Gastric adenocarcinoma (GAC) remains a significant global health challenge, with over a million new cases annually. Peritoneal carcinomatosis (PC), detected in ∼20 % of cases at diagnosis and ∼45 % later, is uniformly fatal, with limited treatment options. This study investigated the role of KAP1 in GAC progression, focusing on its interaction with YAP1 and cancer stemness traits. Analysis of over 596 primary GACs and 72 PC samples revealed that high nuclear KAP1 expression correlates with poor prognosis. KAP1 knockdown reduced oncogenic activity and stemness traits in GAC cells. Mechanistically, KAP1 positively regulates YAP1 transcription by binding to its promoter …

Innovative therapies such as thermoembolization are expected to play an important role in improving care for patients with diseases such as hepatocellular carcinoma. Thermoembolization is a minimally invasive strategy that combines thermal ablation and embolization in a single procedure. This approach exploits an exothermic chemical reaction that occurs when an acid chloride is delivered via an endovascular route. However, comprehension of the complexities of the biophysics of thermoembolization is challenging. Mathematical models can aid in understanding such complex processes and assisting clinicians in making informed decisions. Desmoid tumors are rare mesenchymal neoplasms characterized by a clonal proliferation of fibroblasts and myofibroblasts. Using the novel contrast-enhanced susceptibility-weighted imaging (CE-SWI) for characterizing desmoid tumors can enhance the separation between fibrous T2-hypointense and cellular T1-enhancing components. We aim to evaluate the effectiveness of the CE-SWI signal, volumetric, and radiomics-derived features in assessing desmoid treatment response. This IRB-approved study included 17 single-lesion extremity desmoid fibromatosis patients who underwent standard-of-care MRI, including CE-SWI, from March 2021 to February 2024. The tooth root is a critical component of the tooth anchored to surrounding alveolar bones. Tooth root formation is driven by cells in the apical papilla (AP) that generate new dentin-forming odontoblasts at the root-forming front. Mesenchymal stem cells have been isolated from AP for regenerative use; however, how AP cells physiologically coordinate tooth root formation remains undefined. We find that CXCL12+ cells emerge in AP under hypoxic environments at the onset of tooth root formation. Using Cxcl12-creER-based cell-lineage analysis, we further find that CXCL12+ AP cells contribute not only to odontoblasts but also to cementum-forming cementoblasts of the elongating …

Seasonality and land-use change are key factors influencing forage availability for managed honey bee colonies, yet knowledge of forage identity and how these factors influence forage availability remains limited in Africa. To address these gaps, we used DNA metabarcoding to identify nectar and pollen plant species supporting the nutrition of the African savannah honey bee, Apis mellifera scutellata, across different land-use/land cover types and seasons in Taita Taveta County, Kenya. We identified 224 forage plant species from 65 families, with Asteraceae, Fabaceae, and Myrtaceae being the top contributors. The red spider mites are a major biotic limitation to productivity for Brachiaria (Urochloa spp.) grasses in tropical Eastern Africa, causing severe economic losses mainly in the dry seasons. However, due to the lack of reports of integrated pest management strategies, genetic resistance can play a crucial role in Brachiaria-based livestock systems, minimising the impact of this pest. The emergence and re-emergence of mosquito-borne arbovirus (MBV) diseases pose a rapidly expanding global health threat fueled by the convergence of multiple ecologic, economic, and social factors, including climate change, land use, poverty, deficiencies of water storage and sanitation, and limitations of vector control programs. Memantine is an US Food and Drug Administration (FDA) approved drug that is thought to selectively inhibit NMDA-subtype of ionotropic glutamate receptors (NMDARs). NMDARs enable calcium influx into neurons and are critical for normal brain function. However, increasing evidence shows that calcium influx in neurological diseases is augmented by calcium-permeable AMPA-subtype ionotropic glutamate receptors (AMPARs). Here, we demonstrate that these calcium-permeable AMPARs (CP-AMPARs) are inhibited by memantine. Electrophysiology unveils that memantine inhibition of CP-AMPARs is dependent on their calcium permeability and the presence of their neuronal auxiliary subunit transmembrane AMPAR regulatory proteins (TARPs). Erythropoietic protoporphyria (EPP) is caused by loss-of-function mutations in ferrochelatase (FECH), leading to the accumulation of its substrate, protoporphyrin IX (PPIX). PPIX is primarily produced in the bone marrow and transported to the liver for excretion. Because PPIX is hydrophobic, its elevated levels can cause bile duct blockage, cholestatic liver injury, and even liver failure. However, the specific transporter responsible for PPIX uptake into hepatocytes remains unclear. The OATP1B1/1B3 transporters, which are expressed in hepatocytes, facilitate the uptake of coproporphyrin III, a structural analog of PPIX. Circulating blood platelets are responsible for maintaining hemostasis. They are released into blood vessels from mature megakaryocytes. Although several transcription factors have been reported to orchestrate the transcriptional programs required for platelet production, how chromatin regulators control these processes is still poorly understood. MLL3 and MLL4 are the main lysine methyltransferases responsible for the deposition of H3K4me1 histone marks at enhancers. MLL3 and MLL4 typically form complexes with other co-factors, such as PTIP. Chimeric antigen receptor (CAR) natural killer (NK) cell immunotherapy offers a promising approach against cancer1-3. However, the molecular mechanisms that regulate CAR-NK cell activity remain unclear. Here we identify the transcription factor cyclic AMP response element modulator (CREM) as a crucial regulator of NK cell function. Transcriptomic analysis revealed a significant induction of CREM in CAR-NK cells during the peak of effector function after adoptive transfer in a tumour mouse model, and this peak coincided with signatures of both activation and dysfunction. We demonstrate that both CAR activation and interleukin-15 signalling rapidly induce CREM upregulation in NK cells. Functionally, CREM …

