Precision medicine is a healthcare approach which tailors disease prevention and treatment to an individual, based on their genetics, environment, lifestyle, and physiological state. These factors interact to produce biological changes that can be measured to produce data called omics, and include genomics, lipidomics, and proteomics. Despite the abundance of omics data and analysis techniques, researchers still struggle to identify biological findings that replicate across data sets and translate into clinical applications. Core chlorophytes possess glycerol-3-phosphate dehydrogenases (GPDs) with an unusual bidomain structure, consisting of a glycerol-3-phosphate phosphatase (GPP) domain fused to canonical GPD domains. These plastid-localized enzymes have been implicated in stress responses, being required for the synthesis of glycerol under high salinity and triacylglycerols under nutrient deprivation. However, their regulation under varying environmental conditions is poorly understood. C. reinhardtii transgenic strains expressing constitutively bidomain GPD2 did not accumulate glycerol or triacylglycerols in the absence of any environmental stress. Human kinases play essential and diverse roles in the cellular activities of maintaining homeostasis and growth. Genetic mutations in the genes encoding the kinases (or phosphotransferases) have been linked with various types of cancers. In this study, we cataloged mutations in 500 kinases genes in >65,000 individuals of global populations from the Human Genetic Diversity Project (HGDP) and ExAC databases, and assessed their potentially deleterious impact by using the in silico tools SIFT, Polyphen2, and CADD. The analysis highlighted 35 deleterious non-synonymous SNVs in the ExAC and 5 SNVs in the HGDP project. Notably, a higher number of deleterious mutations …

Demand for reducing energy consumption and detrimental environmental impacts after the effective lifetime of an engineering material are the major driving factors for the development of natural fibre reinforced composites (NFRCs) in various sectors. Compared to synthetic fibre like carbon-fibres or Kevlar, natural fibres provide several benefits including light weight, inexpensive, recyclability, superior specific strength, biodegradable, non-toxic during handling, and less energy intensive during manufacturing. This doctoral work presents observations from mechanical tests and interpretations on the suitability of natural fibre reinforced composites for less heavily loaded engineering applications areas.

Psychosis-spectrum disorders remain a leading cause of disability for both individuals and society, with early identification and prevention efforts representing a promising avenue of research for addressing these concerns. One potential impediment to improving early risk identification is the historical focus on indicators thought to be exclusive to the psychosis-spectrum. This focus often comes at the expense of non-specific risk factors (e.g., disrupted sleep, adverse childhood experiences) which contribute to the risk of developing psychosis as well as other mental illnesses. Previous research shows that consuming violent media can lead to increased aggression in thoughts, feelings, and behaviors. Several other factors also contribute to such behaviors but have not been thoroughly studied including implicit beliefs about aggression in relation to media content. This two-part study consists of (1) an exploratory iterated principal-factor method to create an implicit belief measure about consuming violent media in conjunction with correlational analyses and (2) between subjects hierarchical linear regression models to determine what effects implicit beliefs have on subsequent aggression after participants view either a violent of nonviolent video, while controlling for violent media consumption …

