Investigating The Reversibility Of Protein Adsorption On Gold Nanoparticles And The Role Of Free Thiol, Awosiji Olatunde Awotunde

Theses and Dissertations

Protein-gold nanoparticle (AuNP) bioconjugates have many potential applications in nanomedicine. A thorough understanding of the interaction between the protein and AuNP is critical to engineering these functional bioconjugates with desirable properties. In this work, we investigate the role of free thiols presented by the protein on the stability of the protein-AuNP conjugate. Human serum albumin (HSA) was modified with 2-iminothiolane (Traut’s reagent) to introduce additional thiols on the protein surface, and three variants of HSA were synthesized to present 1, 5, and 20 free thiols by controlling the molar excess of the chemical modifier. Protein exchange studies on AuNPs were …

Presentations, Lectures, Posters, Reports

Participants in the CONVERGE COVID-19 Working Group’s Workshop #6 on Psychological Adjustment breakout sessions discussed the critical threats to psychological health and well-being facing shelter staff, volunteers, and clients in the 2020 hurricane season.

It is widely recognized that people are approaching the 2020 hurricane season under unusual levels of stress. The COVID-19 pandemic has created chronic stressors that individuals are already managing in addition to addressing new acute fears like –will I get sick if I go to a shelter? These new and exacerbated stressors are a risk for higher levels of burnout, compassion fatigue, and ill-being. …

Synthesizing Modified Polymeric Nanoparticles For Biofilm Inhibition, Antibiotic Encapsulation, And Specific Targeting Against Pseudomonas Aeruginosa, Logan Schnorbus

Theses and Dissertations

Widespread usage of antibiotics is a growing concern due to antibiotic resistance development in bacteria. This is due to common use of antibiotics in agricultural, livestock, and clinical usage. Antibiotic resistance is developing at a rate in which it is outpacing new drugs on the market. New strategies in drug development are necessary to combat the increasing resistance. We designed several motifs of sugar-modified nanoparticles to inhibit the biofilm formation of Pseudomonas aeruginosa.

P. aeruginosa is an opportunistic pathogenic bacterium that is responsible for common life-threatening infection in hospitals. This gram-negative opportunistic pathogenic bacterium infects hosts with compromised immune systems …

Racial And Socioeconomic Disparities In Healthcare- And Community-Associated Clostridium Difficile Infection: A Rapid Review, Rutvin Kyada, Amy Leader, Drph, Mph, Rosemary Frasso, Phd, Cph

Master of Public Health Capstone Presentations

Clostridium difficile infection (CDI) is a major cause of morbidity and mortality in the United States and has been termed by the Centers for Disease Control and Prevention as an “urgent threat” in antimicrobial resistance. Traditionally a major healthcare-associated infection, CDIs have become increasingly common in community settings as well. As usage of healthcare services and certain medications are common risk factors for CDI, an individual’s access to such services can impact the CDI burden they experience. Identifying and addressing such disparities is crucial for an equitable healthcare system, but there is a general dearth of literature reviewing sociodemographic disparities …

Michigan Tech Publications, Part 1

This study describes the development of an automated bag valve mask (BVM) compression system, which, during acute shortages and supply chain disruptions can serve as a temporary emergency ventilator. The resuscitation system is based on the Arduino controller with a real-time operating system installed on a largely RepRap 3-D printable parametric component-based structure. The cost of the system is under $170, which makes it affordable for replication by makers around the world. The device provides a controlled breathing mode with tidal volumes from 100 to 800 milliliters, breathing rates from 5 to 40 breaths/minute, and inspiratory-to-expiratory ratio from 1:1 to …

Vaccine Procurement In A Time-Indexed Model With Quantity-Based Discounts, Joseph N. Yeiter Jun 2020

Vaccine Procurement In A Time-Indexed Model With Quantity-Based Discounts, Joseph N. Yeiter

