Ethanolamine (EA) affects the colonization and pathogenicity of certain human bacterial pathogens in the gastrointestinal tract. However, EA can also affect the intracellular survival and replication of host cell invasive bacteria such as Listeria monocytogenes (LMO) and Salmonella enterica serovar Typhimurium (S. Typhimurium). The EA utilization (eut) genes can be categorized as regulatory, enzymatic, or structural, and previous work in LMO showed that loss of genes encoding functions for the enzymatic breakdown of EA inhibited LMO intracellular replication. In this work, we sought to further characterize the role of EA utilization during LMO infection of host …

Advances in perinatal intensive care have significantly enhanced the survival rates of extremely low gestation-al-age neonates but with continued high rates of bronchopulmonary dysplasia (BPD). Nevertheless, as the survival of these infants improves, there is a growing awareness of associated abnormalities in pulmonary vascular development and hemodynamics within the pulmonary circulation. Premature infants, now born as early as 22 weeks, face heightened risks of adverse development in both pulmonary arterial and venous systems. This risk is compounded by parenchymal and airway abnormalities, as well as factors such as inflammation, fibrosis, and adverse growth trajectory. The presence of pulmonary hypertension in …

Hemojuvelin (HJV) is a glycosylphosphatidylinositol-anchored protein of the repulsive guidance molecule family acting as a bone morphogenetic protein (BMP) coreceptor to induce the hepatic iron regulatory protein hepcidin. Hepcidin causes ubiquitination and degradation of the sole known iron exporter ferroportin, thereby limiting iron availability. The detailed signaling mechanism of HJV in vivo has yet to be investigated. In the current manuscript, we used an established model of adeno-associated virus (AAV)-mediated liver-specific overexpression of HJV in murine models of hepatocyte-specific deficiency of the BMP type I receptors Alk2 or Alk3. In control mice, HJV overexpression increased hepatic Hamp messenger RNA (mRNA) …

Recent single-cell RNA sequencing studies have revealed distinct microglial states in development and disease. These include proliferative-region-associated microglia (PAMs) in developing white matter and disease-associated microglia (DAMs) prevalent in various neurodegenerative conditions. PAMs and DAMs share a similar core gene signature. However, the extent of the dynamism and plasticity of these microglial states, as well as their functional significance, remains elusive, partly due to the lack of specific tools. Here, we generated an inducible Cre driver line, Clec7a-CreER

The use of acellular nerve allografts (ANAs) to reconstruct long nerve gaps (>3 cm) is associated with limited axon regeneration. To understand why ANA length might limit regeneration, we focused on identifying differences in the regenerative and vascular microenvironment that develop within ANAs based on their length. A rat sciatic nerve gap model was repaired with either short (2 cm) or long (4 cm) ANAs, and histomorphometry was used to measure myelinated axon regeneration and blood vessel morphology at various timepoints (2-, 4- and 8-weeks). Both groups demonstrated robust axonal regeneration within the proximal graft region, which continued across …

The rent stabilized tenant of record ("landlord" for the purposes of record structure on this project website) initiated a holdover proceeding against their subtenants, seeking to recover possession of the subject premises. The subtenants interposed an answer with affirmative defenses and counterclaims. The court denied the subtenants' motion for discovery and denied the landlord's motion for summary judgment. The court found that the subtenants failed to plead a valid illusory tenancy affirmative defense and that there were disputed issues of fact requiring trial regarding whether a month-to-month tenancy was created after expiration of the sublease agreement. The court also noted …

RNA binding proteins have emerged as central players in the mechanisms of many neurodegenerative diseases. In particular, a proteinopathy of fused in sarcoma (FUS) is present in some instances of familial Amyotrophic lateral sclerosis (ALS) and about 10% of sporadic Frontotemporal lobar degeneration (FTLD). Here we establish that focal injection of sonicated human FUS fibrils into brains of mice in which ALS-linked mutant or wild-type human FUS replaces endogenous mouse FUS is sufficient to induce focal cytoplasmic mislocalization and aggregation of mutant and wild-type FUS which with time spreads to distal regions of the brain. Human FUS fibril-induced FUS aggregation …

Type 2 diabetes mellitus (T2DM) increases the risk of non-alcoholic fatty liver disease (NAFLD) progression to advanced stages, especially upon high-fat diet (HFD). HFD-induced hepatic fibrosis can be marked by oxidative stress, inflammation, and activation of hepatic stellate cells. Sirtuin 1/2 (SIRT1/2), NAD-dependent class III histone deacetylases, are involved in attenuation of fibrosis. In our conducted research, TGF-β1-activated LX-2 cells, free fatty acid (FFA)-treated simultaneous co-culture (SCC) cells, and HFD-induced hepatic fibrosis in Zucker diabetic fatty (ZDF) rats, a widely used animal model in the study of metabolic syndromes, were used to evaluate the protective effect of Tenovin-1, a SIRT1/2 …

