Mucopolysaccharidosis type IVA (MPS IVA, Morquio A syndrome) is a rare inherited disorder characterized by skeletal dysplasia due to deficient N-acetylgalactosamine-6-sulfate sulfatase activity, resulting in glycosaminoglycan (GAG) accumulation. Identifying accurate biomarkers reflecting clinical severity and therapeutic response remains challenging. This study evaluated potential surrogate biomarkers, including N-terminal pro-C-type natriuretic peptide (NT-proCNP), collagen types I and II, mono-sulfated keratan sulfate (KS), di-sulfated KS, and chondroitin-6-sulfate (C6S), in blood and urine samples from 60 patients ranging from 1 to 62 years of age. Irritable bowel syndrome (IBS) is a disorder of gut-brain interaction influenced by biological, psychological, and social factors. Stress and socioeconomic influences on IBS have been studied, however few studies have explored the impact of neighborhood and social environments across racial and ethnic groups and sex at birth. The biopsychosocial framework was used to examine whether built environment and social context contribute to IBS prevalence. Future food production requires sustainable approaches that used both land and water resources efficiently, while remaining economically viable. One approach is aquaponics, the practice which combines hydroponics and aquaculture, as a sustainable food producing system. The extent of genetic variation and its influence on gene expression across multiple tissue and cellular contexts is still being characterized, with germline Structural Variants (SVs) being historically understudied. DNA methylation also represents a component of normal germline variation across individuals. Here, we combine germline SVs (by short-read sequencing) with tumor DNA methylation across 1292 pediatric brain tumor patients. For thousands of methylation probes for CpG Islands (CGIs) or enhancers, rare and common SV breakpoints upstream or downstream associate with differential methylation in tumors spanning various histologic types, a significant subset involving genes with SV-associated differential expression. Objective: Evaluate the use of tranexamic acid (TXA) and observation as a management option for pediatric patients presenting with posttonsillectomy hemorrhage (PTH). Study Design: Retrospective analysis of a prospectively implemented quality improvement initiative with a historical control comparison group. Setting: Tertiary children's hospital. Methods: Patients < 18 years of age who underwent adenotonsillectomy (AT) and returned to the Emergency Department for PTH were included. Patients who were stable without large volume or active bleeding were given intravenous TXA and admitted for overnight observation. Data were compared in a before-and-after analysis: preprotocol (April 2022 to March 2023) versus postprotocol (April 2023 to March 2024). For cost-effectiveness analysis, we analyzed aggregated claims data from a commercial claims database. Results: Preprotocol 1800 adenotonsillectomies were performed, and 40 procedures were performed for control of hemorrhage (2.2 per 100 AT). Postprotocol 2356 adenotonsillectomies were performed, and 30 procedures were performed to control hemorrhage (1.3 per 100 AT) showing a significant reduction in return to the operating room (relative risk [RR] = 0.59, 95% confidence interval [CI] [0.358, 0.916], P-value.020). There were no reported adverse events attributable to TXA. An estimated 21 surgeries were avoided, and 26 additional patients were observed in the hospital during the postprotocol period, for an estimated net cost savings of $174,970. Conclusion: The implementation of a standardized TXA protocol significantly reduced the need for return to the operating room for PTH in pediatric patients, without complications and with net cost savings to the health care system.

Fish oil supplements (FOS) are known to alter circulating levels of polyunsaturated fatty acids (PUFAs) but in a heterogeneous manner across individuals. These varied responses may result from unidentified gene-FOS interactions. To identify genetic factors that interact with FOS to alter the circulating levels of PUFAs, we performed a multi-level genome-wide interaction study (GWIS) of FOS on 14 plasma measurements in 200,060 unrelated European-ancestry individuals from the UK Biobank. From our single-variant tests, we identified genome-wide significant interacting SNPs (p < 5 × 10-8) in the FADS1-FADS2 gene cluster for total omega-3, omega-3%, docosapentaenoic acid (DHA), DHA%, and the omega-6 to omega-3 ratio. Among the interaction signals for omega-3%, the lead SNP, rs35473591 (C>CT, CT allele frequency = 0.34), had a lower association effect size in the FOS-taking group (β = 0.35 for …

