Background: NLRP2 is a subcortical maternal complex (SCMC) protein of mammalian oocytes and preimplantation embryos. SCMC proteins are encoded by maternal effect genes and play a pivotal role in the maternal-to-zygotic transition (MZT), early embryogenesis, and epigenetic (re)programming. Maternal inactivation of genes encoding SCMC proteins has been linked to infertility and subfertility in mice and humans, but the underlying molecular mechanisms for the diverse functions of SCMC proteins, and specifically the role of NLRP2, are incompletely understood.

Unseen Burdens: Exploring Gender Disparities In Infertility Experience And Depressive Symptoms, Khiara Cardoza Brown

This study examines how experiences with infertility relate to mental health outcomes among newly married U.S. couples, emphasizing gender differences and the potential moderating role of gender attitudes. Using data from 3,137 individuals in a nationally representative sample, three Actor-Partner Interdependence Models were estimated: first assessing actor and partner effects of infertility on depressive symptoms and subsequently incorporating gender attitudes as moderators. Results indicated a significant actor effect for wives, where infertility was associated with higher levels of depressive symptoms, and this association was significantly stronger for wives than for husbands. No significant effects were found for husbands' infertility and …

Coronaviruses have evolved a breadth of mechanisms to antagonize host innate immune responses. One widely studied mechanism is the ability to deconjugate ISG15 from target proteins in a process called deISGylation, which is catalyzed by the papain-like protease (PLP) domain of viral non-structural protein (nsp) 3. Since PLP activity is required for viral replication, it has been challenging to generate an infectious virus with a PLP that maintains protease activity but has lost deISGylase activity. INTRODUCTION/OBJECTIVE: Kidney diseases cause high morbidity and mortality worldwide. This study investigated the mechanistic effects of Fisetin (FIS) on nephrotoxicity, kidney injury, and nephropathy induced by drugs, toxic chemicals, diabetes, lupus, diet, ureteral obstruction, and ischemic situations.
METHODS: To identify pertinent articles published before Oct 1, 2024, a comprehensive electronic search was performed across several databases, including MEDLINE/PubMed, Embase, Cochrane Library, Web of Science, and Scopus. After establishing clear inclusion and exclusion criteria, studies that met the research objectives were selected. Data were extracted and analyzed, documenting study characteristics, methodologies, and biological mechanisms.

Microplastics are found in Earth’s atmosphere, lithosphere, hydrosphere, pedosphere, and ecosphere. While there is a growing interest and need to solve this grand challenge in both the academic and policy realms, few have engaged with academics, policymakers, and community partners to co-identify the problem, co-design research, and co-produce knowledge in tackling this issue. Using a convergence research framework, we investigated the perception of microplastic pollution among different end users, delivered educational materials to K-12 teachers and practitioners, and identified key sampling points for assessing environmental microplastic concentrations in the Columbia River Basin, United States. The (Dis)Continuity Of Meaning In John Dewey’S Experience And Nature, Harrison S. Jackson

Experience and Nature proposes a naturalistic empiricism emphasizing continuity. This paper will argue that this continuity is threatened by Dewey’s anthropocentric in its account of meaning, which is only found in humans but is necessary for the perception of objects, communication, and social existence. Given contemporary research in ethology, Dewey’s denotative method requires a modification of this account. I will propose Merleau-Ponty’s notion of sense or the neuroscientific theory of enactivism as supplements preserving embodiment and social communication while being more continuous. Finally, I examine some recent applications of Dewey’s naturalism and show how they modify this account of meaning.

