Consumption Of Negative Media Content Mediates The Association Between Levels Of Trauma Exposure And Insomnia Severity In Hispanic Young Adults, Michiyo Hirai, Laura L. Vernon

Research has shown that individuals who have experienced traumatic events are at greater risk of developing severe insomnia than those without trauma. Further, according to the stress sensitization hypothesis, victims of multiple traumas, compared to those who have experienced a single trauma, are particularly vulnerable to severe insomnia. One potential mechanism underlying the association between trauma experiences and insomnia is media exposure. Although avoidance behaviors are symptoms of posttraumatic stress, victims of traumas also demonstrate attentional bias toward threats and difficulty disengaging attention from threats. Botulinum neurotoxins (BoNT) are the most potent protein toxins for humans, yet how BoNT-Light Chain/A1 (LC/A1) journeys to cleave intracellular SNAP-25 is understudied. Here we use a cell-based assay to measure cytosolic EGFP-LC/A1 intracellular trafficking and SNAP-25 cleavage in Neuro-2A cells. Intracellular LC/A1 associated on microtubules and co-localized with Rab GTPases involved in fast synaptic vesicles and endosome recycling. Multiple Dominant Negative (DN) Rabs GTPases involved in fast synaptic vesicles or endosome recycling inhibited LC/A1 trafficking to the intracellular plasma membrane and SNAP-25 cleavage. Over 95% of ependymomas that arise in the cortex are driven by a gene fusion involving the zinc finger translocation-associated (ZFTA) protein. Here, using super-resolution and lattice light-sheet microscopy, we demonstrate that the most frequent fusion variant, ZFTA-RELA (ZR), forms dynamic nuclear condensates that are required for oncogene expression and tumorigenesis. Mutagenesis studies of ZR reveal a key intrinsically disordered region (IDR) in RELA that governs condensate formation. Condensate-modulating IDR mutations introduced into ZR impaired its genomic occupancy at oncogenic loci and inhibited the recruitment of transcriptional effector proteins, such as MED1, BRD4 and RNA polymerase II.
