Background/aim: Our recent study revealed that the expression of lipoxygenase (LOX) and cyclooxygenase (COX) enzymes in the hypothalamus is activated by nesfatin-1, leading to the liberation of leukotrienes and prostaglandins (PG), respectively. Moreover, our prior report explained that intracerebroventricular (ICV) nesfatin-1 treatment triggers cardiovascular responses mediated by central LOX and COX enzymes. Background/aim: Glioblastoma is one of the most aggressive tumours, resistant to all applied therapy regiments and prone to relapse. Median survival rates are therefore only expressed as months. STING agonists are immunomodulatory molecules that activate type I interferon expression, making them potentially useful in regulating the tumour microenvironment. L1CAM-positive extracellular vesicles (L1EV) are an emerging biomarker that may better reflect ongoing neuronal damage than other blood-based biomarkers. The physiological roles and regulation of L1EVs and their small RNA cargoes following stroke is unknown. We sought to characterize L1EV small RNAs following stroke and assess L1EV RNA signatures for diagnosing stroke using weighted gene co-expression network analysis and random forest (RF) machine learning algorithms. Interestingly, small RNA sequencing of plasma L1EVs from patients with stroke and control patients (n = 28) identified micro(mi)RNAs known to be enriched in the brain. Volumetric data provide unprecedented structural insight to the reproductive tract and add vital anatomical context to the relationships between organs. The morphology of the female reproductive tract in non-avian reptiles varies between species, corresponding to a broad range of reproductive modes and providing valuable insight to comparative investigations of reproductive anatomy. However, reproductive studies in reptilian models, such as the brown anole studied here, have historically relied on histological methods to understand the anatomy. Machine Learning In Time-Lapse Imaging To Differentiate Embryos From Young Vs Old Mice†, Liubin Yang, Carolina Leynes, Ashley Pawelka, Isabel Lorenzo, Andrew Chou, Brendan Lee, Jason D Heaney

Time-lapse microscopy for embryos is a non-invasive technology used to characterize early embryo development. This study employs time-lapse microscopy and machine learning to elucidate changes in embryonic growth kinetics with maternal aging. We analyzed morphokinetic parameters of embryos from young and aged C57BL6/NJ mice via continuous imaging. Our findings show that aged embryos accelerated through cleavage stages (from 5-cells) to morula compared to younger counterparts, with no significant differences observed in later stages of blastulation. Unsupervised machine learning identified two distinct clusters comprising of embryos from aged or young donors. Lake Edku is one of the most important lakes in the northern Nile Delta of Egypt. Studying the physicochemical characteristics and nutrient salts is essential for assessing water quality. pH recorded values fell within internationally permitted limits; the temperature was suitable for the growth of fish and living organisms. Electrical conductivity indicates a source of fresh water; salinity indicates that the lake water is brackish; Total Dissolved Solids (TDS) refers to most of the fresh water in the lake. The study aimed to enhance the durability and flexural strength of fiberglass composites by utilizing water hyacinth and surgical mask fiber ratios, with a focus on reinforcing water hyacinth fibers with polyester-containing surgical mask fibers. Employing a posttest-only controlled group design, the research tested various ratios, including a control group and three experimental groups, to evaluate the mechanical properties of the composites. The results revealed that Experimental 1 (25:25:30) exhibited highest durability (97.33 MPa). Notably, the optimal ratio for the durability was found in Experimental 2 (96.33 MPa) (20:30:50). Studies have shown that academic stress, which comes from overwhelming academic demands, causes various negative effects on the well-being of students. To better represent online health communication modes, an English-language corpus of lay health discourse was expanded to include health-related memes. Using these forms of peer-to-peer lay communication, I propose ways to categorize the illocutionary force of memes when they function as speech acts of advice. The subcorpus contains 200 English-language health-related memes gathered during 2018-2023 through Google image searches. I found the affiliative and disaffiliative strategies that Placencia (2012) noted within Yahoo! Microbial communities comprising bacteria, viruses, fungi, and protists live within and on the surfaces of animal hosts. These microbial communities exist in symbiosis with the host, and heavily influence host physiology, development, health, and fitness. Gut-dwelling microbes (i.e., gut microbiota) contribute to host neurodevelopment through a bidirectional Microbiota-Gut-Brain (MGB) axis. Aim: Alzheimer’s disease (AD) is the most common form of dementia. However, while 150+ animal models of AD exist, drug translation from preclinical models to humans for treatment usually fails. One factor contributing to low translation is likely the absence of neurodegenerative models that also encompass the multi-morbidities of human aging. We previously demonstrated that, in comparison to the PigmEnTed (PET) guinea pig strain which models “typical” brain aging, the Hartley strain develops hallmarks of AD like aging humans. Recent single-cell RNA sequencing studies have revealed distinct microglial states in development and disease. These include proliferative region-associated microglia (PAM) in developing white matter and disease-associated microglia (DAM) prevalent in various neurodegenerative conditions. PAM and DAM share a similar core gene signature. However, the extent of the dynamism and plasticity of these microglial states, as well as their functional significance, remains elusive, partly due to the lack of specific tools. Uveal melanoma (UM) is the deadliest form of eye cancer in adults. Inactivating mutations and/or loss of expression of the gene encoding BRCA1-associated protein 1 (BAP1) in UM tumors are associated with an increased risk of metastasis. To investigate the mechanisms underlying this risk, we explored the functional consequences of BAP1 deficiency. UM cell lines expressing mutant BAP1 grew more slowly than those expressing wild-type BAP1 in culture and in vivo. The ability of BAP1 reconstitution to restore cell proliferation in BAP1-deficient cells required its deubiquitylase activity. Investigating how infants first establish relationships between words is a necessary step towards understanding how an interconnected network of semantic relationships develops in the adult lexical-semantic system. Stimuli selection for these child studies is critical since words must be both familiar and highly imageable. However, there has been a reliance on adult word association norms to inform stimuli selection in English infant studies to date, as no resource currently exists for child-specific word associations. This study explores the ostracism and solitude-seeking link by conceptually replicating the two studies (Ren et al., 2016; 2022) using different methods of inducing the feeling of being ostracized in the participants. In this study, I asked participants to recall a memory of being ostracized to induce the feeling of being ostracized. I am also interested in if shame and guilt mediate this connection. Background: TMPRSS2, a key molecule for SARS-CoV-2 invading human host cells, has an association with cancer. However, its association with lung cancer remains insufficiently unexplored.

Methods: In five bulk transcriptomics datasets, one single-cell RNA sequencing (scRNA-seq) dataset and one proteomics dataset for lung adenocarcinoma (LUAD), we explored associations between TMPRSS2 expression and immune signatures, tumor progression phenotypes, genomic features, and clinical prognosis in LUAD by the bioinformatics approach. Furthermore, we performed experimental validation of the bioinformatics findings.

Results: TMPRSS2 expression levels correlated negatively with the enrichment levels of both immune-stimulatory and immune-inhibitory signatures, while they correlated positively with the ratios …