American youth are leaving organized religion at historical levels. Given that religious and political similarity (consensual solidarity) tends to strengthen affectual solidarity (emotional connection) in parent-adolescent dyads, decreasing adolescent religiousness and increasing political disagreements may threaten family social cohesion. However, during adolescence, youth empathy skills tend to increase and adolescents’ conflict with their parents tends to eventually subside. Based on principles outlined in intergenerational family solidarity theory, I hypothesized that adolescent empathy skills and authoritative parenting style would buffer the negative relational effects of religious and political discrepancies within the parent-adolescent relationship. The Black community has long suffered through a cycle of trauma and pain, with history repeating itself throughout generations. From the civil rights movement to the Black Lives Matter movement, this cycle persists, showing up again in the experiences of Black TikTokers. Despite looking race-neutral on its face, copyright law’s lack of understanding of how Black art can manifest itself has become more obvious in the world of digital creativity. Yellow Perch (Perca flavescens) are widely distributed throughout the Midwest and are popular game fish and table fare. Seasonally, some eastern Lake Michigan Yellow Perch migrate into drowned river mouth lakes. Drowned river mouths are highly productive environments that also hold non-migratory resident Yellow Perch. Migration requires substantial energy expenditure; therefore, we expected migratory Yellow Perch to evolve locomotory traits that optimize energy efficiency for swimming long distances. Institutions of higher education (IHE) are increasingly prioritizing diversity, necessitating the recruitment of more faculty and staff leaders from marginalized populations to better support the university and underrepresented minority student groups. Prenatal exposure to particulate matter with an aerodynamic diameter of less than 2.5 µm (PM2.5) has been linked to altered lung function and respiratory diseases. However, the conclusions of previous studies are controversial. To our knowledge, no study examined whether DNA methylation (DNAm), metabolites, nutrients, and toxins (MNTs) at birth mediate the association between prenatal PM2.5 exposure with multiple respiratory outcomes. This dissertation includes three carefully designed studies focusing on prenatal PM2.5 exposure, and its association with respiratory outcomes. This research assesses the extent to which asynchronous online STEM courses at a public university integrate Merrill's First Principles of Instruction. Through a quantitative content analysis of thirty unique online courses within the College of Arts and Sciences, the study investigates the extent, if any, to which these courses embody principles such as problem-centered learning, activation, demonstration, application, and integration. A validated survey instrument, Course Scan, was used for evaluation and indicated a low extent of Merrill's First Principles of Instruction in the sample courses. The common assumption in regard to metabolic rate scaling with body size is the 3/4 law, which predicts a scaling exponent of 0.75 between log metabolic rate and log body size. Supporting evidence exists for this theory on a large, general scale. However, factors such as temperature, predation, and environment can cause the scaling exponent to deviate from 0.75. This paper takes a closer look at the effects of temperature and predation on metabolic rates via oxygen consumption in freshwater macroinvertebrate populations. The major intracellular polyamines spermine and spermidine are abundant endogenous compounds that are essential for cellular growth and development. Dysregulation of polyamine metabolism has been implicated as a key mechanism of injury across multiple forms of clinically challenging pathologies. Of the enzymes within the polyamine pathway, the catabolic enzyme spermine oxidase (SMOX) is of particular interest as it is subject to induction in response to infection, neuronal excitotoxicity, ischemia, and oxidative stress. Context: Agronomic surveys play a vital role in identifying on-farm yield constraints and opportunities to increase productivity, input-use efficiency, and farmer profit. These surveys typically rely on farmer-reported data, where crop yield and applied inputs per hectare are estimated based on farmer-reported total harvested output and applied inputs per field and associated field size. PARP is a nuclear enzyme with a major function in the DNA damage response. PARP inhibitors (PARPi) have been developed for treating tumors harboring homologous recombination repair (HRR) defects that lead to a dependency on PARP. There are currently three PARPi approved for use in advanced prostate cancer (PCa), and several others are in clinical trials for this disease. Recent clinical trial results have reported differential efficacy based on the specific PARPi utilized as well as patient race. Aspen forests are strongly influenced by disturbances like fire and ungulate herbivory, which can impact plant community composition and succession. Human activities alter these disturbances, affecting their timing, extent, and severity. Chapter 1 explores the effects of large-scale prescribed fire and mechanical thinning treatments on aspen regeneration and vulnerability to ungulate herbivory. Results show that while aspen regeneration was hindered by chronic ungulate herbivory in untreated areas, mechanical thinning and prescribed fire stimulated aspen regeneration. However, mechanically thinned plots were vulnerable to ungulate herbivory, with only prescribed fire having reduced browsing. Given the ubiquity of media use, especially among young children, the current study examines the impact of problematic media use (PMU) on children's (N = 418, M age = 53.62 months, SD = 3.38 months, M and SD are taken from the full sample of 418 children) physiological functioning. With previous studies reporting relations between media use, temperament, and physiological regulation, it was hypothesized that children with greater levels of PMU would have lower levels of baseline respiratory sinus arrhythmia (RSA; a measure of physiological regulatory capacity).
