Comparison Of Two Disaccharides And Their Saccharide Components In Aot Reverse Micelles: Impacts Of D-Trehalose And D-Sucrose, Delaney Collier '25, Bridget L. Gourley, Nancy E. Levinger Oct 2024

Comparison Of Two Disaccharides And Their Saccharide Components In Aot Reverse Micelles: Impacts Of D-Trehalose And D-Sucrose, Delaney Collier '25, Bridget L. Gourley, Nancy E. Levinger

Annual Student Research Poster Session

Reverse micelles, featuring an AOT surfactant with a polar head group and nonpolar tail, encapsulate a polar phase at the nanoscale. With water as the polar phase, the nanoconfined water pool interactions and water-interface interactions mimic biological nanoconfinement. Thus, we investigated the impact of osmolytes on AOT reverse micelles to explore the role they play in biological systems. We investigated two disaccharides, D-trehalose and D-sucrose, and their monosaccharide components, glucose+glucose and glucose+fructose. We chose these molecules for their biological significance and to allow us to investigate whether subtle structural changes in the osmolyte impacts the intermolecular interactions in confinement as …

Bacteriostatic Effects Of Hydroxyl Radical-Generating Device On Klebsiella And Pseudomonas, Caroline Le '26, Philips Akinwole, Nipun Chopra

Annual Student Research Poster Session

Healthcare-associated infections (HAIs) serve as one of the most pressing problems in the medical field. On any given day, one in 31 hospital patients has at least one HAIs and even though the average duration of inpatient admissions has decreased, the frequency of HAIs has increased. The most popular treatment for HAIs is antimicrobial medications such as antibiotics, however, bacteria have been documented to develop resistance to several antibiotics. Hydroxyl radical is being explored as an alternative preventative treatment to HAIs as it has microbicidal effects on planktonic bacteria and fungi over short treatment time, but few studies have looked …

A Comparative Analysis Of Urea Vs. Acetamide In Aot Reverse Micelles, Kate Walsh '26, Bridget Gourley, Nancy E. Levinger

Annual Student Research Poster Session

In our exploration of the interactions between water and amide osmolytes, we focused on the effects of urea and acetamide on the stability of reverse micelles (RMs) formed with Sodium Bis(2-ethylhexyl)sulfosuccinate (AOT) in isooctane. Water, with its unique polarity and its ability to facilitate hydrogen and proton transfer, played a crucial role in our study, as we aimed to uncover how these amides would influence micellar behaviour. Our results revealed a compelling contrast between urea and acetamide. Acetamide, with its lower polarity due to a methyl group substitution, demonstrated superior solubility and significantly reduced precipitate formation compared to urea. This …

Designing An Attachment Assay For Keratinocytes Stimulated By Fibroblast Conditioned Media, Linh "Kas" Do '26, Colleen L. Doçi Oct 2024

Designing An Attachment Assay For Keratinocytes Stimulated By Fibroblast Conditioned Media, Linh "Kas" Do '26, Colleen L. Doçi

Annual Student Research Poster Session

Cell attachment involves the interaction between an individual cell with its neighbors and the extracellular matrix. These interactions are often regulated by the signaling molecules, including a variety of proteins, cytokines and growth factors. It is found in several studies that keratinocytes in the epidermis communicate with dermal fibroblasts in a bilateral paracrine model, in which they were simultaneously stimulated by the byproduct of the others. We developed this protocol with a simple, low-cost, and effective model to examine keratinocyte attachment that could be applied to examine the effects of fibroblasts on cell adhesion. For this method, we collected the …

Comparison Of S100Β Expression In Sk-Mel-25 And A-375, Nev Hommel '26, Ayusha Pokharel '27, Nipun Chopra

Annual Student Research Poster Session

Traumatic brain injury (TBI) affects more than 2 million Americans each year. S100β protein acts as a biomarker for TBI– studies have shown that micromolar levels of S100β are dangerous to neuronal health. Our plan for the summer was to experiment with whether miRNA-4705 could inhibit the production of S100β or not. To get started on the transfection, we had two hurdles to overcome. One of them was to find an efficient s100B primary antibody and the second was to ensure our s100B siRNA worked. By analyzing various research papers and their western blot, we identified and selected two s100B …

