Active Toxoplasmosis Infection: Efficacy And Safety Of Treatment Options, Daniele Krouse Apr 2023

Active Toxoplasmosis Infection: Efficacy And Safety Of Treatment Options, Daniele Krouse

Physician Assistant Scholarly Project Papers

This literature review intends to investigate the safety and efficacy of two different drug treatment regimens, pyrimethamine versus trimethoprim-sulfamethoxazole, for active toxoplasmosis infections. Affecting nearly 11% of the United States population and up to 80% of the populations of tropical countries, Toxoplasmosis infection is the second leading cause of death from food borne illness. Current classic management of active infection with pyrimethamine is correlated with numerous undesirable side effects, high cost and need for complex use of adjunctive medications. It is necessary to identify alternative treatment options with fewer side effects, less complex regimens, lower cost, and easier access for …

For the final review, ten articles were selected. After thorough analyses and comparisons of …

Milton’S Learning: Complementarity And Difference In Paradise Lost, Peter Spaulding Apr 2023

Milton’S Learning: Complementarity And Difference In Paradise Lost, Peter Spaulding

Dissertations (1934 -)

When we consider, in the vein of Golda Werman’s Milton and Midrash, the idea of Milton’s Paradise Lost as self-consciously responding to the Bible, the question of why he makes the changes and additions that he does comes to the fore. This dissertation explores the middle books of Paradise Lost as Milton’s midrashic interventions that, among other things, emphasize the presence of education in the Garden. These scenes shed some light on Milton’s own views of education. Specifically, these interventions show a theory of education that conceives of difference as non-combative, a distinctly non-Hobbesian view of difference. Using Aristotle’s four …

Evaluating The Impact Of Land Use Changes, Drivers Of Tmdl Development, And Green Infrastructure On Stream Impairments, Charitha Jayasri Gunawardana Apr 2023

Evaluating The Impact Of Land Use Changes, Drivers Of Tmdl Development, And Green Infrastructure On Stream Impairments, Charitha Jayasri Gunawardana

Dissertations (1934 -)

Despite the water quality improvements and regulatory advancements over the last 50 years since the enactment of the Clean Water Act, water bodies within the United States are still impaired for a broad range of contaminants from non-point source pollution. Improving watershed management approaches to meet this challenge will require a greater understanding of (1) how changes within a watershed, such as changing land use, impact stream water quality, (2) what influence socioeconomic, spatial and political factors may have on the progress towards meeting water quality goals, such as those set within Total Maximum Daily Loads (TMDLs), and (3) how …

Neuronal Glutamate Transporters Modulate Reward-Based Behaviors In Mice, Corey Nilon Apr 2023

Neuronal Glutamate Transporters Modulate Reward-Based Behaviors In Mice, Corey Nilon

Biological Sciences

Obsessive-compulsive disorder (OCD) is a neuropsychiatric disorder characterized by uncontrollable, intrusive thoughts (obsessions), restlessness, and repetitive behaviors (compulsions). One of the genes associated with OCD encodes the neuronal glutamate transporter EAAC1. EAAC1 is expressed in the striatum, the main input nucleus of the basal ganglia, implicated with reward, which shows patterns of hyperactivity in patients with OCD. What remains unknown is how EAAC1 contributes to the different behavioral abnormalities of OCD. To address this question, we performed a set of behavioral tests in EAAC1-/- mice. Specifically, we asked how the execution of repetitive reward-based behaviors changed in these mice. Our …

A Design, Development, And Evaluation Of Bio-Patch For Myocardial Tissue Repair, Yaya Zhang Apr 2023

A Design, Development, And Evaluation Of Bio-Patch For Myocardial Tissue Repair, Yaya Zhang

Theses and Dissertations (ETD)

Introduction. Ischemic heart disease (IHD) is a major concern of human health issue. The structural repair and functional recovery of injured hearts is a major challenge in the clinical setting. Preclinical studies over last 10 years have demonstrated the potential of using stem cells to treat IHD, but the efficacy of this therapy is jeopardized by uncontrollable migration and low survival of the injected stem cells. An approach based on tissue engineering enabling target-specific stem cell delivery such as cardiac patches has emerged as an alternative solution for stem cell therapy for IHD. It employs scaffold materials to engulf various …

Molecular And Cellular Investigations Of Prader-Willi Syndrome, Anna K. Victor Apr 2023

Molecular And Cellular Investigations Of Prader-Willi Syndrome, Anna K. Victor

Theses and Dissertations (ETD)

