School of Veterinary and Biomedical Sciences: Faculty Publications
Salmonella enterica is, globally, an important cause of human illness with beef being a significant attributable source. In the human patient, systemic Salmonella infection requires antibiotic therapy, and when strains are multidrug resistant (MDR), no effective treatment may be available. MDR in bacteria is often associated with the presence of mobile genetic elements (MGE) that mediate horizontal spread of antimicrobial resistance (AMR) genes. In this study, we sought to determine the potential relationship of MDR in bovine Salmonella isolates with MGE. The present study involved 111 bovine Salmonella isolates obtained collectively from specimens derived from healthy cattle or their environments …

3d Printed Composite Structures For Bone Repair And Drug Delivery Application, Innocent J. Macha
Tanzania Journal of Engineering and Technology (TJET)
Due to the increase in traffic accidents, industrial contingencies, and natural disasters, there is a high demand for bone regeneration biomaterials. Although bone can regenerate and self-repair, it is difficult to do so when the defect area exceeds the critical repair area due to trauma, tumour resection, or congenital diseases. The traditional methods for bone repair are autograft, allograft, and xenograft. However, these methods have flaws and limitations, such as complications in the donor bone area, a limited number of donors, rejection, and infectious diseases. However, in recent years, the use of additive manufacturing technologies in bone tissue engineering has …

Extraction And Characterization Of Collagen From Cattle Horns For Potential Wound Healing, Yeriko Halson
Tanzania Journal of Engineering and Technology (TJET)
The need for bandages and gauze for wound dressing and healing is inevitable due to the high increase in accidents. Most of the material used for the production of bandages and gauzes for wound healing are from non-renewable synthetic material. The sustainability of non renewable resources is at jeopardy due to the health and associated environmental impact. Consequently, the use of naturally occurring and biodegradable material is a promising solution to the sustainability challenges of synthetic material. In this paper, the extraction of collagen films from cattle horns for potential wound healing is reported. Collagen was extracted from cattle horns …

Pusa sibirica, the Baikal seal, is the only extant, exclusively freshwater, pinniped species. The pending issue is, how and when they reached their current habitat—the rift lake Baikal, more than three thousand kilometers away from the Arctic Ocean. To explore the demographic history and genetic diversity of this species, we generated a de novo chromosome-length assembly, and compared it with three closely related marine pinniped species. Multiple whole genome alignment of the four species compared with their karyotypes showed high conservation of chromosomal features, except for three large inversions on chromosome VI. We found the mean heterozygosity of the studied …

The rapid spread and strong immune evasion of the SARS-CoV-2 Omicron subvariants has raised serious concerns for the global COVID-19 pandemic. These new variants exhibit generally reduced fusogenicity and increased endosomal entry pathway utilization compared to the ancestral D614G variant, the underlying mechanisms of which remain elusive. Here, we show that the C-terminal S1 mutations of the BA.1.1 subvariant, H655Y and T547K, critically govern the low fusogenicity of Omicron. Notably, H655Y also dictates the enhanced endosome entry pathway utilization. Mechanistically, T547K and H655Y likely stabilize the spike trimer conformation as suggested by increased molecular interactions in structural modeling and enhanced …

ZNF410 is a highly-conserved transcription factor, remarkable in that it recognizes a 15-base pair DNA element but has just a single responsive target gene in mammalian erythroid cells. ZNF410 includes a tandem array of five zinc-fingers (ZFs), surrounded by uncharacterized N- and C-terminal regions. Unexpectedly, full-length ZNF410 has reduced DNA binding affinity, compared to that of the isolated DNA binding ZF array, both in vitro and in cells. AlphaFold predicts a partially-folded N-terminal subdomain that includes a 30-residue long helix, preceded by a hairpin loop rich in acidic (aspartate/glutamate) and serine/threonine residues. This hairpin loop is predicted by AlphaFold to …

Cellulose-Keratin Supramolecular Composite: Conformation By Combined Fourier Transform Spectroscopy And Partial Least Square Regression Coefficient, Ngesa Ezekiel Mushi Dr
Tanzania Journal of Engineering and Technology (TJET)
Keratin and cellulose are important nano components with excellent mechanical and biological properties. However, the preparation route through dissolution in organic solvents to make manmade materials is challenging because of their intensive bonding interaction characteristics. Structural modification of the natural attributes is difficult to maintain after polymer regeneration. A new technique by a green route was devised to dissolve keratin from wool, hair, and feather with cellulose in situ using 1-Butyl-3-methylimidazolium chloride [BMIm] + Cl- ionic liquid as a solvent. The mixture of keratin and cellulose homogeneously and synergistically resulted in a supramolecular composite film through non-derivatized mechanochemical interactions. The …

Junctional adhesion molecule-like protein (JAML) serves as a co-stimulatory molecule in γδ T cells. While it has recently been described as a cancer immunotherapy target in mice, its potential to cause toxicity, specific mode of action with regard to its cellular targets, and whether it can be targeted in humans remain unknown. Here, we show that JAML is induced by T cell receptor engagement, reveal that this induction is linked to cis-regulatory interactions between the CD3D and JAML gene loci. When compared with other immunotherapy targets plagued by low target specificity and end-organ toxicity, we find JAML to be mostly …

Artificial intelligence (AI) pipelines are complex, heavily parameterized, and expensive to execute in terms of time and computational resources. Consequently, it is onerous to run experiments with all possible parameter combinations to achieve an optimal solution. However, these AI experiments can be optimized by recommending relevant parameters to commence the experiments, reducing search space significantly, which can be fine tuned further. The relevant parameters can be identified by observing the metadata of pipelines executed in the past, and the relevant pipeline with relevant parameters can be recommended to the user. Currently, there are various metadata frameworks that automatically record the …

Cryptosporidium parvum is an enteric pathogen that invades epithelial cells in the intestine, where it resides at the apical surface in a unique epicellular location. Compared with those of related apicomplexan parasites, the processes of host cell attachment and invasion by C. parvum are poorly understood. The streamlined C. parvum genome contains numerous mucin-like glycoproteins, several of which have previously been shown to mediate cell attachment, although the majority are unstudied. Here, we identified the antigens recognized by monoclonal antibody (MAb) 1A5, which stains the apical end of sporozoites and mature merozoites. Immunoprecipitation with MAb 1A5 followed by mass spectrometry …
