PARP2 is a DNA-dependent ADP-ribosyl transferase (ARTs) enzyme with Poly(ADP-ribosyl)ation activity that is triggered by DNA breaks. It plays a role in the Base Excision Repair pathway, where it has overlapping functions with PARP1. However, additional roles for PARP2 have emerged in the response of cells to replication stress. In this study, we demonstrate that PARP2 promotes replication stress-induced telomere fragility and prevents telomere loss following chronic induction of oxidative DNA lesions and BLM helicase depletion. Telomere fragility results from the activity of the break-induced replication pathway (BIR). During this process, PARP2 promotes DNA end resection, strand invasion and BIR-dependent …

Prostate Cancer (PCa) poses a significant clinical challenge, characterized by debates on screening efficacy and continuous issues with distinguishing aggressive from non-aggressive tumors. Despite treatment advancements, poor patient outcome at late stages and clinical overtreatment of indolent disease emphasizes the urgent need for markers of PCa aggression. Aberrant glycosylation and extracellular matrix (ECM) remodeling are two complex molecular processes necessary for PCa progression, highlighting a novel area for biomarker discovery. Leveraging formalin-fixed paraffin-embedded tissues, tissue microarrays (TMAs), and matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI), distinct N-glycan and ECM profiles are identified. This study aims to more deeply understand the role of the ASYMMETRIC LEAVES 2 (AS2) gene in Arabidopsis. During leaf development, the AS2 protein keeps the KNOX genes turned off so that leaves can develop. We use CRISPR technology to create mutations in specific regions of AS2. These mutations are predicted to change the protein structure of AS2, which may result in a mutated leaf. The analysis of the mutation allows us to better understand how AS2 keeps the KNOX genes off.

Difteri merupakan penyakit menular yang disebabkan oleh bakteri Corynebacterium diphtheriae. Bila ditemukan satu kasus difteri di suatu wilayah maka dapat ditetapkan sebagai KLB. Provinsi Lampung terus mengalami kenaikan kasus dan suspek difteri sejak 2020 dan turut menyumbang angka kematian akibat difteri pada tahun 2023. Kasus difteri dipengaruhi oleh beberapa faktor risiko. Penelitian ini bertujuan untuk mendapatkan gambaran distribusi dan faktor risiko kasus difteri di Provinsi Lampung dengan memetakan kabupaten/kota berdasarkan persebaran dan faktor risikonya. Background
Extracellular vesicles in human milk are critical in supporting newborn growth and development. Bioavailability of dietary extracellular vesicles may depend on the composition of membrane lipids. Single-nucleotide polymorphisms (SNPs) in the fatty acid desaturase gene cluster impact the content of long-chain polyunsaturated fatty acids in human milk phospholipids. This study investigated the relation between variation in FADS1 and FADS2 with the content of polyunsaturated fatty acids in extracellular vesicles from human milk.
Methods

