Lung metastases are the primary cause of death for osteosarcoma (OS) patients. We recently validated interleukin-11 receptor α (IL-11Rα) as a molecular target for the inhibition of OS lung metastases. Since there is no clinically approved antibody against this receptor, we sought to identify downstream targets that mediate the effects of IL-11Rα signaling. We used shRNA to deplete IL-11Rα from OS cells; as a complementary approach, we added IL-11 exogenously to OS cells. The resulting changes in gene expression identified EZH2 as a downstream candidate. This was confirmed by knockdown of IL-11Rα in OS cells, which led to increased expression …

The Advanced Scatterometer (ASCAT) is a satellite-based remote sensing instrument designed for measuring wind speed and direction over the Earth's oceans. This thesis aims to expand and improve the capabilities of ASCAT by adding rain detection and advancing wind retrieval. Additionally, this expansion to ASCAT serves as evidence of Artificial Intelligence (AI) techniques learning both novel and traditional methods in remote sensing. I apply semantic segmentation to ASCAT measurements to detect rain over the oceans, enhancing capabilities to monitor global precipitation. Background: Aging significantly elevates the risk of developing neurodegenerative diseases. Neuroinflammation is a universal hallmark of neurodegeneration as well as normal brain aging. Which branches of age-related neuroinflammation, and how they precondition the brain toward pathological progression, remain ill-understood. The presence of elevated type I interferon (IFN-I) has been documented in the aged brain, but its role in promoting degenerative processes, such as the loss of neurons in vulnerable regions, has not been studied in depth.

Resistance to clinical malaria takes years to develop even in hyperendemic regions and sterilizing immunity has rarely been observed. To evaluate the maturation of the host response against controlled repeat exposures to P. falciparum (Pf) NF54 strain-infected mosquitoes, we systematically monitored malaria-naïve participants through an initial exposure to uninfected mosquitoes and 4 subsequent homologous exposures to Pf-infected mosquitoes over 21 months (n = 8 males) (ClinicalTrials.gov# NCT03014258). Global food calorie supply per person is more than 2900 kcal per day on average, but we have failed to ensure equitable access to these calories. Yet how uneven food access is within countries has remained poorly understood, as has the progress made over the past five decades on closing the access gap. Using publicly available data, we developed a theoretically-grounded statistical model to estimate the cross-national relationship between average per capita expenditure and per capita food availability and used this to estimate within-country access to food for income deciles in 135 countries. Jazan Industrial Economic City (JIEC) is located on the Red Sea coast in the province of Jazan, southwest of Saudi Arabia anchors diverse heavy and secondary industries in the energy, water desalination, petroleum, aluminum, copper, refineries, pharmaceuticals and food manufacturing fields. These various industries generate a large quantity of industrial wastewaters containing various toxicants. The present work represents ecologically beneficial alternatives for the advancement of environmental biotechnology, which could help mitigate the adverse impacts of environmental pollution resulting from petroleum refining effluents. Rejection sensitivity indexes individual differences in the expectation of, reaction to, and interpretation of possible rejection. Research on mechanisms underlying these individual differences have identified a reduction in top-down emotion regulatory processes in those exhibiting heightened rejection sensitivity. Most of these studies utilize rejection-specific affective stimuli. This study examined the relationship between rejection sensitivity score, calculated from the 18-question RSQ, and the recruitment of higher level inhibition resources, as indexed by the conflict N2, on three novel Flanker tasks. Current therapies for high-grade TP53-mutated myeloid neoplasms (≥10% blasts) do not offer a meaningful survival benefit except allogeneic stem cell transplantation in the minority who achieve a complete response to first line therapy (CR1). To identify reliable pre-therapy predictors of complete response to first-line therapy (CR1) and outcomes, we assembled a cohort of 242 individuals with TP53-mutated myeloid neoplasms and ≥10% blasts with well-annotated clinical, molecular and pathology data. Key outcomes examined were CR1 & 24-month survival (OS24). Luminal androgen receptor (LAR)-enriched triple-negative breast cancer (TNBC) is a distinct subtype. The efficacy of AR inhibitors and the relevant biomarkers in neoadjuvant therapy (NAT) are yet to be determined. We tested the combination of the AR inhibitor enzalutamide (120 mg daily by mouth) and paclitaxel (80 mg/m2 weekly intravenously) (ZT) for 12 weeks as NAT for LAR-enriched TNBC. Eligibility criteria included a percentage of cells expressing nuclear AR by immunohistochemistry (iAR) of at least 10% and a reduction in sonographic volume of less than 70% after four cycles of doxorubicin and cyclophosphamide. Hyperpolarization and cyclic nucleotide (HCN) activated ion channels are critical for the automaticity of action potentials in pacemaking and rhythmic electrical circuits in the human body. Unlike most voltage-gated ion channels, the HCN and related plant ion channels activate upon membrane hyperpolarization. Although functional studies have identified residues in the interface between the voltage-sensing and pore domain as crucial for inverted electromechanical coupling, the structural mechanisms for this unusual voltage-dependence remain unclear. Dietary protein is a critical regulator of metabolic health and aging. Low protein diets are associated with healthy aging in humans, and dietary protein restriction extends the lifespan and healthspan of mice. In this study, we examined the effect of protein restriction (PR) on metabolic health and the development and progression of Alzheimer's disease (AD) in the 3xTg mouse model of AD. Here, we show that PR promotes leanness and glycemic control in 3xTg mice, specifically rescuing the glucose intolerance of 3xTg females. PR induces sex-specific alterations in circulating and brain metabolites, downregulating sphingolipid subclasses in 3xTg females. PR also …

