The 1,4-dihydropyridines, drugs with well-established bioavailability and toxicity profiles, have proven efficacy in treating human hypertension, peripheral vascular disorders, and coronary artery disease. Every 1,4-dihydropyridine in clinical use blocks L-type voltage-gated calcium channels. We now report our development, using selective optimization of a side activity (SOSA), of a class of 1,4-dihydropyridines that selectively and potently inhibit the intermediate-conductance calcium-activated K+ channel KCa3.1, a validated therapeutic target for diseases affecting many organ systems. One of these 1,4-dihydropyridines, DHP-103, blocked KCa3.1 with an IC50 of 6 nM and exhibited exquisite selectivity over calcium channels and a panel of …

Sesame Street has been around since 1969 and is still creating new episodes. Joan Ganz Cooney created the program in an attempt to use television to educate and entertain children simultaneously. The show set out goals to create smarter, kinder, and stronger kids. It uniquely featured a diverse cast and was targeted towards disadvantaged youth. However, the world has changed a lot since 1969. Remaking Rulemaking: Lessons From The History Of Federal Rule Of Civil Procedure 54(B), Brad Baranowski, Emery G. Lee Iii

This Article uses the lost history of Federal Rule of Civil Procedure 54(b) to provide a framework for rulemaking at the busy intersection of complex litigation and appellate finality. The drafters of the original civil rules spent more time thinking through how to efficiently combine claims and parties in civil actions—a promised benefit of the proposed law-equity merger—than about how combined claims should be dis-assembled. This failure would haunt the original Advisory Committee on Civil Rules for years. Nowhere was this more evident than in the first decades of Rule 54(b)’s existence.

ObjectivesThis study examines the complex dynamics of father-daughter relationships, focusing on how life history factors are associated with paternal behaviors towards daughters. Drawing on Life history Theory and the Daughter-Guarding hypothesis, the research investigates how these factors contribute to father-daughter interactions, such as attachment, protection, support, and control received from fathers.MethodsTwo surveys were conducted: one with 120 daughters aged 18–21 (Study 1) and another with 120 fathers (Study 2), both recruited through online platforms. Alzheimer’s disease is a neurological condition characterized by a significant decline in cognitive function. Due to its severe effects and the absence of a cure, early diagnosis and treatment are crucial for slowing its progression. My project examines how cultural perceptions of Alzheimer’s disease are reflected in digital discourse. Introduction: The vestibular system plays a crucial role in visual and non-visual navigation. Our recent study found that signals from the otolith organs are necessary for mice’s use of distal visual cues to guide navigation to an invisible goal. Somewhat surprisingly, however, performance was not significantly impaired on some spatial tasks (e.g., Barnes maze reference memory task), questioning the role of otolith signals in visual navigation.