A Quantitative Survey Of Goldenrods And Their Galls In Greencastle, Indiana., Chiamaka Ndubuokwu '27, Danielle Sommerman '25, Rose Keith

Annual Student Research Poster Session

During the spring to summer months, galls—an abnormal growth of plant tissue—are commonly found on goldenrod plants. Some of the most common galls that plague goldenrods are the rosette, ball, and elongated galls caused by different flies and midges. This study set out to quantitatively survey goldenrod galls of those three types in Greencastle, Indiana and their potential fitness effects. The literature currently available focuses heavily on the relationships between ball galls and goldenrods, while the other gall types are understudied. This finding was especially surprising considering that the results of our data analysis suggests that the rosette galls were …

Avoidance Behavior Of Oppia Nitens (Oribatid Mites) To Herbicide: Glyphosate Compared To Microplastics In Standard Artificial Soil., Thi Doan '27, Philips Akinwole

Annual Student Research Poster Session

Despite growing public interest in environmental issues, awareness of contaminants affecting soil organisms remains broadly lacking, despite decades of research. This study investigated the impacts of a common herbicide; glyphosate, (Roundup), on the avoidance behavior of Oppia nitens - a soil mite species used as an indicator of soil health, and soil moisture content. The study involved exposing O.nitens to standard artificial soil treated with glyphosate concentrations ranging from 47.8 to 501.6 mg/kg. Avoidance behavior was assessed by observing mite distribution between treated and untreated soil over a 48-hour period. Results demonstrated clear avoidance behavior at higher glyphosate concentrations (501.6 …

Metabolic Adaptation Of Soil Microbial Communities To Metal Contamination: Assessment Of Cadmium Stress, Nina Shaffer '25, Emma Jacobs '25, Philips Akinwole

Annual Student Research Poster Session

Anthropogenic contamination of soil with heavy metals such as Cadmium (Cd2+) alters the metabolic activities and structure of soil microbial communities and has profound impacts on the soil ecosystem. In this study, we examined the effects of Cd2+ exposure on soil microbial communities by amending soil solutions with different concentrations of Cadmium nitrate tetrahydrate Cd(NO3)2 ● 4H2O (0, 0.5, 1.0, 1.5, and 2.5 mM). 150 µL of the Cd2+ amended and control soil solutions were added to BIOLOG 96-well ecoplates and incubated (26 ±1oC) over a 7-day period for metabolic analysis of 31 selected carbon substrates. Average well-color development (AWCD) …

The Origins Of The Major Questions Doctrine, Rachel Rothschild Oct 2024

The Origins Of The Major Questions Doctrine, Rachel Rothschild

Indiana Law Journal

In a series of recent cases, the Supreme Court has invoked the newly named “major questions doctrine” to strike down agency regulations that protect public health and the environment. Several Justices have argued that while the name “major questions” may be new, these decisions are simply the latest iteration in a longstanding effort of the courts to curtail the explosive growth of the administrative state since 1970. The first paradigmatic example of this line of cases is the 1980 “Benzene” case, in which the Supreme Court set aside the Occupational Safety and Health Administration (OSHA)’s new workplace standards for the …

The Rise And Fall Of The Consideration Of Race And Ethnicity In The Admissions Process: The Long-Term Negative Consequences Of The Fall, Kevin Brown Oct 2024

The Rise And Fall Of The Consideration Of Race And Ethnicity In The Admissions Process: The Long-Term Negative Consequences Of The Fall, Kevin Brown

Indiana Law Journal

Chief Justice Roberts wrote the majority opinion for the Supreme Court that struck down the affirmative action policies of Harvard College and the University of North Carolina. While Justices Thomas, Gorsuch, and Kavanaugh each wrote separate concurring opinions, they also joined Chief Justice Roberts’s opinion along with Justices Alito and Barrett. Thus, unlike the Court’s prior significant decisions that narrowly upheld affirmative action by one vote, this decision’s six-Justice majority allowed Chief Justice Roberts to write an opinion that more definitively rejected affirmative action than the Court’s tentative and cautious opinions upholding it. Despite what many may think about the …