Theses

Vaccination is hailed as one of the greatest scientific achievements of the past century. However, affordability and accessibility prevent low-income countries from realizing the full benefit of immunization. This study applies operations research methods to a hypothetically coordinated global vaccine market with quantity-based discounts to minimize procurement and holding costs while ensuring investment recovery of manufacturers. The effects on affordability of fixed-costs, holding costs, cold-chain capacity, and tender length (i.e. the maximum number of years ahead markets are willing to order supply from producers) are analyzed for each income-based segment of the global market. Experimental results show that increases in …

School of Biomedical Sciences

Despite extensive investigations, an efective treatment for sepsis remains elusive and a better understanding of the infammatory response to infection is required to identify potential new targets for therapy. In this study we have used RNAi technology to show, for the frst time, that the inducible lysophosphatidylcholine acyltransferase 2 (LPCAT2) plays a key role in macrophage infammatory gene expression in response to stimulation with bacterial ligands. Using siRNA- or shRNA-mediated knockdown, we demonstrate that, in contrast to the constitutive LPCAT1, LPCAT2 is required for macrophage cytokine gene expression and release in response to TLR4 and TLR2 ligand stimulation but not …

Mice xenotransplanted with human cells and/or expressing human gene products (also known as "humanized mice") recapitulate the human evolutionary specialization and diversity of genotypic and phenotypic traits. These models can provide a relevant in vivo context for understanding of human-specific physiology and pathologies. Humanized mice have advanced toward mainstream preclinical models and are now at the forefront of biomedical research. Here, we considered innovations and challenges regarding the reconstitution of human immunity and human tissues, modeling of human infections and cancer, and the use of humanized mice for testing drugs or regenerative therapy products. As the number of publications exploring …

Membrane vesicles are considered virulence cargoes as they carry capsular and melanin components whose secretory transport is critical for the virulence of the human fungal pathogen Cryptococcus species. However, other components of the vesicles and their function in the growth and virulence of the fungus remain unclear. We have previously found that the cryptococcal intersectin protein Cin1 governs a unique Cin1-Wsp1-Cdc42 endocytic pathway required for intracellular transport and virulence. Using RNA sequencing, we compared the profiles of extracellular RNA (exRNA), including microRNA (miRNA), small interference RNA (siRNA), long noncoding …

Nutritional geometry has advanced our understanding of how macronutrients (e.g.,proteins and carbohydrates) influence the expression of life history traits and theircorresponding trade-offs. For example, recent work has revealed that reproduc-tion and immune function in male decorated crickets are optimized at very differentprotein:carbohydrate (P:C) dietary ratios. However, it is unclear how an individual'smacronutrient intake interacts with its perceived infection status to determine in-vestment in reproduction or other key life history traits. Here, we employed a fullyfactorial design in which calling effort and immune function were quantified formale crickets fed either diets previously demonstrated to maximize calling effort(P:C = 1:8) or immune …

Environmentally mediated changes in body size often underlie population responses to environmental change, yet this is not a universal phenomenon. Understanding when phenotypic change underlies population responses to environmental change is important for obtaining insights and robust predictions of population dynamics in a changing world. We develop a dynamic integral projection model that mechanistically links environmental conditions to demographic rates and phenotypic traits (body size) via changes in resource availability and individual energetics. We apply the model to the northern Yellowstone elk population and explore population responses to changing patterns of seasonality, incorporating the interdependence of growth, demography, and density-dependent …

Unique Case Of Gastroenteritis Presenting As Pneumatosis Intestinalis With Emphysematous Gastritis And Portal Vein Gas: Do Not Take Gastroenteritis Lightly., Asif Mehmood, Rajesh Essrani Md, Swetha Parvataneni, Umair Iqbal

Department of Medicine

Pneumatosis intestinalis (PI) consists of multiple, thin, gas-filled cysts in the wall of the gastrointestinal (GI) tract. It is an uncommon entity that can involve any gastrointestinal site from the stomach to the rectum. Isolated stomach involvement is rare. PI can represent a broad spectrum of diseases with variable prognoses. We present the case of a patient who was admitted with gastroenteritis-like symptoms. He remained hemodynamically stable, and on further imaging with contrast-enhanced computed tomography of the abdomen and pelvis, air was found in the portal vein and gastric wall, with minimal thickening of the proximal small bowel concerning for …
