BACKGROUND: Poorly healing wounds represent the primary health-related burden for hereditary recessive dystrophic epidermolysis bullosa (RDEB) patients. Contribution of wound-associated soluble constituents to wound progression remains not well defined.
OBJECTIVE: To conduct cross-sectional analysis of cytokine, chemokine, and growth factor in exudates from RDEB wounds and define changes associated with wound progression.
METHODS: Concentrations of selected cytokines, chemokines, and growth factors were evaluated by multiplex ELISA in eight blister fluids and 66 exudates from early, established, and chronic RDEB and five chronic venous ulcers (VU). A cross-sectional analysis was performed.

Aim: DNA methylation in brain regions represents a potential mechanism linking genetic variation to Alzheimer's disease (ad) risk, yet most studies have focused on blood-derived methylation markers. In this study, we conducted a systematic Mendelian randomization (MR) study to evaluate associations between predicted brain region-specific DNA methylation levels and ad risk, using methylation quantitative trait loci (mQTL) as genetic instruments.

Water quality assessments are critical for ensuring timely responses to water-related concerns, particularly in low-resource areas with limited water, sanitation, and hygiene (WASH) infrastructure. In collaboration with the Belize Ministry of Health and Wellness and the Ministry of Education, Culture, Science and Technology, we conducted a survey on WASH infrastructure and resources among 221 schools. We identified 65 schools across all six districts of Belize for water quality testing. Among these 65 schools, 83% had at least one water sample that did not meet the WHO's recommended free chlorine residual level for drinking water. Additionally, coliforms and Escherichia coli were …