The nematode, formally known as Caenorhabditis elegans, has served as an excellent model organism in understanding fundamental biological processes, including those governed by conserved genes like pros-1. pros-1 is partially responsible for the formation of tubular structures within the organism. Glial cells and excretory canals are both examples of tubular structures governed by pros-1. pros-1 orchestrates the embryonic development of these cell types by modulating cell proliferation and differentiation, thus ensuring proper morphogenesis and organogenesis. The Americans with Disabilities Act (ADA) was passed in 1990 and protects people with disabilities (PWDs) from discrimination and includes provisions to make society more accommodating. The issue is that the ADA is federal legislation, but most of the implementation occurs at the state-level which means that some states are more accommodating than others. This project attempts to answer the following question: What factors contribute to the discrepancies in state-level disability policy and accommodation? What explains why some cities have developed extensive bike infrastructure while others have not? Bicycling is a low-cost transportation option and is becoming increasingly popular in the 21st century. While there is extensive literature on bike infrastructure in the United States and its relationship with gentrification and advocacy, there is not significant, explicit dedication and focus toward explaining why variation exists across American cities. Due to the increasing prevalence of cannabis product availability and usage in the modern United States, particularly among pregnant individuals, evaluating the bioaccumulation of minor cannabinoids to further understand the adverse effects of cannabinoids during early development is critical. This study quantitated the uptake of eight minor cannabinoids ((∆8-tetrahydrocannabinol (∆8-THC), ∆9-THC, ∆10-THC, cannabidiol (CBD), cannabigerol (CBG), cannabinol (CBN), tetrahydrocannabivarin (THCV), and hexahydrocannabinol (HHC)) in zebrafish larvae following water-borne 2 µM exposures from 6 to 96 hours post fertilization (hpf). While the physical damage induced by the projectiles (typically bullets) and its medical treatment is well-researched, the deformation that the projectile itself undergoes is often underestimated in importance, and the research on this topic is lacking, although such changes can be a valuable source of forensic evidence. Ovarian cancer is the most lethal gynecologic malignancy, with most patients presenting at an advanced stage. While surgical debulking improves survival, nonresectable tumors (<1 cm) contribute significantly to morbidity and remain difficult to treat. Nanocarrier-based drug delivery systems offer an innovative approach to targeted therapy and palliation. In vivo models have demonstrated that intraperitoneally delivered mesoporous silica nanoparticles (MSNs) preferentially accumulate in tumors with low systemic exposure, enabling targeted chemo- and radiotherapy.

MSNs are particularly suited for radiotherapeutic delivery, as their strong interactions with tumor extracellular matrices drive selective uptake. Additionally, they exhibit excellent stability under neutron flux, allowing for loading with stable isotopes and irradiation immediately prior to administration. High radionuclide retention after dilution further minimizes systemic exposure and toxicity.

This study investigates the effectiveness of targeted digital advertising strategies for Company X, a performance motorsports fuel brand. With the goal of optimizing engagement and improving budget allocation, four distinct ad campaigns were developed and tested on Facebook and Instagram: brand awareness, product education, dealer locator promotion, and lifestyle merchandise promotion. Each campaign was evaluated based on reach, impressions, cost per result, and conversion rates. Descriptive statistics revealed significant differences in engagement rates across campaigns, with video-based, emotionally driven ads outperforming static formats. A one-way ANOVA indicated no significant difference in reach across campaigns but confirmed significant differences in spending …

The present thesis aims to determine the effects, if any, of six potential moderators on the effects of acute moderate-intensity exercise on cognitive functioning. The modes of exercise undertaken by the participants included treadmill (Experiment 1) and stationary bike (Experiment 2) exercise. Participants in each experiment either exercised or rested for twenty minutes and then participated in either a memory recall task or cognitive functioning (“Stroop”) task. Synucleinopathies are a group of diseases characterized by neuronal and glial accumulation of α-synuclein (aSyn) linked with different clinical presentations, including Parkinson's disease (PD), Parkinson's disease with dementia (PDD), Dementia with Lewy Bodies (DLB) and Multiple system atrophy (MSA). Interestingly, the structure of the aSyn aggregates can vary across different synucleinopathies. Glioblastoma (GBM) is an aggressive primary brain cancer with few effective therapies. Stereotactic needle biopsies are routinely used for diagnosis; however, the feasibility and utility of investigative biopsies to monitor treatment response remains ill-defined. Here, we demonstrate the depth of data generation possible from routine stereotactic needle core biopsies and perform highly resolved multi-omics analyses, including single-cell RNA sequencing, spatial transcriptomics, metabolomics, proteomics, phosphoproteomics, T-cell clonotype analysis, and MHC Class I immunopeptidomics on standard biopsy tissue obtained intra-operatively. This work proposes a novel framework to detect diseases at earlier stages from medical images, using mathematical model and Machine Learning. Introduce two new quantitative measures for COVID-19 and Tumor disease detection: image uncertainty based on Shannon entropy and image complexity based on fractal dimension.
