Reduced Toxicity Of Centruroides Vittatus (Say, 1821) May Result From Lowered Sodium Β Toxin Gene Expression And Toxin Protein Production, Aimee Bowman, Chloe Fitzgerald, Jeff F. Pummill, Douglas D. Rhoads, Tsunemi Yamashita Nov 2021

Reduced Toxicity Of Centruroides Vittatus (Say, 1821) May Result From Lowered Sodium Β Toxin Gene Expression And Toxin Protein Production, Aimee Bowman, Chloe Fitzgerald, Jeff F. Pummill, Douglas D. Rhoads, Tsunemi Yamashita

Faculty Publications - Biological Sciences

Body tissue and venom glands from an eastern population of the scorpion Centruroides vittatus (Say, 1821) were homogenized and molecular constituents removed to characterize putative sodium β toxin gene diversity, RT-qPCR, transcriptomic, and proteomic variation. We cloned sodium β toxins from genomic DNA, conducted RT-qPCR experiments with seven sodium β toxin variants, performed venom gland tissue RNA-seq, and isolated venom proteins for mass spectrophotometry. We identified greater than 70 putative novel sodium β toxin genes, 111 toxin gene transcripts, 24 different toxin proteins, and quantified sodium β toxin gene expression variation among individuals and between sexes. Our analyses contribute to …

Cytokine Exposure Mediates Transcriptional Activation Of The Orphan Nuclear Receptor Nur77 In Hematopoietic Cells, Orsola Di Martino, Haixia Niu, Gayla Hadwiger, Margaret A Ferris, John S Welch Nov 2021

Cytokine Exposure Mediates Transcriptional Activation Of The Orphan Nuclear Receptor Nur77 In Hematopoietic Cells, Orsola Di Martino, Haixia Niu, Gayla Hadwiger, Margaret A Ferris, John S Welch

Open Access Publications

The orphan nuclear receptor Nur77 is an immediate-early response gene that based on tissue and cell context is implicated in a plethora of cellular processes, including proliferation, differentiation, apoptosis, metabolism, and inflammation. Nur77 has a ligand-binding pocket that is obstructed by hydrophobic side groups. Naturally occurring, cell-endogenous ligands have not been identified, and Nur77 transcriptional activity is thought to be regulated through posttranslational modification and modulation of protein levels. To determine whether Nur77 is transcriptionally active in hematopoietic cells in vivo, we used an upstream activating sequence (UAS)-GFP transgenic reporter. We found that Nur77 is transcriptionally inactive in vivo in …

Recent Advances In Non-Enzymatic Electrochemical Detection Of Hydrophobic Metabolites In Biofluids, Zahra Panahi, Luciana Custer, Jeffrey M. Halpern

New Hampshire EPSCoR

This review focuses on recent advances in non-enzymatic electrochemical biosensors for detection of hydrophobic metabolites. Electrochemical approaches have been widely applied in many established and emerging technologies and a large range of electrochemical biosensors have been used for detection of various hydrophobic metabolites. Despite the progress made in this field, some problems still exist, specifically, electrochemical detection of hydrophobic biomarkers can be challenging in complex biological fluids. In this review, we have highlighted some of the most representative surface modification technologies that have been employed in electrochemical biosensors to counter the problems of poor sensitivity and selectivity towards hydrophobic metabolites. …

Plant Extract Of Limonium Gmelinii Attenuates Oxidative Responses In Neurons, Astrocytes, And Cerebral Endothelial Cells In Vitro And Improves Motor Functions Of Rats After Middle Cerebral Artery Occlusion, Tulendy Nurkenov, Andrey Tsoy, Farkhad Olzhayev, Elvira Abzhanova Nov 2021

Plant Extract Of Limonium Gmelinii Attenuates Oxidative Responses In Neurons, Astrocytes, And Cerebral Endothelial Cells In Vitro And Improves Motor Functions Of Rats After Middle Cerebral Artery Occlusion, Tulendy Nurkenov, Andrey Tsoy, Farkhad Olzhayev, Elvira Abzhanova

Faculty and Student Publications

There are numerous publications demonstrating that plant polyphenols can reduce oxi-dative stress and inflammatory processes in the brain. In the present study we have investigated the neuroprotective effect of plant extract isolated from the roots of L. gmelinii since it contains a rich source of polyphenols and other biologically active compounds. We have applied an oxidative and inflammatory model induced by NMDA, H2O2, and TNF‐α in human primary neurons and astrocytes, and mouse cerebral endothelial cell (CECs) line in vitro. The levels of ROS generation, NADPH oxidase activation, P‐selectin expression, and activity of ERK1/2 were evaluated by quan-titative immunofluorescence analysis, …

Economic Contribution Of Agriculture And Food To Arkansas’ Gross Domestic Product 1997-2020, Leah English, Jennie Popp, Wayne Miller Nov 2021

Economic Contribution Of Agriculture And Food To Arkansas’ Gross Domestic Product 1997-2020, Leah English, Jennie Popp, Wayne Miller

AAES Research Reports and Research Bulletins

Agricultural production, processing, and retail industries are major contributors to Arkansas’ GDP. Agriculture contributes to the state economy through direct agricultural production, value-added processing, and agricultural retail activities. The Agriculture and Food Sector, which is comprised of agricultural production, processing, and retail industries, promotes economic strength through various interactions with other industries. The use of non-agricultural goods and services as inputs into the agricultural sector promotes diversified growth in Arkansas’ economy and thus plays a vital role in maintaining economic stability throughout the state. This report 1) compares the relative size of the Agriculture and Food Sector in Arkansas with …

An Analysis Of Companion Animal Tick Encounters As Revealed By Photograph-Based Crowdsourced Data, Heather L. Kopsco, Roland J. Duhaime, Thomas N. Mather

Plant Sciences and Entomology Faculty Publications

Background: Community science is increasingly utilized to track important vectors of companion animal disease, providing a scalable, cost-effective strategy for identifying new foci, changing phenology, and disease prevalence across wide geographies. Objectives: We examined photographs of ticks found attached to predominately dogs and cats reported to a photograph-based tick surveillance program to identify potential areas for improvements in tick prevention education and risk intervention. Methods: We compared estimated days of tick attachment using a Kruskal–Wallis one-way analysis of variance, and a Pearson's chi-square analysis of variance on the number of submissions by host type submitted for each season. Results: The …

It is challenging to associate features such as human health outcomes, diet, environmental conditions, or other metadata to microbial community measurements, due in part to their quantitative properties. Microbiome multi-omics are typically noisy, sparse (zero-inflated), high-dimensional, extremely non-normal, and often in the form of count or compositional measurements. Here we introduce an optimized combination of novel and established methodology to assess multivariable association of microbial community features with complex metadata in population-scale observational studies. Our approach, MaAsLin 2 (Microbiome Multivariable Associations with Linear Models), uses generalized linear and mixed models to accommodate a wide variety of modern epidemiological studies, including …
