Purpose: Trichodysplasia spinulosa (TS) is a rare, disfiguring skin condition which presents with widespread asymptomatic or pruritic, skin-colored papules with white protruding keratin spiculations in immunocompromised individuals. Due to its rarity, there is little data to guide treatment decisions. The purpose of this article is to report a case of TS that completely resolved after treatment with topical cidofovir.
Materials and methods: A 19-year-old immunosuppressed female presented with widespread painful, itchy bumps on the nose and face. Upon examination, there were erythematous papules with hyperkeratinized spicules affecting the central face. Biopsy of the lesions was consistent with TS which was …

Oxidants produced through endogenous metabolism or encountered in the environment react directly with reactive sites in biological macromolecules. Many proteins, in particular, are susceptible to oxidative damage, which can lead to their altered structure and function. Such structural and functional changes trigger a cascade of events that influence key components of the proteostasis network. Here, we highlight recent advances in our understanding of how cells respond to the challenges of protein folding and metabolic alterations that occur during oxidative stress. Immediately after an oxidative insult, cells selectively block the translation of most new proteins and shift molecular chaperones from folding …

Objective: Determining the pathogenicity of missense variants in clinical genetic tests for individuals with epilepsy is crucial for guiding personalized treatment. However, achieving a definitive pathogenic classification remains challenging, with most missense variants still classified as variants of uncertain significance (VUS) and with the availability of many computational tools which may provide conflicting predictions. Here, we aim to evaluate the performance of state-of-the-art computational tools in pathogenicity prediction of missense variants in epilepsy-associated genes. This will assist in selecting the most appropriate tool and critically assess their use in clinical setting.

Background: Upper respiratory viral infections (URVIs) are responsible for 80% of asthma exacerbation episodes. However, the underlying mechanisms remain poorly understood.
Methods: In this study, we used a mouse model of URVI and examined the impact of URVI on asthma phenotypes and the underlying mechanisms.
Results: Previously, we have reported that nasal-restricted infection with respiratory syncytial virus (RSV) only produces mild sino-nasal inflammation and mucus production, without causing direct lung infection. However, such nasal-restricted infection dramatically enhanced TH2 and TH17 inflammatory responses in the lungs and increased airway hyperresponsiveness (AHR) in mice with house dust mite (HDM)-induced asthma. Additionally, nasal-restricted …

Abstract
This article examines the formation of self-concept in adults growing up in military families in Indonesia that practice authoritarian parenting. Moving away from a binary positive-negative perspective, this article argues that authoritarian parenting in the military context operates as a cultural reproduction, where military institutional values infiltrate the realm of family care at home and shape individual dispositions. Command culture then manifests as doxa, a system that naturalizes parental intervention and limits children's ability to make crucial life choices. Data collection for this article employed a qualitative method with a case study approach and triangulation process through interviews with …

Fish fecundity scales hyperallometrically with body mass, meaning larger females produce disproportionately more eggs than smaller ones. We explore this relationship beyond the species-level to estimate the “reproductive potential” of 1633 coral reef sites distributed globally. We find that, at the site-level, reproductive potential scales hyperallometrically with assemblage biomass, but with a smaller median exponent than at the species-level. Across all families, modelled reproductive potential is greater in fully protected sites versus fished sites. This difference is most pronounced for the important fisheries family, Serranidae. When comparing a scenario where 30% of sites are randomly fully protected to a current …

Fire is a dominant ecosystem process in many Mediterranean climate type ecosystems, and is predicted to increase in severity and frequency, shifting away from previous regimes in many regions. Responses of flora and fauna to fire are relatively well studied, but less is known about the responses of belowground microbiota. We quantified soil fungal dynamics over the first 12–15 months after fire, focusing on attributes of the fire regime (season, interval, severity). Soil samples were collected from three sites in a threatened woodland ecosystem in southwestern Australia, a Mediterranean-type climate region. Fungal taxa were identified via high throughput sequencing of …

Phosphatidylglycerol is a critical membrane phospholipid in microorganisms, synthesized via the dephosphorylation of phosphatidylglycerol-phosphate (PGP) by three membrane-bound phosphatases: PgpA, PgpB, and PgpC. While any one of these enzymes can produce phosphatidylglycerol at WT levels, the reason for the presence of all three in bacteria remains unclear. To address this question, we characterized these phosphatases in vitro to uncover their mechanistic differences. Our assays demonstrated that all three enzymes catalyze the hydrolysis of PGP but exhibit distinct substrate selectivity. PgpB displays a broad substrate range, dephosphorylating various lipid phosphates, while PgpA and PgpC show a higher specificity for lysophosphatidic acid …

