This mixed-methods study investigated the effectiveness of Generative AI (GenAI) powered intelligent tutoring systems (ITS) in undergraduate physics education, specifically comparing learning outcomes between students using Khanmigo (Khan Academy's AI tutor) and Google search engine. The study involved 69 undergraduate students divided into two groups (Khanmigo and Google search engine), with a third Paper-only group emerging during the experiment. Participants completed pre and posttests using the Lunar Phases Concept Inventory (LPCI) and participated in structured interviews about their learning experiences. Quantitative analysis revealed significant learning gains across all conditions but found no statistically significant differences between groups in terms of …

DNA polymerase θ (Polθ) is a polymerase-helicase fusion protein that is synthetically lethal with homologous recombination (HR) factors, such as BRCA1/2, and confers resistance to PARP inhibitors (PARPi) and other genotoxic cancer therapies. Previously developed Polθ polymerase (Polθ-pol) inhibitors (Polθi) exhibited limited pharmacological activity and metabolic stability, warranting the development of a Polθi with improved drug-like properties. Here, we developed RTx-303, a selective allosteric small-molecule Polθ-pol inhibitor that exhibits 5.1 nM IC50, 88% oral bioavailability, and a prolonged half-life along with its equipotent metabolite. X-ray crystallography highlights the development of a solvent-exposed side-chain that is essential for the optimal drug-like …

Humans instinctively adopt methods to reduce their risk of encountering harmful pathogens, yet their adherence to preventive health practices can often be erratic. Condoms exemplify one vital preventive tool against sexually transmitted infections (STIs) that, despite their effectiveness, are not consistently utilized. This pattern of behavior appears to persist even during periods of widespread disease transmission, with varied data from the COVID-19 pandemic indicating that condom usage remained inconsistent. The present study aimed to clarify these inconsistencies by examining changes in condom use cross-culturally. The genome of the hemlock woolly adelgid (Adelges tsugae) is presented in 10 chromosomal pseudomolecules along with 132 unplaced scaffolds for a combined length of 216.56 Mb. The genome is highly contiguous with an N50 of 20.54 Mb and its longest scaffold 37.41 Mb in length. The assembly recovered 97.7% of benchmarked universal single-copy Hemiptera orthologs (BUSCO) with an overall completion score of 98.6%. On the basis of chromosomal synteny with other aphid genomes, the hemlock woolly adelgid’s genome is composed of eight autosomes and two sex (X) chromosomes. Objectives: This study aimed to compare pelvic floor muscle strength (PFMS) in women who had vaginal delivery with episiotomy and those who underwent planned caesarean section 1–3 years postpartum. Methods: This retrospective cohort included 208 women from Taleghani Hospital, Arak, Iran, recruited by purposive sampling between May and August 2022. Participants were allocated to two groups: 104 with vaginal delivery and episiotomy and 104 with planned caesarean section. The intestinal mucosal epithelium forms a barrier between luminal contents and the body. MicroRNAs (miRNAs) regulate mucosal homeostasis by controlling inflammatory responses and structural integrity. Here, we discovered a protective role for miR147 in intestinal inflammation using a miR147tdTomato reporter mouse. miR147 was enriched in the intestines, with the highest expression in the colonic epithelial cells at the luminal surface, with prominent expression in differentiated enterocytes. Mice with general or intestinal epithelial deletion of miR147 showed increased intestinal inflammation and diminished mucosal healing during colitis. Obesity is a major public health concern. Predicting obesity risk from lifestyle data can guide targeted interventions, but current models remain limited. This study first evaluates ensemble learning methods and then combines approaches to improve prediction accuracy and generalizability. Four ensemble techniques-boosting, bagging, stacking, and voting-were tested. Five boosting and five bagging models were constructed alongside voting and stacking models. Hyperparameter tuning optimized performance, and feature importance analysis guided potential feature elemination. In phase two, hybrid stacking and voting models integrated the best-performing boosting and bagging models to enhance predictive capability. The Nebraska Department of Transportation (NDOT) must manage sediment and pollutant loads from roadway runoff to meet stormwater regulations for discharges into impaired waters. The SAFL Baffle, a hydrodynamic separator used by NDOT, depends on accurate estimates of total suspended solids (TSS) and particle size distribution (PSD). However, existing studies show large variability in these characteristics, limiting their relevance to Nebraska roadways.
This study monitored stormwater runoff for 1.5 years at four NDOT-maintained sites—two in Lincoln and two in Beatrice—that received runoff from areas outside the roadway right-of-way. The SAFL Baffle was evaluated using the SHSAM (Sizing Hydrodynamic Separators and …

