This review explores the intricate relationship between phytochemical intake and human health. It first highlights the contrast between the phytochemical-poor Standard American Diet (SAD) and ancestral diets rich in diverse plant and animal foods. The classification, sources, and health benefits of various phytochemicals, including polyphenols, flavonoids, alkaloids, and terpenes are described and discussed. The bioavailability of these compounds and their interactions with the gut microbiome are elucidated, emphasizing on their potential in modulating multiple pathways associated with aging and chronic diseases. This study aimed to (i) characterize municipal solid waste (MSW) sourced from Utah and Michigan transfer stations and (ii) upcycle, produce, and evaluate composites derived from this MSW. Composition analysis showed that the MSW was composed of a variety of commodity plastics, paper/cardboard, and inorganic materials. Detailed chemical analysis for lignin, cellulose, hemicellulose, and lipids was performed. The plastics identified were mainly polyethylene, polypropylene, polystyrene, and poly (ethylene terephthalate). The compoundability of the MSW was assessed by torque rheometry. This paper explores how ancient Greek literature and culture construct autonomous, mobile women as threats to the ideological and spatial order of the polis. Referencing mythological precedents and closely reading Classical texts, I propose the category of “excepted women” to address the threads of female transgression woven throughout this extensive literary corpus. Focusing on figures who transgress imposed spatial boundaries, I examine how such women are positioned as disruptors of gendered norms and civic stability, thus reflecting deep cultural anxieties about female agency and movement into spaces coded as masculine. ABSTRACT Objectives: To identify key trends and influential authors and institutions, provide thematic mapping, and determine recent evolutions and gaps in the disability research in Saudi Arabia. Methods: The study started in June 2024 and the data was analyzed during July 2024 in Al-Ahsa, Saudi Arabia. The analysis employed bibliometric techniques such as bibliometric coupling, and thematic mapping using VOSviewer and RStudio with the Bibliometrix package. Mindfulness and high-quality sleep can lessen nightmare frequency and severity. Sleep quality and nightmares are negatively correlated, and some studies have shown that mindfulness lessens nightmares. However, less is known about the combined effect of mindfulness and sleep quality on nightmare frequency and severity. This study aims to fill this gap by asking the question, does the combination of mindfulness and high-quality sleep have a stronger effect in lessening nightmare frequency and severity than either variable alone? This thesis examines the perspectives of educators working with unaccompanied migrant minors (MNAs) in eastern France, focusing on how they perceive and navigate the care system and creates their own communities of care within the gaps of the bureaucratic French system. Through qualitative analysis, it explores how educators interpret the structural challenges MNAs face, including access to healthcare, social integration, and legal protections. In addition to energy storage, adipose tissue communication to other organs plays a key role in regulating organismal physiology. While the link between adipose tissue dysfunction and pathophysiology, including diabetes, chronic inflammation, and infertility, is clear, the molecular mechanisms that underlie these associations have not been fully described. We use Drosophila melanogaster as a model to better understand how adipose tissue communicates to the ovary. Objective: Granulomatosis with polyangiitis (GPA) and microscopic polyangiitis (MPA) are chronic life-threatening vasculitides requiring substantial immunotherapy. Adult trials identified rituximab (RTX) as an alternative to cyclophosphamide (CYC) for remission induction of GPA and MPA. Disease rarity has limited feasibility of similar trials with pediatric patients. We aim to evaluate the relative efficacy and toxicity of CYC and RTX for patients with childhood GPA and MPA through registry-based comparative evaluation.

Mitochondrial dysfunction fuels vascular inflammation and atherosclerosis. Mitochondrial calcium uptake 1 (MICU1) maintains mitochondrial Ca2+ homeostasis. However, the role of MICU1 in vascular inflammation and atherosclerosis remains unknown. Here, we report that endothelial MICU1 prevents vascular inflammation and atherosclerosis by maintaining mitochondrial homeostasis. We observed that vascular inflammation was aggravated in endothelial cell–specific Micu1 knockout mice (Micu1ECKO) and attenuated in endothelial cell–specific Micu1 transgenic mice (Micu1ECTg). Furthermore, hypercholesterolemic Micu1ECKO mice also showed accelerated development of atherosclerosis, while Micu1ECTg mice were protected against atherosclerosis. Purpose: To explore the destructive and protective effects and therapeutic targets of IL-36 cytokines in dry eye disease using a murine dry eye model.

Methods: A dry eye model was established in C57BL/6 mice exposed to desiccating stress (DS) with untreated mice as controls. A topical challenge model was performed in normal mice with exogenous rmIL-36α, rhIL-38 and 2 % ectoine, or PBS vehicle. IL-36 cytokine expression was assessed by RT-qPCR and immunofluorescent (IF) staining. Corneal epithelial damage was evaluated by corneal smoothness score, Oregon Green Dextran (OGD) fluorescent staining, and tight junction barrier.

DNA damage promotes mutations that fuel cancer, aging, and neurodegenerative diseases13, but surprisingly, the causes and types of damage remain largely unknown. There are three identified mechanisms that damage DNA during transcription: RNA polymerase (RNAP) colliding with DNA-replication machinery head-on and co-directionally46, and R-loop-induced DNA breakage710. Background: Guideline-directed medical therapy (GDMT) with multiple risk factor goals is recommended for patients with chronic coronary disease (CCD), yet achieving all GDMT goals is uncommon. The relative importance of these goals and timing of their attainment on cardiovascular events is uncertain.