P2X receptors (P2X1-7) are non-selective cation channels involved in many physiological activities such as synaptic transmission, immunological modulation, and cardiovascular function. These receptors share a conserved mechanism to sense extracellular ATP. TNP-ATP is an ATP derivative acting as a nonselective competitive P2X antagonist. Understanding how it occupies the orthosteric site in the absence of agonism may help reveal the key allostery during P2X gating. However, TNP-ATP/P2X complexes (TNP-ATP/human P2X3 (hP2X3) and TNP-ATP/chicken P2X7 (ckP2X7)) with distinct conformations and different mechanisms of action have been proposed. Whether these represent species and subtype variations or experimental differences remains unclear. Here, we show …

In-depth comparative genomic analysis was conducted to predict carbon, nitrogen, and phosphate assimilation pathways in the halotolerant, acidophilic genus Acidihalobacter. The study primarily aimed to understand how the metabolic capabilities of each species can determine their roles and effects on the microbial ecology of their unique saline and acidic environments, as well as in their potential application to saline water bioleaching systems. All four genomes encoded the genes for the complete tricarboxylic acid cycle, including 2-oxoglutarate dehydrogenase, a key enzyme absent in obligate chemolithotrophic acidophiles. Genes for a unique carboxysome shell protein, csoS1D, typically found in halotolerant bacteria but not …

Research outputs 2022 to 2026
Asthma is a highly prevalent but heterogenous condition. Uncontrolled disease is relatively common and may be due to ongoing inflammation and/or persisting bronchial hyper-reactivity. Bronchial thermoplasty has been in use for many years but optimal case selection and post-procedure assessment remain uncertain. We present a case of a gentleman in his 50s with lifelong asthma who experienced a persistent loss of control following influenza A in 2017. Despite multidisciplinary assessment and guideline-based escalation of therapy, he had persisting symptoms. In the context of his atopy, he was trialled unsuccessfully on dupilumab. Bronchial thermoplasty (BT) was undertaken with good clinical benefit. …

Crosstalk Between Cytokines And Igg N-Glycosylation: Bidirectional Effects And Relevance To Clinical Innovation For Inflammatory Diseases, Zhixian Chen, Xiaojia Xu, Manshu Song, Ling Lin
Research outputs 2022 to 2026
The crosstalk between cytokines and immunoglobulin G (IgG) N-glycosylation forms a bidirectional regulatory network that significantly impacts inflammation and immune function. This review examines how various cytokines, both pro- and anti-inflammatory, modulate IgG N-glycosylation, shaping antibody activity and influencing inflammatory responses. In addition, we explore how altered IgG N-glycosylation patterns affect cytokine production and immune signaling, either promoting or reducing inflammation. Through a comprehensive analysis of current studies, this review underscores the dynamic relationship between cytokines and IgG N-glycosylation. These insights enhance our understanding of the mechanisms underlying inflammatory diseases and contribute to improved strategies for disease prevention, diagnosis, monitoring, …

Anthropogenic activities are causing detrimental changes to coastal plants– namely seagrass, mangrove, and tidal marshes. Looking beyond recent times to past vegetation dynamics is critical to assess the response and resilience of an environment to change. Here, we develop a high-resolution multi-proxy approach, providing a new evidence base to decipher long-term change in coastal plant communities. Combining targeted environmental DNA analysis with chemical analysis of soils, we reconstructed 4,000 years of change at a temperate wetland on Torrens Island South Australia and identified an ecosystem shift that occurred ~ 1000 years ago. What was once a subtidal seagrass system shifted …

Studies suggest links between diabetes and gastrointestinal (GI) traits; however, their underlying biological mechanisms remain unclear. Here, we comprehensively assess the genetic relationship between type 2 diabetes (T2D) and GI disorders. Our study demonstrates a significant positive global genetic correlation of T2D with peptic ulcer disease (PUD), irritable bowel syndrome (IBS), gastritis-duodenitis, gastroesophageal reflux disease (GERD), and diverticular disease, but not inflammatory bowel disease (IBD). We identify several positive local genetic correlations (negative for T2D – IBD) contributing to T2D’s relationship with GI disorders. Univariable and multivariable Mendelian randomisation analyses suggest causal effects of T2D on PUD and gastritis-duodenitis and …

To improve the corrosion resistance of TiNi alloy fabricated by laser powder bed fusion (LPBF), a porous oxidation layer was synthesized by micro-arc oxidation in a sodium aluminate and sodium silicate electrolyte. The influences of the applied voltage and the processing time on the morphology of oxidation layer were investigated, and the corrosion behavior of the oxidation layer in artificial saliva was evaluated and compared with that of the as-fabricated LPBF alloy. The results indicate that, as increasing the applied voltage and the processing time, the oxidation layer becomes uniform and integrated. The optimum parameters are with an applied voltage …

The Disappearing Fishermen is one of a series of ethnographic journalism stories about the violence embedded in the web of power structures shaping our global market system. In the Honduran wetlands of the Central American Gulf of Fonseca, these structures result in multiple forms of harm and damage to artisanal fishing communities. During the eighties and the nineties, the shrimp farming industry that settled in these wetlands was conceived to meet the Global North’s “need” for shrimp. Powerful international institutions including the World Bank and development agencies such as USAID provided loans to private companies, while the Honduran government enabled …