Mutations in ATP8B1 cause progressive familial intrahepatic cholestasis, with symptoms including pruritus, pancreatitis, fat malabsorption, intestinal inflammation, and failure to thrive. High-throughput studies showed interconnection between ATP8B1 and phosphoinositide (PIPs), but the mechanism linking ATP8B1, lipid metabolism, and inflammation remains unclear. Atp8b1G308V/G308V mouse model, unbiased RNAseq, high-resolution-stimulation emission depltion (STED)-microscopy, and Crispr-Cas9 generated ATP8B1-/- knockouts in hepatocytes/monocytes/macrophages were used to determine role of ATP8B1 in phosphatidylinositol,4-5-bisphosphate (PIP2) trafficking and inflammation. Degenerative joint disease remains a leading cause of global disability, with early diagnosis posing a significant clinical challenge due to its gradual onset and symptom overlap with other musculoskeletal disorders. This review focuses on emerging diagnostic strategies by synthesizing evidence specifically from studies that integrate biochemical biomarkers, advanced imaging techniques, and machine learning models relevant to osteoarthritis. We evaluate the diagnostic utility of cartilage degradation markers (e.g., CTX-II, COMP), inflammatory cytokines (e.g., IL-1β, TNF-α), and synovial fluid microRNA profiles, and how they correlate with quantitative imaging readouts from T2-mapping MRI, ultrasound elastography, and dual-energy CT. A system and method for controlling active ion transporters, such as sodium-potassium (Na/K) pumps to actively transports 3 Na+ and 2 K+ ions by consuming one ATP. The system and method provide an oscillating electric field that is effective in running the active ion-transport at their physiological running mode by consuming one ATP molecule to actively transport ion and to synthesize one ATP for each pumping cycle. The ATP molecule that is synthesized compensates for the ATP molecule that is consumed in actively transporting ions, resulting in a net-consumption of ATP that is significantly reduced, theoretically to zero.

Immune checkpoint inhibition (ICI) has emerged as a critical treatment strategy for squamous cell carcinoma of the head and neck (HNSCC) that halts the immune escape of the tumor cells. Increasing evidence suggests that the onset, progression, and lack of/no response of HNSCC to ICI are emergent properties arising from the interactions within the tumor microenvironment (TME). Deciphering how the diversity of cellular and molecular interactions leads to distinct HNSCC TME subtypes subsequently governing the ICI response remains largely unexplored. Multiple system atrophy (MSA) is a rare, fast progressing, debilitating and ultimately fatal movement disorder. The pathological hallmark of MSA disease is the aggregation and misfolding of alpha-synuclein (α-syn) protein in the cytoplasm of oligodendrocytes, termed glial cytoplasmic inclusions (GCIs). Accumulation of these GCIs result in progressive demyelination and neurodegeneration of the striatonigral and olivopontocerebellar regions of the brain. Currently, no therapeutics have been approved that slow or halt the progression of this devastating disease. We evaluated inflammatory markers among 389 surgically confirmed endometriosis cases and 505 controls from the Women's Health Study: From Adolescence to Adulthood (A2A) cohort. Participants reported dysmenorrhea, acyclic pelvic pain, dyspareunia, and pain with bowel movements. Using multiplex assays, we measured their levels of plasma interleukin (IL)-1β, -6, -8, -10, and -16, tumor necrosis factor (TNF)-α, monocyte chemotactic protein (MCP)-1 and -4, thymus and activation-regulated chemokine (TARC), and interferon gamma-induced protein (IP)-10. For each symptom, we computed biomarker-level geometric means (GMs) with 95% confidence intervals (95% CI) using multivariate linear regression among the endometriosis cases and controls, with interactions with …

