Faculty, Staff and Students Publications

Background: Contrast-induced acute kidney injury (CI-AKI) continues to pose a pressing clinical challenge during invasive cardiovascular procedures due to the limited availability of preventative strategies. We aimed to demonstrate that irisin, a myokine induced by exercise, protects against CI-AKI by inhibiting the cGAS-STING inflammatory pathway.

Methods and results: We explored the relationship between serum irisin levels and CI-AKI incidence in patients administered the contrast media iohexol. Notably, lower serum irisin levels were strongly associated with an increased incidence of CI-AKI following contrast media administration. To establish a causal link between serum irisin levels and CI-AKI, we utilised a mouse model …

Anesthetic Management For Proximal Aortic Repair, Thomas R Powell, Emily B Shah, Ali Khalifa, Vicente Orozco-Sevilla, Daniel A Tolpin

Faculty, Staff and Students Publications

Surgical repair of the proximal aorta is a complex endeavor, requiring cardiopulmonary bypass (CPB) and often the use of hypothermic circulatory arrest (HCA). In addition to the normal considerations for patients undergoing cardiopulmonary bypass, additional challenges include cerebral and end-organ protection during periods of circulatory arrest. This review aims to provide an up-to-date, evidence-based review on anesthetic management for proximal aortic repair.

Visible Light Polarization-Sensitive Optical Coherence Tomography With Balanced Detection, Bahar Baradaran, Adam J Black, Sarah R Heilbronner, Taner Akkin

Faculty, Staff and Students Publications

Significance: We introduce a visible-light polarization-sensitive optical coherence tomography (PS-OCT) system that operates in the spectral domain with balanced detection (BD) capability. While the BD improves the signal-to-noise ratio (SNR), the use of shorter wavelengths improves spatial resolution and birefringence sensitivity.

Aim: We aim to implement a new optical design, characterize its performance, and investigate the imaging potential for biological tissues.

Approach: The design utilizes a unique interferometer and a custom spectrometer that captures four highly aligned spectra with a single area/multi-line camera. Each pair of spectral lines is highly aligned, and their subtraction yields balanced detected spectra of the …

Thalamic Deep Brain Stimulation Improves Movement In A Cerebellar Model Of Lesion-Based Status Dystonicus, Megan X Nguyen, Amanda M Brown, Tao Lin, Roy V Sillitoe, Jason S Gill Mar 2025

Thalamic Deep Brain Stimulation Improves Movement In A Cerebellar Model Of Lesion-Based Status Dystonicus, Megan X Nguyen, Amanda M Brown, Tao Lin, Roy V Sillitoe, Jason S Gill

Faculty, Staff and Students Publications

Dystonia is the third most common movement disorder and an incapacitating co-morbidity in a variety of neurologic conditions. Dystonia can be caused by genetic, degenerative, idiopathic, and acquired etiologies, which are hypothesized to converge on a "dystonia network" consisting of the basal ganglia, thalamus, cerebellum, and cerebral cortex. In acquired dystonia, focal lesions to subcortical network regions lead to dystonia that can be difficult to manage with canonical treatments, including deep brain stimulation (DBS). While studies in animal models have begun to parse the contribution of individual nodes in the dystonia network, how acquired injury to the cerebellar outflow tracts …

Cerebrotendinous Xanthomatosis Occurs At High Frequency In Ashkenazi Jews, Jennifer Hanson, Penelope E Bonnen

Faculty, Staff and Students Publications

Cerebrotendinous Xanthomatosis (CTX) is a treatable, inborn error of bile acids metabolism caused by pathogenic variants in CYP27A1. CTX is a multi-organ system disorder that progresses over decades. Clinical features include cerebellar dysfunction, pyramidal tract dysfunction, cognitive deficits and decline, peripheral neuropathy, chronic diarrhea, bilateral cataracts, and tendon xanthomas. Treatment is effective when started early, but diagnostic delays often result in individuals not being diagnosed until after the window of highest treatment efficacy. CTX is documented to occur in most global populations, however, no CTX-causing genetic variants have been reported in Ashkenazi Jews. We conducted a systematic review of every …