Oxygen (O2) is vital for cellular development, function, proliferation, and repair, underscoring its critical role in organogenesis. Both hypoxia (reduced tissue O2) and hyperoxia (excess tissue O2), when prolonged, can trigger inflammation and oxidative stress, contributing to acute and long-term cardiopulmonary and neurodevelopmental morbidity. In sick neonates, immature defense mechanisms and coexisting morbidities demand nuanced and sometimes opposing strategies for O2 saturation targets and therapeutic titration. Most current neonatal O2 targeting guidelines are based on animal models or small clinical studies, resulting in recommendations with limited evidence. During the COVID-19 pandemic, African American adolescents experienced reduced access to needed resources and may have suffered poorer mental health outcomes as a result. The purpose of this quantitative study was to examine the association between race and the availability of mental health resources, food, medical health resources and mental health outcomes among African American adolescents in the United States during the COVID-19 pandemic compared to their White counterparts. Background: Methotrexate is a critical component of pediatric acute lymphoblastic leukemia (ALL) therapy that can result in neurotoxicity which has been associated with an increased risk of relapse. We leveraged machine learning to develop a neurotoxicity risk prediction model in a diverse cohort of children with ALL.
Methods: We included children (age 2-20 years) diagnosed with ALL (2005-2019) and treated in Texas without pre-existing neurologic disease. Clinical information was obtained by medical record review. Neurotoxicity occurring post-induction and prior to maintenance therapy was defined as neurologic episodes occurring within 21 days of methotrexate. Vascular endothelial growth factor receptors (VEGFRs) are key regulators of angiogenesis, lymphangiogenesis, and vascular permeability, playing essential roles in both physiological and pathological processes. The VEGFR family, including VEGFR-1, VEGFR-2, and VEGFR-3, interacts with structurally related VEGF ligands (VEGFA, VEGFB, VEGFC, VEGFD, and placental growth factor [PlGF]), activating downstream signaling pathways that mediate critical cellular processes, including proliferation, migration, and survival. To explore the potential of site-selectively radiolabeled antibody-drug conjugates (ADC) against solid tumors, we constructed and evaluated radiolabeled ADCs equipped with lutetium-177 (177Lu) and a membrane-permeable antimitotic agent. Site-selective 177Lu-labeled ADCs [anti-trophoblast cell-surface antigen 2 (TROP2) 177Lu-DTPA ADCs or anti-HER2 177Lu-DO3A ADCs], a 177Lu-labeled homogeneous radioimmunoconjugate (homogeneous RIC), and 177Lu-labeled conventional RIC (heterogeneous RIC) were constructed. We confirmed that 177Lu-labeled ADCs and the homogeneous RIC were obtained with high homogeneity and defined chelator/payload-to-antibody ratios. The REnal cancer consortium for Focused Investigation of Novel biomarkers and Expression (REFINE) consortium represents an important initiative in integrating clinical data with molecular sequencing in patients with advanced renal cell carcinoma (RCC) treated with immunotherapy-based approaches. By leveraging real-world evidence and genomic analysis, this consortium aims to explore putative predictive biomarkers with the potential to inform personalized treatment strategies. This qualitative phenomenological study explored how the intersection of identities related to race and gender shapes the lived experiences of Black female kinship caregivers raising youth aged 13–18. Drawing on Intersectionality Theory, the study sought to understand the caregiving challenges, emotional resilience, and systemic barriers encountered by this underrepresented population. Nine participants were recruited through purposive and snowball sampling and engaged in in-depth semi-structured interviews. Thematic analysis revealed that cultural norms and racialized gender expectations often compelled Black females to assume caregiving roles without institutional recognition or support. Previous studies have found that silver nanoparticle (SNP) addition into conventional PMMA resin improves fungal resistance without interfering with its flexural strength. Pesticides are ubiquitous in aquatic environments, alongside numerous contaminants and other stressors that can result in combined exposure to target and non-target organisms. Nevertheless, traditional toxicity studies quantify the effects of these compounds at an individual level, and to a narrow range of traditional test organisms. Through this dissertation, I explored the presence and effects of commonly detected pesticides of concern in the Pacific Northwest in order to realistically quantify potential effects from combined stressors. Transition metal-catalyzed arylation, using catalysts such as palladium, nickel, or copper, has revolutionized C-C, C-O, and C-N bond formation, offering broad substrate scope and high efficiency. Marker-assisted breeding can increase the response to selection; however, it requires the identification of marker associations with different traits. Animal genetic resources are recognized as an indispensable component of humanity’s natural heritage and are essential for ensuring environmental sustainability for future generations. The aim of this study was to investigate the genetic diversity, population structure, and presence of genetic bottlenecks in three indigenous Turkish sheep breeds: Dağlıç, Karagül, and Sakız, all of which are conserved as genetic resources in Türkiye. The sensitivity of ram semen to cryobiological procedures has motivated many researchers to investigate this issue. In clinical practice, one of the main objectives is to reduce the damage caused by freezing cells through the addition of preservatives to semen diluents. In our study, fetal calf serum (15% FCS) and 3–6 mM cysteamine were added to ram semen diluents, and their effects on semen after freeze-thawing were investigated. PARP inhibitors sensitize pancreatic ductal adenocarcinoma (PDAC) to radiation by inducing DNA damage and replication stress. These mechanisms also have the potential to enhance radiation-induced type I interferon (T1IFN) mediated anti-tumoral immune responses. We hypothesized that the PARP inhibitor olaparib would also potentiate radiation-induced T1IFN to promote anti-tumor immune responses and sensitization of otherwise resistant PDAC to immunotherapy. To test this hypothesis, we assessed the effects of olaparib and radiation on T1IFN production and sensitivity to αPD-L1 immunotherapy, as well as on the tumor microenvironment by single-cell RNA sequencing (scRNA-seq). Background: Obstructive sleep apnoea (OSA) is a common chronic condition, with obesity its strongest risk factor. Polygenic scores (PGSs) summarise the genetic liability to phenotype and can provide insights into relationships between phenotypes. Recently, large datasets that include genetic data and OSA status became available, providing an opportunity to utilise PGS approaches to study the genetic relationship between OSA and other phenotypes, while differentiating OSA-specific from obesity-specific genetic factors.

