BACKGROUND: Organoids have great potential to revolutionize various aspects of biomedical research and healthcare. Researchers typically use the fluorescence-based approach to analyse their dynamics, which requires specialized equipment and may interfere with their growth. Therefore, it is an open challenge to develop a general framework to analyse organoid dynamics under non-invasive and low-resource settings.
RESULTS: In this paper, we present a knowledge-driven deep learning system named TransOrga-plus to automatically analyse organoid dynamics in a non-invasive manner. Given a bright-field microscopic image, TransOrga-plus detects organoids through a multi-modal transformer-based segmentation module. Suicide remains a critical public health issue in the United States, with rates continuing to rise despite national prevention initiatives. Primary care settings serve as a pivotal point for early detection, yet inconsistent use of standardized screening and safety planning tools contributes to under-recognition of suicide risk. This Doctor of Nursing Practice (DNP) project aimed to enhance the identification and management of suicide risk in primary care by developing and implementing an evidence-based clinical practice guideline (CPG). While the Language Essentials for Teachers of Reading and Spelling (LETRS) professional development provided elementary teachers with a new form of instruction, third-grade reading scores remain below proficiency levels on the Alabama state test. Guided by Vygotsky’s theory of social constructivism, the purpose of this basic qualitative study was to explore elementary teachers’ challenges of implementing LETRS in a local Alabama school district. Bedside nurses in acute care settings face intensified job demands amid persistent low nursing staff census and rising patient acuity. The purpose of the study, guided by the job demands–resources (JD-R) model, was to examine the relationship between job demands, years of nursing experience, and perceived usefulness of virtual nursing technology among bedside nurses in acute care settings. A cross-sectional online survey was administered to bedside registered nurses (N = 102) currently using virtual nursing technology. Background: Pediatric specialist shortages and rapidly rising autism prevalence rates have compelled primary care clinicians to consider playing a greater role in the autism diagnostic process. The ECHO Autism: Early Diagnosis Program (EDx) prepares clinicians to screen, evaluate, differentiate, diagnose, and provide longitudinal care for children with autism in primary care settings. Despite the Americans with Disabilities Act of 1990 promoting equality for people with disabilities, parents with developmental and intellectual disabilities experience high rates of involvement with child welfare services due to outdated terminology, information about disabilities, and policies. The purpose of this study was to identify the challenges of parents with intellectual and developmental disabilities and determine if they differ from those of parents without a disability. Minority students are often omitted from social, economic, cultural, and political life due to ethnicity or poverty, which increases their risk of adverse childhood experiences, such as abuse or neglect, and increases their likelihood of discrimination that can obstruct their normal childhood development, including academic performance. Child-centered play therapy (CCPT) is a multiculturally appropriate approach that creates a safe and nonjudgmental environment, supporting adjustment and coping. Passive acoustic monitoring techniques are useful for studying vocally active marine species, particularly in remote and difficult to access areas. In this study, three months of acoustic recordings were collected off Egmont Atoll, Chagos Archipelago in the central Indian Ocean to detect, characterise, and investigate temporal variability of delphinid whistles. A subsample (3.6% of the total dataset) was used to manually annotate whistles and test automated whistle detectors. Higher frequency whistles (6-25 kHz, n = 126) were manually classified into six categories: upsweep (56%), convex (19%), downsweep (13%), concave (5%), sine (4%), and constant frequency (3%). Objective: The objective of this work is to develop a standard-based taxonomy of features that might affect user response to alerts using evidence from literature and public alert logic repositories.
Methods: We developed a taxonomy of features using multiple sources: (1) the Agency for Healthcare Research and Quality (AHRQ) CDS Connect Repository, (2) alert logic from commercial electronic health record (EHR) customers, and (3) published literature. Three categories (patient, provider, environment/context) were used a priori to develop the taxonomy. The final taxonomy was mapped to the Fast Healthcare Interoperability Resources (FHIR) standard for development of standardized CDS services.

Developmental processes extend beyond embryogenesis to support lifelong tissue adaptations. Avian feather follicles, with their resident stem cells and capacity for cyclic regeneration, provide a dynamic model for postnatal tissue remodeling. Here, we propose the Mandarin duck (Aix galericulata) as an ideal model to study lifelong developmental modulation, focusing on the sexually dimorphic "sail feather"-a secondary flight feather in males that undergoes seasonal transformation into a strikingly asymmetric, ornamented phenotype during the breeding season. We identified asymmetric morphogen expression in regenerating male sail feathers and used transcriptome and H3K27ac ChIP-seq to uncover male and female signaling pathways and regulatory elements. …

Grafting is an effective horticultural technique that significantly enhances disease resistance and stress tolerance in watermelon. This review systematically summarizes the types of rootstocks used in watermelon grafting and analyzes the effects of grafting on plant responses to biotic stresses, such as viral and fungal pathogens, root-knot nematodes infections, and abiotic stresses, including drought, temperature extremes, and salinity. Furthermore, it discusses the changes in fruit quality and explores the underlying mechanisms associated with graft-induced resistance. Fort Brooke, established in 1824 at the mouth of the Hillsborough River, shaped the growth of Tampa and Florida’s west coast for nearly sixty years and served as the nucleus for the forced removal of Indigenous people. Though the Fort itself has long since vanished beneath the modern city, archaeology has preserved its legacy. Excavations since the 1970s have uncovered rich assemblages of ceramics, glass, military equipment, faunal remains, and domestic refuse. These materials reveal daily life at the post, highlighting the interactions of soldiers, Seminoles, African Americans, white settlers, and Cuban fishermen, while also uncovering the deeper prehistoric, referred …