High throughput screening (HTS) is routinely used to identify bioactive small molecules. This requires physical compounds, which limits coverage of accessible chemical space. Computational approaches combined with vast on-demand chemical libraries can access far greater chemical space, provided that the predictive accuracy is sufficient to identify useful molecules. Through the largest and most diverse virtual HTS campaign reported to date, comprising 318 individual projects, we demonstrate that our AtomNet® convolutional neural network successfully finds novel hits across every major therapeutic area and protein class. In this dissertation, we propose two clustering methods to study DNA methylation (DNAm) data in one-dimensional and stochastic optical reconstruction microscopy (STORM) image data in two-dimensional. Background: African ancestry is a known factor associated with the presentation and aggressiveness of prostate cancer (PC). Hispanic/Latino populations exhibit varying degrees of genetic admixture across Latin American countries, leading to diverse levels of African ancestry. However, it remains unclear whether genetic ancestry plays a role in the aggressiveness of PC in Hispanic/Latino patients. We explored the associations between genetic ancestry and the clinicopathological data in Hispanic/Latino PC patients from Colombia. This article explores the benefits of transitioning from traditional agricultural irrigation systems to more cost-effective alternatives powered by renewable energy sources. By integrating solar photovoltaic technology into irrigation processes, significant reductions in resource wastage, estimated at 45-50%, are achievable through the implementation of drip irrigation techniques. This results in enhanced agricultural productivity. Moreover, the utilization of solar panels in green zones mitigates surface temperature increases, contributing to a 2-5% boost in electricity production. Currently, irrigation facilities primarily rely on the main energy grid. The purpose of this work is the chemical modification of cotton fabrics with natural globular protein. Modification was carried out in the process of finishing cotton fabrics, after decoction and bleaching in a continuous mode. The effect of modification on the strength, protein content and functional groups of cotton fabric has been studied. According to the results obtained using the AUTOGRAPH AGS-N machine, it was found that the modification of cotton fabrics with a solution of globular protein increases the strength of the treated fabrics, depending on the concentration of the modifier and the temperature regime. The Biological Weapons Convention, initially ratified in 1975, banned the production and stockpiling of biological weapons; however, it has faced considerable modern criticism for being unenforceable and not strong enough to ensure states' compliance. These modern critiques are based on the knowledge that the Soviet Union was in violation of the Convention, which was not confirmed until 1989. Purpose
Artificial intelligence (AI) technology is being rapidly adopted into many different branches of medicine. Although research has started to highlight the impact of AI on health care, the focus on patient perspectives of AI is scarce. This scoping review aimed to explore the literature on adult patients’ perspectives on the use of an array of AI technologies in the health care setting for design and deployment.

Building a culture of conceptual inquiry in psychiatric training requires the development of conceptual competence: the ability to identify and examine assumptions that constitute the philosophical foundations of clinical care and scientific investigation in psychiatry. In this article, we argue for the importance of such competence and illustrate approaches to instilling it through examples drawn from our collective experiences as psychiatric educators.

Metastatic disease remains the leading cause of death due to cancer, yet the mechanism(s) of metastasis and its timely detection remain to be elucidated. Neutrophil elastase (NE), a serine protease secreted by neutrophils, is a crucial mediator of chronic inflammation and tumor progression. In this study, we used the PyMT model (NE+/+ and NE-/-) of breast cancer to interrogate the tumor-intrinsic and -extrinsic mechanisms by which NE can promote metastasis. Our results showed that genetic ablation of NE significantly reduced lung metastasis and improved metastasis-free survival. Traditional proteomics studies can measure many protein biomarkers simultaneously from a single patient-derived sample, promising the possibility of syndromic diagnoses of multiple diseases sharing common symptoms. However, precious cellular-level information is lost in conventional bulk-scale studies that measure tissues comprising many types of cells. As single cells are the building blocks of organisms and are easier to biopsy than traditional bulk samples, performing proteomics on a single-cell level would benefit clinicians and patients. Single-cell proteomics, combined with mass spectrometry imaging, can be used to analyze cells in their microenvironment, preserving spatial information. The 2014–2016 Ebola outbreak left thousands of Liberian survivors with severely diminished quality of life. Applying a social determinants framework, this mixed method study investigates to what extent Ebola virus disease (EVD) survivors suffer long-haul psychosocial stress, diminished quality-of-life factors, and the impact of EVD-related service provisions on their ongoing healing. We present the results of a quantitative analysis survey of data collected from 19 Liberian EVD survivors in 2022 using snowball sampling. Additionally, a qualitative analysis of survivor statements helps triangulate key statistical findings and inform causal mechanisms. Water scarcity is a problem that faces many regions. In a developing and arid countries, the need for fresh water is exponentially increasing. One of the most water consuming industries is the construction industry, especially in concrete manufacturing. This research is an evaluation of the Reliability of recycled wastewater to be used in concrete manufacturing. Two different wastewater treatment regimens are implemented: secondary and tertiary treated wastewater. The treated wastewater, either secondary or tertiary, replaces potable water with percentages of 25%, 50%, and 100%. The mixture of the two types of water is used in concrete mixing and curing. Fibromyalgia is a chronic, widespread pain disorder that is strongly represented across the affective and cognitive dimensions of pain, given that the underlying pathophysiology of the disorder is yet to be identified. These affective and cognitive deficits are crucial to understanding and treating the fibromyalgia pain experience as a whole but replicating this multidimensionality on a preclinical level is challenging. To understand the underlying mechanisms, animal models are used.