Human brain organoid models have emerged as a promising tool for studying human brain development and function. These models preserve human genetics and recapitulate some aspects of human brain development, while facilitating manipulation in an in vitro setting. Despite their potential to transform biology and medicine, concerns persist about their fidelity. To fully harness their potential, it is imperative to establish reliable analytic methods, ensuring rigor and reproducibility. Here, we review current analytical platforms used to characterize human forebrain cortical organoids, highlight challenges, and propose recommendations for future studies to achieve greater precision and uniformity across laboratories.

We aimed to investigate the therapeutic potential of ibuprofen against type 2 diabetes (T2D) using obese Zucker diabetic fatty (ZDF) rats as type 2 diabetes model. ZDF rats were hyperglycemic, dyslipidemic and expressed pro-inflammatory markers in contrast to lean controls, thus reflecting the relationship between obesity and chronic inflammation promoting T2D. Chronic treatment with ibuprofen (2-(4-Isobutylphenyl)propanoic acid) was used to study the impact on pathological T2D conditions as compared to metformin (1,1-dimethylbiguanide) treated ZDF as well as lean controls. Ibuprofen decreased A1c but induced a high insulin release with improved glucose tolerance only after early time points (i.g., 15 and …

40 Years After The Moratorium On Commercial Whaling: Assessing The Competence Of The International Whaling Commission To Confront Critical Threats To Cetaceans, Chris Wold
Pace International Law Review
With Japan’s withdrawal from the International Convention for the Regulation of Whaling (ICRW), no member of the International Whaling Commission (IWC) hunts whales commercially except by objection or reservation, thus intensifying the debate over what the IWC should do and what it may do. For decades, some IWC members have questioned the mandate of the IWC to manage small cetaceans, regulate whaling in coastal State exclusive economic zones, and make recommendations concerning entanglement, bycatch, and other threats to whale conservation. An analysis of the ordinary meaning of the ICRW and the practice of the IWC shows that the IWC has …

Tuareg Women’S Writing: The Works Of Zakiyatou Oualett Halatine, Cheryl Toman
Journal of Amazigh Studies
In 2013, Tuareg and Malian writer, Zakiyatou oualett Halatine, was forced to flee a conflict that rages on in Mali to this day, and her creative work, Passions du désert, along with a collection of proverbs and a book-length essay, became a means of reconstructing memory and identity of a people ultimately blamed for Mali’s divisions. Zakiyatou’s texts provide us with a rare look at the desert region of Northern Mali from a woman’s perspective. Zakiyatou’s writings are unique in a corpus of Malian literature mostly generated by Southern Malian authors. In many ways, she is the lone literary …

The Secrets Of Christian Others: Hungarian Catholic Intellectuals Debate Ecumenism At A Transylvanian Pilgrimage Site, Marc Roscoe Loustau
Journal of Global Catholicism
Claims about a shared Christian tradition animate European debates about religious otherness, but more remains to be known about how Catholics on Europe’s near-margins understand ecumenical unity among churches. I analyze contemporary Hungarian Catholic intellectuals’ publications about a controversy at the Hungarian national shrine, Our Lady of Csíksomlyó, in Transylvania. When a priest wrote that Csíksomlyó’s annual pilgrimage commemorated sixteenth-century Catholics’ victory over an invading Unitarian army, Transylvania’s Unitarian bishop denounced the origin as an undocumented myth. Prominent Catholic ethnologists, historians, and theologians agreed that, in the name of ecumenism, intellectuals should not publicly mention the origin narrative. But they …

Socialization Procedure For An Adult Feral Cat In A Home Setting, Erica Serito
Thesis Projects
Of the nearly sixty million feral cats, most are deemed unadoptable and thereby unable to be rescuable given their aggressive behaviors towards humans. There are currently no evidence-based socialization training procedures used to socialize adult feral cats with the main interventions for the fearful adult feral cats being trap-neuter-return (TNR) or euthanasia. The purpose of the current study was to decrease undesirable behaviors of an adult feral cat to facilitate safe and reciprocal interactions between her and humans. The study evaluated the use of a differential reinforcement of other behavior (DRO) procedure without extinction on an adult feral cat to …