Purpose: Dr. Richard Hill performed pioneering work in the field of radiation-induced normal tissue injury to the lung including noninvasive imaging studies aimed at identifying imaging biomarkers of radiation-induced lung injury (RILI). RILI is a life-threatening toxicity of radiation exposure relevant to both cancer patients undergoing thoracic radiation therapy (RT) and victims of accidental radiation exposure. The ability to detect RILI noninvasively has the potential to guide treatment planning for RT and, in the case of victims of acute radiation exposures, inform the decision to start mitigative therapies. Once considered a nuisance with little medical importance, the lone star tick (Amblyomma americanum, Acari:Ixodidae) has emerged as a formidable vector driving a new era of public health threats across the United States of America. This dissertation traces the rise of the lone star tick from obscurity to infamy, exploring its expanding geographic range, unique immunologic impacts on humans, and role in the transmission of emerging tickborne viruses. The pathogenic fungi Aspergillus fumigatus requires metals to survive and infect humans, but metal overload can be detrimental. Metallothioneins (MTs) are small, cysteine-rich proteins that alleviate metal toxicity by binding excess metals (typically Zn or Cu) for storage or delivery. A recent report indicates that the Fe-S cluster binding glutaredoxin GrxD and the putative Cu-MT CmtA were interacting under high iron conditions in A. fumigatus. We hypothesize that this MT may have a role in binding iron or Fe-S clusters under iron overload conditions to prevent iron toxicity. We also aim to characterize this new MT by determining its …

Evolutionary success is defined by the ability of a species to adapt and survive across many generations. This success is not determined by any single attribute but rather includes the entrenched, complex system that could be associated with biological rhythms, reproductive strategies, and regulations of epigenetic and genetic components. This study examines how the Scooby-Doo franchise employs narrative complexity within an episodic formula, balancing accessibility with layered storytelling. Analyzing two pivotal iterations, Scooby-Doo, Where Are You! (1969) and Scooby-Doo! Mystery Incorporated (2010), the research challenges the perception that episodic animation is inherently simplistic.
While the original series established a consistent mystery-of-the-week format, characterized by predictable structures and comedic relief, it also demonstrated narrative ingenuity through character dynamics and tonal balance. In this project, I notice how we listen and respond to animals, both through marine bioacoustics studies and rhetorical theory, and consider how we might develop new ways of noticing and interacting with animals. I hold together rhetorical theory, marine bioacoustics studies, and my observations nest sitting sea turtles to see what kind of multispecies ethic emerges from messy encounters with nonhuman others – specifically, with sea turtles nesting on the beaches of the Outer Banks in the Summer of 2020. Regression is a ubiquitous and fundamental method that can be found in any ele- mentary statistics course. The simplicity and self evidently useful nature of linear regression beguiles a non-negligible portion of researchers into disrespecting assump- tions required by these models, namely in terms of accuracy of covariates and the underlying nature of the data. This disregard can at best lead to meaningless results and at worse cause significant misunderstandings in scientific pursuit.

The concepts of primordial incomprehensibility, ordered creation, and the liminal chaos between them, form a foundational framework in Mesopotamian intellectual culture. The distinctive ways in which these concepts manifest in Sumero-Akkadian cultures suggest a systematic cosmological substructure — characterized by the paradoxical, cyclical, and co-creative relationships between forces such as Infinity and measure, generation and destruction, and definition and obscurity — that remains insufficiently examined in current scholarship. Two free-breeding dog populations, one in Chernobyl, Ukraine, and the other on the Galápagos Islands, Ecuador, exist on opposite sides of the globe, yet both hold unique scientific potential to further our understanding of free-breeding dog population dynamics and evolutionary processes related to their environments. In the following studies, blood samples from more than 400 individual Chernobyl dogs were collected and their genomes were analyzed alongside over 1500 purebred and 200 global free-breeding dogs, describing genetic relationships and the extent of admixture with purebred migrants. Based on a summer of ethnographic research in a subsection of Appalachian Ohio colloquially referred to as “The Valley,” this research challenges three narratives of this place that formulate a harmful Appalachian imaginary. The first narrative of industrycontrolled coal heritage is complicated by the way local people negotiate control over their own stories of coal and its existence outside of industry domain. During the 1857-58 rebellion in India, British soldiers and civilians saw ample evidence of Indian belief in “magic”. Prophecies about the end of British rule were implicated in inciting rebellion. When a solar eclipse occurred during the British army’s campaign to regain control of Delhi, victory was attributed partially to Indian superstition. Much of the historiography of the 1857-58 rebellion has uncritically accepted British sources’ conceptions of Indian “magic”, dismissing it as superstitious, politically hindering, and divorced from material reality. Traumatic brain injury (TBI) is a global health concern, with over 69 million cases annually. TBI risk depends on subject-specific neuroanatomical variation, yet research often only considers generic finite element (FE) models, based on the geometry and material composition of a 50th percentile male head. Peer interaction is essential for adult English as a Second or Other Language (ESOL) students' oral skills development. However, it is more challenging to offer peer interaction in online synchronous classes. To support new teachers who begin online teaching, the author reviewed research on peer learning and online classroom interactional competencies. Based on a foundation of second language acquisition theories and communicative teaching practices, oral skills are developed more with specific types of activities in online breakout rooms: task-based, cooperative, and collaborative activities. 4-H uses the “Experiential Learning Model” as a way to create high quality youth programs. This model, commonly known as “Do-Reflect-Apply” focuses not only on the particular 4-H experience, but also how youth are interpreting their learning, and if they can apply it to real world scenarios. In the context of the showring, the goal is for youth to dive into their animal projects and learn about industry standards with agriculture, while also developing into capable young adults.
