DNA polymerase θ (Polθ) plays a central role in a DNA double-strand break repair pathway termed theta-mediated end joining (TMEJ). TMEJ functions by pairing short-sequence "microhomologies" (MHs) in single-stranded DNA at each end of a break and subsequently initiating DNA synthesis. It is not known how the Polθ helicase domain (HD) and polymerase domain (PD) operate to bring together MHs and facilitate repair. To resolve these transient processes in real time, we utilized in vitro single-molecule FRET approaches and biochemical analyses. Potassium-competitive acid blockers (P-CABs) provide potent acid inhibition, yet studies on P-CAB-based quadruple therapy for H. pylori eradication are limited. We theorized that integrating bismuth subsalicylate into a quadruple therapy regimen could enhance eradication rates. However, data on the efficacy of vonoprazan bismuth quadruple therapy are notably scarce. Therefore, the aim of this study was to evaluate the efficacy of vonoprazan-based bismuth quadruple therapy in areas with high clarithromycin and levofloxacin resistance. Vaccination is the best defense against infection, but highly variable and evolving pathogens like influenza virus pose a significant immunological challenge for vaccine design. An ideal influenza virus vaccine should elicit a durable and broadly cross-reactive antibody response. However, the development of cross-reactive antibodies is hindered by the high degree of antigenic variation among influenza viruses, particularly in the surface glycoprotein hemagglutinin (HA), which is the main target of vaccine-induced responses. Cdk8 in Drosophila is the orthologue of vertebrate CDK8 and CDK19. These proteins have been shown to modulate transcriptional control by RNA polymerase II. We found that neuronal loss of Cdk8 severely reduces fly lifespan and causes bang sensitivity. Remarkably, these defects can be rescued by expression of human CDK19, found in the cytoplasm of neurons, suggesting a non-nuclear function of CDK19/Cdk8. Here we show that Cdk8 plays a critical role in the cytoplasm, with its loss causing elongated mitochondria in both muscles and neurons. We find that endogenous GFP-tagged Cdk8 can be found in both the cytoplasm and nucleus. …

Measuring body composition in children is important due to the rise of childhood obesity and changes that occur during pre-pubertal years. Sarcopenia, defined as a muscle mass loss and function, is increasingly recognized in pediatric populations, particularly in childhood obesity. Therefore, it is necessary to have measurements which not only provide an overall picture of body composition, but that can distinguish between muscle and fat mass. Viral infections remain a major risk in immunocompromised pediatric patients, and virus-specific T cell (VST) therapy has been successful for treatment of refractory viral infections in prior studies. We performed a phase II multicenter study (NCT03475212) for the treatment of pediatric patients with inborn errors of immunity and/or post allogeneic hematopoietic stem cell transplant with refractory viral infections using partially-HLA matched VSTs targeting cytomegalovirus, Epstein-Barr virus, or adenovirus. Primary endpoints were feasibility, safety, and clinical responses (>1 log reduction in viremia at 28 days). Reverse transcription polymerase chain reaction (RT-PCR) is the most sensitive technique commonly used to characterize gene expression in cells and tissues. While a standard PCR requires hours to complete, a modified “fast” PCR takes ~30 minutes which could allow for advanced integration into undergraduate laboratory courses. Therefore, studies were performed to flashlight putative “fast” PCR efficacy by detecting anti-Müllerian hormone (AMH). AMH is a peptide hormone regulating sex development. Two primary problems are posed by the production of BPA: It is environmentally and toxicologically damaging. If a molecule that mimicked the properties and structure of BPA yet lacked its toxicologically damaging effects could be manufactured from renewable sources these concerns could be greatly reduced. One such monomer is bisguaiacol F (BGF, 4-[(4-Hydroxy-3-methoxyphenyl)methyl]-2-methoxyphenol). Previous research by Yuan et al. has shown that labeling fluorescent dyes with amino acids is a useful method for fluorescent microscopy because it does not significantly alter the size of the fluorophore. Data by Yuan et al. utilized three different neutral amino acids (glycine, lysine, and proline) and individually linked them to the same fluorophore, resulting in three individual fluorophores. These three fluorophores were tested in the plant, Arabidopsis thaliana, which was able to absorb the fluorophore. Amino acid absorption and transport could be visualized. In-vitro studies have demonstrated the role of serine hydrolase, specifically a/b hydrolase domain-containing 2 (ABHD2) in hepatitis B viral propagation. The downregulation of ABHD2 via antisense oligonucleotides (ASODNs) decreased HBV mRNA and protein levels, implicating the role of ABHD2 in HBV replication and expression. However, the downregulation of ABHD2 was not demonstrated to effectively prevent HBV propagation in further studies. The Peaks of Otter salamander, Plethodon hubrichti, is an endemic species located exclusively in a 15km range of the Blue Ridge Mountains in central Virginia. The Peaks of Otter salamander is only found at elevations between 440m and up to the highest elevation in that area around 1100m. These specific parameters render P. hubrichti highly susceptible to habitat loss and population declines, two factors which have heavily influenced the Virginia Department of Wildlife Resources to label these salamanders a species of concern. Baraminology is the study of God’s created kinds as described in Genesis 1. Holobaramins (groups of known organisms sharing continuity and bounded by discontinuity) are scientific approximations of these kinds identified by building up monobaramins (smaller, continuous groups) and dividing apobaramins (larger, discontinuous groups). The lungless salamanders (Caudata: Plethodontidae), composed of over 500 species in 29 genera, have been grouped together by herpetologists for almost 200 years.