Safeguarding Children's Voices In Child Protective Proceedings, Stephanie L. Tang Oct 2024

Safeguarding Children's Voices In Child Protective Proceedings, Stephanie L. Tang

Denver Law Review

In child protective proceedings, courts face the difficult task of determining the permanent placement of a child that promotes their well-being and safety, even if it means terminating their parents’ rights. Every year, authorities wrongfully and forcibly remove thousands of children from their families and homes. It is imperative that courts and attorneys adopt steps to provide children a voice and to guarantee children’s rights in these proceedings. Courts hear children’s preferences in child protective proceedings through four primary avenues: (1) appointment of a representative for the child, (2) direct in-court testimony, (3) in camera judicial interviews, and (4) admission …

Shocking Sentences, John B. Meixner Jr. Oct 2024

Shocking Sentences, John B. Meixner Jr.

Indiana Law Journal

Harsh recidivist sentencing penalties, like three-strikes laws, have been criticized heavily among both academics and practitioners on a number of different grounds. Most arguments focus on how sentences arising from these penalties are disproportionate—that there is no sensible relationship between the wrong committed and the sentence imposed. Those critiques are valid, but there’s another important problem with recidivist sentencing penalties that has been overlooked: They lead to sentences that are totally unexpected—indeed, shocking—to the defendants who face them. Many recidivist sentencing penalties cause large leaps in sentencing exposure that amount to exponential growth when compared with a defendant’s prior sentences. …

Identification Of Obstructive Sleep Apnea Symptoms Among Veterans In The Primary Care Setting, John Dominic Rosella Iv Oct 2024

Identification Of Obstructive Sleep Apnea Symptoms Among Veterans In The Primary Care Setting, John Dominic Rosella Iv

Doctor of Nursing Practice Scholarly Projects

Untreated obstructive sleep apnea (OSA) has long-term cardiovascular and metabolic health implications, impacting quality of life and increasing health expenditures. Veterans have a higher prevalence rate of OSA attributed to an aging population and higher rates of comorbid conditions. The problem of under recognition of OSA symptoms partly relates to clinicians needing to elicit relevant symptoms routinely. At the same time, the lack of primary-care-based screening protocols contributes to the percentage of undiagnosed OSA. The purpose of this evidence-based project was to implement routine screening to identify OSA risk among veterans in primary care settings in South Carolina. The method …

Animal, Dairy, and Veterinary Science Faculty Publications

Somatic cell nuclear transfer (SCNT) is one of the primary methods for production of genetically engineered sheep, which allows for gene editing or transgene introduction in somatic cells. The use of SCNT eliminates the risk of genetic mosaicism in embryos and animals that is commonly observed after zygote micromanipulations. This retrospective analysis of SCNT in sheep performed at Utah State University, spanning from 2016 to 2021, examined parameters that may impact pregnancy and full-term development, including donor oocytes (donor age), donor cell lines, SCNT parameters (time of oocyte activation following SCNT, number of transferred embryos, in vitro maturation and culture …

Zirconium-based metal-organic frameworks (UiO-66) have gained considerable attention owing to their versatile application. In the present research, UiO-66 was synthesized via a defect engineering approach, and its toxicity profile was explored. The synthesized nanomaterial was extensively characterized via spectroscopic methods such as FTIR and Raman spectroscopy, which confirmed the formation of the framework. X-ray diffraction (XRD) and transmission electron microscopy (TEM) were used to determine the crystallinity, shape and size of the nanoformulations. Thermal gravimetric analysis, 1H NMR spectroscopy and Brunauer–Emmett–Teller (BET) surface area analysis were used to identify the differences between pristine and defective UiO-66. Furthermore, the synthesized …

