Investigating The Multiple Enzyme Complex In The Carotenoid Biosynthesis Pathway, Mercy Mormor Aug 2025

Investigating The Multiple Enzyme Complex In The Carotenoid Biosynthesis Pathway, Mercy Mormor

Electronic Theses and Dissertations

In nature, plant carotenoids are 40-carbon isoprenoids with polyene chains that may have double bonds. They are vital components of all photosynthetic organisms, playing several roles in the plant cell, including photosynthesis, pigmentation, phytohormone synthesis, signaling and photoprotection. In this study, we investigated protein-protein interactions among key enzymes involved in the carotenoid biosynthesis pathway using the Bimolecular Fluorescent Complementation (BiFC) assay. Our results revealed several interactions that suggest a coordinated regulation of enzymatic activity within this pathway, highlighting potential regulatory complexes that influence carotenoid production. The study also highlights the pivotal role of Zeta-Carotene Isomerase (Z-ISO) as a key regulatory …

Gut Passage Time In Small Mammals: Influences Of Small Mammal Traits, Digesta Phase, And Particle Morphology, Noah G. Hale Aug 2025

Gut Passage Time In Small Mammals: Influences Of Small Mammal Traits, Digesta Phase, And Particle Morphology, Noah G. Hale

Electronic Theses and Dissertations

Dispersal is crucial in maintaining healthy ecosystems, but little is known about mechanisms influencing fungal spore dispersal by animals. We aimed to develop a more mechanistic understanding of the influence of fungal spore traits and small mammal traits on gut retention time. We fed three species of small mammals (shrew, mouse, vole), which differ in gut morphologies, digestive strategies, and activity a combination of markers including: liquid digesta phase (CrEDTA), small particle (fungal spores with diverse morphologies), and large particles (glitter and beads, representing plant cells and seeds, respectively). We found that hindgut fermenters selectively retained the liquid markers, spores …

Cattle Regenerative Grazing Effects On Bat Activity In Southern Appalachia, Seth Morelock Aug 2025

Cattle Regenerative Grazing Effects On Bat Activity In Southern Appalachia, Seth Morelock

Electronic Theses and Dissertations

Livestock farming accounts for about 70% of agricultural activity, with continuous grazing as the dominant conventional practice. However, this method often degrades biodiversity and ecosystem health through overgrazing. Regenerative grazing practices offer a potential ecologically sustainable approach to cattle management. Insectivorous bats are known to use agroecosystems for foraging and roosting habitats, offering an opportunity to assess their response to regenerative grazing. Using passive acoustic monitors, we evaluated the response of bats to cattle within regenerative grazed farms in East Tennessee, USA. Overall bat activity declined when cattle entered and exited paddocks, except for the tri-colored bat (Perimyotis subflavus …

Graduate Theses and Dissertations

Having a scientifically literate citizenry is an asset nationally and globally, and knowledge of the nature of science (NOS) is a crucial element in such literacy. Science teachers are a vital link if students are to understand key elements of the NOS, yet teachers themselves often lack knowledge of NOS and how to integrate NOS content into science lessons. Investigating the impact of a series of inservice professional development experiences using three distinct delivery modes (face-to-face, hybrid, online) for teachers in my home country of Bhutan is the overarching goal of the study reported here. The study used a mixed-methods …

Critical Communication Pedagogy At The End Of The World, Tyler S. Rife Aug 2025

Critical Communication Pedagogy At The End Of The World, Tyler S. Rife

Journal of Communication Pedagogy

This essay draws upon my experiences teaching the undergraduate course, “Communication at the End of the World,” to argue for an approach to critical communication pedagogy (CCP) contextualized by and reflexively responsive to planetary-scale ecological crises. I outline how five commitments have come to guide my classroom’s explorations through our topic of “end-times” communication: (1) Anthropocentrism and speciesism participate in the maintenance of social hierarchy; (2) environmental justice and social justice are intimately entangled projects; (3) ecological crisis is (re)made concrete, embodied, proximal, and implicative; (4) critique of language involves critique of humanism and humanist historicization; and, (5) ecologically just …

Comparative Phylogeography Of Three Widely Distributed Amazonian Poison Frogs, Sofia Del Milagro Granados Martinez Aug 2025

Comparative Phylogeography Of Three Widely Distributed Amazonian Poison Frogs, Sofia Del Milagro Granados Martinez

Theses

Amazonia has gone through extensive changes in its climates, geology, hydrology and drainage systems over the past 10 mya; these processes have been postulated as key drivers of diversification. Contrary to the trend where species once broadly distributed are subdivided into several species with restricted distributions, many taxa span the Amazon basin without noticeable genetic differentiation. In my thesis, I studied the phylogenetic and phylogeographic patterns of three widely distributed Amazonian poison frogs: Ameerega hahneli, Ameerega trivittata and Ranitomeya variabilis. I examined phylogenetic relationships based on two different types of genomic data—nuclear and mitochondrial—and I carried out population structure analyses. …

Center for Translational Medicine Faculty Papers

Endoplasmic reticulum (ER)-mitochondrial (ER-Mito) interface, termed mitochondrial-ER contacts (MERCs), plays significant roles in the maintenance of bioenergetics and basal cell functions via the exchange of lipids, Ca 2 þ , and reactive oxygen species (ROS) in various cell types/tissues. Genetic deletion of mitofusin 2 (Mfn2), one of the key components of ER-Mito tethering, in cardiomy- ocytes (CMs) in vivo revealed the importance of the microdomains between mitochondria and sarcoplasmic reticulum (SR), a dif- ferentiated form of ER in muscle cells, for maintaining normal mitochondrial Ca 2 þ (mtCa 2 þ ) handling and bioenergetics in the adult heart. However, key …

