The fifth annual workshop on Principles and Techniques for Improving Preclinical Translation of Alzheimer's Disease Research was held in May 2023 at The Jackson Laboratory in Bar Harbor, Maine, USA. The workshop was established in 2018 to address training gaps in preclinical translational studies for Alzheimer's disease (AD). In addition to providing fundamental knowledge and hands-on skills essential for executing rigorous in vivo studies that are designed to facilitate translation, each year the workshop aims to provide insight on state-of-the-field technological advances and new resources including novel animal models, publicly available datasets, novel biomarkers, and new medical imaging tracers. Investigating The Antimicrobial Potential Of Bovine Lactoferrin Against The Neonatal Pathogen, Staphylococcus Capitis, Anna Desmond, Lesley Cotter, Des Field, Fiona O'Halloran

Lactoferrin is an antimicrobial glycoprotein that demonstrates a broad-spectrum of activity against a wide variety of clinical pathogens. This study investigated the potential of bovine lactoferrin (bLf) against multidrug resistant Staphylococcus capitis (S. capitis) strains. Growth curve analysis and time-kill curves demonstrated that at 750 µg ml−1 lactoferrin significantly inhibited (50.6%, P < 0.05) the growth of most isolates tested (90%), and this effect was based on a bacteriostatic mechanism. At the same concentration, bLf also significantly inhibited (30%, P < 0.05) biofilm formation in 40% of strains tested. Combinations of bLf with selected antibiotics were assessed for enhanced antimicrobial activity using growth curves. BLf combined with β-lactam antibiotics reduced the growth of S. capitis strains, however, the effects were not significant. BLf displays antimicrobial effects against multidrug resistant S. capitis isolates, but with strain-specific effects.

Background: Ocular gene transfer clinical trials are raising hopes for blindness treatments and attracting media attention. News media provide an accessible health information source for patients and the public, but are often criticized for overemphasizing benefits and underplaying risks of novel biomedical interventions. Overly optimistic portrayals of unproven interventions may influence public and patient expectations; the latter may cause patients to downplay risks and over-emphasize benefits, with implications for informed consent for clinical trials. Background: Preterm births are the leading cause of death worldwide. Preterm births not only have devastating effects on the newborn, but they also have psychological effects on the mothers. Identifying stress related to preterm births, and the factors associated with the stress among mothers with preterm infants is of great importance, for providing sound care to the newborns and their mothers. Background: Antenatal care (ANC) is critical for achieving Sustainable Development Goals (SDG) 3.1 and 3.2, aiming to reduce the global maternal mortality ratio to less than 70 per 100,000 live births by 2030, and to decrease neonatal and under-5 mortality rates. Despite its importance, approximately 830 women die daily from preventable pregnancy and childbirth causes, predominantly in low-resource settings.

We recently developed a dynamically coupled hydrological-ocean modeling system that provides seamless coverage across the land-ocean continuum during hurricane-induced compound flooding. This study introduced a local inertial equation and a diagonal flow algorithm to the overland routing of the coupled system’s hydrology model (WRF-Hydro). Using Hurricane Florence (2018) as a test case, the performance of the coupled model was significantly improved, evidenced by its enhanced capability of capturing backwater and increased water level simulation accuracy and stability. A coach's behavior significantly influences both athlete and team morale. As a generational change occurs, the behaviors exhibited by coaches may also require change. As members of Generation Z (Gen Z) begin to fill collegiate sports rosters, understanding their unique characteristics and preferences becomes critical for effective coaching strategies. Drawing upon generational studies and the interviews of six collegiate athletes within the Gen Z cohort, how coaching behavior influences collegiate athletes' athletic performance and team morale was analyzed within this cohort. Cognitive theories have been proposed to explain the development and maintenance of posttraumatic stress disorder (PTSD). Social-cognitive theories, as one of two major categories of cognitive theories (Brewin et al., 1996), emphasize the role of trauma-related beliefs in the development of PTSD, which led to the incorporation of modifying maladaptive trauma-related cognitions in psychological treatments of PTSD. Although extant studies have provided empirical evidence for the efficacy of these treatments, questions regarding active therapeutic components still exist. Phosphorus (P) is a life-essential element aiding the deterioration of water quality. Stormwater runoff transports nonpoint source P pollution to waterbodies, contributing to excessive P loading which may result in eutrophication. Green infrastructure (GI) is implemented to solve stormwater runoff pollution, but the P removal performance of these systems is variable. Moreover, P is arguably being unsustainably mined for fertilizer production and animal feed supplements to support demand from increasing global populations. This dissertation explored advancing the circular P economy utilizing additive manufacturing and GI soil amendments to remove and recover P from stormwater runoff. This research was an exploration of the impact of community trauma in urban communities. The focal participants were educators who have worked 10 or more years in the profession and in a school building. The study surveyed 20 educators using open-ended questions to understand their experiences with trauma, strategies for coping, and the effects on their personal and professional lives. Community trauma, including gun violence, mass and school shootings, homelessness, educational neglect, was common among participants. Notably, traumatic events such as the deaths of Michael Brown, George Floyd, and Trayvon Martin, and the COVID-19 pandemic, had significant emotional personal and …

