Understanding the relationship between circuit connectivity and function is crucial for uncovering how the brain computes. In mouse primary visual cortex, excitatory neurons with similar response properties are more likely to be synaptically connected18; however, broader connectivity rules remain unknown. Here we leverage the millimetre-scale MICrONS dataset to analyse synaptic connectivity and functional properties of neurons across cortical layers and areas. Our results reveal that neurons with similar response properties are preferentially connected within and across layers and areas—including feedback connections—supporting the universality of ‘like-to-like’ connectivity across the visual hierarchy. Using a validated digital twin model, …

Cl-Amidine And Rapamycin Combination As A Novel Synergistic Co-Therapy For Ovarian Cancer, Victoria Walden Apr 2025

Cl-Amidine And Rapamycin Combination As A Novel Synergistic Co-Therapy For Ovarian Cancer, Victoria Walden

Longwood Senior Theses

Peptidyl arginine deiminases (PADs) are a family of calcium-binding enzymes that catalyze the citrullination of arginine and methylarginine residues as a form of gene regulation. The overexpression of PAD4 in cancer cells contributes to tumorigenesis and metastasis through the accelerated formation of neutrophil extracellular traps (NETs) and the downregulation of the p53 pathway. Significantly elevated PAD4 levels have been observed in various ovarian cancer subtypes, and it has been proposed that this elevation is a significant contributor to the malignancy of these cancers. Thus, the purpose of this research was to explore the effect of Cl-amidine, a small organic compound …

Rethinking Reverse Logistics: Neutrosophic Strategies For Warehouse Management Challenges, Thanh- Ngan Le, Quoc-Hieu Phan, Phi Hung Nguyen, Lan-Anh Thi Nguyen

Neutrosophic Sets and Systems

Implementing warehouse management platforms (WMPs) in Vietnam's smart reverse logistics (SRL) sector faces numerous barriers, hindering operational efficiency and competitiveness. This study develops a robust multi-stage decision-making framework to identify, analyze, and address these barriers, filling a critical research gap in SRL practices. The proposed model integrates Neutrosophic Sets (NS) for managing uncertainties in expert evaluations with the Decision-Making Trial and Evaluation Laboratory (DEMATEL) method to examine cause-and-effect relationships among barriers. Additionally, the Neutrosophic Combined Compromise Solution (NS COCOSO) method ranks solutions for overcoming these challenges. In the first stage, key barriers—such as insufficient strategic planning, lack of expertise in …

Articles

This Article introduces the new interdisciplinary field of Environmental Geography and Law, which has deep roots in ecology, social science, and law. Environmental and natural resources laws are situated in specific times and places where the climate, ecosystems, history and political economy influence both the land and the law. These places drive and constrain the way law develops. In turn, the law shapes places, from the dispossession and forced migration of indigenous groups, to land development via railroad land grants, to patterns of resource extraction and infrastructure development. Past efforts to integrate law and geography have focused more on critical …

17β-oestradiol (E2) inhibits overeating and promotes brown adipose tissue (BAT) thermogenesis, whereas prolactin (PRL) does the opposite. During lactation, the simultaneous decline in E2 and surge in PRL contribute to maternal metabolic adaptations, including hyperphagia and suppressed BAT thermogenesis. However, the underlying neuroendocrine mechanisms remain unclear. Here, we find that oestrogen receptor alpha (ERα)-expressing neurons in the medial basal hypothalamus (MBH), specifically the arcuate nucleus of the hypothalamus and the ventrolateral subdivision of the ventromedial hypothalamus (vlVMH), are suppressed during lactation. Deletion of ERα from MBH neurons in virgin female mice induces metabolic phenotypes characteristic of lactation, including hyperprolactinemia, hyperphagia …

Alphafold 3, Ai, Antibody Patents, The Future Of Broad Pharmaceutical Patent Claims, And Drug Development, Brendan Bargmann, Robert A. Bohrer

Faculty Scholarship

Artificial intelligence (AI) will have an enormous impact both on pharmaceutical development and patent protection, particularly for antibody therapeutics. In Amgen Inc. v. Sanofi, the U.S. Supreme Court limited the scope of Amgen’s therapeutic antibody patent to only those antibodies that were specifically described in Amgen’s patent application and that had been shown to bind to a particular region of the target antigen, blocking the activity of the antigen that caused disease. The reason for this limitation was the patent requirement of enablement: that potentially millions of antibodies could be generated to the target antigen but that not all …

The Genetic In Alzheimer, Elaine Vanterpool, Cristy Jimenez Apr 2025

APOE gene (apolipoprotein E gene) can be associated with pathogenic effects, particularly in the context of Alzheimer's disease and cardiovascular diseases, though it’s not inherently pathogenic in everyone. APOE comes in different alleles (variants), with APOE ε2, ε3, and ε4 being the most common. The protein encoded by this gene (Apolipoprotein) is a major apoprotein of the chylomicron. It binds to a specific liver and peripheral cell receptor, and is essential for the normal catabolism of triglyceride rich lipoprotein constituents. This gene maps to chromosome 19 in a cluster with the related apolipoprotein C1 and C2 genes. Mutations in this …

Brca1 Variants And Associations With Breast Cancer, Elaine Vanterpool, Tessa Tsoka

