Of all the claims in the Declaration of Independence, its surety about the existence of an intentional British “design to reduce” the colonists “under absolute Despotism” is perhaps the most questionable one to modern ears. Contemporary historians have largely dismissed such language, and the accompanying concerns about an alleged British plot to “enslave” its Atlantic possessions. However, this paper argues that such a view fails to properly consider the role of “the Irish precedent” of English imperial exploitation in sparking American resistance and rebellion. Polyploidy is common among plants and can contribute to physiological and morphological differences, altering how plants respond to environmental changes, promoting genetic diversification, and even species radiation. Quaking aspen (Populus tremuloides), a keystone species associated with high plant and animal diversity is frequently found in mixed diploid/triploid populations in the Intermountain West. High mortality rates and widespread population declines in aspen are of increasing concern in the Intermountain West, often ascribed to changing climates and drought stress events. The mesolimbic dopamine (DA) system originates in the ventral tegmental area (VTA) and projects to the nucleus accumbens (NAc) and other areas including the basolateral amygdala (BLA), prefrontal cortex, and the hippocampus. Drug use induces reward and leads to dysregulation in these brain areas and eventually to substance use disorders (SUDs). Chapter 1 introduces the mesolimbic DA system and its relationship to drug use and their relevance to each chapter. Rotavirus (RV) is the main pathogen that causes severe diarrhea in infants and children under 5 years of age. No specific antiviral therapies or licensed anti-rotavirus drugs are available. It is crucial to develop effective and low-toxicity anti-rotavirus small-molecule drugs that act on novel host targets. In this study, a new anti-rotavirus compound was selected by ELISA, and cell activity was detected from 453 small-molecule compounds. The anti-RV effects and underlying mechanisms of the screened compounds were explored. Eye infections from bacterial contamination of bulk-refillable liquid soap dispensers and artificial tear eye drops continue to occur, resulting in adverse health outcomes that include impaired vision or eye enucleation. Pseudomonas aeruginosa (P. aeruginosa), a common cause of eye infections, can grow in eye drop containers and refillable soap dispensers to high numbers. Using mixed methods and multimedia approaches, I investigated the multigenerational perspectives and seasonal changes in Utqiaġvik’s spring whaling. As an Iñupiaq living in my home community of Utqiaġvik, it was important to me to center my community’s knowledge and Iñupiaq-led entities. I used various tools and resources such as conducting semi-directed interviews, hiring a local Iñupiaq videographer to take photos and videos of spring whaling preparations, and using observational data from the Alaska Arctic Observatory and Knowledge Hub. Counselors and qualitative researchers have the honor of hearing peoples’ stories and thus have a great responsibility to explore and use clinical and research methodologies that are anti-oppressive, liberatory, and healing. Therefore, in 2019 we began a photovoice project alongside seven queer womxn of color (QWoC) that collaboratively explored their experiences of microaggressions in counseling. Through this journey, we recognized that to be fully present with the “co-researchers’” (participants’) narratives and experiences, we needed to remain attuned and grounded. This phenomenological study explored the lived experiences of teachers when enrolled in an asynchronous pre-service educator certification program. This study was guided by John Keller’s theory of motivation, as it explained the motivational factors that influence the Attention, Relevance, Confidence, and Satisfaction (ARCS) model for teachers who have completed an asynchronous educator certification program. A qualitative hermeneutical phenomenology design was combined with snowball and convenience sampling to enroll 10 participants from a school district in the Midwest. The Scriptures contain a theology of dung. When the semantic and conceptual cognates that pertain to dung are synthesized, the result is a scriptural scatology whereby dung is presented as a symbol for sin. To clarify, the biblical exhibition of dung presents excrement as a central symbol employed by Scripture to concretize the abstract nature of sin. When each semantic and conceptual cognate that pertains to dung is examined, in nearly every instance where dung is mentioned, sin is in close proximity. Autism Spectrum Disorder (ASD) is a neurodevelopmental disorder that is diagnosed when an individual has persistent deficits in social communication and restrictive repetitive patterns of behaviors, interests, or activities. Restricted repetitive behaviors (RRBs) are typically triggered by anxiety and can cause significant detriment to an individual's life. The purpose of this embedded single-case study was to understand how participation in work-based learning experiences (WBLE) prepares students with intellectual disabilities (ID) for successful post-school competitive employment. The theory guiding this study was Kolb’s experiential learning theory, as it systematically explains the four stages of learning that learners go through during WBLE. The mainstays of lung cancer pathogenesis are cell cycle progression dysregulation, impaired apoptosis, and unregulated cell proliferation. While individual microRNA (miR) targeting or delivering is a promising approach that has been extensively studied, combination of miR targeting can enhance therapeutic efficacy and overcome limitations present in individual miR regulations. We previously reported on the use of a miR-143 and miR-506 combination via transient transfections against lung cancer. Transfected stem cells and T cells are promising in personalized cell therapy and immunotherapy against various diseases. However, existing transfection techniques face a fundamental trade-off between transfection efficiency and cell viability; achieving both simultaneously remains a substantial challenge. This study presents an acoustothermal transfection method that leverages acoustic and thermal effects on cells to enhance the permeability of both the cell membrane and nuclear envelope to achieve safe, efficient, and high-throughput transfection of primary T cells and stem cells. Photoreceptors are electrically coupled to one another, and the spatiotemporal properties of electrical synapses in a two-dimensional retinal network are still not well studied, because of the limitation of the single electrode or pair recording techniques which do not allow simultaneously measuring responses of multiple photoreceptors at various locations in the retina. A multiple electrode recording system is needed. The human medial temporal lobe (MTL) plays a crucial role in recognizing visual objects, a key cognitive function that relies on the formation of semantic representations. Nonetheless, it remains unknown how visual information of general objects is translated into semantic representations in the MTL. Furthermore, the debate about whether the human MTL is involved in perception has endured for a long time.
