Enhanced Triacylglycerol Production In Chlorella Vulgaris By Flux Balance Analysis On Metabolic Network, Issalapap Kaothong
Chulalongkorn University Theses and Dissertations (Chula ETD)
Triacylglycerol (TAG) is a type of lipid that can be produced by a microalga called Chlorella vulgaris, which is a promising source of biodiesel. However, there is a lack of research and models explaining how to directly produce this lipid. To address this, we propose using linear minimization of metabolic adjustment (LMOMA), which predicts gene manipulation effects based on Flux Balance Analysis (FBA), to enhance TAG production. We conducted single-gene deletion experiments on the genome-scale metabolic model of C. vulgaris UTEX 395 (iCZ843) under different growth conditions: heterotrophy, photoautotrophy, and mixotrophy conditions. We found that the TAG production increased by …

Comparative Analysis Of Mutational Landscapes Identified From Exome Sequencing Of Tumor Tissues And Circulating Tumor Dna, Julanee Leenanitikul
Chulalongkorn University Theses and Dissertations (Chula ETD)
Next-generation sequencing of circulating tumor DNA (ctDNA) has been used as a noninvasive alternative for cancer diagnosis and characterization of tumor mutational landscape. However, low ctDNA fraction and other factors can limit the ability of ctDNA analysis to capture tumor-specific and actionable variants. In this study, whole-exome sequencings (WES) were performed on paired ctDNA and tumor biopsy in 15 cancer patients to assess the extent of concordance between mutational profiles derived from the two source materials. We found that up to 16.4% ctDNA fraction can still be insufficient for detecting tumor-specific variants and that high concordance with tumor biopsy is …

Chulalongkorn University Theses and Dissertations (Chula ETD)
Brassinosteroid (BR) plays a complex role in root growth and development by regulating the expression of downstream target genes through the BR signaling pathway. In this scRNA-seq study of Arabidopsis roots, tissue- and developmental-specific roles of BR were identified. Atrichoblast, an epidermal cell that develops into a non-hair cell, displayed the highest enrichment of BR-induced genes in the elongation and maturation zones among all cell types. Trajectory analysis revealed distinct expression patterns of BR biosynthesis and signaling genes between atrichoblasts and trichoblasts. Atrichoblasts exhibited higher expression levels of BR biosynthesis genes (DET2 and ROT3) and BR signaling genes (BRI1 and …

The T Cell Senescence In Breast Cancer, Sikrit Denariyakoon
The T Cell Senescence In Breast Cancer, Sikrit Denariyakoon
Chulalongkorn University Theses and Dissertations (Chula ETD)
Background: The cancer-immune cell crosstalk is a common immune-evasion mechanism of cancer. This crosstalk can result in epigenetic changes in immune phenotypes to accelerate tumor growth and metastasis. Senescent T lymphocytes are formed during T cell differentiation, and are recently described as one of the dysfunctional T cells in cancer. In breast cancer, estrogen does not only regulate normal physiology in females, but is also involved in cancer development and cancer treatment. It may also attenuate senescent phenotypes, and increase DNA damage in several cell-lines. This thesis therefore examined for the presence of senescent T lymphocytes in breast cancer patients, …

Development Of High Protein Beverage From Hydrolyzed Egg White, Nuntorn Charoensak
Chulalongkorn University Theses and Dissertations (Chula ETD)
The primary objective of this study was to examine the impact of enzymatic treatment on pasteurized egg white protein (EWP) and assess its influence on various physicochemical properties and antioxidant activity. Our findings revealed that the addition of SDAY10 enzymes led to a significant increase in lightness (L*), redness (a*), and yellowness (b*) parameters. Furthermore, higher concentrations of the enzyme resulted in a considerable reduction in pH, viscosity, and volume mean diameter (D4,3) of EWP protein compared to the control group (P < 0.05). Additionally, the application of SDAY10 enzyme (0.1-0.3% w/v) for EWP hydrolysis exhibited a noteworthy enhancement in antioxidant activity (measured by DPPH, ABTS, and FRAP assays) compared to lower enzyme concentrations. Notably, the 0.3% concentration of SDAY10 exhibited the highest release of amino acids during in vitro digestion and received the highest sensory evaluation score. Consequently, employing SDAY10 enzymes at a concentration of 0.3% w/v for EWP hydrolysis demonstrates significant potential for improving the physical properties of EWP and enhancing antioxidant activity. However, it is imperative to investigate the sensory evaluation with regards to the bitterness of the product developed using such enzymes.

Influence Of Tidal Current On Microplastic Dynamics In Chao Phraya River Estuarine Ecosystem, Jiradet Tang-Siri
Chulalongkorn University Theses and Dissertations (Chula ETD)
Microplastics (MPs) can be found almost everywhere in marine ecosystems. That is one of the issues of concern around the world, including in Thailand. Estuary ecosystems are also among the most disturbed. By the estuary ecosystem affected by both rivers and oceans, such as the mouth of the Chao Phraya River in Thailand. Due to the change in tide, it is necessary to study the effects of tidal currents on the depth of MPs. Water samples were collected at approximately 60 water levels in 24 hours. Samples and three water quality indicators were collected at Pak Nam Chao Phraya. The …

Wastewater-Based Epidemiology And Quantitative Microbial Risk Assessment Associated With Wastewater Contact Of Norovirus In Bangkok, Thailand, Nonnarit Sriporatana
Chulalongkorn University Theses and Dissertations (Chula ETD)
Norovirus is one of the leading causes of acute gastroenteritis worldwide. Wastewater surveillance offer significant benefits such as enhancing our understanding of the disease occurrence in the population, evaluating the effectiveness of pathogen reduction in the treatment process, and tracking the spread of pathogens in the environment which pose a health risk to humans. Hence, wastewater surveillance is crucial. In this study, the influent and effluent samples were collected bi-monthly from March 2020 to March 2021 from three distinct wastewater treatment plants in Bangkok. Wastewater samples were concentrated and subjected to quantitative polymerase chain reaction (qPCR) to assess norovirus concentrations. …
