Objective: To develop a fast image reconstruction method for stroke monitoring with electrical impedance tomography with image quality comparable to computationally expensive nonlinear model-based methods. Methods: A post-processing approach with graph convolutional networks is employed. Utilizing the flexibility of the graph setting, a graph U-net is trained on linear difference reconstructions from 2D simulated stroke data and applied to fully 3D images from realistic simulated and experimental data. An additional network, trained on 3D vs. 2D images, is also considered for comparison. Results: Post-processing the linear difference reconstructions through the graph U-net significantly improved the image quality, resulting in images …

Selenium (Se)-deficient soils result in Se-deficient forages that do not meet the nutritional requirement for this trace mineral in grazing cattle. Supplementation of Se conventionally occurs through a basal vitamin/mineral mix containing Se in an inorganic form (ISe), although organic Se (OSe) is available when cattle graze on forage. We previously reported that the provision of supplemental Se as a 1:1 mixture of ISe:OSe (MIX), in contrast to the industry standard of ISe alone, increases early luteal phase concentrations of systemic progesterone (P4) and increases the length of the conceptus at day 17 of pregnancy. Ultrasonic vocalizations (USVs) are critical for understanding rodents' emotional states and social behaviors. However, manual analysis of USVs is time-consuming, subjective, and prone to errors. This thesis presents an automated pipeline that addresses these challenges by performing efficient USV detection and clustering. The proposed approach significantly reduces the time and effort needed to analyze USV data while improving accuracy and reproducibility.
To address this gap, we introduce ContourUSV, a five-step pipeline for USV detection. First, it begins with generating spectrograms from audio recordings, which are then pre-processed to enhance the contrast between USVs and background noise. Key steps include median …

As digital technologies redefine the definition of literacy, elementary teachers face the challenge of effectively integrating digital literacies into their classroom spaces. This dissertation examines how professional development can enhance teachers’ understanding and implementation of high-quality digital literacy practices. Grounded in the New Literacies framework, the study identifies key characteristics of effective digital literacy instruction and explores how professional development that is contextualized, meaningful, and sustained can foster these practices. Using data from classroom observations, professional learning communities, and teacher interviews, the research highlights how individualized coaching and collaboration empower teachers to create authentic, engaging, and critical digital literacy experiences. …

Writing requires the ability to encode phonemes to separate sounds to spell. Students who are mystified by print often find writing a laborious task. This action research investigated the impact of Heggerty's Bridge the Gap phonological instruction on third grade writing development, specifically focusing on elaboration. In this thesis will provide a brief history of polymer bound reagents, their uses, and advancements. The importance of polymer microenvironments will be explored using a model kinetic resolution reaction utilizing polymer bound reagents to observe how changes in a polymer’s microenvironment effects the yield and selectivity of a reaction. It was found that there are copious factors that affect polymer bound reagents, and no direct conclusion could be drawn from the study.