After New York State Rifle & Pistol Association v. Bruen, history is the touchstone of Second Amendment analysis. Thus, this Note explores an understudied part of America's long and complicated history with weapons: Catholic disarmament. By undertaking a detailed historical analysis of three Catholic disarmament measures in the late colonial United States, this Note attempts to determine what the history means for present day firearms law. It concludes that even though courts frequently cite America’s history of Catholic disarmament, they rarely use it in a historically accurate way. Stopping then restarting the blood flow to the heart can cause ischaemia reperfusion (IR) injury. This can happen during revascularisation following a myocardial infarction and during on pump cardiac surgery using cardioplegic arrest. Despite extensive studies to identify cardioprotective interventions, the myocardium continues to sustain significant injury. Therefore, there is a need to identify agents that can be used during IR. This review focuses on the potential cardioprotective role for acidic amino acids and natural dipeptides using evidence from experimental studies and clinical trials with particular emphasis on their membrane transport. Background: Germline pathogenic variants in TP53 cause Li-Fraumeni syndrome, with significantly elevated cancer risk from infancy. Accurate classification of TP53 variants is essential to guide clinical management and surveillance, yet many variants remain classified as variants of uncertain significance (VUS). To improve classification accuracy and reduce the proportion of VUS, the ClinGen TP53 Variant Curation Expert Panel (VCEP) has updated its specifications.
Methods: The updated specifications incorporate the latest ClinGen recommendations and methodological advances, providing greater granularity for multiple evidence types, and also introduce the novel use of variant allele fraction as evidence of pathogenicity, particularly in the context of …

B-cell acute lymphoblastic leukemia (B-ALL) is the most common pediatric malignancy. Given racial/ethnic differences in incidence and outcomes, B-ALL genome-wide association studies among children of African ancestry are needed. Leveraging multi-institutional datasets with 840 African American children with B-ALL and 3360 controls, nine loci achieved genome-wide significance (P < 5 × 10−8) after meta-analysis. Two loci were established trans-ancestral susceptibility regions (IKZF1, ARID5B), while the remaining novel loci were specific to African populations. Five-year overall survival among children carrying novel risk alleles was significantly worse (83% versus 96% in non-carriers, P = 4.8 × 10−3). Background: Lower extremity amputations (LEAs) are medically, psychologically, and functionally devastating. The objective of our study was to evaluate region- and sex-specific differences of unilateral and bilateral LEAs across the United States (US) from 1990 to 2019.
Methods: The Global Burden of Disease database was used to analyze years lived with disability (YLDs), prevalence, and incidence rates per 100,000 people for LEAs in the US from 1990 to 2019. Data were stratified into four US Census Bureau-defined regions: Northeast, Midwest, South, and West. Differences between regions and sexes were assessed, with statistical significance defined as p < 0.05.

Smoke plume dynamics involve various smoke processes and mechanics in the atmosphere and provide the scientific foundation for the development of tools to simulate and predict smoke and its environmental and human impacts. BACKGROUND: Incorporating people who use substances into a community-engaged research process can support the implementation and evaluation of evidence-based harm reduction programs. Attending to their voice ensures those who need these programs will use them. Tetanus and diphtheria (tetanus/diphtheria) infections are rare in the United States (U.S.), but adequate vaccination is essential to prevent disease and associated morbidity and mortality. Compared with U.S.-born persons, immigrant and refugee adults have lower completion rates for tetanus/diphtheria vaccination series. However, most electronic health record (EHR)-generated recommendations only advise tetanus/diphtheria vaccines every 10 years for persons ≥18 years. This analysis compared documented completion of tetanus/diphtheria 3-dose vaccination series with EHR-generated recommendations for immigrant and refugee patients at three U.S. health systems seen during 2017-2022. The dominance of "WEIRD" (Western, Educated, Industrialized, Rich, and Democratic) authors might indicate significant differences in research practices between them and non-WEIRD researchers or highlight issues such as inequality and publishing bias. In this survey, using a convenience sample of researchers worldwide (N = 72), we examined current research practices and researchers' views on the feasibility of advancements in employed methodologies among and between researchers in both WEIRD and Non-WEIRD countries. Happiness is a state that is constantly strived for amidst the complexity of life. Previous research suggests that the student population is prone to psychological problems due to high workloads and a scarce resource of time that could affect happiness. Referring to previous studies, this study seeks to examine the relationship between feelings of awe and subjective well-being which is influenced by perceptions of time availability and perceived workload. The aim of this study was to explore the relationship between social identity, industry role, and implicit biases in fitness professionals. Participants (n=296) were randomly assigned two implicit association tests (IATs): disability, weight, gender, skin-tone, or age.
