Proteins Associated With The Early Endosome Contribute To De Novo [Psi+] Prion Formation, Jacqueline N. Kivila
Proteins Associated With The Early Endosome Contribute To De Novo [Psi+] Prion Formation, Jacqueline N. Kivila
Master's Theses (2009 -)
Prion diseases are infectious neurodegenerative disorders associated with Creutzfeldt-Jakob disease, bovine spongiform encephalopathy (“Mad cow” disease) and kuru. In prion disease, a native protein can misfold and further convert a normal version of itself to the misfolded conformation. These misfolded proteins can form aggregates which are associated with these diseases. While much work has been dedicated to understanding how protein aggregates impact cellular health, less is known about the cellular mechanisms that contribute to the formation of the prion. To dissect these mechanisms, we use the budding yeast, Saccharomyces cerevisiae, which provides a powerful tool for studying prion formation. The …

Despite advancements made in treatment options, cancer continues to be one of the leading causes of death worldwide. Photodynamic therapy (PDT) has gained attention as a minimally invasive and highly selective treatment option for cancer. However, challenges due to the hydrophobicity of photosensitizers and their poor tumor selectivity have limited their use in cancer therapy. Recent developments in nanotechnology, particularly the use of gold nanoparticles (AuNPs), help overcome these challenges. AuNPs provide a stable and biocompatible platform to deliver photosensitizers, improving their solubility, stability, and ability to target tumors while reducing side effects. Functionalized AuNPs take advantage of mechanisms like …

The adrenocorticotropic hormone (ACTH) test diagnoses relative adrenal insufficiency (RAI) or critical illness–related corticosteroid insufficiency (CIRCI). Initially, guidelines recommended corticosteroid/glucocorticoid (GC) therapy for septic patients with RAI, but later trials did not show a survival benefit, leading to updated guidelines that abandon targeting RAI or CIRCI. Recent studies with an RAI mouse model showed a clear survival benefit from GC therapy in mice with RAI, suggesting that inconclusive GC clinical trials might be due to issues with the ACTH test rather than targeting RAI. To investigate, we performed the ACTH test in septic mice. Interestingly, the ACTH test identified most …
Interspecific Sources Of Resistance For Neisseria Gonorrhoeae: Investigating Horizontally Transferred Antimicrobial Resistance Markers, Makenna Swartz
Interspecific Sources Of Resistance For Neisseria Gonorrhoeae: Investigating Horizontally Transferred Antimicrobial Resistance Markers, Makenna Swartz
Theses
Antibiotic resistance is a growing public health crisis, limiting treatment options and increasing the burden of infectious diseases. Bacteria can rapidly gain resistance through mutations and horizontal gene transfer (HGT). Understanding these mechanisms is critical for developing strategies to mitigate resistance and preserve antibiotic efficiency. Neisseria gonorrhoeae, the causative agent of gonorrhea, has developed resistance to multiple antibiotics, and has only one remaining treatment option. Commensal species are constantly exposed to antibiotics in humans, and over time acquire resistant mutations that then transfer to pathogenic species through HGT. Investigating these exchanges can provide insight into how resistance evolves and spreads …

Gender Differences In Perceived Organizational Support Among Veterans With Posttraumatic Stress Disorder, Elizabeth Gomez
Dissertations
Posttraumatic stress disorder (PTSD) is a well-studied issue within the U.S. military, with data showing military personnel face a higher risk of trauma than the civilian population. Many veterans and service members suffer from PTSD related to their military experiences (Renshaw, 2011). As research has largely focused on protective factors such as social and familial support, little attention has been given to organizational support (Meredith et al., 2011). Perceived organizational support (POS) refers to how individuals perceive the support they receive from their employers, with the military as the primary employer for veterans. This study was designed to explore whether …

Transforming Optical Vernier Effect Into Coherent Microwave Interference Towards Highly Sensitive Optical Fiber Sensing, Ruimin Jie, Jie Huang, Chen Zhu
Electrical and Computer Engineering Faculty Research & Creative Works
The optical Vernier effect has garnered significant research attention and found widespread applications in enhancing the measurement sensitivity of optical fiber interferometric sensors. Typically, Vernier sensor interrogation involves measuring its optical spectrum across a wide wavelength range using a high-precision spectrometer. This process is further complicated by the intricate signal processing required for accurately extracting the Vernier envelope, which can inadvertently introduce errors that compromise sensing performance. In this work, we introduce a novel approach to interrogating Vernier sensors based on a coherent microwave interference-assisted measurement technique. Instead of measuring the optical spectrum, we acquire the frequency response of the …

Production Of Biofertilizer And Biohydrogen Via Anaerobic Fermentation Of Ulva Spp.: A Feasibility Study For Reducing Coastal Eutrophication And Generating Clean Energy, Katie Graves
Capstone Projects and Master's Theses
This feasibility study investigates the potential of anaerobic fermentation of Ulva spp., a locally sourced macroalga, using Clostridium acetobutylicum to produce biofertilizer and biohydrogen. It further examines the viability of Ulva as fermentation biomass by identifying seasonal trends in nutrient concentrations of biofertilizer and gas production while determining the impact of the preservation method on gas yield. The biofertilizer used for nutrient analysis was produced from fresh Ulva harvested within 24 hours prior to fermentation. Orthophosphate (PO4-P), nitrite (NO2-N), nitrate (NO3-N), and ammonia (NH3-N) concentrations were analyzed post-fermentation using spectrophotometric flow injection analysis (FIA). Potassium (K+) concentrations were measured using …

