Statistical Methods For Investigating Cell-Type Specificity And Genetic Regulation Of Dna Methylation In Complex Traits, Youshu Cheng Jan 2025

Statistical Methods For Investigating Cell-Type Specificity And Genetic Regulation Of Dna Methylation In Complex Traits, Youshu Cheng

Yale Graduate School of Arts and Sciences Dissertations

DNA methylation is one of the most widely studied epigenetic modifications that capture the cumulative effects of environmental exposures and inheritable effects and regulates gene expression. DNA methylation occurs at CpG sites, where the cytosine and guanine bases are connected by a phosphate group. Over the past decades, DNA methylation have been useful resources to understand the etiology and mechanisms of complex diseases. Multiple studies have linked DNA methylation with diseases to identify differentially methylated CpGs that associated with complex traits. In addition, another research direction is to focus on the single nucleotide polymorphisms (SNPs) associated with DNA methylation levels, …

Revues de littérature, synthèses de connaissances

Research organizations and academics often seek to map the development of scientific fields, identify research gaps, and guide the direction of future research. In cobot-related research, the scientific literature consulted does not propose any comprehensive research agenda. Moreover, cobots, industrial robots inherently designed to collaborate with humans, bring with them emerging issues. To solve them, interdisciplinary research is often essential (e.g., combination of engineering, ergonomics and biomechanics expertise to handle safety challenges). This paper proposes an exhaustive study that employs a scoping review and bibliometric analysis to provide a structured macro perspective on the developments, key topics, and trends in …

An Evaluation Of Data Protection And Privacy Issues Introduced By Byod In Financial Institutions, Andy Miguel Santana Jan 2025

An Evaluation Of Data Protection And Privacy Issues Introduced By Byod In Financial Institutions, Andy Miguel Santana

CCAC Theses and Dissertations

The idea of “bring your own device” (BYOD) allows organizational employees to conduct their tasks or processes on their own personal devices, has increased organizational efficiency significantly while allowing employees more flexibility. However, this approach also introduces major concerns about the security of organizational data as employees take their devices everywhere with them, opening more opportunities for unauthorized access to important data. Another major concern is the privacy of employee personal data. As many organizations implement BYOD, employees worry that with organizational monitoring and device management, their personal data is at risk as well. The problem this study tackles is …

Architectural Technical Debt Migrating Object Oriented Systems To Modular Architectures, Lionel Standridge Jan 2025

Architectural Technical Debt Migrating Object Oriented Systems To Modular Architectures, Lionel Standridge

CCAC Theses and Dissertations

Architectural Technical Debt (ATD), a subset of Technical Debt (TD), arises when outdated architectural decisions present significant challenges to the maintenance and evolution of legacy object-oriented monolithic systems. These systems tend to have tightly coupled components and rigid dependencies, making it difficult to scale, adapt, and modernize. This dissertation investigated strategies for managing ATD during the transition from monolithic architectures to modular systems. By identifying the root causes of ATD in a legacy objectoriented system and evaluating various decomposition strategies, this research proposed a framework to guide practitioners in reducing ATD and improving a system’s modularity. Using quantitative metrics, the …

Tau-Connectome Predictive Modeling And Subtyping In Preclinical Alzheimer's Disease, Hamid Abuwarda

Yale Graduate School of Arts and Sciences Dissertations

Alzheimer's disease (AD) is characterized by the accumulation of amyloid-? and tau, yet clinical progression varies widely across individuals. The central aim of this dissertation was to evaluate whether functional brain connectivity derived from resting-state fMRI can serve as a clinically meaningful predictive tool in the asymptomatic phase of the disease, preclinical AD. In Chapter 2, I develop connectome-based predictive models linking whole-brain functional connectivity to regional tau positron emission tomography (PET) signal in the Anti-Amyloid Treatment in Asymptomatic Alzheimer's Disease (A4) Study. These models predicted tau burden, particularly in regions vulnerable to tau deposition in Braak stages IV/V. In …

Genetic Variants Linked To Neurodevelopmental Disorders Within The Beta3-Beta4 Loop Of The Trio Ph 2 Domain Release Autoinhibition Of The Trio Gef2 Activity, Melissa Carrizales

Yale Graduate School of Arts and Sciences Dissertations

The TRIO family of proteins, which includes Trio, Kalirin, UNC-73 (in Caenorhabditis elegans), and dTrio (in Drosophila), are key regulators of cell morphogenesis, migration, tissue organization, and neuronal development through their regulation of Rho GTPases. These proteins feature multiple catalytic and scaffolding domains that mediate GTPase activation and cellular localization. TRIO proteins are essential for processes like actin dynamics, cell migration, and synaptic function, and disruptions in their activity have been linked to neurodevelopmental and neuropsychiatric disorders. This study focuses on Trio, specifically its regulatory mechanisms and how disease-related mutations disrupt its catalytic activity, contributing to various human diseases.

