The major histocompatibility complex (MHC) is the most polymorphic gene
region in jawed vertebrates. Its gene products play a critical role in determining
individual and population fitness by presenting foreign antigens to immune system
cells, thereby initiating immune response to pathogen and parasite infections and
cancer. I surveyed the literature to investigate how selection maintains such
important adaptive diversity in mammalian populations and how MHC diversity
affects individual and population fitness. Balancing selection in the form of either
heterozygote advantage or rare allele advantage has long been proposed as the key
mechanism for maintaining the extreme diversity in MHC gene …

Metabolic switches play a critical role in the pathophysiology of cardiac diseases, including heart failure. Here, we describe an assay for long-chain fatty acid oxidation in neonatal mouse cardiomyocytes by using a SeaHorse Flux Analyzer (Agilent). This protocol is a simplified but robust adaptation of the standard protocol that enables metabolic measurements in cells isolated from transgenic mouse models, which can be timesaving and informative. Cell isolation and culture represent a critical point that may require bench optimization.
For complete details on the use and execution of this protocol, please refer to Angelini et al. (2021).

Hypertension and Vascular Research Articles
Exposure to environmental pollutants, including dioxin-like polychlorinated biphenyls (PCBs), play an important role in vascular inflammation and cardiometabolic diseases (CMDs) by inducing oxidative stress. Earlier, we demonstrated that oxidative stress-mediated lipid peroxidation derived 4-hydroxy-2-nonenal (4HNE) contributes to CMDs by decreasing the angiogenesis of coronary endothelial cells (CECs). By detoxifying 4HNE, aldehyde dehydrogenase 2 (ALDH2), a mitochondrial enzyme, enhances CEC angiogenesis. Therefore, we hypothesize that ALDH2 activation attenuates a PCB 126-mediated 4HNE-induced decrease in CEC angiogenesis. To test our hypothesis, we treated cultured mouse CECs with 4.4 µM PCB 126 and performed spheroid and aortic ring sprouting assays, the ALDH2 activity …
Sleep And Eating Behavior Among Adolescent Females With Overweight Or Obesity: The Role Of Appetite-Related Cognitive Processes, Kirandeep Kaur
Sleep And Eating Behavior Among Adolescent Females With Overweight Or Obesity: The Role Of Appetite-Related Cognitive Processes, Kirandeep Kaur
Theses and Dissertations
Insufficient sleep duration and poor sleep quality can potentiate weight gain and obesity in adolescents. Furthermore, overweight and obese females are at unique risk for insufficient sleep and associated health complications. We examined self-reported sleep duration and self-reported adequacy of sleep duration as potential moderators of the relationship between eating behavior and several cognitive processes including hedonic hunger, executive dysfunction, and self-control. We used a multisystemic conceptual framework to highlight the pathways that may explain the relationship between sleep behaviors and the Healthy Eating Index (HEI). The study employed a cross-sectional design. Participants completed baseline measures of height and weight, …

Aging is a naturally occurring decline of physiological processes and biological pathways that affects both the structural and functional integrity of the body and brain. These physiological changes reduce motor skills, executive function, memory recall, and processing speeds. Aging is also a major risk factor for multiple neurodegenerative disorders including Alzheimer’s disease (AD). Identifying a biomarker, or biomarkers, that signals the transition from physiological to pathological aging would aid in earlier therapeutic options or interventional strategies. Considering the importance of glutamate signaling in synaptic plasticity, motor movement, and cognition, this neurotransmitter serves as a juncture between cognitive health and disease. …
Long Chain Fatty Acids And Virulence Repression In Intestinal Bacterial Pathogens, Mary K. Mitchell, Melissa Ellermann
Long Chain Fatty Acids And Virulence Repression In Intestinal Bacterial Pathogens, Mary K. Mitchell, Melissa Ellermann

When bacterial pathogens enter the gut, they encounter a complex milieu of signaling molecules and metabolites produced by host and microbial cells or derived from external sources such as the diet. This metabolomic landscape varies throughout the gut, thus establishing a biogeographical gradient of signals that may be sensed by pathogens and resident bacteria alike. Enteric bacterial pathogens have evolved elaborate mechanisms to appropriately regulate their virulence programs, which involves sensing and responding to many of these gut metabolites to facilitate successful gut colonization. Long chain fatty acids (LCFAs) represent major constituents of the gut metabolome that can impact bacterial …

Acute myocardial infarction (MI) is one of the leading causes of death worldwide. Early identification of ischemia and establishing reperfusion remain cornerstones in the treatment of MI, as mortality and morbidity can be significantly reduced by establishing reperfusion to the affected areas. The aim of the current study was to investigate the metabolomic changes in the serum in a swine model of MI induced by ischemia and reperfusion (I/R) injury, and to identify circulating metabolomic biomarkers for myocardial injury at different phases. Female Yucatan minipigs were subjected to 60 min of ischemia followed by reperfusion, and serum samples were collected …

Loose smut (LS) disease is a serious problem that affects barley yield. Breeding of resistant cultivars and identifying new genes controlling LS has received very little attention. Therefore, it is important to understand the genetic basis of LS control in order to genetically improve LS resistance. To address this challenge, a set of 57 highly diverse barley genotypes were inoculated with Egyptian loose smut race(s) and the infected seeds/plants were evaluated in two growing seasons. Loose smut resistance (%) was scored on each genotype. High genetic variation was found among all tested genotypes indicating considerable differences in LS resistance that …

Facilitators And Barriers To Farmers’ Market Use In A Rural Area, Soghra Jarvandi, Kristen Johnson, Karen Franck
Journal of Extension
Farmers’ markets may improve access to healthful foods in rural areas. Our objective was to identify facilitators and barriers to farmers’ market use in a rural county. We collected data via surveys, focus group sessions, and key informant interviews. Study participants identified the two existing farmers’ markets as community assets. Barriers to use farmers’ markets included inconvenient market hours, not accepting nutrition assistance program benefits, limited transportation, and limited variety. Interventions to improve food access should include ways to meet the needs of specific populations such as low-income residents and residents living in outlying areas without farmers’ markets.

Topics in Exercise Science and Kinesiology
Topics in Exercise Science and Kinesiology Volume 3: Issue 1, Article 5, 2022. A simple way to gauge daily physical activity levels is to use a wearable technology device to count the number of steps taken during the day. However, it is unknown whether these devices return accurate step counts for persons with different body fat percentages or body mass index scores. The purpose was to determine if there is a correlation between either body fat percentages and/or body mass index values and the percent error calculated between a manual step count and values recorded by a wearable technology device. …