Amyloid Accelerator Polyphosphate Fits As The Mystery Density In Α-Synuclein Fibrils, Philipp Huettemann, Pavithra Mahadevan, Justine Lempart, Eric Tse

Open Access archive

Aberrant aggregation of α-Synuclein is the pathological hallmark of a set of neurodegenerative diseases termed synucleinopathies. Recent advances in cryo-electron microscopy have led to the structural determination of the first synucleinopathy-derived α-Synuclein fibrils, which contain a non-proteinaceous, “mystery density” at the core of the protofilaments, hypothesized to be highly negatively charged. Guided by previous studies that demonstrated that polyphosphate (polyP), a universally conserved polyanion, significantly accelerates α-Synuclein fibril formation, we conducted blind docking and molecular dynamics simulation experiments to model the polyP binding site in α-Synuclein fibrils. Here, we demonstrate that our models uniformly place polyP into the lysine-rich pocket, …

Approaching A U.S. Regulatory Framework For Sporeless Seaweed Biotechnology, Gabriela Martinez, Terra Bowling, Catherine M. Janasie, Charlie Doering-Powell, Amalia Aruda Almada

William & Mary Environmental Law and Policy Review

Seaweed farming is the fastest-growing aquaculture sector in the United States. The development of sterile, selectively bred (sporeless) seaweed aims to support the economic viability of future large offshore seaweed operations and minimize the environmental risks of these operations to wild seaweed beds. Farmers have implemented analogous breeding techniques to control and enhance most terrestrial crops and common aquaculture products (e.g., oysters and carp). This Article draws from existing federal and state aquaculture policies to explore four pillars critical to a future sporeless seaweed regulatory framework: ensuring maintained sterility, tracing the geographic origin of seed, securing offshore permits, and facilitating …

Dignity: A Journal of Analysis of Exploitation and Violence

The prevalence of non-partner sexual violence among college students is a pressing concern that demands urgent attention. Although an integral part of the education system, international college students are often underrepresented in various studies. This calls attention to understanding the cultural nuances and multiple factors affecting this population. The current research delves into the experiences of sexual abuse among international students within the last 12 months, focusing specifically on incidents where sexual penetration or unwanted sexual touching occurred without consent. The study sheds light on the frequency and demographics associated with these distressing incidents using data from a large-scale survey …

Enzyme Encapsulation Within The Hk97 Virus-Like Particle: An Investigation Of Substrate Inhibition Kinetics Within A Confined And Crowded Environment, Joseph B. Lively Oct 2024

Enzyme Encapsulation Within The Hk97 Virus-Like Particle: An Investigation Of Substrate Inhibition Kinetics Within A Confined And Crowded Environment, Joseph B. Lively

Chemistry Theses

Substrate inhibition is a paradoxical phenomenon observed in enzyme kinetics where increasing substrate concentrations lead to a marked decrease in the rates of enzyme-catalyzed reactions. Affecting an estimated 20% of studied enzymes, substrate inhibition poses significant challenges to the understanding of their function in essential biological processes and to their exploitation in industrial and therapeutic contexts. Studies show substrate inhibition to be a real limitation in vitro and rational conclusions have been drawn to explain the relevance of substrate inhibition in the self-regulation of biological pathways. However, there is currently no consensus on what role substrate inhibition plays in vivo …

Morphology Of The Normative Human Cone Photoreceptor Mosaic And A Publicly Available Adaptive Optics Montage Repository, Robert F. Cooper, Snega Kalaparambath, Geoffrey K. Aguirre, Jessica I. W. Morgan

Biomedical Engineering Faculty Research and Publications

Adaptive optics ophthalmoscopy has enabled visualization of the in vivo human photoreceptor mosaic in health, disease and its treatment. Despite this, the clinical utility of the imaging technology has been limited by a lack of automated analysis techniques capable of accurately quantifying photoreceptor structure and a lack of an available normative image database. Here, we present a fully automated algorithm for estimating cone spacing and density over a complete adaptive optics montage along with a database of normative images and cone densities. We imaged the cone mosaics surrounding the fovea and along the horizontal and vertical meridians of fifty normal-sighted …
