The development of therapies that can be applied in human health care to promote longer and healthier lives is one of the main goals of aging research. However, many of the interventions currently available have serious adverse effects that limit their translational potential. Thus, new strategies are urgently needed to promote healthy aging in humans. Here, we reveal a functional interaction between the insulin signaling pathway and mTOR pathway that promotes healthy aging in C. elegans via lysosomal tubulation. Their respective transcription factors, DAF-16/FOXO and HLH-30/TFEB cooperate, to drive the formation of tubular lysosomes (TLs) under various contexts, which contributes …

An early event after lung injury is extracellular matrix (ECM) remodeling and the formation of a provisional matrix. While megakaryocytes and platelets (Mgk/plt) play important roles in hemostasis, their impact on the matrix in lung injury is not fully understood. Using lung intravital microscopy, 3D large area scanning with multiphoton confocal hybrid imaging, and label-free quantitative proteomics, the matricellular protein thrombospondin-1 (TSP1) arising from Mgk/plt protects the lung from alveolar injury. Mgk/plt cell-specific Thbs1 knockout (cKO) mice show increased alveolar barrier disruption with exaggerated neutrophil-mediated injury. Organisms commonly respond to stressors in their environment by increasing the secretion of corticosterone (CORT). While increased CORT is acknowledged as a response to predation risk by organisms inhabiting a landscape of fear, new research raises the potential for changes in CORT as a response to the risk of infection by organisms in their environment. An increase in CORT of a healthy, uninfected individual in response to their perception of the risk of infection in their environment could change their susceptibility to infection, highlighting a significant role for CORT to alter host–pathogen dynamics. Recent advancements in single-cell DNA methylation (scDNAm) sequencing technologies have enabled the profiling of epigenetic landscapes at unprecedented resolution, offering insights into cellular heterogeneity, differentiation and evolution. Trajectory inference, which orders cells along pseudotime, allows researchers to track genomics changes across continuous cell states and identify key loci exhibiting differential methylation. However, no methods currently exist to model methylation changes along pseudotime in scDNAm data. Here, we present a hierarchical Bayesian framework for scDNAm data analysis. How can the molecules that strengthen synaptic connections maintain memory in the face of molecular turnover? Our previous work showed that persistent interaction between the postsynaptic scaffolding protein, KIBRA, and the autonomously active PKC isoform, PKMζ, is crucial for maintaining synaptic long-term potentiation (LTP) and memory lasting at least a month. This duration is longer than the lifespans of individual KIBRA and PKMζ molecules. Biophysical modeling of the interaction suggests oligomers of KIBRA-PKMζ dimers, but not individual dimers or monomers, can overcome molecular turnover by continuously incorporating newly synthesized KIBRA and PKMζ, replacing those that have degraded. Although autophagy, which is the process by which cells reallocate nutrients and eliminate unwanted materials, has been studied for decades, the connections between autophagy, inflammation, and viral immunity are still under investigation. Our studies focus on two genes: murine Epg5, which is involved in fusion of the autophagosome and lysosome, and C. elegans atg-3, which is important for the elongation of the phagophore. Here, we find that Epg5 mutation protects against enteric viruses including norovirus and rotavirus. We identify IFN- responsive genes as a key alteration and observe that Epg5 mutant mice exhibit substantial alterations in the intestinal microbiota. Surprisingly, …

Household Expenditure And Insurance Adoption As Determinants Of Financial Risk In Urban Companion Animal Healthcare, Hong Yun Wu, Chee Vui Wong, Changh Hao Han

