Background: Lung adenocarcinoma (LUAD) has several histologically distinct subtypes that differ by a number of clinical features including patient survival. Molecular mechanisms underlying histological and clinical differences between subtypes remain poorly understood.

Methods: We conducted a comparative analyses of gene expression in acinar, lepidic, papillary and solid subtypes, as well as mucinous adenocarcinoma. We used a novel, more efficient approach to identify subtype-specific genes. We compared the mean gene expression level separately for tumors and adjacent normal tissue with pure or a highly represented (≥ 75%) subtype of interest to the mean expression in tumors where the subtype of interest …

Background/objectives: The tumor microenvironment (TME) plays a critical role in cancer progression by shaping immune responses and influencing patient outcomes. We hypothesized that the relative proximity of specific immune cell pairs to cancer cells within the TME could help predict their pro- or anti-tumor functions and reflect clinically relevant immune dynamics.

Methods: We analyzed imaging mass cytometry (IMC) data from lung adenocarcinoma (LUAD) and triple-negative breast cancer (TNBC) cohorts. For each immune cell pair, we calculated a relative distance (RD) score, which quantifies the spatial difference in proximity to cancer cells. Research on children's technology and digital media (TDM) is hampered by a lack of robust approaches for assessing TDM use. This study assessed the feasibility of passively measuring children's TV screens and mobile devices (TDM) in a naturalistic setting. In the three-day feasibility study, FLASH-TV was set up on one to two TVs the child (5-12 year olds) typically used in the home (n=20). Children's mobile device use was assessed with either the Chronicle App or ScreenTime screenshots. Parents completed three TDM diaries. Immune dysfunction and late mortality from multiorgan failure are hallmarks of severe sepsis. Arginine, a semi-essential amino acid important for protein synthesis, immune response, and circulatory regulation, is deficient in sepsis. However, arginine supplementation in sepsis remains controversial due to the potential to upregulate inducible nitric oxide synthase (iNOS)-mediated excessive nitric oxide (NO) generation in macrophages, leading to vasodilation and hemodynamic catastrophe. Citrulline supplementation has been considered an alternative to replenishing arginine via de novo synthesis, orchestrated by argininosuccinate synthase 1 (ASS1) and argininosuccinate lyase (ASL). Transcranial electrical stimulation (tES) has shown potential to modulate attention and enhance learning, yet its effects in applied, real-world contexts, remains underexplored. This study investigated the impact of transcranial direct current stimulation (tDCS) and high-frequency transcranial random noise stimulation (hf-tRNS with DC offset) on performance in a synthetic aperture radar (SAR) visual search task requiring both target identification and change detection. Participants (N = 63) were randomly assigned to tDCS, hf-tRNS, or sham conditions, with stimulation applied to the right inferior frontal gyrus (F10) during the training phase. Retrieval-augmented generation (RAG) has become integral to large language models (LLMs), particularly for conversational AI systems where user questions may reference knowledge beyond the LLMs' training cutoff. However, many natural user questions lack well-defined answers, either due to limited domain knowledge or because the retrieval system returns documents that are relevant in appearance but uninformative in content. In such cases, LLMs often produce hallucinated answers without flagging them. With lower detection limits for forensic DNA analysis, it is important to have a better understanding of background DNA levels unrelated to the crime to facilitate accurate data interpretation. Duct tape is used in crimes, e.g. to bind victims and seal drug packaging, and may be tested for DNA. To determine DNA background levels, twelve volunteers were recruited to donate a used roll of duct tape from their home. Neural IR has advanced through two distinct paths: entity-oriented approaches leveraging knowledge graphs and multi-vector models capturing fine-grained semantics. We introduce QDER, a neural re-ranking model that unifies these approaches by integrating knowledge graph semantics into a multi-vector model. QDER's key innovation lies in its modeling of query-document relationships: rather than computing similarity scores on aggregated embeddings, we maintain individual token and entity representations throughout the ranking process, performing aggregation only at the final scoring stage-an approach we call "late aggregation." We first transform these fine-grained representations through learned attention patterns, then apply carefully chosen mathematical operations for precise matches. …