I wanted to shed light on the situation of Indigenous women in Colombia and Venezuela, focusing specifically on the situation of Wayuu women living in the northern border of both countries. The identity of an Indigenous woman carries a dual burden—defined by both gender and ethnicity—that determines living conditions and access to rights within a worldview rooted in a collective sense of belonging to an Indigenous people. As a Wayuu Indigenous woman and a human rights researcher, my objective was to identify forms of discrimination by listening to Wayuu women in the territories of both countries. This approach allowed us …
Hydraulic Boundary Conditions Determine Biofilm Growth Patterns And Permeability In Groundwater, Jason Tyler Mick
Hydraulic Boundary Conditions Determine Biofilm Growth Patterns And Permeability In Groundwater, Jason Tyler Mick
Boise State University Theses and Dissertations
Biofilms are naturally occurring consortia of bacteria that colonize porous media such as streambed sediments, soils, and aquifers. Biofilm growth leads to clogging (i.e., bioclogging), which reduces the porosity and permeability of the porous medium and directly influences how nutrients and contaminants are transported and transformed in groundwater. To improve our understanding of how bioclogging regulates nutrient and contaminant fluxes, we must better characterize how flow conditions influence the spatial and temporal progression of biofilm growth within natural porous media geometries, as well as understand how this progression alters fluid flow.
In this study, I conducted microfluidic experiments to test …

As Moore's law ends, the conventional von Neumann computer architecture with binary-coded data representation has reached its bottleneck because of having separate computing and memory modules. In this architecture, continuous power is required due to sequential processing, making it challenging to improve efficiency further. The human brain can be regarded as the most energy-efficient computer architecture. In the brain, data is represented as small voltage pulses as spikes. That is why the neural units are called spiking neural networks (SNN). In SNNs, energy is required only when there is a spike, making it more energy efficient than the von-Neumann computer …

Quantum entanglement occupies a central role in the application of quantum mechanics towards emerging technologies of quantum information systems. Meanwhile, quantum entanglement is believed to be observed at room temperature in photosynthetic complexes in plants and algae through the energy transfer of dye molecules. The goal of this project is to engineer dye properties and their arrangement when dyes and aggregates are covalently attached to DNA so that the emergent property of exciton delocalization is as coherent, predictable, detectable and controllable. The creation of such a platform would enable the continued study of near room-temperature quantum entanglement and the more …

Seagrasses are marine flowering plants that form extensive meadows from the inter-tidal zone up to ~50 m depth. As biological and ecological Essential Biodiversity Variables, seagrass cover and composition provide a wide range of ecosystem services. Inter-tidal seagrass meadows provide services to many ecosystems, so monitoring their occurrence, extent, condition and diversity can be used to indicate the biodiversity and health of local ecosystems. Current global estimates of seagrass extent and recent reviews either do not mention inter-tidal seagrasses and their seasonal variation, or combine them with sub-tidal seagrasses. Here, using high-spatial and high-temporal resolution satellite data (Sentinel-2), we demonstrate …

The changes in air quality brought about by the COVID-19 lockdowns can provide valuable insight into how longer-term reductions in emissions might affect atmospheric composition. In urban Leicester UK, the 2020 COVID “lockdown” brought about several notable changes in atmospheric composition, including a considerable decrease (20–60%) in concentrations of non-methane volatile organic compounds (NMVOCs). Given their varied emission sources and lifetimes, NMVOC data can give valuable insights into how behavioural change impact atmospheric composition. The total concentration of 48 NMVOCs at roadside (RS) sites decreased from a pre-lockdown (sampled 16th-24th March 2020) concentration of 181.3 ± 41.4 μg/m3 to …

Large herbivorous mammals strongly influence vegetation structure by creating and maintaining open areas and causing disturbance within closed woody habitats. The herbivores alive today in Europe are only a small remnant of the large species that existed in high diversity and abundance before modern humans. The extinction of so many large herbivores during the last 50,000 years, and the loss of megaherbivores (body weight ≥1000 kg) from most of the continent before the Pleistocene–Holocene transition, is likely to have had cascading effects on vegetation structure and composition. To evaluate these effects within the European temperate forest biome, we examine the …

Background: Available treatment options for pediatric alopecia areata are limited and may even exacerbate the physical and psychosocial burden. Objective: This article aimed to provide a treatment classification system and counseling recommendations. Recommendations: Topical corticosteroids are appropriate for initial treatment due to their ease of use and safety. Topical minoxidil and dithranol are often used as adjunctive treatments based on mixed data. Intralesional corticosteroids are effective for localized disease, but injections may be challenging in children. Contact immunotherapy, systemic corticosteroids, and immunosuppressive agents such as topical and oral Janus kinase (JAK) inhibitors, dupilumab, and low-dose methotrexate demonstrate varying effectiveness, adverse …