Effects Of Density On Morphometrics And Reproductive Physiology In The Bull Kelp (Nereocystis Luetkeana), An Annual Foundation Species, Bennett M. Bugbee
Capstone Projects and Master's Theses
The bull kelp, Nereocystis luetkeana, is the predominant canopy forming kelp along much of the northeastern Pacific. However, it remains largely understudied due to the dynamic nature of its niche coupled with its annual life history. For populations to persist, reproductive sporophytes must release substantial spores to facilitate successful sexual fertilization among male and female gametophytes that will develop into the next cohort of sporophytes. High densities of spores increase the likelihood of completing the alternation of generation life cycle characteristic of kelp genera. Yet those high densities of spores have the potential to result in a high density of …
Seasonality Of Groundfish Community Structure And Habitat Associations On A Mesophotic Reef In Monterey Bay National Marine Sanctuary: Implications For Conservation And Management, Lauren Parker
Capstone Projects and Master's Theses
Management of marine ecosystems requires accurate, quantitative, and spatially explicit information on species distributions at scales that are relevant to the management process (J. Ward et al. 1999). This study investigates fish assemblages at depths between 30 and 40 meters, which are relatively understudied compared to shallower ( < 30 m) waters, surveyed extensively via SCUBA, and deeper (>40 m) waters, which are surveyed using submersibles. This research addresses ecological and management-based questions about seasonal and depth-related shifts in groundfish community structure, the role of oceanographic variables in predicting these shifts, and microhabitat selectivity as predictors of community assemblage. To evaluate species abundance, biomass, diversity, and microhabitat associations associations on …

Diet selection informs the health, fitness, and behavior of wild predators. Due to assumptions that vertebrate prey contains similar compositions of macronutrients (i.e., protein, carbohydrates, and lipids), whole prey items traditionally define carnivore diets. However, increasing evidence suggests that prey differ in terms of their macronutrient compositions, particularly relative to body size. Furthermore, omnivorous predators, like coyotes (Canis latrans), integrate both prey and nonprey diet items whose macronutrient compositions vary. This is particularly important in urbanized systems, which introduce or alter the distributions of prey (e.g., domestic pets) and nonprey (e.g., ornamental plants) foods in ways that contribute …
Analyzing The Inhibition Of Chemical Compounds On Collagenase In P. Aeruginosa, Elaine Vanterpool
Analyzing The Inhibition Of Chemical Compounds On Collagenase In P. Aeruginosa, Elaine Vanterpool
Student Posters
Pseudomonas aeruginosa is an opportunistic pathogen that poses significant challenges in clinical treatment due to its production of collagenase. This bacterium can cause several infections including pneumonia, meningitis, septicemia, and a host of other diseases. Collagenase acts as a key virulence factor by breaking down collagen in the host’s extracellular matrix, allowing bacteria to invade tissues. This study hypothesized that the effects of cell secreted collagenase would be inhibited by metronidazole, vanillin, and silver nanoparticles to reduce the pathogenicity of Ps. aeruginosa related infections. With the use of a spectrophotometric assay, collagenase activity was measured in treated and untreated bacterial …

Jenifer Michaels was a legendary subcultural star within the gay liberation film milieu of 1970s Los Angeles. Taking three paths to approaching and interpreting Michaels’s filmic and archival image, this article is structured by an analysis of Michaels’s documentary appearances, her performance in sex films, and her statements published in a textual interview. Working against what I call historiographic gentrification, the article utilizes archival and micro-historical methods to think through Michaels’s subcultural stardom as a nexus for complicating biography, documentary, and the politics of veracity.

The soluble variant of the ectopeptidase CD13 (sCD13), released from the cell surface by matrix metalloproteinase 14 (MMP14), is a potent pro-inflammatory mediator, displaying chemotactic, angiogenic, and arthritogenic properties through bradykinin receptor B1 (B1R). We revealed a link between sCD13 and amplified neutrophil-mediated inflammatory responses in SARS-CoV-2 infection. sCD13 was markedly elevated in patients with COVID-19 and correlated with disease severity and variants, ethnicity, inflammation markers, and neutrophil extracellular trap formation (NETosis). Neutrophils treated with sCD13 showed heightened NETosis and chemotaxis, which were inhibited by sCD13 receptor blockade. Meanwhile sCD13 did not induce platelet aggregation. Single-cell analysis of COVID-19 lungs …

In Savanna ecosystems, cycles of sustenance include periods of food abundance and deficits. In rain-fed agricultural systems, forests are a vital food source that helps households close this cycle. This paper uses cross-sectional data to explore the association between provisioning ecosystem services and timing to food insecurity in forest fringe communities (N = 500). The time to food insecurity analysis revealed a significant difference in the onset of food insecurity among households with varying levels of access to forest provisioning services. Households with access to more than four forest products experienced a slower timing to seasonal insecurity. In contrast, …

Valuing Worker Authenticity, Dallan F. Flake
Valuing Worker Authenticity, Dallan F. Flake
William & Mary Law Review
Title VII of the Civil Rights Act of 1964 protects workers from discrimination because of their race, color, religion, sex, or national origin. Controversially, courts do not typically extend this protection to individual expression tied to a protected trait, such as a Black employee who wears their hair in dreadlocks or a Latino employee who speaks Spanish on the job. Judicial unwillingness to protect employee self-expression tied to a protected trait causes employees who identify with marginalized groups to suppress their identities to conform with mainstream culture—often at tremendous costs to themselves and the organizations they work for.
