Despite the long history of transgender people and transgender care, state legislatures suddenly rushed to pass a wave of bans on gender-affirming medical care for trans youth over a three-year period beginning in 2021—an alarming surge within a flurry of legal attacks on the transgender community. In analyzing the constitutionality of these bans, courts and scholars have focused their attention on how to characterize the nature of the rights implicated, and therefore the level of scrutiny warranted. Cognitive flexibility is an executive function that involves dynamically adapting behavior in the face of changing environmental circumstances to achieve a particular goal. Disrupted in numerous psychiatric disorders such as Alzheimer’s disease, schizophrenia, OCD, and depression, the neural correlates of cognitive flexibility are not well understood. However, the prefrontal cortex (PFC) is strongly implicated in executive function and attentional control processes. This paper investigates the relationship between biomass composition and hydrocarbon yield as well as chain length distribution produced by Fischer-Tropsch synthesis. Through an Aspen Plus model, the conversion of three different biomasses – digestate, sawdust, and corn stover – into hydrocarbon fuel was simulated. The model converts biomass into gaseous and liquid hydrocarbons via various unit operations and reactions. Simulation results highlight the product distribution and hydrocarbon output, allowing for an analysis of hydrocarbon yield and comparison of chain length distributions amongst the three biomasses. Leptospirosis is a neglected zoonotic disease with global distribution caused by infection with pathogenic spirochetes of the genus Leptospira. Bacterial adherence to host cells is considered a crucial step in leptospiral infection and dissemination. However, the role of host-specific cellular interactions in disease outcome remains poorly understood. In this study, the adherence of pathogenic Leptospira interrogans serovar Manilae strain L495 and saprophytic Leptospira biflexa serovar Patoc I to kidney epithelial cell lines was investigated. Antifolates disrupt one-carbon metabolism (OCM), which stops nucleotide synthesis and therefore slows down cell proliferation. Our experimental antifolate, UCP1162, stops de novo purine synthesis and thymidine synthesis by targeting the OCM pathway. We investigated whether the downstream metabolites of OCM pathway: methionine, thymidine, and inosine, affect the cell death caused by UCP1162 using MV4-11 leukemia cells. Thymidine was found to partially rescue MV4-11 cells from UCP1162, in contrast, inosine was found to accelerate the cell death caused by UCP1162. These cytotoxic effects of UCP1162 alone and in combination with inosine were irreversible, as removal of treatment did not restore cell …

Sentinel lymph nodes are the first nodes that receive drainage from a tumor and are commonly used to evaluate whether cancer has begun to spread, but they also provide an important window into how a patient’s immune system is reacting to tumor-derived antigens. This project examines immune activity within SLNs, with particular attention to germinal centers, where B cells mature, undergo clonal expansion and somatic hypermutation, and generate improved antibody responses. Pneumocystis jirovecii is an opportunistic, host-obligate fungal pathogen responsible for causing P. jirovecii pneumonia (PJP) in humans. PJP is treated with trimethoprim-sulfamethoxazole (TMP-SMX), a strong broad-spectrum antibiotic that carries risk of renal and hepatotoxicity. Therefore, rapid diagnosis or exclusion of PJP is crucial to ensure PJP-positive patients receive effective treatment and PJP-negative patients avoid unnecessary antibiotics. Unlike most microorganisms, P. jirovecii cannot be cultured, presenting complications for the typical methods of diagnosing fungal pathogens. Animals utilize predictive processing to improve the processing of sensory information by reducing neural responses to anticipated stimuli and increasing responses to unexpected ones. Prior studies use simple, single-prediction, head-fixed behavioral paradigms, leaving it unclear how the brain stores and processes multiple predictions. Animals must be taught sound generating behaviors to study multiple, simultaneous predictions; however, learning in head-fixed conditions is inefficient. Therefore, we developed a two-lever behavioral task with auditory feedback and exposed mice to the task in home cages prior to switching to head-fixed training. Lynch syndrome is a hereditary disease caused by the inheritance of a mismatch repair gene variant. Individuals with this condition are predisposed to cancer development, most commonly colorectal cancer. Current guidelines for variant classification are based on numerous evidence categories, including functional evidence. However, many novel clinical variants are not well characterized, and evidence is difficult to obtain if functional assays are not calibrated.