Triple-negative breast cancer (TNBC) and inflammatory breast cancer (IBC) are the most aggressive molecular subtypes of breast cancer. Poor clinical outcomes highlight the pressing need to discover novel targets for the effective treatment of these diseases. LMP7 (β5i/PSMB8), a proteolytic subunit of the immunoproteasome, is implicated in the pathogenesis of multiple myeloma, autoimmune and inflammatory diseases, and inflammation-related cancers. Immunosuppressive myeloid cells are critical obstacles to T cell-centered immune checkpoint blockade therapies, which have been successful in treating a fraction of patients with cancer. How tumor cells interact with myeloid cells to regulate immune responses and tumor development is unclear. In this study, we report that certain membrane tyrosine kinase Eph receptors, including EphA7 and EphB1, specifically bind the immune inhibitory receptors leukocyte Ig-like receptor family B 5 (LILRB5) and LILRB2. These Eph receptors induce LILRB5-mediated signaling activation, and LILRB5 also activates Eph receptor signaling. Recent research advances have resulted in an experimental benchtop X-ray fluorescence computed tomography (XFCT) system that likely meets the imaging dose/scan time constraints for benchtop XFCT imaging of live mice injected with gold nanoparticles (GNPs). For routine in vivo benchtop XFCT imaging, however, additional challenges, most notably the need for rapid/near-real-time handling of X-ray fluorescence (XRF) signal extraction and XFCT image reconstruction, must be successfully addressed. California's Imperial Valley, with lithium-rich geothermal brines, extensive flat agricultural fields with abundant desert sunshine, and access to transmission lines, is a leading case to explore interconnected themes around just energy transitions. Despite being the poorest county in California and one of the smallest, Imperial already provides 15% of the state’s solar electricity, and the region as a whole represents on the order of 25% of the state’s electric power capacity. Microvascular proliferation (MVP) is a disease-defining hallmark of glioblastoma and other World Health Organization grade 4 gliomas. MVP also serves as a poor prognostic marker in various solid tumors. Despite its clinical significance, the mechanisms and biological consequences of MVP are controversial and remain unclear. In this study, we performed single-cell RNA sequencing on paired CD45-CD105+ vascular/perivascular stromal cells (PVSC) and CD45+CD105± immune cells from 16 primary glioma patient samples, both with and without MVP. This analysis revealed the presence of developmentally related mesenchymal stem cells alongside cancer-associated fibroblasts, pericytes, fibromyocytes, and smooth muscle cells within the CD45-CD105+ compartment. Current prevalence rates of autism spectrum disorder (ASD) in transgender and gender diverse (TGD) populations range between 4 and 13% (Thrower et al., 2020). Studies into autistic TGD individuals’ experiences with healthcare have primarily collected qualitative data, identifying themes including barriers to healthcare, invalidation of both autistic and TGD identities, difficulty finding competent providers, limited support and resources, and refusal of care (Cooper et al., 2022; Maroney & Horne, 2022; Hall et al., 2020). Black mold rot of tomato (Solanum lycopersicum Mill) fruits, incited by Alternaria alternata, is a major constraint in tomato production worldwide. The fungus invades unripe fruits in the field, remains dormant and then cause symptoms as the fruits begin to ripe. In this study, the effectiveness of the antagonistic yeast Meyerozyma guilliermondii SQUCC-33Y and Tamarindus indica leaf extract was assessed for controlling post-harvest black mold fruit rot of tomato. Greenhouse and field tests revealed that pre-harvest application of M. guilliermondii at weekly intervals starting from fruit set to harvest stage significantly reduced the development of post-harvest fruit rot when challenge-inoculated …

Sudden decline syndrome (SDS) poses a significant threat to the date palm (Phoenix dactylifera), a crop of critical agricultural and cultural importance in arid regions.