Mentorship has emerged as a potential solution to reduce social isolation and provide support, guidance, and encouragement to young people. Dissolved organic matter (DOM) has important roles in many biogeochemical cycles. DOM is primarily composed of dissolved organic carbon (DOC), nitrogen, and phosphorus, and the processing of DOM is important for nutrient regulation and carbon cycling. In the coastal plains of the southeastern United States, there are many rivers and streams known as “blackwaters” with exceptionally high concentrations of chromophoric dissolved organic matter (CDOM) leached from terrestrial soils, coinciding with a region experiencing some of the nation’s highest rates of development. Though acknowledging that Persius alludes to Horace so frequently that he creates a dialogue with his elder, many commentators fail to see that Persius would challenge his predecessor. Especially evident in Satire 4, Persius' challenge to Horace stems from the younger poet's Stoicism, which philosophy he sees Horace as lampooning throughout the elder poet's Satires. Persius not only defends his Stoicism in Satires 4 but also engages with perceived weaknesses in the Epicureanism he finds in Horace. Childhood adversity (CA) impacts a large proportion of US youth with far reaching consequences for development and subsequent health outcomes (McLaughlin et al., 2012). Given the widespread impact of CA, it is imperative to understand how these exposures impact development, as well as identify opportunities for intervention. McLaughlin and Sheridan (2016) proposed the Dimensional Model of Adversity of Psychopathology (DMAP), providing a framework for understanding the heterogeneity of outcomes related to exposure to CA. The present study seeks to compare requirements for orchestral auditions with season programming for 15 American orchestras to gauge the perception that the most frequently requested percussion excerpts are from works that are rarely, if ever, performed. Examination of required repertoire lists for 35 auditions held by the surveyed orchestras from 2000-2025 shows the relative frequency of each excerpt among eight instrument categories and demonstrates that certain selections are more firmly entrenched in the audition canon than others. HER2-positive (HER2⁺) breast cancer is aggressive and often develops resistance to anti-HER2 therapies. CDK8 and CDK19, transcription-associated cyclin-dependent kinases (tCDKs) in the Mediator complex, are dysregulated in HER2⁺ breast cancer, correlate with poor survival, and support transcriptional escape driving therapeutic resistance. Previous studies Previous studies in immune-deficient models demonstrated that CDK8/19 inhibition (CDK8/19i) reduces pro-tumor macrophages (M2Φs), but its role in immune-competent tumors and innate immune–mediated tumor control remains unclear. The growing prevalence of cyanobacterial blooms and associated cyanotoxins, such as microcystin-LR (MC-LR), presents a significant threat to water quality and public health. Conventional detection methods, including ELISA and PCR, are often time-consuming, expensive, and require trained personnel, limiting their application in field settings. Hence, a miniaturized, quick testing sensor is proposed to circumvent the current sensing challenges for real time, in-situ diagnostics of MC-LR. Supercritical water oxidation (SCWO) has been a topic of immense interest as an effective technique/tool for hazardous aqueous waste disposal. SCWO can be achieved by introducing an oxidizer (e.g., H2O2), or a hydrothermal flame as an internal source of both heat and oxidizer species in an aqueous environment at conditions above the critical point of water (P > 218 atm and T > 647 K). The Taliban emerged as a movement in 1994, toppled the government of the Mujahideen, and ruled Afghanistan from 1996 to 2001. They finally recaptured power in 2021 after the US withdrawal from the region. This paper refers to the first Taliban rule as Taliban 1.0 and their second as Taliban 2.0. Taliban 1.0 imposed draconian laws and regulations on women and minorities while harboring regional and international terrorist organizations. The role of midbrain dopamine (DA) activity in reward seeking and reinforcement learning is well-documented, contrasting the lesser-understood impact of inhibitory signaling on DAergic activity, especially in the context of aversive learning and avoidance. The rostromedial tegmental nucleus (RMTg) is a GABAergic midbrain structure activated by aversive stimuli and exerts inhibitory influence over ventral tegmental area (VTA) DAergic activity in response to aversive consequences. Stimulation of the RMTg decreases the firing rate of VTA DA neurons and direct inhibition of the RMTg – VTA circuit enhances persistent reward seeking despite aversive consequences, a major criterion of substance use disorder (SUD). …

Conjure practices in the nineteenth-century American South, such as “Hoodoo” or “Voodoo,” are commonly portrayed as primarily “black” beliefs or entrenched in “Africanistic” cultures. For the enslaved, their black spiritual systems, including conjure, were a powerful tool of resistance against the white enslaver. Black conjure practitioners established themselves as an affordable and pervasive medical alternative to often ineffective or harmful professional medicines for black and white Southerners, regardless of class. In this way, conjure challenged the authority of racial capitalism and destabilized the perceived moral and intellectual superiority of whiteness. Membrane protein topology studies offer guidance to membrane protein structure, folding, and function, serving as a credible scaffold for designing site-directed mutagenesis and biochemical experiments, helping to identify functionally significant extracellular and intracellular regions, modeling three-dimensional structures, and building reliable mechanistic models. Membrane protein structure as a function of given lipid composition and physiological state of the cell is best probed in whole intact cells. Subcellular targeting of signaling enzymes influences where and when various modes of intracellular communication operate. Macromolecular complexes of signal transduction and signal termination elements favor reversible control of repetitive processes. This includes adrenergic stimulation of excitation–contraction coupling in the heart. Long isoforms of A-kinase anchoring protein 18 (AKAP18γ and δ) modulate this process via regulation of calcium uptake into the sarcoplasmic reticulum through the Ca2+ATPase 2a (SERCA2a). AKAP18 proximity-proteomic screening in cardiomyocytes identifies networks for protein kinase A (PKA) and ubiquitin-specific proteinases (USPs). Smooth muscle cell-specific myosin heavy chain, encoded by MYH11, is selectively expressed in smooth muscle cells (SMCs). Pathogenic variants in MYH11 predispose to a number of disorders, including heritable thoracic aortic disease associated with patent ductus arteriosus, visceral myopathy, and megacystis-microcolon-intestinal hypoperistalsis syndrome. Rare variants of uncertain significance occur throughout the gene, including MYH11 p.Glu1892Asp, and we sought to determine if this variant causes thoracic aortic disease in mice. Background: Complex Regional Pain Syndrome (CRPS) is a challenging and often disabling condition marked by persistent pain, most commonly in a limb following injury or surgery. It presents with a wide array of symptoms, including intense pain, swelling, alterations in skin color and temperature, motor dysfunction, and trophic changes such as skin and tissue atrophy. While the precise cause of CRPS is not fully understood, it is thought to stem from abnormal nervous system activity, leading to heightened pain sensitivity and inflammatory responses. The ways that parents respond to their children’s initiations and guide their learning are associated with subsequent language development. Responsive parenting interventions have shown positive impacts on parent behaviors and child outcomes. However, less is known about how intervention effects vary for families from different linguistic and ethnic backgrounds. Introduction: Impaired insulin (INS) and insulin-like growth factor 1 (IGF-1) signaling are features of both brain aging and late-onset Alzheimer's disease (LOAD). However, their exact underlying mechanisms and cause-and-effect linkages, including the downstream regulation of endocytosis and autophagy, are still not well understood.
Methods: We investigated INS/IGF-1 signaling and its connection with endocytic and autophagic processes in fibroblasts from LOAD patients and healthy young or old control individuals.