Store-operated calcium entry (SOCE) through calcium release-activated

calcium (CRAC) channels is the dominant pathway for sustained Ca2+ influx in

non-excitable cells and is essential for T cell activation, driving downstream

signaling through the calcineurin-NFAT transcriptional axis. Dysregulation of

SOCE produces multi-systemic disease phenotypes, establishing that precise,

spatiotemporally controlled modulation of this pathway is a fundamental

therapeutic requirement. ML-9 is one of the only characterized small molecules

capable of targeting SOCE upstream at the level of STIM1 localization, but lacks

the selectivity, pharmacokinetic properties, and spatiotemporal control necessary

Women leave STEM not because of a lack of ability, but because of the classroom and institutional conditions they encounter once they arrive. This study examines how classroom experiences shape women’s decisions to persist in or leave STEM majors at the college level. Three deposits of perinates, infants, and young children within two non-residential superstructures of Buenavista del Cayo, Belize, display evidence of daily maintenance and care. A total of ten subadult individuals, aged 39.4 weeks gestation to 3 years, from the archaeological site of Buenavista were recovered during 2018 excavations. Signs of care and daily maintenance include survivorship through the vulnerable neonatal period (first twenty-eight days) and transition into infancy, survivorship with potential pinprick lesions on external and internal zygomatic, parietal, and pars basilaris, and intentional maintenance of the body after death. In this autoethnographic study, vignettes of a female high school teacher and graduate student with invisible disabilities are analyzed through the lens of Critical Disability Theory. This study provides teachers, administrators, and policy makers with a first-hand account of some of the barriers individuals with invisible disabilities face. Policies, environments, and practices in K-12 education, higher education, and broader society are analyzed. Advancements in inclusivity and accessibility as well as improvements in teacher education programs are suggested. Reflection throughout this study, three core themes emerged: listen, believe, and support individuals with invisible disabilities.

Institutional Strain And Substance Coping: Analyzing Perceived Racial Discrimination As A Predictor Of Adolescent Marijuana Use, Fahad Bin Islam Khan

This study examines the relationship between perceived racial/ethnic discrimination, psychological distress, and youth marijuana consumption through the lens of the General Strain Theory. It uses the complete-case sample from the 2023 Youth Risk Behavior Survey (YRBS), which surveyed 17,268 high school students across the United States, in a cross-sectional study. The study used logistic regression analyses to assess the association between perceived racial or ethnic bias (e.g., actual discriminatory experiences), psychological distress, and substance use (marijuana), and how closely emotional stress linked these two phenomena. Chronic infected wounds remain a major clinical challenge because elevated reactive oxygen species (ROS) in the wound microenvironment sustain inflammation, damage regenerating tissue, and limit the effectiveness of conventional antimicrobial and anti-inflammatory therapy. To address this problem, this study aimed to develop ROS-responsive thioketal-capped mesoporous silica nanoparticles (TK-MSNs) for the dual delivery of moxifloxacin (MOX) and curcumin (CUR) to support the management of infected wounds. The traditional memory theory of personal identity relies on an implicit storage model of memory to explain how the self persists over time. This model assumes that memories endure as stable representations that can be accurately retrieved, and that such representational continuity grounds diachronic identity. However, contemporary cognitive science demonstrates that remembering is an active and reconstructive process. Each act of recall reorganizes and transforms prior experience during the window of ecphory. This reconstructive reality poses a significant philosophical challenge. This thesis examines the formation, service, post-war life, and impact of the 6th United States Volunteer Infantry Regiment. This regiment and the five preceding it consisted of Confederate POWs who enlisted in the Union Army between 1864 and 1865. The project intersects with a few different historiographies, including Confederate nationalism/dissent and the military’s role in westward expansion. The 6th was formed from soldiers in Camp Douglas, Illinois, Camp Morton, Indiana, and Camp Chase, Ohio. The soldiers were predominantly young and poor southern farmers. The Tugaloo River is a 50-mile-long stretch of river that serves as an important political and biogeographical boundary in far northeastern Georgia and northwestern South Carolina. The purpose of this mixed-methods study was to examine women’s intra-gender experiences in higher education workplaces and their perceived influence on future interactions and behaviors, as well as to explore potential relationships between relational aggression (RA) and the Queen Bee Phenomenon (QBP). This mixed-methods study examines how generational cohorts (Generation X, Millennials, and Generation Z) and gender influence attitudes and responses to workplace sexual harassment (SH). Grounded in the bystander intervention model, moral responsibility theory, critical theory, and critical feminist theory, the study explores perceptions of verbal, visual, physical, and digital SH. Data were collected through a survey instrument administered to professionals in higher education and related fields, which included quantitative measures and open-ended questions to capture lived experiences. Quantitative analyses explored differences across generational cohorts and gender identities, while qualitative responses were analyzed thematically.