Objectives: This study aims to describe the relationship between achieving specific GDMT goals, when they are achieved, and clinical outcomes.

The vascular endothelium features unique molecular and functional properties across different vessel types, such as between arteries, veins and capillaries, as well as between different organs, such as the leaky sinusoidal endothelium of the liver versus the impermeable vessels of the brain. However, the transcriptional networks governing endothelial organ specialization remain unclear. Background and aims: Enhanced inflammatory signalling causally contributes to atrial fibrillation (AF) development. Gasdermin D (GSDMD) is an important downstream effector of several inflammasome pathways. However, the role of GSDMD, particularly the cleaved N-terminal (NT)-GSDMD, in non-immune cells remains elusive. This study aimed to elucidate the function of NT-GSDMD in atrial cardiomyocytes (ACMs) and determine its contribution to atrial arrhythmogenesis.

Methods: Human atrial appendages were used to assess the protein levels and localization. A modified adeno-associated virus 9 was employed to establish ACM-restricted overexpression of NT-GSDMD in mice.

Background: Immune checkpoint therapy (ICT) has significantly impacted the treatment of malignant pleural mesothelioma (MPM). Despite some promising results from combination therapies, nearly half of MPM patients do not benefit, underscoring the urgent need for reliable predictive biomarkers. This study assesses the prognostic value of serum soluble mesothelin-related peptide (SMRP) and PD-L1 levels in MPM patients receiving ICT.

Background: Approximately 1.25 million individuals died of tuberculosis in 2023, in part due to ineffective treatment. In patients with paucibacillary tuberculosis, microbiologic confirmation and drug resistance testing via respiratory specimens is challenging; hence, stool samples are increasingly used for microbiologic confirmation. Targeted next-generation sequencing (tNGS) of stool DNA may improve detection of drug-resistant (DR) tuberculosis, helping patients receive appropriate treatments.

Prostate cancer exhibits significant racial disparities, with African American (AA) individuals showing ∼64% higher incidence and 2.3 times greater mortality rates compared to their Caucasian (CA) counterparts. Understanding the complex interplay of genetic, environmental, lifestyle, socioeconomic, and healthcare access factors is crucial for developing effective interventions to reduce this disproportionate burden.

This study aims to uncover the genetic and transcriptomic differences driving these disparities through a comprehensive analysis using RNA sequencing (RNA-seq) and exome sequencing of prostate cancer tissues from both Black and White patients.

We investigated ferroptosis, a type of programmed cell death mechanism, in human hearts donated after brain death (DBD) and those donated after circulatory death (DCD), focusing on warm ischemia time (WIT) and cold storage. A total of 24 hearts were procured, with six from the DBD group and 18 from the DCD group. The DCD group was divided into three subgroups, each containing six hearts, based on different WITs of 20, 40, and 60 min. All procured hearts were placed in cold storage for up to 6 h. The Playbook Workflow Builder (PWB) is a web-based platform to dynamically construct and execute bioinformatics workflows by utilizing a growing network of input datasets, semantically annotated API endpoints, and data visualization tools contributed by an ecosystem of collaborators. Via a user-friendly user interface, workflows can be constructed from contributed building-blocks without technical expertise. Genetic counselors (GCs) practice in critical care settings. Some GCs have full-time inpatient roles, while most GCs who see inpatients do so as needed or on a rotating schedule in addition to seeing patients in an outpatient setting. Few studies have tracked and compared the amount of time it takes GCs to perform tasks in the inpatient and outpatient settings. Whether subclinical hypothyroidism should be treated in elderly patients (≥ 65 years) is controversial. The authors argue for a personalized, wait-and-see approach rather than universal treatment, pointing out that randomized clinical trials have not shown that levothyroxine treatment makes any difference in terms of hard clinical end points, quality of life, or hypothyroid symptom relief in elderly patients with this condition.

Foraging theory predicts animal behavior in many contexts. In patch-based foraging behaviors, the marginal value theorem (MVT) gives the optimal strategy for deterministic environments whose parameters are fully known to the forager. In natural settings, environmental parameters exhibit variability and are only partially known to the animal based on its experience, creating uncertainty. Models of uncertainty in foraging are well established. However, natural environments also exhibit unpredicted changes in their statistics. As a result, animals must ascertain whether the currently observed quality of the environment is consistent with their internal models, or whether something has changed, creating meta-uncertainty. Understanding the relationship between circuit connectivity and function is crucial for uncovering how the brain computes. In mouse primary visual cortex, excitatory neurons with similar response properties are more likely to be synaptically connected18; however, broader connectivity rules remain unknown. Here we leverage the millimetre-scale MICrONS dataset to analyse synaptic connectivity and functional properties of neurons across cortical layers and areas. Our results reveal that neurons with similar response properties are preferentially connected within and across layers and areas—including feedback connections—supporting the universality of ‘like-to-like’ connectivity across the visual hierarchy. Peptidyl arginine deiminases (PADs) are a family of calcium-binding enzymes that catalyze the citrullination of arginine and methylarginine residues as a form of gene regulation. The overexpression of PAD4 in cancer cells contributes to tumorigenesis and metastasis through the accelerated formation of neutrophil extracellular traps (NETs) and the downregulation of the p53 pathway. Significantly elevated PAD4 levels have been observed in various ovarian cancer subtypes, and it has been proposed that this elevation is a significant contributor to the malignancy of these cancers.
