Consumption Of Negative Media Content Mediates The Association Between Levels Of Trauma Exposure And Insomnia Severity In Hispanic Young Adults, Michiyo Hirai, Laura L. Vernon

Research has shown that individuals who have experienced traumatic events are at greater risk of developing severe insomnia than those without trauma. Further, according to the stress sensitization hypothesis, victims of multiple traumas, compared to those who have experienced a single trauma, are particularly vulnerable to severe insomnia. One potential mechanism underlying the association between trauma experiences and insomnia is media exposure. Although avoidance behaviors are symptoms of posttraumatic stress, victims of traumas also demonstrate attentional bias toward threats and difficulty disengaging attention from threats. These cognitive tendencies may result in heightened vigilance to negative media content, particularly trauma-related content, …

Botulinum Neurotoxin Light Chain/A1 Uses Fast Synaptic Vesicle Cycling To Cleave Plasma Membrane Bound Snap-25, Alexander Gardner, Molly Sneller, William H. Tepp, Joseph T. Barbieri, Sabine Pellett

Botulinum neurotoxins (BoNT) are the most potent protein toxins for humans, yet how BoNT-Light Chain/A1 (LC/A1) journeys to cleave intracellular SNAP-25 is understudied. Here we use a cell-based assay to measure cytosolic EGFP-LC/A1 intracellular trafficking and SNAP-25 cleavage in Neuro-2A cells. Intracellular LC/A1 associated on microtubules and co-localized with Rab GTPases involved in fast synaptic vesicles and endosome recycling. Multiple Dominant Negative (DN) Rabs GTPases involved in fast synaptic vesicles or endosome recycling inhibited LC/A1 trafficking to the intracellular plasma membrane and SNAP-25 cleavage. Over 95% of ependymomas that arise in the cortex are driven by a gene fusion involving the zinc finger translocation-associated (ZFTA) protein. Here, using super-resolution and lattice light-sheet microscopy, we demonstrate that the most frequent fusion variant, ZFTA-RELA (ZR), forms dynamic nuclear condensates that are required for oncogene expression and tumorigenesis. Mutagenesis studies of ZR reveal a key intrinsically disordered region (IDR) in RELA that governs condensate formation. Condensate-modulating IDR mutations introduced into ZR impaired its genomic occupancy at oncogenic loci and inhibited the recruitment of transcriptional effector proteins, such as MED1, BRD4 and RNA polymerase II. Understanding the complex factors influencing mammalian metabolism and body weight homeostasis is a long-standing challenge requiring knowledge of energy intake, absorption and expenditure. Using measurements of respiratory gas exchange, indirect calorimetry can provide non-invasive estimates of whole-body energy expenditure. However, inconsistent measurement units and flawed data normalization methods have slowed progress in this field. This guide aims to establish consensus standards to unify indirect calorimetry experiments and their analysis for more consistent, meaningful and reproducible results. By establishing community-driven standards, we hope to facilitate data comparison across research datasets. This advance will allow the creation of an in-depth, machine-readable data …

Aims: There is a paucity of information on the pharmacokinetics (PK) and pharmacodynamics (PD) of menthol, the active ingredient in peppermint oil (PMO). We studied the PK of menthol (540 and 900 mg) in children and measured their effects on gut transit/contractility.
Methods: Children 7-12 years of age with functional abdominal pain underwent wireless motility capsule (WMC) testing. One week later, they were randomized to 540 mg or 900 mg of oral enteric-coated PMO. Blood was sampled over 24 h to determine menthol PK. Peanut allergies pose significant health risks globally, with an increasing prevalence observed over the past two decades. Accidental exposures to peanuts can lead to severe allergic reactions, including anaphylaxis, prompting the exploration of innovative therapeutic approaches to mitigate these risks. This literature review examines the potential use of peanut proteins embedded within toothpaste formulations as a novel avenue for inducing tolerance in individuals with peanut allergies. It includes recent studies and clinical trials in oral immunotherapy, focusing on mechanisms of action, safety profiles and long-term outcomes associated with controlled allergen exposure. Gorlin-Goltz syndrome is a hereditary autosomal dominant disorder. It has multiple clinical manifestations caused by mutation in the patched gene (PTCH), which is responsible for the growth and development of healthy tissue and for regulating the cell cycle. Early diagnosis of the condition and an extended duration of follow-up are essential to prevent complications, such as basal cell carcinomas and facial deformities, leading to a more favorable prognosis. The prevalence of physical aggression among women has increased, challenging traditional gender norms that portray women as nurturing and nonviolent. While most research on aggression has focused on men, children, or intimate partner violence, research is sparse concerning female aggression outside familial contexts. In this qualitative multiple-case study, the contextual antecedents of nonfamilial aggression in women arrested for aggressive behavior were explored using the general aggression model as the theoretical framework. Data were obtained from 20 publicly documented cases using archival sources such as media stories, court documents, and video recordings. Objective: Hyperglycemic states (prediabetes and diabetes) are associated with heart failure (HF) risk. We aimed to identify distinct metabolites for subclinical cardiac dysfunction, a precursor of HF, in hyperglycemic or euglycemic individuals.
Research design and methods: We conducted cross-sectional and prospective analyses of 2,492 HF-free participants from the Atherosclerosis Risk in Communities (ARIC) study visit 5, 2011-2013. A total of 1,297 participants were hyperglycemic (assessed on the basis of hemoglobin A1c >5.7%, fasting glucose >100 mg/dL, use of diabetes medication, or diagnosis), and 1,195 were euglycemic.