Individual-Level Large-Scale Circuit Modelling Of Human Cortex, Rachel Ann Cooper Jan 2025

Individual-Level Large-Scale Circuit Modelling Of Human Cortex, Rachel Ann Cooper

Yale Graduate School of Arts and Sciences Dissertations

Individuality is a defining feature of human existence. Yet the neural mechanisms underlying such individual variation are not fully understood. Prior research has suggested that variation in the balance between excitation and inhibition within the brain may contribute to these differences. To investigate this hypothesis, we employed a large-scale, biophysically-based model of the human cortex. This model is governed by a small set of biologically interpretable parameters, allowing for a focused analysis of how shifts in excitation and inhibition shape individual brain function. Using this modelling framework, this thesis examined patterns of variation across both healthy individuals and clinical populations. …

Situating Standing In Environmental Litigation, Wyatt Balius Jan 2025

Situating Standing In Environmental Litigation, Wyatt Balius

Undergraduate Theses, Professional Papers, and Capstone Artifacts

Controversies over issues of standing in federal civil litigation are hotly contested, as they strike at the heart of separation of powers issues and broader notions of the intent of civil action. Here I identify two competing theories of the purpose of standing and its requirements as such. Restrictive standing, as argued by Professor Ann Woolhandler, seeks to inherently link civil redress with an injury-in-fact requirement for both public and private law claims. Expansive standing, as argued by Professor Cass R. Sunstein, seeks to ground civil redress largely within violations of Congressionally designated causes of action. I argue that the …

Rejuvra's Effect On Soil Seed Banks, Blake J. Roeslein, Bryan Endress, Abby Mcmurtry, Akasha Faist

Undergraduate Theses, Professional Papers, and Capstone Artifacts

Invasive annual grasses are a major driver of grassland degradation globally. In perennial grasslands, invasion can heavily impact biodiversity, including potential changes to the soil seed banks. Seed banks, the living seeds stored in the upper soil column, play an important role in healthy systems, or can harbor invasive species. To reduce negative invasion outcomes, whether in the seed bank, or aboveground vegetation, pre-emergent herbicides are a commonly used tool for land managers. Pre-emergent herbicides work by targeting seeds as they germinate and begin emerging from the soil. Thus, our study asks 1) how does pre-emergent herbicide influence germination and …

Transcending Boundaries: Integrating Scientific Perspectives In Liberal Arts Education, Zlata Božac Jan 2025

Transcending Boundaries: Integrating Scientific Perspectives In Liberal Arts Education, Zlata Božac

GHEA21 Open Educational Resources

Transcending Boundaries: Integrating Scientific Perspectives in Liberal Arts Education confronts the enduring divide between the sciences and humanities, a split famously critiqued by C.P. Snow in his essay "The Two Cultures". While liberal arts education now includes natural sciences alongside humanities and social sciences, many Core Curriculum courses still fail to resonate with STEM students. These students often find such courses disconnected from their academic interests, perceiving them as irrelevant or lacking transferable skills.
This guide aims to bridge that gap by providing practical strategies and class exercises that integrate scientific thinking into the broader liberal arts framework. Drawing on …

North Carolina A&T: Perfect Storms, Charles Euchner Jan 2025

North Carolina A&T: Perfect Storms, Charles Euchner

GHEA21 Open Educational Resources

If the problem of race is the “American dilemma,” as the Swedish sociologist Gunnar Myrdal argued in a classic 1944 study, North Carolina’s response to this dilemma presents a complex puzzle. Unlike other states of the old Confederacy, North Carolina has a long tradition of reform in education, labor, business, and voting rights. North Carolina also has robust competition between Republicans and Democrats. Because of this competition, both parties have battled intensely over voting rights.