This research aims to develop a STEM-based virtual microscope module. It follows a 4D development model, comprising define, design, develop, and disseminate stages. Interview were employed as the primary data collection techniques. The data analysis techniques utilized both descriptive and quantitative methods. The feasibility of developed module validated by expert validators in material content, language, and learning design. This qualitative research employing participatory action protocols (PAR) engages a limited number of gender minority adolescents (current transgender, nonbinary, and gender-fluid secondary students) and allied staff members at one large Midwestern public high school district. The principal purpose is to improve transgender secondary student schooling experiences. One of the most common sites of cancer metastasis is to the bone. Bone metastasis is associated with substantial morbidity and mortality, and current therapeutic interventions remain largely palliative. Metastasizing tumor cells need to reprogram their metabolic states to adapt to the nutrient environment of distant organs; however, the role and translational relevance of lipid metabolism in bone metastasis remain unclear. Purpose: To evaluate the pharmacology, efficacy, safety, and dosing and administration considerations (including adjusted ideal body weight [AIBW] dosing) for mirvetuximab soravtansine-gynx, a first-in-class folate receptor alpha (FRα)-directed antibody-drug conjugate for platinum-resistant ovarian cancer (PROC).
Summary: A literature search was conducted in PubMed using the terms "ovarian cancer" and "mirvetuximab soravtansine" of articles published from inception to April 16, 2024. Relevant publications, abstracts, and clinical trials were reviewed. Mirvetuximab soravtansine-gynx is dosed at 6 mg/kg AIBW every 3 weeks and comprises an FRα-binding antibody, a hydrophilic disulfide linker, and a maytansinoid DM4 payload. Vocational schools equip students with practical skills and expose them to industry-related hazards, making Occupational Health and Safety (OHS) a crucial aspect of the school environment. This study aims to identify key factors behind the weak implementation of OHS in vocational schools. A literature review used articles published between 2017–2024 from national and international journals. The review found poor OHS implementation stems from lack of awareness, inadequate policies, insufficient infrastructure, weak safety culture, and negligence in accident prevention. Dry eye disease (DED) is a multifactorial disease that drives tear film instability and hyperosmolarity, directly damaging the ocular surface epithelium. Lysophosphatidic acid receptor (LPAR) is a G-protein coupled receptor (GPCR) responsible for Ca2+ and cAMP mobilization via PLC and adenylyl cyclase activation, respectively. LPAR signaling is involved in cell proliferation and wound healing in human corneal epithelial cells (HCECs) as well as in neuropathic pain. Despite FDA-approved amyloid-beta (Aβ) therapies in Alzheimer’s disease (AD), Tau pathology remains untreated and is closely linked to cognitive decline. Mitophagy, the process of mitochondrial degradation, is vital for maintaining neuronal health, and its dysregulation is implicated in AD. Recently, our lab identified a mitophagy inducing compound (MIC) that extends the lifespan of C. elegans. We hypothesized that MIC would significantly mitigate neuropathology in an AD mouse model. Introduction: Amblyomma americanum is a medically important tick species established in the southeastern United States, with populations gradually spreading northward into New England. This study aimed to evaluate the current presence, distribution, and density of A. americanum in Maine.
Methods: Surveillance efforts focused on Ixodes scapularis have been conducted by the Vector-Borne Disease Laboratory of MaineHealth Institute for Research since 1989. This surveillance uses tick flagging, examination of hunter-harvested game and trapped wild hosts for feeding ticks, and a statewide passive surveillance program. Introduction: Maine has experienced a rise in tick-borne diseases over the past several decades, reflecting broader trends in the northeastern United States. Ixodes scapularis is now found statewide and is the primary vector of Borrelia burgdorferi, Anaplasma phagocytophilum, Babesia microti, Borrelia miyamotoi, and Powassan virus. Other emerging tick species pose additional threats.
Methods: The University of Maine Cooperative Extension Tick Lab conducts a public tick-testing program. Introduction: Vector-borne lymphadenitis in children is uncommon. A detailed exposure and travel history can facilitate diagnosis.
Clinical Findings: The pediatric inpatient service admitted a 6-year-old patient after multiple acute care visits for right posterior auricular tenderness, erythema, and swelling.
Clinical Course: Upon admission, the patient received intravenous ceftriaxone and clindamycin for progressive suppurative lymphadenitis. After a review of the patient’s history, which included the removal of an engorged tick and recent travel to Martha’s Vineyard, Massachusetts, the treatment was transitioned to intravenous gentamicin for ulceroglandular tularemia.

Sunflower (Helianthus annuus L.) is considered as one of the most important oilseed crops in Egypt and worldwide. It is being infected with many pathogens, among these pathogens Macrophomina phaseolina (Tassi) Goid, the causal agent of charcoal rot is the most prevalent one, and responsible for severe economic losses on sunflower production. Fourteen isolates of M. phaseolina were collected from naturally infected sunflower plants. Pathogenicity tests revealed that tested isolates varied significantly in their pathogenic capabilities. But all of the tested isolates were pathogenic and incited the symptoms of pre- and post-emergence damping-off as well as symptoms of charcoal rot. …

Honey, a natural product with effective medicinal properties and exceptional energy content, holds a significant place in the hearts of consumers. This study evaluated the pH, free acidity, conductivity, sugar composition, and total antioxidant content of eight types of rare honey produced in Oman. Additionally, well-known types of honey such as Sidr and Sumur, along with some commercial honey samples, were studied for comparison purposes. A simple and innovative paper-based analytical device, recently developed by our group, was applied as an appropriate alternative to time-consuming chromatography-based methods. Because of their ubiquitous presence, ions interact with numerous macromolecules in the cell and affect critical biological processes. Here, we discuss how cations including Mg2+ alter the enzymatic activity of a DNA glycosylase by tuning its affinity for DNA. The response of uracil DNA glycosylase (UNG2) to Mg2+ ions in solution is biphasic and paradoxical, where low concentrations of the ion stimulate the enzyme, but high concentrations inhibit the enzyme. We analyzed this phenomenon by modeling experimental data with a statistical framework that we empirically derived to understand molecular systems that display biphasic behaviors. During the Fall 2023 semester, a group of students in their first semester of the elementary education program at The University of Texas Rio Grande Valley (UTRGV) was required to use OneNote to complete a Community Case Study, which is a capstone project in the teacher education program at UTRGV. The projectconsists of three sections, which include a study of the school and district, visits to area attractions in the community, interviews with members of the community, and a final reflection on the project.