Schizophrenia (SCZ) is a complex psychiatric disorder with unclear biological mechanisms. Spectrins, cytoskeletal proteins linked to neurodevelopmental disorders, are regulated by the AKT/GSK3 pathway, which is implicated in SCZ. However, the impact of SCZ-related dysregulation of this pathway on spectrin expression and distribution remains unexplored. Here, we show that βIV spectrin protein levels were reduced in neurons of the dorsolateral prefrontal cortex in SCZ postmortem samples compared to healthy control (HC) from the Human Brain Collection Core (HBCC). Introduction: The dysregulation of cyclin-dependent kinases (CDKs) is a key driver of cancer progression, making them attractive therapeutic targets. In CDK4/6 inhibitor (CDK4/6i)-resistant breast cancer, targeting CDK2 offers a promising approach. CDK2 is frequently hyperactivated due to cyclin E1 overexpression or retinoblastoma protein loss, acting as a mechanism that sustains proliferation despite CDK4/6 inhibition. CDK2 inhibitors (CDK2i) show strong anti-tumor activity, particularly in combination with CDK4/6i or immune checkpoint inhibitors.

Background: The granular methods by which centers can safely implement and effectively expand robotic distal pancreatectomy (RDP), including those related to appropriate patient selection during the learning curve period, remain unclear. This study aimed to verify that our strategic robotic surgical oncology program effectively implemented RDP as standard practice and to identify factors associated with prolonged operation time.

Acute myeloid leukemia (AML) accounts for only about 15-20% of pediatric leukemia and an overall incidence of 1.4 cases per 200,000 children under the age of 15 years. The majority of pediatric AML occurs de novo, often as the result of somatic first hits in utero. A minority of pediatric AML occurs in response to a predisposition syndrome, such as a bone marrow failure syndrome, or other inherited mutations and copy number changes. Significance: This review investigates how radiation therapy (RT) increases the risk of delayed cardiovascular disease (CVD) in cancer survivors. Understanding the mechanisms underlying radiation-induced CVD is essential for developing targeted therapies to mitigate these effects and improve long-term outcomes for patients with cancer.
Recent Advances: Recent studies have primarily focused on metabolic alterations induced by irradiation in various cancer cell types. Gastric adenocarcinoma (GAC) remains a significant global health challenge, with over a million new cases annually. Peritoneal carcinomatosis (PC), detected in ∼20 % of cases at diagnosis and ∼45 % later, is uniformly fatal, with limited treatment options. This study investigated the role of KAP1 in GAC progression, focusing on its interaction with YAP1 and cancer stemness traits. Analysis of over 596 primary GACs and 72 PC samples revealed that high nuclear KAP1 expression correlates with poor prognosis. KAP1 knockdown reduced oncogenic activity and stemness traits in GAC cells. Mechanistically, KAP1 positively regulates YAP1 transcription by binding to its promoter …