Positive parent-teacher relationships are vital for student success in pre-kindergarten through eighth grade, particularly among low-income, African-American parents and guardians. Research suggests respect, trust, and open communication as key factors for cultivating positive parent-relationships; however, race and social class have influenced the development of African-American parents and guardians’ relationships with their children’s teachers. This instrumental case study investigated how six low- income, African-American parents and guardians in Midwestern PK-8 public schools perceive their relationships with their children’s teachers. Adoption functions as both an identity status and a source of unique social benefits and challenges for teenagers, informing their experience of friendship. While prior research primarily focuses on individual outcomes and difficulties that adoptees face, such as trauma-related disorders and social skill deficits, it often overlooks how adoptees and their parents perceive these impacts on peer relationships. This study draws on twenty-one in-depth interviews with adoptees ages thirteen to twenty-one and their adoptive parents to explore the role that adoption and adverse childhood experiences have on an adopted teen’s experience of friendships. OBJECTIVE: To evaluate differences in study engagement in diverse racial/ethnic groups that have been significantly underrepresented in atrial fibrillation and digital clinical trials.
PATIENTS AND METHODS: This was a secondary analysis of participants from the Apple Heart Study, a prospective, siteless, single-arm pragmatic clinical trial from November 29, 2017, to January 31, 2019. Black, Hispanic, Asian, and White participants were monitored using an irregular rhythm notification algorithm designed to detect atrial fibrillation on a smartwatch. Purpose: To evaluate the effect of endodontic access hole preparation and choice of luting cement on the fracture resistance of translucent zirconia (5Y) crowns. Materials and Methods: Polymethylmethacrylate (PMMA) dies, representing a prepared maxillary molar tooth, were milled. Translucent zirconia crowns were milled from 5Y (Cercon XTML) zirconia discs and sintered. Forty crowns were divided into four groups (n = 10 per group) based on access hole preparation (accessed, non-accessed) and type of luting cement (resin cement, resin modified glass ionomer (RMGI) cement). Bacteriophages, or phages, are viruses that target and infect bacteria. Due to a worldwide rise in antimicrobial resistance (AMR), phages have been proposed as a promising alternative to antibiotics for the treatment of resistant bacterial infections. Up to this point in history, phage use in preclinical animal studies, clinical trials, and emergency-use compassionate care cases has centered around the original observation from 1915 showing phage as lytic agent, and thus a treatment that kills bacteria. The MacAB-TolC tripartite efflux pump is widely distributed in Gram-negative bacteria, extruding antibiotics and virulence factors that lead to multidrug resistance and pathogenicity. However, the in situ structure and assembly mechanism of MacAB-TolC remain unclear. Here, we resolve the in situ structures of the MacAB-TolC efflux pump in Escherichia coli by electron cryo-tomography and subtomogram averaging. In the cells without antibiotic treatment, we observe a fully assembled MacAB-TolC pump. When Escherichia coli cells are treated with erythromycin, in addition to the tripartite pumps, we also discover the emergence of MacA-TolC subcomplexes without MacB, indicating flexible binding of MacB in the …

Background: Morbidity and mortality of heart failure with preserved ejection fraction (HFpEF) is increased in metabolic disorders. However, options for preventing and treating these prevalent outcomes are limited. Intramyocardial lipotoxicity contributes to cardiac dysfunction. Here, we investigate the mechanisms underlying EDEM2 (endoplasmic reticulum degradation-enhancing alpha-mannosidase-like protein 2) regulation of cardiac lipid homeostasis and assess strategies that inhibit the incidence and progression of HFpEF.

Loss of function in the chromatin remodeler CHD7 causes CHARGE syndrome, characterized by variable penetrance and diverse abnormalities. However, establishing genotype-phenotype correlations has been challenging, as most CHD7 inactivating mutations are null alleles. Through CHD7 missense variant analysis at potential phosphorylation sites, we identified T730 (T720 in mice) as a critical residue associated with pathogenesis. Using a CHD7 T730 missense variant (Chd7T720A) and a frameshift null allele (Chd7fs) in a mouse model, we found that Chd7fs/fs mice were non-viable, while Chd7fs/+ mice exhibited haploinsufficiency-related circling behavior. The aim of this study was to describe psychological inflexibility for hearing difficulty for a sample of adults seeking hearing help and explore associations with demographic and audiological characteristics.Method:
Archival data from 226 clinical electronic medical records at one university clinic were analyzed for this retrospective exploratory study. Audiometric test findings and results from the Acceptance and Action Questionnaire–Adult Hearing Loss (AAQ-AHL) were extracted for adult patients seen between June 1, 2022, and February 29, 2024.Results:

This dissertation provides a comprehensive analysis of the interaction between urban heat and green infrastructure (GI) in terms of dynamic aspects, spatiotemporal variation across various spatial scales, temporal phases, and socioeconomic contexts. By integrating a national-scale longitudinal analysis, street-level structural assessments, and neighborhood-specific resilience frameworks, this study enhances our understanding of how cities can utilize existing ecological assets to mitigate increasing heat risks while addressing systemic inequities.

Mental health disorders impact approximately 14% of persons aged 65 and older and often are undertreated or underrecognized. Assessment of factors that link environmental, sociocultural, behavioral, and biological factors to overall health and disease is integral to guiding research on health disparities in older populations.
