Some significant incidents like the Chernobyl disaster (1986), the Fukushima Daiichi hydrogen explosion (2011), and some frequent military and terror threats to the nuclear Power plant structures have led to studying the capacity of power plant structures to withstand blast loadings. From this research, even one-third of the loss of the coolant is enough to damage the power plant structure. Crack starts from the wall and propagates toward the top of the dome and the foundation. Despite the success of targeted inhibitors in cutaneous melanoma, therapeutic responses are limited by the aged tumor microenvironment and drug-tolerant residual cells. Given the similarities between drug tolerance and cellular dormancy, we studied the dormancy marker, nuclear receptor subfamily 2 group F member 1 (NR2F1), in response to BRAF-V600E inhibitors (BRAFi) plus MEK inhibitors (MEKi) in BRAF-mutant melanoma models. Transcriptomic analysis of melanoma patient samples treated with BRAFi + MEKi showed increased NR2F1. NR2F1 was highly expressed in the drug-tolerant invasive cell state of minimal residual disease in patient-derived and mouse-derived xenografts on BRAFi + MEKi. In 1974 the University of Arizona renovated their football stadium through a bonds bill passed by the state legislature. The renovations allowed the university to increase the stadium’s capacity by 17,000 seats. While in the state house, an Arizona Congressman attached a rider to the bonds bill banning the university’s medical school from performing abortions in most circumstances, effectively barring the university from teaching the medical students how to perform the procedure as well. Boundary exchanges dominate the cost of both stenciled codes and those that rely on sparse matrix operations. The performance of large boundary exchanges is limited by synchronization overheads and injection bandwidth limitations. Irregular boundary exchanges incur additional overheads due to the large number of required messages. This thesis investigates multiple methods for improving the performance and scalability of both Cartesian and irregular boundary exchanges. Since boundary exchanges are typically performed iteratively, persistent communication presents an opportunity for optimization by sharing and amortizing setup costs. Partitioned communication is also explored to increase asynchrony, reducing bottlenecks from synchronization overheads and data congestion. …

The adaptive immune response is a complex defense mechanism that develops over time to recognize and eliminate pathogens with remarkable precision and durability. This dissertation investigates the dynamics, scaling, and efficiency of the adaptive immune response through a synthesis of computational modeling, mathematical analysis, and agent-based simulations. First, we analyze the topology of the lymphatic network and investigate the T cell search time to find the lymph node that is containing the matching dendritic cell. The problem addressed in this study was the lack of women in K–12 educational technology leadership positions in the United States. The purpose of this basic qualitative study was to explore the experiences of women in K–12 educational technology leadership regarding the challenges and successes they encountered in achieving and maintaining their positions. Feminist standpoint theory formed the framework for this study. Semistructured interviews were conducted with 10 women who were active or former K–12 educational technology leaders in public schools across the country. Sometime in the mid-tenth century on the Jutland peninsula of Denmark, the Viking Age Queen Thyra (d. ca. 958/9) passed away and the epithet Danmarkarbót—Denmark’s Adornment—was skillfully carved onto her memorial runestone. Yet Thyra’s story has been clouded by few and unreliable textual sources, by differing assessments of her familial background that have emerged from those sources, and by misrepresentations of the material culture relating to her. Modern organizations often reward “looking busy” rather than genuinely contributing, prompting employees to engage in superficial displays of productivity. This type of behavior can be considered as a previously unexplored yet important form of impression management. While impression management research has predominantly examined outcome-oriented tactics, it has not fully captured the subtler, non-contributory forms of performative behavior that permeate everyday work life. Glucose metabolism is a pivotal hub for cellular energy production and the generation of building blocks that support cell growth, survival, and differentiation. Cancer cells undergo metabolic reprogramming to sustain rapid proliferation, survive in harsh microenvironments, and resist therapies. Beyond producing energy and building blocks to meet cancer cell demands, glucose metabolism generates numerous metabolites that serve as signaling molecules, orchestrating signaling pathways and epigenetic modifications that regulate cancer cell phenotypes and immunity. Background: High-grade gliomas are highly invasive and respond variably to chemoradiation. Accurate, patient-specific predictions of tumor response could enhance treatment planning. We present a novel computational platform that assimilates MRI data to continually predict spatiotemporal tumor changes during chemoradiotherapy.

Methods: Tumor growth and response to chemoradiation was described using a two-species reaction-diffusion model of enhancing and non-enhancing regions of the tumor. Two evaluation scenarios were used to test the predictive accuracy of this model. The prevalence of type 2 diabetes is increasing among the Hispanic/Latino population. Type 2 diabetes incidence rates vary between neighborhoods, but no single aspect of the neighborhood environment is known to cause type 2 diabetes. Using data from the Hispanic Community Health Study/Study of Latinos cohort of 16,415 Hispanic/Latino adults in four major US cities, we conducted a neighborhood environment-wide association study to identify neighborhood measures or clusters of measures associated with diabetes incidence. Background: Prior clinical trials established the safety, but not the efficacy of bone marrow-derived mesenchymal stromal cells (MSCs) in the acute respiratory distress syndrome (ARDS).

Methods: We conducted a prospective, double-blind, multi-center randomized phase 2b clinical trial of one dose of intravenous MSCs (10 x 106/kg predicted body weight) versus placebo in 120 ventilated patients with ARDS (PaO2/FiO2 < 250 mmHg). The primary endpoint was change in oxygenation index (OI) over 36 hours from baseline.

Findings: Enrollment began in January 2020. Due to the Coronavirus 2019 (COVID-19) pandemic, the majority of subjects (101/120, 84%) developed ARDS from COVID-19. There were no significant baseline differences in severity of illness between patients treated with MSCs and those who received …

Neutrophils play a key role in autoimmune diseases like rheumatoid arthritis, contributing to tissue damage through rapid recruitment and activation. In this study, we investigated the regulatory properties of two receptor-like tyrosine phosphatases (RPTPs), CD45 and CD148, in inflammatory arthritis. Using an in vivo mouse model of K/BxN serum transfer-induced arthritis, we found that CD45 and CD148 feature distinct regulatory properties during inflammatory arthritis. CD45 is required for neutrophil infiltration, cytokine release, and reactive oxygen species production, whereas CD148 deficiency leads to a delayed onset of arthritis but unaltered overall neutrophil infiltration and reduced ROS production. Classifying Heterogeneous Cooperation In Social Dilemmas: Experimental Evidence And Simulation Insights, Sining Wang

In this paper, we introduce a revealed strategy method to capture heterogeneous behavioral patterns in repeated public goods experiments. This method identifies an individual’s strategic type—a stable decision-making tendency in interactive settings—by analyzing behavioral profiles. We conduct a repeated public goods game to collect participants’ behavioral data and then apply both k-medians and hierarchical clustering algorithms to classify them into five distinctive types: free riders, strong cooperators, above-average conditional cooperators, below-average conditional cooperators, and hump-shaped players. These classifications are then used to construct and calibrate an agent-based simulation model. Our simulation results suggest that policies targeting how conditional cooperators respond …