Onchocerca volvulus is the agent of onchocerciasis (river blindness) and targeted by WHO for elimination though mass drug administration with ivermectin. A small percentage of adult female worms develop pleomorphic neoplasms (PN) which occur more frequently after ivermectin treatment. Worms with PN have a lower life expectancy and improved understanding of proteins expressed in PN and their impact on different tissues could help elucidate the mechanisms of macrofilaricidal activity of ivermectin. Within paraffin embedded nodules removed after ivermectin treatment, we detected 24 (5.6%) O. volvulus females with PN. To assess the protein inventory of the PN and identify proteins potentially …

Rest And Rinse: Sleeping Rhythms Drive Brain Detox, Leon C D Smyth, Benjamin A Plog, Jonathan Kipnis

2020-Current year OA Pubs

Sleep is a major driver of waste clearance from the brain, but the mechanisms underpinning brain cleansing during sleep, which are also important for immunological functions, are poorly understood. Recent mouse work by Hauglund et al. shows how oscillatory surges in norepinephrine (NE) during sleep drive vascular pulsation and cerebrospinal fluid (CSF) movement to cleanse the brain.

Decreased Voluntary Alcohol Intake And Ventral Striatal Epigenetic And Transcriptional Remodeling In Male Acss2 Ko Mice, Gabor Egervari, Greg Donahue, Natalia A Quijano Cardé, Desi C Alexander, Connor Hogan, Jessica K Shaw, Erica M Periandri, Vanessa Fleites, Mariella De Biasi, Shelley L Berger

2020-Current year OA Pubs

Metabolic-epigenetic interactions are emerging as key pathways in regulating alcohol-related transcriptional changes in the brain. Recently, we have shown that this is mediated by the metabolic enzyme Acetyl-CoA synthetase 2 (Acss2), which is nuclear and chromatin-bound in neurons. Mice lacking ACSS2 fail to deposit alcohol-derived acetate onto histones in the brain and show no conditioned place preference for ethanol reward. Here, we further explored the role of this pathway during voluntary alcohol intake. We found that Acss2 KO mice consume significantly less alcohol in a model of binge drinking, an effect primarily driven by males. Genome-wide transcriptional profiling of 7 …

Aims/hypothesis: Upregulation of serum leucine-rich α-2-glycoprotein 1 (LRG1) has been implicated in diet-induced obesity and metabolic disorders. However, its specific hormonal actions remain unclear. This study aimed to determine whether diet-enhanced serum LRG1 levels promote hyperinsulinaemia by directly stimulating insulin secretion from pancreatic beta cells.

Methods: Human serum samples were obtained from individuals (both male and female) undergoing plastic surgery. Male C57BL/6 wild-type and Lrg1 whole-body knockout (Lrg1KO) mice were fed a 45% high-fat diet, with serum samples collected every 2 weeks to monitor LRG1 and insulin levels throughout diet-induced obesity. MIN6 beta cells were used to investigate the effects …

Occlusion-Insensitive Talking Head Video Generation Via Facelet Compensation, Yuhui Deng, Yuqin Lu, Yangyang Xu, Yongwei Nie, Shengfeng He

Research Collection School Of Computing and Information Systems

Talking head video generation involves animating a still face image using facial motion cues derived from a driving video to replicate target poses and expressions. Traditional methods often rely on the assumption that the relative positions of facial keypoints remain unchanged. However, this assumption fails when keypoints are occluded or when the head is in a profile pose, leading to inconsistencies in identity and blurring in certain facial regions. In this paper, we introduce Occlusion-Insensitive Talking Head Video Generation, a novel approach that eliminates the reliance on spatial correlation of keypoints and instead leverages semantic correlation. Our method transforms facial …

Research Collection School Of Computing and Information Systems

Personalized image generation has made significant strides in adapting content to novel concepts. However, a persistent challenge remains: balancing the accurate reconstruction of unseen concepts with the need for editability according to the prompt, especially when dealing with the complex nuances of facial features. In this study, we delve into the temporal dynamics of the text-to-image conditioning process, emphasizing the crucial role of stage partitioning in introducing new concepts. We present PersonaMagic, a stage-regulated generative technique designed for high-fidelity face customization. Using a simple MLP network, our method learns a series of embeddings within a specific timestep interval to capture …