Urban households increasingly face financial exposure associated with companion animal healthcare in market-based systems where costs are largely borne out of pocket. This study examines how household expenditure and insurance adoption shape financial risk in urban companion animal healthcare. Using survey data from 500 urban households, the analysis investigates expenditure patterns, predictors of pet insurance adoption, and spatial variation in healthcare spending across urban districts. Regression results show that household income, number of pets owned, educational attainment, and age are significantly associated with variation in annual healthcare expenditure. Porphyromonas gingivalis is described as a keystone pathogen associated with periodontal disease (PD), which exhibits enhanced representation upon microbial dysbiosis in such a chronic inflammatory disease. This oral pathogen drives and contributes to a dysregulated immune response, resulting in stages of aggressive destructive immune activation and inflammation punctuated by immune suppression, which underlies the relapsing–remitting nature of this disease. The understanding of key mechanisms and balance between protective innate, adaptive immune responses and dysregulated responses, linked to changes in the oral mucosal microbial environment, will afford researchers the potential to manipulate oral mucosal environments for clinical benefit. We aim to develop an in vivo hematopoietic stem cell (HSC) gene therapy approach for the prevention and control of HIV-1 infection. Toward this goal, we engineered helper-dependent adenovirus (HDAd) 6/3+ vectors to directly transduce HSCs in vivo, enabling progeny cells to secrete eCD4-Ig, a decoy protein that broadly neutralizes HIV/simian immunodeficiency virus (SIV) isolates by mimicking the primary viral receptor CD4 and coreceptors such as CCR5. In rhesus macaques, the HDAd 6/3+ platform achieved long-term expression of an enhanced eCD4-Ig variant (“eCD4-Ig-Emm06”) that retained potent neutralization efficacy in vivo. Purpose – This study explores perceptions and reactions to earthquake preparedness education, earthquake early warning (EEW) systems and earthquake drill protocols in Mexico, as well as potential strategies to make these earthquake preparedness tools more effective and meaningful. Design/methodology/approach – Eleven Focus Groups were conducted with 109 students from diverse academic and social backgrounds of the two largest public universities in Mexico. The findings were validated on a broader participant group using questionnaires, focused solely on the drill, administered to two populations at two distinct institutions following the 2025 National Earthquake Drill in Mexico City. Cosplay is a fun way to express oneself through costumes and roleplay that has given rise to a vibrant global community. While various studies have explored certain elements of this phenomenon, the cosplay community remains partially unexplored. This research aims to further investigate the dynamics of the cosplay community, providing meaningful insights into the identity processes, motivations, sense of belonging, and creativity that define it. Using in-depth interviews and online questionnaires, the research employs an inductive thematic analysis to examine how cosplayers interact with community-building, creativity, identity, and self-expression. Streptococcus gallolyticus subsp. gallolyticus (Sgg) is a gram-positive, non-beta-hemolytic bacterium that belongs to the Lancefield group D of streptococcal pathogens. For the past several decades, associations between Sgg bacteremia and colorectal cancer (CRC) have been apparent, with gastroenterologists routinely recommending colonoscopies for positive cases of Sgg in either blood or stool samples. However, current research has not wholly established whether Sgg acts as a causative agent of CRC or if pre-existing cancers make the lumen of the colon more hospitable to its colonization. A system for interactive visual disambiguation can visually identify and highlight uncertain terms within a user’s natural language input. Red blood cells (RBCs) transport oxygen but accumulate oxidative damage over time, reducing function in vivo and during storage, critical for transfusions. To explore the genetics of RBC resilience, we profiled proteins, metabolites, and lipids from fresh and stored RBCs from 350 genetically diverse mice. Our analysis identified over 6,000 quantitative trait loci (QTLs). Compared to other tissues, the prevalence of trans genetic effects over cis ones reflects the absence of de novo protein synthesis in anucleated RBCs. QTL hotspots at Hbb, Hba, Mon1a, and (storage-specific) Steap3 linked ferroptosis to hemolysis. Proteasome QTLs clustered at multiple loci, underscoring the importance …

In vitro models have tremendous utility for improving our understanding of complex physiological and pathophysiological processes in a more controlled environment than animal models, in addition to incorporating human cells to overcome species-specific disparities. This dissertation describes innovations in biomaterial-based fabrication processes that advance in vitro models of the blood-brain barrier, providing the opportunity to evaluate the effects of cell-matrix interactions on barrier integrity, additives to reduce viscous drag of blood flow, and molecular mechanisms underlying the cellular response to amyloid beta in the context of Alzheimer’s disease. This report explores how screen production educators in the ASPERA community are responding to the rapid integration of generative Artificial Intelligence (genAI) in teaching and learning. Framed by Sean Cubitt’s (2020) notion of Common Intelligence, we investigate two guiding questions: How are students engaging with genAI tools in their productions, and what are the implications for educators? Methodologically, we engaged in qualitative professional dialogue with screen practice educators through three mechanisms during the 2024 ASPERA Conference: lightning provocations, an anonymous Mentimeter survey, and a facilitated roundtable discussion. Xerostomia, the subjective sensation of dry mouth, is a debilitating consequence of head and neck radiotherapy and autoimmune disorders such as Sjögren’s syndrome. It severely impairs oral health and quality of life by promoting mucosal ulceration, infection, malnutrition, and speech difficulties, yet effective regenerative treatments remain limited. Macrophages have recently emerged as critical regulators of salivary gland repair through their roles in coordinating inflammation, fibrosis, and epithelial regeneration; however, the molecular mechanisms governing macrophage activation and function in the injured salivary gland remain poorly defined.