Prader-Willi syndrome (PWS) is a complex multigenic neurodevelopmental disorder resulting in hypotonia, developmental delay, hypogonadism, sleep dysfunction and childhood onset obesity affecting 1 in 10,000 to 30,000 individuals. PWS is an imprinting disorder that is caused by a loss of expression of maternally imprinted genes in the 15q11.2-q13 region including NDN, MAGEL2, SNRPN/SNURF, and a cluster of snoRNAs. The majority of cases are caused by inheriting a paternal allele deletion of this region (65-75%) and a smaller number are caused by chromosome 15 maternal uniparental disomy (UPD) (20-30%) or imprinting center defects (1-3%). Here, we used dental pulp stem cells …

Modulation Of Cerebral Artery Smooth Muscle Bk Channel Function By Neurosteroids, Kelsey Caroline Cleland North Apr 2023

Modulation Of Cerebral Artery Smooth Muscle Bk Channel Function By Neurosteroids, Kelsey Caroline Cleland North

Theses and Dissertations (ETD)

Voltage-gated and Ca2+ activated K+ channels of large conductance (BK) channels are essential for many key physiological processes, such as controlling the contraction of smooth muscle, including cerebrovascular smooth muscle. Importantly, BK channels are made up of a homotetramer of α subunits which associate with accessory β subunits. These β subunits alter the activity of the channel; this modulation is subunit-specific. Moreover, these accessory subunits have tissue-specific expression, such that the β1 subunit is highly expressed within cerebrovascular smooth muscle and greatly increases the open probability of BK channels. Increased BK channel open probability is linked to increased dilation of …

Vascular Mechanisms Of Rhabdomyolysis-Induced Acute Kidney Injury, Jeremiah Moyinolu Afolabi Apr 2023

Vascular Mechanisms Of Rhabdomyolysis-Induced Acute Kidney Injury, Jeremiah Moyinolu Afolabi

Theses and Dissertations (ETD)

In patients with rhabdomyolysis, the overwhelming release of myoglobin into circulation is the primary cause of kidney injury. Myoglobin causes direct kidney injury as well as severe renal vasoconstriction. An increase in renal vascular resistance (RVR) results in renal blood flow (RBF) and glomerular filtration rate (GFR) reduction, tubular injury, and acute kidney injury (AKI). The mechanisms that underlie rhabdomyolysis-induced AKI are not fully understood but may involve the local production of vasoactive mediators in the kidney. Myoglobin stimulated endothelin-1 (ET-1) production in proximal tubular cells, an effect driven by NADPH oxidase-dependent oxyradical generation. Rats subjected to glycerol-induced rhabdomyolysis also …

It’S What Our Grandmas Did: Decoding Black Wellness, Joy, And Liberation In Contemporary Digital Hoodoo Practice Through A Cooperative Healing Literacies Framework, Patrick Li , '23

Senior Theses, Projects, and Awards

Hoodoo, Conjure, and Rootwork refer to the constellation of African American traditions of spiritual and physical healing, ancestral devotion, retaliation, and protection created in response to chattel slavery. The current study investigates the multimodal potentials of Hoodoo for holistic healing in the contemporary digital age. Expanding on Erika Gault’s definition of digital Black religion studies as “the study of Black folks’ digital pathways to healing and wholeness (freedom) and their religious contribution to the development of digital technology,” I triangulate a framework of Hoodoo Healing Literacies (HHLs) using practices of Rootwork, Conjure, and CyberHoodoo. These practices correlate to bio-psycho, psycho-spiritual, …

Swimming Robots Using Memory Metal, Joshua R. Heckman , '23 Apr 2023

Swimming Robots Using Memory Metal, Joshua R. Heckman , '23

Senior Theses, Projects, and Awards

While in the very early stages of life, Barnacle nauplii are only a few micrometers in length, which causes them to experience a very low Reynolds number while swimming through water. It is important to understand how these biological creatures swim, so their movements can be mimicked and incorporated into robotics. Currently, there are no autonomous underwater vehicles (AUV) that are capable of carrying out missions in fluids with low Reynolds numbers. This project aimed to replicate the movement of a Barnacle Napuli with the use of muscle memory wire, in hopes that one day a AUV that can operate …

Cov And Underspecified Nouns: A Syntactic And Semantic Analysis Of Hmong Classifiers, William L. Ball , '23 Apr 2023

Cov And Underspecified Nouns: A Syntactic And Semantic Analysis Of Hmong Classifiers, William L. Ball , '23