Biomedical Sciences

The chemical composition of wood plays a pivotal role in the adaptability and structural integrity of trees. However, few studies have investigated the environmental factors that determine lignin composition and its biological significance in plants. Here, we examined the lignin syringyl-to-guaiacyl (S/G) ratio in members of a Populus trichocarpa population sourced from their native habitat and conducted a genome wide association study to identify genes linked to lignin formation. Our results revealed many significant associations, suggesting that lignin biosynthesis is a complex polygenic trait. Additionally, we found an increase in the S/G ratio from northern to southern geographic origin of …

Differential Inhibition Of Tumor Oxidative Metabolism While Sparing T Cells Through A Novel Genetic Approach, Jean Schneider, Michael Curran Phd, Priyamvada Jayaprakash Phd, Krithikaa Rajkumar Bhanu

Students Summer in Oncology at Anderson Research (SOAR) 2025

In pursuit of identifying new potential targets for immunotherapy, there has been renewed focus on Oxidative Phosphorylation (OxPhos) metabolism that cancers rely on for growth and, to some degree, immune suppression. Previous research has shown selective deletion of the mitochondrial Complex I (CI) subunits Ndufs4 and Ndufs6 increases the efficacy of immunotherapy through induction of genes related to MHC class-I expression leading to improved tumor antigen presentation. Based on these studies, we hypothesize that selectively targeting CI subunits differentially expressed in tumor cells versus immune effector cells will significantly hinder tumor cell viability and growth without compromising anti-tumor immunity.

Sccosmix: A Mixed-Effects Framework For Differential Coexpression And Transcriptional Interactions Modeling In Single-Cell Rna-Seq, Anderson Bussing, Giampiero Marra, Daping Fan, Russell Shinohara, Danni Tu, Yen-Yi Ho

Faculty Publications

Advancements in single-cell RNA-sequencing (scRNA-seq) technologies generate a wealth of gene expression data that provide exciting opportunities for studying gene-gene interactions systematically at individual cell resolution. Genetic interactions within a cell are tightly regulated and often highly dynamic in response to internal cellular signals and external stimuli. Evidence of these dynamic interactions can often be observed in scRNA-seq data by examining conditional co-expression changes. Existing approaches for studying these dynamic interaction changes in scRNA-seq data do not address the multi-subject hierarchical design commonly considered in single-cell experiments. In this paper, we propose a Mixed-effects framework for differential Coexpression and transcriptional …

Master's Theses

Pierce’s disease (PD) and black rot disease cause severe economic losses for grape production under humid and warm conditions. Identifying resistance genes is crucial for controlling these disease by developing resistant cultivars. Muscadine grape (Vitis rotundifolia) cultivar ‘Pam’ has been identified to possess a high level of resistance to diseases. To elucidate the genetic basis of the resistance to the causal pathogens Xylella fastidiosa and Guignardia bidwellii in this cultivar, parents and 141 F1 individuals developed from open pollinated ‘Pam’ were evaluated for two seasons under natural disease pressure. The mapping population was genotyped using genotyping-by-sequencing (GBS) and …

Projecting Echoes: Valeria Luiselli's Writing On Immigration, Jacob C. Cisneros

Electronic Theses, Projects, and Dissertations

The growing concerns of immigration has been a fraught topic in both U.S. society and the American political landscape. Certain political narratives are formed which fuel the debate for and against immigration. This coincides with how immigrants are treated and how legal processes change throughout each term of presidency within the United States. This has sparked outrage from both advocates and adversaries of immigrants coming into the United States. Papers, speeches, news articles, and more have been created from both sides of the argument concerning how to handle the immigration crisis along with the question of how immigrants and asylum …

Medical Surveillance In Custodial Jsas: Preventing Bloodborne Pathogen Exposure, Kiara Gadea Aug 2025

Medical Surveillance In Custodial Jsas: Preventing Bloodborne Pathogen Exposure, Kiara Gadea

Electronic Theses, Projects, and Dissertations

Custodians play an important role in maintaining our surroundings safe and sanitary. However, in doing so, they are their exposure to health hazards that put them at risk, yet these exposures are often overlooked. One major risk they face is contact with bloodborne pathogens, which put custodial staff at risk for infections such as hepatitis B. While Job Safety Analyses (JSA’s) outline various protective measures, many fail to incorporate medical surveillance as part of their risk management program. Overlooking this important detail leaves custodians vulnerable to health risks that can be prevented.

Adding a medical surveillance section to custodial JSA’s, …

Improved Autofluorescence Lifetime Decay Analysis For Identification Of Cellular Autofluorescence Biomarkers For Label-Free, Single-Cell Assessment Of Mitochondrial Membrane Potential, Olivia Isabel Kolenc

Graduate Theses and Dissertations

Optical imaging of metabolism using the autofluorescence of the metabolic cofactors NADH, NADPH, and FAD has a wide range of sensitivities and the capability to collect a great deal of metabolic information. Multiphoton microscopy (MPM) is a non-invasive, label-free optical imaging technique capable of performing fluorescence lifetime imaging (FLIM) using time correlated single photon counting (TCSPC). However, autofluorescence FLIM presents a challenge in collecting sufficient photons to provide adequate signal to noise ratios (SNR) for analysis. Additionally, the specificity of autofluorescence biomarkers of metabolism has not been well-defined. Contextualizing these autofluorescence measures using established functional biomarkers can improve interpretability of …