The PLATO network, a collection of mainframe computers, terminals, and educational software, has received an increasing amount of scholarly attention in the last decade as a social precursor to the modern internet. Existing histories have neglected to investigate the ways in which PLATO and its related business enterprise worked to accelerate the shift in university governance away from classical liberal ideas centering the public good and towards a more profit-centered neoliberal rationality. There is a general consensus among literary critics that the Romantic Era stands for a celebration of Imagination and an elevation of the latter to a status of a superior mental faculty. This generalization, however, fails to acknowledge that the project of the Romantics was not so much to extol Imagination over Reason, as to unify the two faculties. The delivery of culturally relevant content in K-12 computing education is widely recognized as essential for engaging students and fostering meaningful learning experiences. However, in many African contexts, K-12 computing teachers feel unprepared to address this need. Course materials often fail to reflect the cultural and linguistic diversity of the students they serve. In this study, we conducted participatory design sessions with education stakeholders, including primary and secondary school teachers, to develop culturally relevant programming activities for a national programming outreach initiative in Botswana. Contextualizing computer science education has been recognized as a key factor in enhancing student engagement and learning outcomes. This study investigates the initial perceptions of university computer science faculty in Africa regarding the benefits, adoption challenges, and institutional support required for the successful integration of contextually relevant materials into introductory computer science (CS1) courses. Faculty then assessed a set of previously developed contextually tailored materials, grounded in Banks' Additive Approach to curriculum reform and aligned to the CS curricula 2023. Achieving sustained local release of small hydrophilic drugs is challenging and is particularly important when the drugs are toxic. To address these challenges, we developed a hybrid system comprising drug-containing microparticles embedded within a nanoliposomal hydrogel matrix. This system forms through salt-induced gelation using physiologically relevant sodium chloride concentrations (0.9%), allowing for microparticle encapsulation without harsh chemical processes. In vitro, the hybrid system exhibited a slower release of encapsulated cargo compared to microparticles or hydrogel alone. In vivo proof of principle was provided with tetrodotoxin (TTX), a small hydrophilic and ultrapotent local anesthetic, which can cause systemic toxicity if the …

The lymphatic system maintains tissue fluid balance, and its dysfunction can result in lymphedema. Although cholesterol is essential for cellular function, its role in lymphatic development has remained unknown. Here, we identify APOA1 binding protein (AIBP) as a key regulator that promotes lymphatic endothelial cell fate specification and lymphangiogenesis. Microsurgical testicular sperm extraction (microTESE) with intracytoplasmic sperm injection (ICSI) represents the current standard treatment for nonobstructive azoospermia (NOA). However, cures remain unavailable for NOA patients lacking retrievable haploid cells. mRNA supplementation could be a potential treatment for genetic defects leading to impaired spermatogenesis. Lipid nanoparticles (LNPs) have emerged as mRNA delivery vehicles with minimal risk of genome integration; however, their ability to selectively deliver mRNA to specific cell types remains limited. Importance: Studies suggest that everolimus may reduce the risk of rejection, cardiac allograft vasculopathy (CAV), chronic kidney disease (CKD), and cytomegalovirus (CMV) after heart transplant. Everolimus use is controversial because of data demonstrating higher infection deaths when everolimus is introduced de novo after transplant. It is unclear whether everolimus is safe and effective when initiated at 6 months posttransplant in children, a population in which median graft survival is limited to 15 years and randomized clinical trials are lacking.

Introduction: The rapidly growing population of older adults (individuals aged 65 years and older) presents a new set of challenges for healthcare providers in the emergency department (ED), given the prevalence of severe and life-threatening conditions among this group, such as chronic cancer, Alzheimer's disease/dementia and congestive heart failure. Within the software engineering context, agile approaches encourage customer collaboration, iterative development, and flexibility. However, the mass exodus of highly skilled professionals, known as the “brain drain” or "Japa Syndrome," has emerged as a significant challenge, especially in Nigeria. This phenomenon has particularly impacted agile software development practitioners by undermining project continuity, knowledge transfer, and team dynamics. In this article, I describe an original teaching activity that explores the intersection of generative AI, poetic inquiry, and posthuman pedagogy, examining how students can critically and creatively engage with disruptive technologies. Using Rosi Braidotti’s posthuman framework (2013, 2019), I consider how generative AI might be used to challenge traditional notions of creativity, authorship, and knowledge production. Through a structured learning activity, students analyze a human poet’s work, generate AI-assisted poetry, and compose their own creative pieces, fostering non-linearity, memory and imagination, and defamiliarization—three key elements of Braidotti’s posthuman thinking. Teaching Social Studies in the Peach State
These classroom-based activities focus on Jimmy Carter, the Heritage Trust Act, and Ossabaw Island. We developed two short classroom experiences based on Georgia social studies standards with supporting English language arts standards: one classroom experience for second grade and the other for eighth grade. Teachers can adapt content and sequence for their grade levels, schedules, and other school-specific factors. Students will explore the history, culture, and geography of Ossabaw Island, Georgia’s third-largest barrier island. Classroom experiences include different sources, photo analysis, and a range of learning activities with modifications; figures and appendices are included.

A survey was conducted in eight major range science departments in the U.S. to determine the extent of appropriate training for international students, following recommendations made during the past twenty years. The average department was found to have little ability to teach in a language other than English, has few faculty who have worked in a developing country more than one year, and does not equally reward faculty for international work compared with domestic work.