This report describes the aims, approaches, and findings of an exploratory research study that researchers in the Institute for Asian American Studies at the University of Massachusetts Boston conducted in collaboration with the Bureau of Substance Addiction Services in the Massachusetts Department of Public Health. The study aimed to shed light on substance use and its associated harms among Asian Americans in Massachusetts. The researchers used focus group and individual interviews to learn about the participants’ perceptions, observations, and opinions about substance use in Asian American communities. Cercidiphyllum japonicum (katsura) is a dioecious tree native to China and Japan. Although imperiled in parts of its natural distribution, the species has become quite common as a cultivated tree in temperate North America. The species is noted as occasionally escaping cultivation in the eastern United States, but few records of established incursions (and the conditions under which they might recruit) exist in the literature. We here report an incursion of C. japonicum in central Pennsylvania, along with notes on seedling recruitment on the Bucknell University campus.

The development of an ideal membrane for membrane distillation (MD) is of the utmost importance. Enhancing the efficiency of MD by adding nanoparticles to or onto a membrane’s surface has drawn considerable attention from the scientific community. It is crucial to thoroughly examine state-of-the-art nanomaterials-enabled MD membranes with desirable properties, as they greatly enhance the efficiency and reliability of the MD process. This, in turn, opens up opportunities for achieving a sustainable water–energy–environment nexus. By introducing carbon-based nanomaterials into the membrane’s structure, the membrane gains excellent separation abilities, resistance to various feed waters, and a longer lifespan. Symbols For Schools: Types Of School Nicknames, How They Are Formed, And Implications For Leaders, Andrew Hudacs

School nicknames are powerful symbols representing the shared values and culture of their respective communities. This descriptive mixed method-study attempts to illuminate patterns in current school nicknames and examine ways that school leaders and communities approach changes to their nicknames. Nicknames from 1108 postsecondary schools were identified and analyzed with mixed methods to form a classification system that was then used to measure relationships between nicknames and school characteristics. The migratory and matrix-invading capacities of the cumulus–oocyte complex have been shown to be important for the ovulatory process. In metastatic cancers, these capacities are due to increased expression of proteases, however, there is limited information on protease expression in the cumulus–oocyte complexes. The present study examined cumulus–oocyte complex expression of plasmins, matrix metalloproteases, and A Disintegrin and Metalloproteinase with Thrombospondin Motifs family members in the rat and human. In the rat, human chorionic gonadotropin (hCG) administration increased cumulus–oocyte complex expression of Mmp2, Mmp9, Mmp13, Mmp14, Mmp16, Adamts1, and the protease inhibitors Timp1, Timp3, and Serpine1 by 8–12 h. Effective planning for natural resource management and wildlife conservation requires detailed information on vegetation structure at landscape scales and how structure is influenced by land- use practices. In many forested landscapes, the largest impacts of land use on forest structure are driven by forest management activities, which can include invasive species control, prescribed fire, partial harvests (e.g. shelterwood harvests) or overstory removals and clearcuts. Active timber management is often used to achieve forest conservation objectives, but to be used effectively, managers require knowledge of harvest frequency and extent in adjacent forests and at landscape scales. INTRODUCTION: Genome-wide association studies have identified over 70 genetic loci associated with late-onset Alzheimer's disease (LOAD), but few candidate polymorphisms have been functionally assessed for disease relevance and mechanism of action.
METHODS: Candidate genetic risk variants were informatically prioritized and individually engineered into a LOAD-sensitized mouse model that carries the AD risk variants APOE ε4/ε4 and Trem2*R47H. The potential disease relevance of each model was assessed by comparing brain transcriptomes measured with the Nanostring Mouse AD Panel at 4 and 12 months of age with human study cohorts.

BACKGROUND: Alzheimer's disease (AD) is the most common cause of dementia worldwide, with apolipoprotein Eε4 (APOEε4) being the strongest genetic risk factor. Current clinical diagnostic imaging focuses on amyloid and tau; however, new methods are needed for earlier detection.
METHODS: PET imaging was used to assess metabolism-perfusion in both sexes of aging C57BL/6J, and hAPOE mice, and were verified by transcriptomics, and immunopathology.
RESULTS: All hAPOE strains showed AD phenotype progression by 8 months, with females exhibiting the regional changes, which correlated with GO-term enrichments for glucose metabolism, perfusion, and immunity. Uncoupling analysis revealed APOEε4/ε4 exhibited significant Type-1 uncoupling (↓ …