BACKGROUND: Most adults with sickle cell disease will experience a silent cerebral infarction (SCI) or overt stroke. Identifying patient subgroups with increased stroke incidence is important for future clinical trials focused on stroke prevention. Our 3-center prospective cohort study tested the primary hypothesis that adults with sickle cell disease and SCIs have a greater incidence of new stroke or SCI compared with those without SCI. A secondary aim focused on identifying additional risk factors for progressive infarcts, particularly traditional risk factors for stroke in adults.

Myxine limosa is a burrowing species of hagfish that occurs in the western North Atlantic in areas with muddy substrate and at depths generally greater than 100 meters. Burrowing of M. limosa has been observed from submersibles, but little is known about the behavior of these animals within the substrate or the biomechanical mechanisms involved. Here, we investigated burrowing in M. limosa by observing individuals as they burrowed through transparent gelatin. A photoelastic setup using crossed polarizers allowed us to visualize stress development in the gelatin as the hagfish moved through it. We found that M. limosa created U-shaped burrows …

Sexual violence is a societal epidemic and disproportionally affects disabled women. Disabled women belong to an intersectional identity that also experiences more negative attitudes, stigma, stereotypes, and discrimination than nondisabled individuals. In sexual assault cases, juror biases may translate into more detrimental attitudes toward disabled victims. The purpose of this study was to examine how these attitudes may impact outcomes in sexual assault trials when comparing disabled and nondisabled female victims. Purpose in life is an important dimension of psychological well-being with associated health benefits. The current study used a path analysis model to examine how perceived cognitive control, health locus of control- self, engagement in physical activity, and engagement in challenging cognitive activities relate to purpose in life longitudinally. Multimorbidity status was also examined as a potential moderator of paths in the model. This article introduces a novel method for combating microbial infections in textiles using advanced polymer coatings, particularly emphasizing formulations incorporating piperazine. We outline the synthesis and characterization of a copolymer consisting of piperazine, jeffamine, and bisphenol-a-diglycidyl ether, engineered to demonstrate robust antibacterial properties. Through rigorous testing, encompassing assessments of structural integrity and thermal resilience, we highlight the remarkable efficacy of this polymer in preventing microbial colonization. This work explores the fabrication route of curcumin-loaded carboxymethyl cellulose sodium/sodium alginate/palladium chloride (CMC-Na/SA/PdCl2) composite hydrogel as localized delivery system for cancer treatment. Herein, an approach was established for improving encapsulation capacity and exploiting therapeutic efficiency. CMC-Na/SA/PdCl2 composite hydrogels were prepared by gelation method using ionic crosslinker (PdCl2). Results proved that the optimized formula (CMC-Na:SA (4:1)/PdCl2 0.4%) recorded a gel fraction of 91% with a swelling ratio ~ 3000% after 96 h, while XRD analysis exhibited no remarks of sharp peaks which confirmed an amorphous phase of tested hydrogel.