Medetomidine, a veterinary α2-adrenergic agonist, has recently emerged as an adulterant in the non-medical opioid supply, yet human exposure has remained poorly characterized. We conducted a pragmatic retrospective cohort analysis utilizing chart review and liquid chromatography-tandem mass spectrometry (LC-MS/MS) toxicology testing on available urine samples from patients presenting to two hospitals in Philadelphia, PA, who fit two clinical phenotypes, intoxication or withdrawal. Samples also underwent glucuronidase pre-treatment to assess impact on the yield of medetomidine and xylazine metabolite detection. Spermatozoa acquire fertilizing competence during epididymal transit through proteolytic, chaperone-mediated, and post-translational modifications. Ovochymase 2, an epididymis-specific trypsin-like serine protease, has emerged as a central regulator of this maturation process. Here, we integrate targeted gene disruption, comprehensive proteomic profiling, and affinity-based proteome enrichment to delineate how Ovochymase 2 influences sperm functionality. Deletion of Ovochymase 2 disrupts the proteome of epididymal sperm, resulting in diminished levels of core fertility-related factors-including a disintegrin and metalloprotease domain 3, β-defensins, and protease-inhibitor complexes-while inducing compensatory upregulation of alternate proteases and chaperones. The Cre/loxP system is widely used for site-specific genetic manipulation in mice. The PgrCre mouse model, where Cre recombinase is driven by the progesterone receptor promoter, is commonly used for gene ablation in Pgr-positive uterine cells. However, the PgrCre is active in the neonatal uterus and does not allow temporal control. To enhance the functionality of the PgrCre mouse, we generated and characterized an inducible PgriCreERT2 mouse, in which iCreERT2 is inserted downstream of the endogenous Pgr promoter. PgriCreERT2 mice crossed with Rosa26-CAG-LSL-Sun1-sfGFP-myc reporter mice demonstrated tamoxifen-dependent recombination in uterine stromal fibroblasts and a subset of epithelial cells. Objectives: Cortical reorganization has been proposed as a mechanism for motor recovery of the upper limb. The objective of this study was to determine if intervention-related clinical improvement in post-stroke hemiparesis is associated with enlarged motor cortex representation. Methods: Data were analyzed from a single site subset of the NICHE trial. This study explored the impact of Holistic Admissions Review (HAR) on demographic characteristics, multicultural experiences, and cultural humility (CH) levels among newly admitted pre-licensure nursing students. Fostering CH and diversity among healthcare providers may help reduce health disparities. Traditional admissions methods may not identify candidates with traits supporting CH development, whereas HAR considers diverse backgrounds and experiences. Two groups were surveyed: one admitted through HAR and the other through traditional methods. No significant differences in CH scores were found, but significant differences were found in age, living abroad, and volunteering with diverse populations. We examine the relationship between mindfulness and eudaimonic well-being in Thai adults, a population largely overlooked in mindfulness research despite the strong influence of Buddhism within this culture. Specifically, we hypothesized that hope and meaning will account for variance in this relationship. Utilizing self-report measures for mindfulness, hope, meaning in life, and eudaimonic well-being, findings from a sample of 269 Thai adults revealed significant positive correlations between all of these constructs and eudaimonic well-being. Regression analyses also indicated that hope and meaning in life accounted for statistically significant variance in the relationship between mindfulness and eudaimonic well-being and that mindfulness, …

African feminist theories provide essential insights into the everyday lived experiences of women within African spaces, allowing for a contextual analysis of the impacts of climate change. Punctuated Equilibrium Theory, as applied by Niles Eldredge and Stephen Jay Gould (1941-2002) to the study of species evolution, examined the infrequency of morphological change or stasis discontinuity.1 While groundbreaking in the field of Paleontology and the study of fossil change, the theory has been well-fitting to the decision sciences as introduced by Baumgartner and Jones2, in their 1993 and 2010 publications.
