The opioid crisis disturbs several bioethics principles as indicated by the fact that it burgeoned out of the health care system as opposed to the illicit environments of the underground market. The book Dreamland by investigative journalist and storyteller, Sam Quinones encapsulates the complexity of the opioid epidemic through research and interviews with important figures related to the crisis such as physicians, people with opioid use disorder, and pharmaceutical sales representatives, among others. Background Complex social determinants of health may not be easily recognized by health care providers and pose a unique challenge in the vulnerable pediatric population where patients may not be able to advocate for themselves. Experiments examined the effect of photoperiod on the expression of adult diapause in North American populations of two Palearctic ladybird beetle species, Propylea quatuordecimpunctata and Hippodamia variegata.

Comparative data for the reproductive responses to different photoperiod conditions for H. variegata were taken from Obrycki (2018). Additionally, comparisons of responses to the four photoperiods between P. quatuordecimpunctata from Jefferson County, NY, USA to P. quatuordecimpunctata from Montreal, Quebec, Canada were also conducted. Data for the Montreal, Quebec, Canada population are from Obrycki et al., (1993).

Glioblastoma (GBM) presents a significant public health challenge as the deadliest and most common malignant brain tumor in adults. Despite standard-of-care treatment, which includes surgery, radiation, and chemotherapy, mortality rates are high, underscoring the critical need for advancing GBM therapy. Over the past two decades, numerous clinical trials have been performed, yet only a small fraction demonstrated a benefit, raising concerns about the predictability of current preclinical models. Traditionally, preclinical studies utilize treatment-naïve tumors, failing to model the clinical scenario where patients undergo standard-of-care treatment prior to recurrence. There have been major breakthroughs in clinical cancer research leading to some effective treatments, whereas the field of dementia has achieved comparatively limited success in clinical research. Alzheimer’s disease (AD) is a neurodegenerative disease that commonly causes dementia. Identifying biomarkers for the early detection of AD is an emerging need, as brain dysfunction begins two decades before the onset of clinical symptoms. To this end, we reanalyzed untargeted metabolomic mass spectrometry data from 905 patients enrolled in the AD Neuroimaging Initiative (ADNI) cohort using MS-DIAL, with 1,304,633 spectra of 39,108 unique biomolecules. Metabolic profiles of 93 hydrophilic metabolites were determined. Additionally, we integrated targeted lipidomic data (4873 samples from 1524 patients) to explore candidate biomarkers for predicting progressive mild cognitive impairment (pMCI) in patients diagnosed with AD …

Parents of young children play a significant role in their child’s communication development during early childhood. Many parents of children with or at risk for disabilities could benefit from learning ways to promote their child’s communication. This study implemented a rapid coaching intervention to teach a parent of a child with Down syndrome how to implement naturalistic language strategies such as expanding on their child’s play, providing linguistic input during play, and arranging the environment to create an opportunity for their child to communicate.

The novel biofluorescent glands of the Eastern Red-backed Salamander (Plethodon cinereus), have been shown to be sexually dimorphic where males have larger and greater numbers of glands than females. Interestingly, a seasonal pattern of increased numbers of fluorescent glands, which mirrors the peak in testosterone levels, has been described. Currently, the function and regulation of the fluorescent glands is unknown. I predicted that the glands function as a form of male ornamentation used in intersexual communication, and perhaps act as an honest signal of male quality. The kappa opioid receptor has been identified as a promising therapeutic target for promoting remyelination. In the current study, we evaluated the ability of nalfurafine to promote oligodendrocyte progenitor cell (OPC) differentiation and myelination in vitro, and its efficacy in an extended, cuprizone-induced demyelination model. Primary mouse (C57BL/6J) OPC-containing cultures were treated with nalfurafine (0.6–200 nM), clemastine (0.01–100 μM), T3 (30 ng/mL), or vehicle for 5 days. Using immunocytochemistry and confocal microscopy, we found that nalfurafine treatment increased OPC differentiation, oligodendrocyte (OL) morphological complexity, and myelination of nanofibers in vitro. Two important factors that contribute to resistance to immune checkpoint inhibitors (ICI) are an immune-suppressive microenvironment and limited antigen presentation by tumor cells. In this study, we examine whether in- hibition of the methyltransferase enhancer of zeste homolog 2 (EZH2) can increase ICI response in lung squamous cell carcinomas (LSCC). Our in vitro experiments using two-dimensional human cancer cell lines as well as three-dimensional murine and patient-derived organoids treated with two inhibitors of the EZH2 plus IFNγ showed that EZH2 inhibition leads to expression of both MHC class I and II (MHCI/II) expression at both the mRNA and protein levels. …

Introduction: Eastern equine encephalitis virus (EEEV), a mosquito-borne alphavirus, is detected throughout the eastern United States. The primary enzootic vector, Culiseta melanura (Blacktailed mosquito), allows transmission among 50+ avian host species resulting in complex disease patterns. Spillover to dead-end host mammals, such as equines and humans, rarely occurs, however, human cases can cause severe neurological deficits and have a 30% mortality rate. Eco-epidemiology and avian host immunity patterns are not well characterized for EEEV and require ongoing surveillance to develop more targeted interventions. Obesity is an epidemic that affects over 40% of adults in America. Obesity can be divided into metabolically healthy and metabolically unhealthy obesity, with the latter being characterized by symptoms closely associated with metabolic syndrome, type 2 diabetes, and metabolic dysfunction-associated steatotic liver disease (MASLD). What causes the transition from metabolically healthy to metabolically unhealthy obesity is not fully known. Further, it is unclear how this transition correlates with the onset of co-morbid conditions. Lake Tanganyika, in central Africa, contains a diverse and endemic fauna under threat from global climate change, overfishing, and nearshore sediment pollution. Previous studies of sediment pollution focused justifiably on impacts along rocky shorelines where diversity is high, but Lake Tanganyika also contains widespread shelly accumulations (shell beds) unprecedented in the modern East African lakes, but where impacts are less con- strained. Introduction: Deletion or mutation of members of the spectrin gene family contributes to many neurologic and neuropsychiatric disorders. While each spectrinopathy may generate distinct neuropathology, the study of βΙ spectrin’s role (Sptb) in the brain has been hampered by the hematologic consequences of its loss.

Methods: Jaundiced mice (ja/ja) that lack βΙ spectrin suffer a rapidly fatal hemolytic anemia. We have used exchange transfusion of newborn ja/ja mice to blunt their hemolytic pathology, enabling an examination of βΙ spectrin deficiency in the mature mouse brain by ultrastructural and biochemical analysis.

INTRODUCTION: In September 2022, The Jackson Laboratory Center for Alzheimer's and Dementia Research (JAX CADR) hosted a workshop with leading researchers in the Alzheimer's disease and related dementias (ADRD) field.

METHODS: During the workshop, the participants brainstormed new directions to overcome current barriers to providing patients with effective ADRD therapeutics. The participants outlined specific areas of focus. Following the workshop, each group used standard literature search methods to provide background for each topic.