Adversarial Attacks On Event-Based Pedestrian Detectors: A Physical Approach, Guixu Lin, Muyao Niu, Qingtian Zhu, Zhengwei Yin, Zhuoxiao Li, Shengfeng He, Yinqiang Zheng

Research Collection School Of Computing and Information Systems

Event cameras, known for their low latency and high dynamic range, show great potential in pedestrian detection applications. However, while recent research has primarily focused on improving detection accuracy, the robustness of event-based visual models against physical adversarial attacks has received limited attention. For example, adversarial physical objects, such as specific clothing patterns or accessories, can exploit inherent vulnerabilities in these systems, leading to misdetections or misclassifications. This study is the first to explore physical adversarial attacks on event-driven pedestrian detectors, specifically investigating whether certain clothing patterns worn by pedestrians can cause these detectors to fail, effectively rendering them unable …

A Comparison Of Training Procedures On The Emergence Of Intraverbal-Tacts, Mary Halbur, Tiffany Kodak, Jessi Reidy Mar 2025

A Comparison Of Training Procedures On The Emergence Of Intraverbal-Tacts, Mary Halbur, Tiffany Kodak, Jessi Reidy

Psychology Faculty Research and Publications

Vocal exchanges are often comprised of responses under multiple sources of stimulus control. For example, a picture may contain multiple components, and an instructor may ask a learner to respond differentially to questions about the picture (e.g., “who,” “what,” “where,” “color,” “number,” “shape”). The format of teaching may affect the development of verbal behavior under multiple sources of stimulus control. Therefore, the present investigation compared teaching stimuli in isolation to teaching with compound stimuli on the emergence of verbal behavior to evaluate methods that assist with correctly answering questions about compound stimuli. This study used a translational model with undergraduate …

Leveraging Artificial Intelligence And Machine Learning To Accelerate Discovery Of Disease-Modifying Therapies In Type 1 Diabetes., Melanie R. Shapiro, Erin M. Tallon, Matthew E. Brown, Amanda L. Posgai, Mark A. Clements, Todd M. Brusko

Manuscripts, Articles, Book Chapters and Other Papers

Progress in developing therapies for the maintenance of endogenous insulin secretion in, or the prevention of, type 1 diabetes has been hindered by limited animal models, the length and cost of clinical trials, difficulties in identifying individuals who will progress faster to a clinical diagnosis of type 1 diabetes, and heterogeneous clinical responses in intervention trials. Classic placebo-controlled intervention trials often include monotherapies, broad participant populations and extended follow-up periods focused on clinical endpoints. While this approach remains the 'gold standard' of clinical research, efforts are underway to implement new approaches harnessing the power of artificial intelligence and machine learning …

Computational Model Of Coarctation Of The Aorta In Rabbits Suggests Persistent Ascending Aortic Remodeling Post-Correction, Ashley A. Hiebing, Matthew A. Culver, John F. Ladisa, Colleen M. Witzenburg

Biomedical Engineering Faculty Research and Publications

Coarctation of the aorta (CoA) is a common congenital cardiovascular lesion that presents as a localized narrowing of the proximal descending aorta. While improvements in surgical and catheter-based techniques have increased short-term survival, there is a high long-term risk of hypertension and a reduced average lifespan despite correction. Computational models can be used to estimate aortic remodeling and peripheral vascular compensation, potentially serving as key tools in developing a mechanistic understanding of the interplay between pre-treatment dynamics, post-treatment recovery, and long-term hypertension risk. In this study, we developed a lumped-parameter model of the heart and circulation to simulate CoA. After …

Can Targeting The Fgf23-Αklotho Signaling System Delay Phosphate-Driven Organ Damage?, Mohammed S. Razzaque, Moosa Mohammadi

School of Medicine Publications

Introduction: Inexorable high serum phosphate levels in chronic kidney disease (CKD) patients deteriorate the functionality of the musculoskeletal, renal, and cardiovascular systems, thereby contributing to increased morbidity and mortality. Higher phosphate balance has also been correlated with increased mortality rates in individuals with normal renal function, independent of other comorbidities. Clinical and epidemiological studies of CKD patients and healthy subjects, alongside evidence of accelerated aging in murine models induced by excessive phosphate loading, indicate that phosphate toxicity is a driver of premature aging and age-related organ damage.

Area covered: This article briefly discusses the causes and consequences of phosphate toxicity …
