine analogues on the OVCAR3, OVSAHO, and KURAMOCHI HGSOC cell lines using the sulforhodamine B assay. To investigate the mechanism of action, we conducted flow cytometry and western blot analyses, focusing on DNA replication and apoptosis.

This study analyzes the self-perception of digital teaching competence (DTC) among primary education teachers in Andalusia, using an instrument based on the European DigCompEdu framework. Data were collected through a self-administered online questionnaire applied to a sample of 416 primary education teachers working in Andalusia. The study employed a mixed-methods design, combining quantitative analyses (descriptive, correlational, and comparative) with qualitative analysis of open-ended responses. Employing a mixed-methods approach, the research assesses teachers’ competence levels and examines differences according to professional experience, school ownership, teaching specialization, and territorial distribution. How does immigrant ownership influence small firm survival in comparison with that of family ownership? While it is generally accepted that socioemotional wealth (SEW) preservation motivates family firms to avoid taking risks, this effect has not been considered in immigrant-owned firms. Many of the same affinity factors are likely to influence the survival of both ownership types, but we argue that immigrant-owned firms may experience them more strongly as they reside at the intersection of the liabilities of smallness and foreignness. Background: Upper extremity return to sport (RTS) assessments are not standardized with respect to which metrics to use, if the metrics are appropriate for all populations and levels of competition, and what constitutes good or poor test performance. Subsequently, clinicians may utilize suboptimal metrics to evaluate RTS readiness, or forgo objective criteria altogether. The purpose of this study was to examine the psychometric properties (reliability, agreement/measurement error, hypothesis testing/construct validity, criterion/predictive validity, responsiveness) of upper extremity functional tests used to assess RTS readiness.

An analysis was conducted on the variation in the counts of seven meristic traits of the eastern mosquito fish, Gambusia holbrooki, using samples collected from ten different sites in the Middle East. These sites are located along the Tigris, Euphrates, and Shatt al-Arab Rivers, spanning across Iraq, Iran,Türkiye, and Syria. The study revealed a northward increase in the average values of these meristic traits, with the highest recorded in Türkiye and the lowest in the southern region of Iraq. Graduate nursing programs increasingly rely on adjunct faculty to sustain enrollment growth; however, inconsistent onboarding processes may contribute to variability in faculty preparedness and job satisfaction. The purpose of this quality improvement project was to implement a standardized adjunct faculty education program and evaluate its impact on adjunct faculty knowledge and job satisfaction within an online Psychiatric–Mental Health Nurse Practitioner (PMHNP) program. Synthesis of six new curcumin analogues was performed through condensation reaction of 3-methyl-2, 4-pentandione with substituted aromatic aldehyde derivatives and boron oxide in the presence of trimethylborate and butylamine. The reaction was carried out under ultrasonic irradiation. The synthesized compounds were identified using 1H-, 13C-NMR, FT-IR spectroscopy, and electron impact mass spectrometry. The Swiss ADME website was used to predict the physicochemical properties of the new compounds using computational techniques like ADME studies. Proposed herein are techniques for a system that automatically generates Agent-to-Agent (A2A) capability cards by scanning Model Context Protocol (MCP) servers and semantically analyzing the tool inventories of the servers. The system transforms low-level tool specifications into high-level capability descriptions through a multi-stage pipeline that extracts schemas, infers dependencies, detects relationships, clusters related tools into skills, and abstracts skills into comprehensive capabilities. The output is a standardized A2A agent card that enables efficient agent discovery and collaboration in multi-agent systems.

Solar energy is a promising alternative, but its intermittency and inefficiency limit its widespread adoption. This study explores the role of advanced concrete-based thermal energy storage systems in addressing these challenges. Key aspects examined include packing density optimization, high-temperature performance, and hydrothermal durability of concrete.