Student Posters

Breast cancer continues to be one of the most life-threatening diseases throughout the world, and various genetic factors play a critical role in its development. While there are a number of gene variants associated with hereditary breast cancer, BRCA1 is an extremely important tumor suppressor gene which is responsible for the maintenance of genomic stability. BRCA1, in addition to BRCA2, is crucial for DNA repair by method of homologous recombination. Together, these genes function to prevent the accumulation of DNA damage which could cause uncontrolled cell growth, leading to the development of tumors. The tumor suppressor protein that the BRCA1 …

An Analysis Of Dguok Variants Associated With Mitochondrial Dna Depletion Syndrome, Elaine Vanterpool, Ethan Harris Apr 2025

An Analysis Of Dguok Variants Associated With Mitochondrial Dna Depletion Syndrome, Elaine Vanterpool, Ethan Harris

Student Posters

The DGUOK is a nuclear gene that codes for the enzyme deoxyguanosine kinase, an essential protein in maintaining mitochondrial DNA and producing mitochondria. Mutagens in this gene can result in deoxyguanosine kinase deficiency, an inherited disorder that disrupts liver, muscle, and cerebral function. Manifesting from biallelic pathogenic variants, Mitochondrial DNA Depletion Syndrome ((MDDS) is a clinically heterogeneous group of autosomal recessive mitochondrial disorders that significantly reduce the number of mitochondrial DNA (mtDNA) in the affected cells. This results in stifled ATP synthesis, decreased energy reserves, and cellular dysfunction. As such, energy-dependent tissues such as the liver, heart, skeletal muscles, and …

Impact Of Smad 4 Gene To Juvenile Polyposis Syndrome, Elaine Vanterpool, Robert Lister Apr 2025

Impact Of Smad 4 Gene To Juvenile Polyposis Syndrome, Elaine Vanterpool, Robert Lister

Student Posters

The SMAD family job is to aid in signal transduction of transforming growth factor (TGF ), which is used as a growth hormone that aids in hyperpermeability of blood vessels, epithelial cell production, and cell differentiation. For the specific SMAD4 protein it has been to be a tumor suppressor gene that regulates the production of cells so that the abnormal production does not occur. However, with a mutated SMAD gene, these functions could be altered and cause drastic damage to the body. Learning how the SMAD 4 gene can cause diseases such as juvenile polyposis syndrome (JPS) and gastric …

An Analysis Of Androgen Receptor (Ar) Variants In Breast Cancer, Elaine Vanterpool Apr 2025

An Analysis Of Androgen Receptor (Ar) Variants In Breast Cancer, Elaine Vanterpool

Student Posters

The androgen receptor gene is more than 90 kb long and codes for a protein that has 3 major functional domains: the N terminal domain, DNA-binding domain, and androgen-binding domain. The protein functions as a steroid-hormone activated transcription factor. Upon binding the hormone ligand, the receptor dissociates from accessory proteins, translocates into the nucleus, dimerizes, and then stimulates transcription of androgen responsive genes. This gene contains 2 polymorphic trinucleotide repeat segments that encode polyglutamine and polyglycine tracts in the N-terminal transactivation domain of its protein. Expansion of the polyglutamine tract from the normal 9-34 repeats to the pathogenic 38-62 repeats …

Student Posters

Diabetes is a chronic condition characterized by high blood sugar levels resulting from the body's inability to produce or effectively use insulin. Type 2 diabetes, the most common form, is often linked to insulin resistance, where cells do not respond properly to insulin. The APPL1 gene encodes a multifunctional adaptor protein that plays a key role in insulin and adiponectin signaling, both of which are crucial for glucose metabolism. APPL1 enhances insulin stimulated glucose uptake by activating Akt, a major component in the insulin signaling pathway. Deficiencies in APPL1 have been linked to impaired insulin signaling, contributing to insulin resistance …

Early Motor And Language Development In Infants At Elevated Likelihood For Autism, Natalie C. Deal Apr 2025

Early Motor And Language Development In Infants At Elevated Likelihood For Autism, Natalie C. Deal

Senior Theses

During the early stages of infant development, various emerging skills set the foundation for more complex abilities. One such connection is the development of motor skills, which play a crucial role in influencing language outcomes later in the infants' first year. Although these developmental cascades are well understood in typically-developing populations, less is known about the cascade in infants at elevated likelihood for autism spectrum disorder (ASD). Many infants who are later diagnosed with ASD have motor delays or language delays, suggesting that the cascade from motor to language may not be typical in this population. Additionally, little is known …

The Isolation And Characterization Of Acidovorax And Brevundimonas Bacteriophages With Preliminary Biocontrol Study On Arabidopsis Thaliana, Minh Quynh N. Vu

Senior Theses

As antibiotic-resistance becomes an increasing global concern, researchers are looking for alternatives to combat phytopathogenic rhizobacteria – one of which is through the application of phage biocontrol. To elucidate the complex tripartite relationships between phage, bacteria and plant interactions, nine novel phages were isolated from potential phytopathogenic strains of Acidovorax (RBW13) and Brevundimonas (RBW18). These phages were isolated from root samples obtained from the Rocky Branch Creek and the Broad River located in Columbia, SC. Using Pulsed-Field Gel Electrophoresis (PFGE), we identified seven “medium sized” phages (~40 kbp), one “large-sized” phage (~200 kbp), and one “small-sized” phage ( < 25 kbp). Three phages were determined to be lambda-like due to their “ladder-building” patterns on PFGE. Although spot-tests revealed that all phages seemed to have broad host ranges that spanned over multiple genera, plaque-formations must be observed in future experiments to determine if these phages can infect their hosts productively. Preliminary in planta experiments were performed on Arabidopsis thaliana …
