Chulalongkorn University Theses and Dissertations (Chula ETD)
Hepatocellular carcinoma (HCC) is the most common type of primary liver cancer. HCC has a replicative immortality and sustained proliferation rate. In addition, cell cycle-related protein regulating proliferation in cancer are often found dysregulated, allowing cancer cells to proceed their proliferation uncontrollably. Recently, a small non-coding RNA, microRNA (miRNA), was found to play an important role in numerous biological functions. Specific miRNA may ameliorate or promote cancer progression through different target mRNA. MiR-372-3p has been explored in various cancers such as colon cancer, colorectal cancer, and glioma. However, its functions have been rarely studied in HCC, especially in the aspect …

Effects Of Chitosan Oligosaccharide And Probiotics On Chronic Kidney Disease Rats, Weerapat Anegkamol
Chulalongkorn University Theses and Dissertations (Chula ETD)
Chronic kidney disease (CKD) patients suffer from the accumulation of toxic substances in their blood due to the loss of kidney function, which results in hyperphosphatemia. This condition contributes to hyperparathyroidism, leading to the development of chronic kidney disease-related mineral bone disorder (CKD-MBD). Additionally, CKD patients experience changes in their gut microbiota, disrupting epithelial tight junctions and allowing excessive absorption of dietary phosphate. In this study, we aimed to investigate the effects of various oligosaccharides and probiotics on the gut microbiota, intestinal barrier, hyperphosphatemia, and hyperparathyroidism in CKD rats. We isolated Lactobacillus and Bifidobacterium strains from healthy participants and tested …

Study Of Thermodynamic Binding Between Ader And Ades In Two-Component Antibiotic Sensor Of Antibiotic Resistant Strains Of Acinetobacter Baumannii, Konrawee Thananon
Chulalongkorn University Theses and Dissertations (Chula ETD)
The proteins AdeR (response regulator) and AdeS (histidine kinase sensor) play a crucial role in regulating the efflux pump, which is responsible for removing antibiotics from bacterial cells. This regulation occurs through a two-component regulatory system (TCS) involving autophosphorylation. Upon receiving external signals, AdeS undergoes autophosphorylation, leading to the activation of AdeR, which in turn initiates the expression of target genes related to the efflux pump. In this study, both enzymes from the reference strain of Acinetobacter baumannii (ATCC19606) and antibiotic resistant strains (H1074 and G560) were cloned and expressed in E. coli. The enzymes were purified using an immobilized …

Chulalongkorn University Theses and Dissertations (Chula ETD)
B-mode ultrasound imaging is the standard method for hepatocellular carcinoma (HCC) screening. If a lesion has a unique appearance, a conventional detector can be applied. However, if the lesion shares its appearance with other lesion, a large training dataset is required, which may be unavailable. Therefore, a two-stage method is proposed. In the first stage, lesions are detected but not differentiated into any particular class. The detected lesions are then classified in the second stage using a conventional convolutional neural network (CNN). The aims of the dissertation are to design an artificial intelligent system model and to investigate the most …

We synthesized a new derivative of renieramycin T (RT), named DH_22, and tested it for anti-cancer activities in human lung cancer cells. In the cell viability and nuclear staining tests, DH_22 was discovered to be cytotoxic, with an IC50 of around 13 μM; and found to mediate apoptosis in the lung cancer cells. Meanwhile, in a mechanistic sense, DH_22 contributed to the activation of p53-dependent apoptosis and decreased the cellular level of c-Myc. The p53-dependent mechanism was indicated by an increase of p53, an induction of …

Development Of Surrogate Sars-Cov-2 Virus Neutralization Test Method Using Plant-Produced Recombinant Proteins, Perawat Jirarojwattana
Chulalongkorn University Theses and Dissertations (Chula ETD)
Detecting immunity against SARS-CoV-2 is vital for evaluating vaccine response and natural infection, but a conventional virus neutralization test (cVNT) requires BSL3 and live viruses, and a pseudo-virus neutralization test (pVNT) needs specialized equipment and trained professionals in BSL2. The surrogate virus neutralization test (sVNT) was developed to overcome these limitations. This study explored the use of angiotensin converting enzyme (ACE2) produced from Nicotiana benthamiana for the development of an affordable neutralizing antibodies detection assay. The results showed that the plant-produced ACE2-His can bind to the receptor binding domain (RBD) of the SARS-CoV-2, and was used to develop sVNT with …

Since the emergence of the coronavirus pandemic in 2019 (COVID-19), the development of effective vaccines to combat the infection has accelerated worldwide. While the severe acute respiratory syndrome-coronavirus 2 (SARS-CoV-2) with mutations in the receptor-binding domain (RBD) with high transmissibility, enhanced infectivity, and immune escape from vaccination is also predominantly emerging. Effective vaccines against variant of concern (VOC) and optimized booster vaccination strategies are thus highly required. Here, the gene encoding seven different RBD (ancestral (Wuhan), Alpha, Beta, Gamma, Kappa, Delta, and Epsilon variants) fused with the fragment crystallizable region (Fc) of human IgG1 (RBD-Fc) was constructed and cloned into …
Predictors Of Fertility Quality Of Life In Infertile Patients Visiting Infertility Center In Kathmandu, Nepal : A Cross-Sectional Study., Shital Shakya
Chulalongkorn University Theses and Dissertations (Chula ETD)
In Nepal, infertility is a rising reproductive health issue with an estimated prevalence of 15%. There are few studies on quality of life in infertile women, but none on quality of life in infertile men and women although infertility is a shared condition and has effects on couples. The study objective is to describe the various predictors of infertile patients seeking infertility treatment and find association between these predictors and the quality of life. A cross-sectional study using a self-administered disease specific FertiQoL questionnaire was conducted among 409 infertile men and women seeking infertility treatment in an infertility center in …