Senior Theses, Projects, and Awards

Hmong nominal classifiers are quite complex. One clear rule however is that double classifiers are not allowed. Surprisingly, then, there is a double classifier construction involving the classifier cov and underspecified nouns that appears to break these rules. The goal of this thesis is to resolve this problem, syntactically and semantically modeling Hmong classifiers along the way. After giving background on the literature on Hmong classifiers, I develop a syntactic model for Hmong classifiers based on the Minimalist Program and Distributed Morphology, and use Link’s semantic model of plurality to make sure the syntactic model works out semantically. Then I …

The Future Of Property, Yael R. Lifshitz, Maytal Gilboa, Yotam Kaplan Apr 2023

The Future Of Property, Yael R. Lifshitz, Maytal Gilboa, Yotam Kaplan

Cardozo Law Review

Property law focuses predominantly on spatial conflicts of interest between neighbors but neglects temporal conflicts between generations. This lack of attention to the temporal dimension leads to a troubling mismatch in property law: while property rights last forever, the corresponding duties that require property holders to respect the interests of others are remarkably short lived. The result is that property law currently does not adequately protect the rights of future generations. In this Article, we offer a blueprint for correcting this anomaly. We advocate a change in the current conception of property and propose that property law focus more on …

Chronic liver disease is a major public health burden worldwide1. Although different aetiologies and mechanisms of liver injury exist, progression of chronic liver disease follows a common pathway of liver inflammation, injury and fibrosis2. Here we examined the association between clonal haematopoiesis of indeterminate potential (CHIP) and chronic liver disease in 214,563 individuals from 4 independent cohorts with whole-exome sequencing data (Framingham Heart Study, Atherosclerosis Risk in Communities Study, UK Biobank and Mass General Brigham Biobank). CHIP was associated with an increased risk of prevalent and incident chronic liver disease (odds ratio = 2.01, 95% confidence interval (95% CI) [1.46, …

Mutations in a diverse set of driver genes increase the fitness of haematopoietic stem cells (HSCs), leading to clonal haematopoiesis1. These lesions are precursors for blood cancers2-6, but the basis of their fitness advantage remains largely unknown, partly owing to a paucity of large cohorts in which the clonal expansion rate has been assessed by longitudinal sampling. Here, to circumvent this limitation, we developed a method to infer the expansion rate from data from a single time point. We applied this method to 5,071 people with clonal haematopoiesis. A genome-wide association study revealed that a common inherited polymorphism in the …

Upon stimulation by extrinsic stimuli, stem cells initiate a programme that enables differentiation or self-renewal. Disruption of the stem state exit has catastrophic consequences for embryogenesis and can lead to cancer. While some elements of this stem state switch are known, major regulatory mechanisms remain unclear. Here we show that this switch involves a global increase in splicing efficiency coordinated by DNA methyltransferase 3α (DNMT3A), an enzyme typically involved in DNA methylation. Proper activation of murine and human embryonic and haematopoietic stem cells depends on messenger RNA processing, influenced by DNMT3A in response to stimuli. DNMT3A coordinates splicing through recruitment …

WWOX gene loss-of-function (LoF) has been associated with neuropathologies resulting in developmental, epileptic, and ataxic phenotypes of varying severity based on the level of WWOX dysfunction. WWOX gene biallelic germline variant p.Pro47Thr (P47T) has been causally associated with a new form of autosomal recessive cerebellar ataxia with epilepsy and intellectual disability (SCAR12, MIM:614322). This mutation affecting the WW1 protein binding domain of WWOX, impairs its interaction with canonical proline-proline-X-tyrosine motifs in partner proteins. We generated a mutant knock-in mouse model of Wwox P47T mutation that phenocopies human SCAR12. Wwox

Cerebellar hypoplasia and dysplasia encompass a group of clinically and genetically heterogeneous disorders frequently associated with neurodevelopmental impairment. The Neuron Navigator 2 (NAV2) gene (MIM: 607,026) encodes a member of the Neuron Navigator protein family, widely expressed within the central nervous system (CNS), and particularly abundant in the developing cerebellum. Evidence across different species supports a pivotal function of NAV2 in cytoskeletal dynamics and neurite outgrowth. Specifically, deficiency of Nav2 in mice leads to cerebellar hypoplasia with abnormal foliation due to impaired axonal outgrowth. However, little is known about the involvement of the NAV2 gene in human disease phenotypes. In …
